      • Age-Associated Changes in the Vascular Renin-Angiotensin System in Mice

        <P><I>Background</I>. This study evaluated whether the change in the renin-angiotensin system (RAS) is associated with arterial aging in mice.<I> Methods</I>. Histologic changes and expressions of transforming growth factor-<I>β</I> (TGF-<I>β</I>), collagen IV,<I> fibronectin</I>, angiotensin II (Ang II), angiotensin-converting enzyme (ACE), angiotensin-converting enzyme 2 (ACE2), angiotensin II type 1 receptor (AT1R), angiotensin II type 2 receptor (AT2R), prorenin receptor (PRR), Mas receptor (MasR), endothelial nitric oxide synthase (eNOS), NADPH oxidase 2 and oxidase 4 (Nox2 and Nox4), 8-hydroxy-2′-deoxyguanosine (8-OHdG),<I> 3-nitrotyrosine</I>, and superoxide dismutase 1 and dismutase 2 (SOD1 and SOD2) were measured in the thoracic aortas from 2-month-old, 12-month-old, and 24-month-old C57/BL6 mice.<I> Results</I>. Twenty-four-month-old mice showed significantly increased aortic media thickness and expressions of TGF-<I>β</I>, collagen IV, and fibronectin, compared to 2-month-old and 12-month-old mice. The expressions of PRR, ACE, and Ang II, and AT1R-positive area significantly increased, whereas expressions of ACE2 and MasR and AT2R-positive area decreased with age. The expressions of phosphorylated serine<SUP>1177</SUP>-eNOS, SOD1, and SOD2 decreased, and the 8-OHdG-positive area and the 3-nitrotyrosine-positive area increased with age. The expression of Nox2 significantly increased with age, but that of Nox4 did not change.<I> Conclusions</I>. The enhanced PRR-ACE-Ang II-AT1R axis and reduced ACE2-MasR axis were associated with arterial aging in mice. </P>

        Time-Related Changes in Detrusor Overactivity in Awake Rats with Spinal Cord Injury Observed by Simultaneous Registrations of Intravesical and Intraabdominal Pressures

        <P><B>Purpose</B></P><P>To compare the physical characteristics of detrusor overactivity (DO) induced by intravesical infusion of saline in awake, sham rats and rats with chronic spinal cord injury (SCI), by simultaneous registrations of intravesical and intraabdominal pressures.</P><P><B>Methods</B></P><P>Male Sprague-Dawley rats, normal or with a spinal vascular clip at the level of Th9, were investigated cystometrically 1 and 4 weeks after SCI. Intravesical pressure (IVP) and intraabdominal pressure (IAP) were recorded simultaneously to evaluate true DO. During the filling phase, the event of IVP rises, defined as increments that exceeded 2 cmH<SUB>2</SUB>O from baseline, were determined as DO according to the absence of simultaneous changes in IAP.</P><P><B>Results</B></P><P>All SCI rats exhibited DO during the filling phase, which was not shown in sham rats. The frequency and pressure of DO had a tendency to decrease with time. The DO frequency of SCI rats after 4 weeks (0.9±0.2 min<SUP>-1</SUP>) was decreased compared with that after 1 week (2.1±0.4 min<SUP>-1</SUP>; P<0.05). The DO pressure of SCI rats after 4 weeks (8.4±1.9 cmH<SUB>2</SUB>O) was decreased compared with that after 1 week (11.6±2.9 cmH<SUB>2</SUB>O; P>0.05).</P><P><B>Conclusions</B></P><P>Cystometric studies in awake male SCI rats showed some significant changes in bladder function after SCI. All SCI rats exhibited DO during the filling phase, and showed different physical characteristics of DO over the course of time. The neurological basis of these time-related changes remains poorly understood, but may provide important prognostic information about long-term urological management in SCI patients.</P>

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        Soymilk를 이용한 요구르트 제조 및 저장기간 동안의 품질변화에 관한 연구

        This study was performed for analyzing the general composition and the change in the quality of soymilk-derived yogurts manufactured by adding skim milk and whey powder to soymilk heat-treated at 95℃/5 min and 120℃/10 min, respectively. 1. During the storage of soymilk yogurt, the concentrations of total solids, protein, fat, and lactose slightly decreased, whereas viscosity, content of ash and NPN, and the number of lactic acid bacteria remained unchanged. 2. The pH and titratable acidity changed rapidly in all soymilk yogurts after 3 h of incubation. 3. We found 7.8×108 lactic acid bacteria in the control sample, 4.7×108 and 5.02×108 in soymilk yogurt with skim milk, respectively, and 5.9×108 and 5.5×108, respectively in soymilk yogurt with whey powder according to degree of heat treatment with 95℃/5 min and 120℃/10 min. 4. The viscosity of yogurt samples became lower as the heat treatment increased in temperature and in the length of time. 5. The value of sensory evaluation was relatively high in soymilk yogurt with the added skim milk, which was heat-treated 95℃/5 min; however, the value was significantly lower than that of the control sample. 6. Lactose, glucose, and galactose were detected in all samples because lactose is degraded into glucose and galactose within 3 h of inoculation.

      • Correlating Structural Changes and Gas Evolution during the Thermal Decomposition of Charged Li<sub><i>x</i></sub>Ni<sub>0.8</sub>Co<sub>0.15</sub>Al<sub>0.05</sub>O<sub>2</sub> Cathode Materials

        <P>In this work, we present results from the application of a new in situ technique that combines time-resolved synchrotron X-ray diffraction and mass spectroscopy. We exploit this approach to provide direct correlation between structural changes and the evolution of gas that occurs during the thermal decomposition of (over)charged cathode materials used in lithium-ion batteries. Results from charged Li<SUB><I>x</I></SUB>Ni<SUB>0.8</SUB>Co<SUB>0.15</SUB>Al<SUB>0.05</SUB>O<SUB>2</SUB> cathode materials indicate that the evolution of both O<SUB>2</SUB> and CO<SUB>2</SUB> gases are strongly related to phase transitions that occur during thermal decomposition, specifically from the layered structure (space group <I>R</I>3̅<I>m</I>) to the disordered spinel structure (<I>Fd</I>3̅<I>m</I>), and finally to the rock-salt structure (<I>Fm</I>3̅<I>m</I>). The state of charge also significantly affects both the structural changes and the evolution of oxygen as the temperature increases: the more extensive the charge, the lower the temperature of the phase transitions and the larger the oxygen release. Ex situ X-ray absorption spectroscopy (XAS) and in situ transmission electron microscopy (TEM) are also utilized to investigate the local structural and valence state changes in Ni and Co ions, and to characterize microscopic morphology changes.       • Using Real-Time Electron Microscopy To Explore the Effects of Transition-Metal Composition on the Local Thermal Stability in Charged Li<sub><i>x</i></sub>Ni<sub><i>y</i></sub>Mn<sub><i>z</i></sub>Co<sub>1–<i>y</i>–<i>z</i></sub>O<sub>2</sub

        <P>In this work, we use in situ transmission electron microscopy (TEM) to investigate the thermal decomposition that occurs at the surface of charged LixNiyMnzCo1-y-zO2 (NMC) cathode materials of different composition (with y, z = 0.8, 0.1, and 0.6, 0.2, and 0.4,and 0.3), after they have been charged to their practical upper limit voltage (4.3 V). By heating these materials inside the TEM, we are able to directly characterize near surface changes in both their electronic structure (using electron energy loss spectroscopy) and crystal structure and morphology (using electron diffraction and bright-field imaging). The most Ni-rich material (y, z = 0.8, 0.1) is found to be thermally unstable at significantly lower temperatures than the other compositionsthis is manifested by changes in both the electronic structure and the onset of phase transitions at temperatures as low as 100 degrees C. Electron energy loss spectroscopy indicates that (i) the thermally induced reduction of Ni ions drives these changes, and (ii) this is exacerbated by the presence of an additional redox reaction that occurs at 4.2 V in the y, z = 0.8, 0.1 material. Exploration of individual particles shows that there are substantial variations in the onset temperatures and overall extent of these changes. Of the compositions studied, the composition of y, z = 0.6, 0.2 has the optimal combination of high energy density and reasonable thermal stability. The observations herein demonstrate that real-time electron microscopy provide direct insight into the changes that occur in cathode materials with temperature, allowing optimization of different alloy concentrations to maximize overall performance.</P>

        사염화탄소 투여 흰쥐에서 간섬유화 진행에 따른 Propranolol 의 약동학적 지표들의 변화

        Background/Aims : This study was designed to determine the effect of hepatic fibrotic severity on pharmacokinetics of propranolol in CC1q - treated rats. Methods - 1 mL/kg of 10% CC14 in olive oil was injected intramuscularly to rats twice weekly for 4, 6, 8 and 10 weeks, respectively (n=6). Control (n=6) was a sham - injected equal dose of olive oil for 10 weeks. After intravenous bolus injection of 2 mg/kg propranolol to rats, the serum propranolol concentrations were analyzed for 4 hours at various time points by a HPLC - fluorimetric system, and pharmacokinetic parameters such as Co, MRT, AUC, Vdss, tl/2(p) and CLp were determined. Then, a small amount of hepatic tissue was obtained and subjected to determination of the hepatic 4 - hydroxyproline content, which confirmed the hepatic fibrotic severity. Results : The serum concentrations of propranolol at 0.5, 1, 2 and 4 hours were significantly increased in CC1q-treated rats (p$lt;0.01). In proportion to the duration of CC4 treatment, Co and AUC were significantly increased, and Vdss and CLp were significantly decreased (p$lt;0.001). But MRT and t1/2(β) were not significantly changed. The hepatic 4 -hydroxyproline content was gradually increased in CC4-treated rats (p$lt;0.001). Conclusion - Gradual changes in pharmacokinetic parameters of propranolol were seen to be dependent on the hepatic fibrotic severity. We suggest that gradual dosage modification, according to their hepatic fibrotic severity, is necessary for many drugs administered to patients with chronic liver disease.(Korean J Hepatol 2001;7:181-188)

      • 강원도지역에서 발생한 발진열과 쯔쯔가무시병

        In Korea, there has been an increasing concern on rickettsiosis as one of most prevailing acute febrile illness, since murine typhus and scrub typhus had not been reported for 2 decades until 1986 and thereafter a few studies were reported on these diseases. A microimmunofluorescence test was used to study antibodies to R. typhi and R. tsutsugamushi in urban and rural inhabitants students, and patiants suspected of leptospirosis or rickettsiosis from 1986 to 1987 in Kangwon-do Korea. 43% of inhabitatnts and 17% of students reacted to R. typhi. Only one inhabitant among inhabitants and students reacted to R. tsutsugamushi. Among 169 patients, 43(25%) reacted to R. typhi, 38(22%) to R. tsutsugamushi and 25(15%) to Leptospira interrogans. Sexual rate(male; female) in murine typhus was 1.7:1, that in scrub typhus was 1:2.2 and that in leptospirosis was 1.5;1. The age ditribution of patients with murine typhus was generally even over 20 years old. But 71% of patients with scrub typhus were over 40 years old. Scrub typhus was most prevailing in female over 40 years old. Murine typhus was relatively more prevailing in Choonchun city than rural area as compared with scrub typhus and leptospirosis. Most of scrub typhus occurred in rural area. Most of each disease occurred between September and November (murine typhus; 93%, scrub typhus; 95%, leptospirosis; 84%) with a peak in October. Scrub typhus considerably occurred not only October(58%) but also November(28%). All patients with scrub typhus were immediately diagnosed by antibody when they visited hospital. 53% of patients with murine typhus were diagnosed when they visited hospital within 7 days after onset of illness, and others were done by second sera obtained 8 days after. The patients in whom IgM was seronegative but antibodies of other class were seropsitive by 20 days after onset of illness, were 14% in murine typhus and 26% in scrub typhus. At least those patients were considered to be infected second. Murine typhus and scrub typhus were suggested to be endemic diseases in Kangwon-do as antibodies of inhabitants and characteristics of patients' sera were considered. By serologic finding of patients with scrub typhus in Kangwon-do, Gilliam was 59'% of etiologic strain of scrub typhus, Karp was 41q% and Kato was none. Most scrub typhus occurred in person to be related to agriculture, but murine typhus was not related to job and occurred more in small city than rural area. Rash was scarcely detected in murine typhus and scrub typhus, and eschar was in 50% of scrub typhus only. Serum transaminase elevated in many patients with scrub typhus and murine typhus. Leukocytosis and abnormal finding in chest X-ray were showed in some patients with scrub typhus but were scarcely in murine typhus.
