        금불초 꽃 추출물의 항균 및 항산화 활성

        본 연구에서는 금불초 꽃 추출물의 피부 상재균에 대한 항균작용과 항산화, tyroinase 저해 효과에 관한 조사를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, P. acnes에 대한 ethyl acetate 분획의 MIC는 0.25 %로 나타났으며, methyl paraben과 비슷한 활성을 나타냈고 quercetin보다는 큰 항균활성을 나타내었다. 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성(FSC<sub>50</sub>)은 ethyl acetate 분획에서 8.55 μg/mL로 측정되었다. Luminol-의존성 화학발광법을 이용한 Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub>계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 금불초 꽃 추출물의 총 항산화능은 ethyl acetate 분획 0.24 μg/mL로, 50 % EtOH extract와 aglycone 분획 보다 ethyl acetate 분획에서 가장 큰 활성을 나타내었다. 금불초 꽃 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였다. 금불초 꽃 추출물의 경우 5 ~ 100 μg/mL의 농도에서 광용혈을 억제하였다. 특히 당을 제거시킨 플라보노이드 aglycone 분획은 25 μg/mL 농도에서 τ<sub>50</sub>이 164.15 min으로 매우 큰 세포보호 효과를 나타내었다. 미백 효과측정으로는 각각 tyrosinase의 활성 저해 효과를 측정하였다. Tyrosinase의 활성 저해 효과 (IC<sub>50</sub>)는 ethyl acetate 분획에서 87.03 μg/mL 로 나타났다. 이상의 결과들은 금불초 꽃 추출물이 <sup>1</sup>O<sub>2</sub> 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, 금불초 꽃 성분 분석, ethyl acetate 분획의 tyrosinase 저해활성 그리고 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성을 확인하였다. In this study, the antibacterial activity, antioxidative effects, inhibitory effects on tyrosinase of Inula britannica flower extracts were investigated. MIC values of ethyl acetate fraction from Inula britannica flower on P. acnes 0.25 %, respectively. The results showed that the antibacterial activity of the ethyl acetate fraction was the highest in the P. acnes. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities (FSC<sub>50</sub>) of ethyl acetate fraction of Inula britannica flower was 8.55 μg/mL. Reactive oxygen species (ROS) scavenging activities (OSC<sub>50</sub>) of some fInula britannica flower extracts on ROS generated in Fe<sup>3+</sup>- EDTA/H<sub>2</sub>O<sub>2</sub> system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activities were ethyl acetate fraction 0.24 μg/mL. Ethyl acetate fraction showed the most prominent ROS scavenging activity. The protective effects of extract/fractions of Inula britannica flower on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Inula britannica flower extracts suppressed photohemolysis in a concentration dependent manner (5 ∼ 100 μg/mL), particularly deglycosylated flavonoid aglycone fraction exhibited the most prominent celluar protective effect (τ<sub>50</sub>, 164.15 min at 25 μg/mL). The inhibitory effect of Inula britannica flower extracts on tyrosinase was investigated to assess their whitening efficacy. Inhibitory effects (IC<sub>50</sub>) on tyrosinase of some Inula britannica flower extracts were high. Ethyl acetate fraction has IC<sub>50</sub> of 87.03 μg/mL. These results indicate that extract/ fractions of Inula britannica flower can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging <sup>1</sup>O<sub>2</sub> and other ROS, and protect cellular membranes against ROS. And inhibitory activity on tyrosinase of the ethyl acetate fraction and high potential as bactericide against the skin pathogenic bacteria could be applicable to new functional cosmetics for antioxidant, antiaging, antibacterial activity.

        대황의 항산화 효과 및 타이로시네이즈 저해 활성

        본 연구에서는 대황 추출물의 항산화 활성, 타이로시네이즈(tyrosinase) 저해 활성을 확인하였다. 대황의 50 % 에탄올 추출물, 에틸아세테이트(ethyl acetate) 분획, 아글리콘(aglycone) 분획으로 실험을 진행하였다. 대황 추출물들의 DPPH (1,1-diphenyl-2-picrylhydrazyl) 소거활성(FSC<sub>50</sub>)은 대표적인 항산화제인 (+)-α-tocopherol보다 낮은 것으로 나타났다. Luminol 발광법을 이용한 Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> 계에서 생성된 활성산소종에 대한 아글리콘 분획의 소거활성(총 항산화능, OSC<sub>50</sub>)은 0.265 μg/mL로 매우 큰 활성을 나타내었다. 대황 추출물의 rose-bengal로 증감된 <sup>1</sup>O<sub>2</sub>에 의한 적혈구 파괴에 대한 세포보호 효과는 모든 분획에서 농도 의존적(1∼50 μg/mL)으로 증가하였으며, 특히 아글리콘 분획은 10 μg/mL 농도에서 τ<sub>50</sub>이 757.0 min으로 높은 세포 보호 활성을 나타내었다. 대황 추출물 중 아글리콘 분획의 타이로시네이즈 저해활성(IC<sub>50</sub>)은 11.20 μg/mL으로 226.88 μg/mL인 알부틴(arbutin)보다 큰 활성을 보여주었다. 이상의 결과들로부터 대황 추출물은 활성산소종을 소거하는 항산화제로 이용가능하며, 특히 아글리콘 분획의 현저한 항산화 작용 및 큰 타이로시네이즈 저해 효과로부터 이들 분획 또한 화장품원료로서 응용 가능성이 큼을 알 수 있었다. In this study, the antioxidative effects and inhibitory activities on tyrosinase of Rheum undulatum (R. undulatum) L. extracts were investigated. 50 % ethanol extract, ethyl acetate and aglycone fractions of R. undulatum L. were used in experiments. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activities (FSC<sub>50</sub>) of R. undulatum L. extracts was lower than (+)-α-tocopherol, known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities (OSC<sub>50</sub>) of aglycone fraction on ROS generated in Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> system using the luminol-dependent chemiluminescence assay showed the most prominent effect at a concentration of 0.265 μg/mL. The cellular protective effects of extract/fractions of R. undulatum L. on the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner (1 ~ 50 μg/mL). Especially, aglycone fraction in 10 μg/mL concentration showed the most protective effect among extracts (τ<sub>50</sub>, 757.0 min). The inhibitory effects (IC<sub>50</sub>, 11.20 μg/mL) on tyrosinase of aglycone fraction was much higher than arbutin (226.88 μg/mL), known as a whitening agent. These results indicate that R. undulatum L. extracts can be used as antioxidant. Particularly, aglycone fraction of R. undulatum L. showed superior antioxdative activity and high inhibitory effect on tyrosinase. Therefore, aglycone fraction of R. undulatum L. could be applicable to new functional cosmetics.

        대장직장 종양에 대한 내시경 점막하 절제술의 연대순 임상 결과와 학습 곡선

        Background/Aims: Endoscopic submucosal dissection (ESD) is an effective procedure for en-bloc curative resection of the colorectal tumor. As it requires high technical skills and experience in therapeutic endoscopy, it is important to understand learning curve of ESD technique. The aim of this study was to retrospectively describe the clinical results of ESD and to evaluate learning curve for the ESD of colorectal tumors. Methods: A total of 90 patients with 90 colorectal neoplasms, who had undergone ESD at a tertiary referral hospital from July 2009 to December 2012, were enrolled. The ESD was performed by a single endoscopist. All ESD cases were divided into three periods: first, cases 1-30; second, cases 31-60; and third, cases 61-90. Results: The en-bloc resection rates in third period (100%) was significantly higher than that of the first (93.3%) and second period (80%) (p=0.025). The perforation rate in third period (0%) also significantly decreased compared with that of the first (13.3%) and second period (20%) (p=0.032). To calibrate the difference of tumor size among periods, proficiency was calculated, as the procedure time per specimen area (min/cm2). The proficiency in third period (4.3) was significantly shorter than that of the first (16.8) and second period (10.2) (p=0.004). Conclusions: The learning curve of colorectal ESD in our study shows that at least 60 cases of ESD have to be conducted to acquire sufficient skill of degree without perforation.         The relationship between aerobic capacity and the recruitment of activated sweat gland density with passive heating

        This study investigated whether the recruitment of activated sweat gland density during passive heating [immersion of the lower body into hot water, 41℃ for 30 min (PH), room temperature 23 ± 0.5℃ with 60 ± 3% relative humidity] is different in endurance-trained (Trainee), compared with that of untrained subjects (Control), as indicated by VO2max. Eight trainees and seven control female subjects (38.31 ± 4.91 ml·kg-1·min-1 and 31.8 ± 0.92 ml·kg-1·min-1, respectively, p < 0.01) were similar on all other physical characteristics. To compare the changes between the trainees and controls, serum cortisol and prolactin (PRL), tympanic temperature (TYMP), local activated sweat gland density (L-ASGD) and the mean whole body sweat loss volume (M-WBSLV) changes were measured after PH. The cortisol was significantly higher (p < 0.05) and the PRL tended to be higher in the Trainee than in the Control. The TYMP increased more in the Trainee than in the Control (p < 0.001). The L-ASGD showed a higher tendency in the Trainee than in the Control, there was a significant difference in the mean activated sweat gland density (p < 0.01) and the L-ASGD was significantly correlated with the VO2max (p < 0.001). The M-WBSLV was significantly higher in the Trainee (p < 0.001) and significantly correlated with the VO2max (p < 0.001). The results suggest that in humans, a high aerobic capacity is associated with a greater recruitment of activated sweat glands, whole body loss of volume and TYMP. Therefore, the decline in the heat loss responses, due to decrease in maximal oxygen consumption, may be masked by repeated endurance exercise training.

      • 본태성 고혈압 환자의 적혈구 Na+-Li+ Countertransport 와 적혈구막 지방산 구성에 대한 연구

        Since the changes of Na^+-Li^+ countertransport (SLC) Vmax in essential hypertension have been reported. it has been known that hyperlipidemia could affect SLC independently of blood pressure. To reveal the relation between SLC Vmax and RBC membrane fatty acid conposition in the hypertensive patients, we divided patients into two groups according to their levels of SLC Vmax of erythrocytes(elevated SLC Vmax: n=4, normal SLC Vamx: n=6). The results were as follows: 1) There was no difference in body mate index, waist-hip ratio, blood pressure, serum cholesterol level between two groups. Triglyceride were higher in elevated group (278.0±180.0 ㎎/㎗) than normal group (124.3±60.7 ㎎/㎗) but there was no statistical significance. HDL-cholesterol levels were significantly lower in elevated group (32.0±3.6 vs. 44.8±8.9 ㎎/㎗) and free fatty acid concentrations, intracellular (RBC) Na^+ concentrations were similar between two groups. 2) The results of oral glucose tolerance test showed no differences in the glucose and insulin area, however the incremental insulin areas of elevated group (108.8±31.3, μU. min/㎖×10²) were significantly higher compared with those of normal group (56.3±20.7 μU. min/㎖×10²). 3) In the elevated group, palmitic acid proportions of erythrocytes (22.1±0.7%) were higher than those of normal group (20.1±0.7%) and SLC Vmax showed significant correlation (r=0.81, p$lt;0.01) with palmitic acid proportion. These results suggest that the abnormal lipid metabolism may change the RBC Na^+ transport system through the alterations of RBC membrane fatty acid composition.

      • 딸기 와인 개발을 위한 최적 발효 조건

        Optimal fermentation condition for development of new strawberry wine were investigated. Commercial yeast, Fermivin was good for brewing of strawberry wine and its optimal addition concentration was also 1%. Maximal ethanol content of strawberry wine was obtained when raw strawberry juice(24 brix) was fermented by 1% Fermivin at 25℃ for 7 days and its antioxidant activity was 78.5%. 새로운 딸기 와인을 개발하기 위한 최적 발효조건을 연구하였다. 시판효모인 페미빈이 딸기와인 제조에 제일 좋았고 1%가 최적 첨가 농도이었다. 딸기와인의 최대 에탄올 생성량은 생딸기주스 (24brix)를 1% 페미빈으로 25℃에서 7일 발효시켰을 때 얻어졌고 이 발효액의 항산화활성은 78.5% 이었다.

        2003년 국내 중증급성호흡기증후군 진료 현황 및 문제점 분석

        목적 : 2002년 말 중국에서 SARS가 발생한 이후 국내에서도 2003년 10월까지 총 3명의 추정환자, 17명의 의심환자가 보고되었다. 향후 추가적인 SARS의 유행이 우려되는 상황에서, 그간의 SARS 환자 진료에 있어서의 실질적인 준비사항, 진료 현황 등에 대한 조사를 통해 문제점을 파악하여, 향후 더 나은 대비가 될 수 있도록 개선점을 제시하고자 하였다. 재료 및 방법 : SARS로 의심되는 환자를 진료 경험이 있는 병원의료진을 대상으로 2003년 10월에 설문조사를 실시하였다. 설문에는 SARS 환자 진료 시의 실질적인 조치, 진료 현황, 병실, 응급실 및 외래에서의 격리 시설과 준비사항, 보건당국의 관리와 지원에 관한 사항을 포함하였다. 결과 : 대상이 되는 22개 병원 중 17개(17/22, 77.2%) 병원이 설문에 응하였다. SARS 환자를 위한 격리실은 응급실, 외래, 일반병실 및 중환자실에서 각각 9개(9/17, 52.9%), 5개(5/17, 29.4%), 15개(15/16, 93.7%), 4개(4/16, 25%) 병원에서 음압처리가 되어있지 않은 일인실 혹은 다인실이 사용되었고, 1개(1/16, 6.3%) 병원에서만 일반병실에서 음압격리실이 운영되었다. 입원환자의 진찰 시 개인보호구의 착용은 거의 모든 의료기관에서 이루어졌다. 보건당국에서 SARS지정병원의 시설 등을 사전에 확인한 곳은 1곳(1/12, 8.3%)이였고, 14개 병원(14/15, 93.3%)에서는 보건당국에 의뢰한 검사결과를 통하 받지 못하였다. 결론 : 의료기관에서 SARS 환자용 격리실뿐만 아니라 기존의 격리실 설비 등이 미흡하였으며, 특히 중환자실 및 외래의 준비가 더욱 부족하였다. 보건당국의 의료기관에 대한 종합적인 지원이 부족하였고, 병원과의 원활한 연계가 잘 이루어지지 않았다. SARS 만이 아닌 격리를 필요로 하는 질환의 적절한 진료를 위해 향후 병원 시설의 정비와 정부차원에서의 보다 구체적이고 실질적인 대책마련이 필요하다. Background : There was an worldwide outbreak of the Severe Acute Respiratory Syndrome (SARS) originated from China in late 2002. During that period three cases of suspected SARS and 17 cases of probable SARS were reported in Korea. With the concerns about the reemergence of SARS-coV transmission, it is important to be prepared for any possibility. So, this study is aimed to analysis the past measures in managing SARS and propose the amendatory plans to improve the preparedness. Materials & Methods : Questionnaires were collected among clinicians with any experience in managing the probable or suspected SARS cases in Oct. 2003. 17 out of 22 hospitals responded to the questionnaire. The contents in the questionnaire were practical activities, personal equipments, response plans, isolation facilities in emergency centers, outpatient clinics, general wards and intensive care units, and relationship with the public health department. Results : The dedicated isolation rooms in emergency centers, outpatient clinics, general wards, and intensive care units were prepared in 9 (9/17, 52.9%), 5 (5/17, 29.4%), 15 (15/16, 93.7%), and 4 (4/16, 25.0%) hospitals, respectively. Except for one hospital that newly made negative pressure room for SARS, single or multi-bed rooms without airborne infection control were used in all the other hospitals. The personal precaution principles were kept quite well in general wards. Before the designation of SARS hospital by the public health department prior evalution to see if the hospital was suitable for managing SARS was conducted in only 1 (1/12, 8.3%) hospital. The results of laboratory diagnosis were reported back in 1 (1/15, 6.6%) hospital. Conclusions : The isolation facilities which can control airborne infection were almost deficient not only for SARS but also for other respiratory transmissible diseases. For the infection control of transmissible diseases including SARS, more investment is needed on medical facilities and comprehensive support from the public health department required.

      • 인체 자궁암 세포에서 천연 성분이 P-당단백질의 활성에 미치는 영향

        Multidrug resistance (MDR) of cancer cells is, at least in part, associated with the overexpression of P-gly-coprotein (P-gp). Many studies hve demonstrated that natural compounds obtained from fruits, vegetables, teas and medicinal plants may modulate P-gp activity. The objective of the present investigation was to examine the effect of seven natural compounds on the P-gp activity in human uterine sarcoma cell line, MES-SA/DX5. Daunomycin uptake was significantly increased by biochanin A and silymarin (p<0.0001) whereas it was reduced by morin (p<0.01). The efflux of daunomycin from the cells was significantly inhibited by biochanin A, morin, cephalotaxine, berberine (p<0.05) and silymarin (p<0.0001). Biochanin A, berberine and silymarin significantly decreased IC_(50) value of daunomycin (p<0.05) while morin increased it (p<0.05). These results suggest that some natural compounds such as biochanin A and silymarin may inhibit P-gp funciton and can by developed as MDR reversing agents to improve the efficacy of chemotherapeutic drugs when administered concomitantly.

      • 한약재료(단삼, 도인, 당귀미, 솔잎) 추출물이 지방산화에 미치는 영향

        지방산화 촉진인자인 활성산소종 (Hydrogen peroxide, Superoxide, Hydroxyl radical)과 Iron sources 들이 고혈압 치료제로 사용되고 있는 한약재료추출물들과의 반응효과를 in vitro 상에서 검토한 결과 oil emulsion 상태에서 활성산소종은 OH, H₂O₂, KO₂ 순으로 지방산화 촉진작용이 나타났으며, 한약재료추출물 자체도 산화반응을 어느정도 촉진시키는 경향이었다. 한약재료추출물의 활성산소 포집능은 KO₂에 비해 H₂O₂와 OH은 나타내지 않는 경향이었다. 그러나, 전반적으로 지방산화를 촉진시키는 ?? ion binding 능력은 추출물 모두 탁월하였다. ?? ion의 함량은 도인, 솔잎에 비해 단삼, 당귀미가 높은 수치를 나타내었으며, total iron 함량 역시 같은 경향이었다. 항산화작용을 하는 ascorbic acid 함량은 솔잎이 26.97ppm으로 가장 높은 함량을 나타내었으며, 단삼과 당귀미는 19.74, 22.14ppm으로 유사하며, 도인이 5.50ppm으로 낮은 값을 나타내었다.(P<0.05). 전자공여능은 솔잎, 단삼이 각각 79.54%,77.11%로 도인, 당귀미보다 높게 나타났고, SOD 유사활성측정에서는 도인이 0.16으로 가장 낮은 흡광도 수치를 나타내어, pyrogallol의 자동산화를 억제하는 SOD 유사활성이 다른 추출물에 비해 높은 것으로 나타났다(P<0.05). 아질산염소거작용은 pH에 따라 소거능이 다르게 나타났으며, 솔잎과 당귀미가 pH 1.2에서 각각 99.8%, 98.6%의 강한 소거능을 나타내었고, pH의 증가에 따라 전반적으로 감소하는 경향이었으며, 특히, pH6.0에서는 단삼과 도인의 경우 아질산염 소거작용이 나타나지 않았다. 이상의 결과로 한약재료 추출물 4종의 기능성은 솔잎 추출물이 가장 우수한 것으로 사료된다. This study was carried out to investigate the effects of herb extracts on lipid oxidation and free radical reaction in iron sources reacted with active oxygen species. The results were summarized as follow; The catalytic effects of active oxygen on lipid oxidation in oil emulsion tended to be showed OH, H₂O₂ and KO₂ in order. At the same time, herb extracts itself were tended to be showed a little catalytic effects. Active oxygen scavenging ability of herb extracts didn't show, but herb extracts played role as a strong chelating agents to bind iron ion if ?? ion exist in oil emulsion. The content of ?? ion Salvia miltiorrhiza Bge, and also, the content of total iron was the same persica Stokes and Pinus strobus. And also, the content of total iron was the same tendancy. The content of ascorbic acid of Pinus strobus showed the highest amounts as 26.97ppm among all herb extracts, but Salvia miltiorrhiza Bge., Angelica gigas Nakai and Prunus persica Stokes were 19.74ppm, 22.14ppm and 5.50ppm, respectively(P<0.05). Electron donating ability of Pinus strobus and Salvia miltiorrhiza Bge. were 79.54% and 77.11%, respectively, which was showed higher content than those of Prunus persica Stokes and Angelica gigas Nakai. The SOD-like activity of Prunus persica Stokes was showed optical density(O.D.) 0.16, compared to other berb extracts which means the most strong antioxidant reaction. The nitrite scavenging effects were tended to be different, depending on pH value as pH value was increased. They were decreased overall. Especially, they didn't show the nitrite scavenging effect in pH 6.0. In conculsion, the Pinus strobus extract among herb extracts were the most effective by evaluation as functional sources.

        The vernacular houses in Jeju island are never found in any other places and they also make it a particular living culture. The wind is acted as the binding force of the obvious culture formation in Jeju island with stone, especially the physical characteristics in the living style can be influenced by the wind. The conclusions are as follows. 1) As Mt.Hanra is located in the middle of Jeju, it is expressed by the difference in the arragement and style of vernacular houses. In the Southern Jeju, most of houses facing south and their style is the 'ㄱ-type'. In the other hand, there are houses facing east, west and having style of the '二-type' in the Northern Jeju. 2) The size of lumber used in vernacular houses in Jeju island is small but strong, '2-Goju 7-Ryang' is very available construction. 3) The height of stylobate and floor is lower than that of the Korean Peninsula, the height of them in the Northern Jeju is far lower than that of the southern Jeju. 4) The slope of roof is lower than that of the Korean Peninsula. This puts the stress on the defense against the wind rather than the rain. 5) The windbreaks in the Northern Jeju are three times as many as that of the Southern Jeju, stone walls in Jeju island are so higher than those of the Korean Peninsula and the windbreak forest is so much developed that the vernacular houses in Jeju island are characteristic of closing.
