      • Heterosigma akashiwo의 psbA 유전자 구조 특성

        Heterosigma akashiwo로부터 광합성에 중요한 psbA 유전자를 찾고 그 구조를 분석하였다. H.akashiwo에서 전체 DNA를 추출하고 psbA유전자에 특이적인 primer들을 사용하여 PCR로 증폭시켰다. 증폭된 약 1.2kb 크기의 DNA를 pGEM-3Z vector에 클로닝시키고 염기서열을 결정하여 유전자 구조를 조사하였다. 결과, 360개의 아미노산으로 구성된 D1단백질의 암호 지역과 원핵생물의 특징적인 promoter 구조 그리고 ribosome 결합자리를 확인하였으며 D1 단백질의 카르복시 말단 부위에는 엽록소 b를 포함하지 않은 cyanobacteria처럼 특징적인 7개 아미노산이 첨가된 특징을 알 수 있었다. 다른 광합성 생물들의 psbA 유전자와 비교하였을 때 고등식물과는 DNA 염기서열에서 83.3%, 아미노산 서열은 91%의 유사성을 보였으며, cyanobacteria와는 DNA 염기서열은 75.2%, 아미노산 서열은 82%의 유사성을 보였다. 또한 psbA mRNA의 5'-말단 부위에는 번역과정의 조절 부위로 작용할 수 있는 이차구조를 확인하였다. The psbA gene was isolated from Heterosigma akashiwo and its structure was characterized. The psbA gene of about 1.2kb size was amplified from genomic DNA of H. akashiwo by PCR and the nucleotide sequences were determined. A coding region for D1 protein containing 360 amino acids, a promoter element like prokaryotic promoter, and ribosome binding site exist in psbA gene of H. akashiwo. There was addition of the motif consisting of seven amino acids at carboxy terminus region like in cyanobacteria. As the nucleotide and amino acid sequences of H.akashiwo psbA gene were compared with those of higher plant, Spinacia and cyanobacteria, homologies in nucleotide and amino acid sequences between H.akashiwo and Spinacia were 83.3% and 91%, respectively. Those between H.akashiwo and cyanobacteria were 75.2% and 82%, respectively. A secondary structure being act as cis-acting element in translational regulation of psbA gene was showed at 5'-terminus of mRNA.

      • 문자 단위 매칭과 유닛 단위 매칭을 이용한 온라인 필기 한자 인식

        This paper presents on-line Chinese character recognition methods based on DP-Matching. The recognition system consists of character matching and unit matching methods. The character matching method adopts DP-matching between the input data and models as a major matching method, while unit matching method uses relation matching between consecutive units (part of character) in addition to DP-matching between units. A couple of methods combining character matching and unit matching methods were examined also. We found that we got best recognition result applying unit matching methods to the recognition candidates from character matching method.

      • DP-Matching과 HMM을 이용한 온라인 필기 한자 인식

        On-line handwritten Chinese character recognition systems should be fast and yield accurate results in order to be practical systems. DP-matching method for variable length input requires building several allographs (multiple models) per class, which inevitably reduces the recognition speed. HMM is a good alternative, since one HMM per class is usually sufficient to model large variation of data. We proposed a recognition system in which HMM and DP-matching method are combined to improve the recognition speed and recognition accuracy at the same time. The HMM method using structure code features produces recognition candidates, then DP-matching method using chain code features reorders the candidates to get more accurate results. In doing so, we accomplished fast and accurate recognition results for 2,362 Chinese character classes. We also adopted result verification method to discriminate confusable character classes. We got 97.44% recognition accuracy using the proposed method in 113 ms per character in average.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

      • 새만금 단벌기 목재에너지림 식재 포플러 묘목의 생육특성 및 바이오매스 생산량

        본 연구는 새만금 간척지 배후도시용 유보용지를 이용하여 2012년에 조성된 포플러 단벌기 바이오매스 생산림을 대상으로 식재 시 대절처리에 따른 묘목의 생육특성과 목질계 바이오매스 생산성을 조사하기 위해 수행되었다. 식재 2년차 목재에너지림 포플러 묘목의 총 수고와 직경은 무대절처리구(347.8cm/19.5mm)와 1m 대절 처리구(324.5cm/18mm)에서 유사한 수준이었으며, 지제부 대절처리구(259.8cm/15.1mm)에서 가장 낮은 경향을 보였다. 그러나 식재 이후 2년간 순수고생장량에 있어서는 1m 대절 처리구가 241.4cm로 가장 높았으며, 다음으로 무대절 처리구(212.7cm) 및 지제부대절 처리구(181.6cm) 순으로 나타났다. 2년 동안의 지상부 목질계 바이오매스 생산량을 추산한 결과, 무대절 처리구(약 4.3ton/ha)에서 가장 높았고, 다음으로 1m 대절 처리구(약 4.0ton/ha) 및 지제부 대절 처리구(약 2.6ton/ha) 순으로 나타났다. 고사율의 경우 1m 대절 처리구가 약 10.0%로 무대절 처리구(26.2%) 및 지제부 대절 처리구(30.1%)에 비해 현저히 낮았으며, 수관활력도 및 잎 피해도 역시 가장 양호한 것으로 조사되었다. 종합적으로 식재 후 2년 간 지상부 목질계 바이오매스의 총 생산량은 무대절 처리구에서 가장 높은 것으로 추산되었으나 간척지 환경을 고려한 적응지수는 1m 대절 처리구에서 가장 높은 것으로 평가되었다. This study was conducted to investigate the growth characteristic and wood biomass production for a short rotation coppice plantation with differently coppiced poplar established in Saemangeum reclaimed land in 2012. Total tree hight and DBH after two growing seasons were highest in non-coppiced plantation (347.8㎝/19.5㎜) followed by 1m hight-coppiced (324.5㎝/18㎜) and ground level-coppiced plantation (259.8㎝/15.1㎜). However, net growth of tree hight during the two growing seasons was highest in 1m hight-coppiced plantation (241.4㎝) followed by non-coppiced (212.7㎝) and ground level-coppiced plantation (181.6㎝). Above ground wood biomass production during the 2 years after plantation was estimated to be highest in non-coppiced plantation (about 4.3 ton/ha) followed by 1m hight-coppiced (about 4.0 ton/ha) and ground level-coppiced plantation (about 2.6 ton/ha). However, the best performance in terms of survival rate, vitality of tree crown and degree of leaf injury was showed in 1m hight-coppiced plantation. Overall adaptability concerning the environmental conditions of reclaimed land was also highest in 1m hight-coppiced plantation.

        A Case of Inadvertent Anterior Chamber and Corneal Stromal Injection with Antibiotics during Cataract Operation

        Purpose: To report a case of inadvertent anterior chamber and cornea stromal injection with high dose antibiotics and steroids during cataract operation. Methods: During cataract operation on a 78 year-old female patient, high dose gentamicin (20 mg/0.5 ml) and dexamethasone (2 mg/0.5 ml) were inadvertently injected into the anterior chamber and cornea stroma when making cornea edema for sealing of the incision sites. Anterior chamber irrigation with balanced salt solution (BSS) was immediately administered. On postoperative day one, extensive cornea edema was noted, and best-corrected visual acuity was 0.2. Descemet`s membrane folds were observed around the corneal incision sites. Topical 5% NaCl and 1% prednisolone were started. Results: Four weeks postoperatively, corneal edema began to reduce significantly. At four months postoperatively, corneal edema fully resolved, and best-corrected visual acuity was 0.8. However, some Descemet`s membrane folds still remained, and a decrease in the number of endothelial cells was noted by specular microscope. Conclusions: In this case involving anterior chamber and cornea stromal injection with high dose antibiotics and steroids, immediate anterior chamber irrigation with balanced salt solution seemed an appropriate management, and the patient`s long-term visual acuity appears good. To prevent such mistakes, precise labeling of all solutions and use of different syringe needles should be considered.

        Mitomycin C Does Not Inhibit Exacerbation of Granular Corneal Dystrophy Type II Induced by Refractive Surface Ablation

        PURPOSE:: To evaluate the effect of mitomycin C (MMC) on the exacerbation of corneal opacity that occurs in patients with granular corneal dystrophy type II (GCD II) after refractive corneal surface ablation. METHODS:: Ten eyes of patients with GCD II who underwent refractive corneal surface ablation with MMC were compared with 10 eyes that were not treated with MMC. Best spectacle-corrected visual acuity, the degree of corneal opacity, and contrast sensitivity were evaluated at least 3 years after surgery. Corneal opacities were quantified using Pentacam densitometry maps. RESULTS:: No measured between-group value showed a statistically significant difference. CONCLUSION:: Simultaneous application of MMC does not prevent exacerbation of GCD II after refractive corneal surface ablation.
