      • Species Diversity of Caterpillars on Feeding on the foliage Willow Trees (Salix spp.) in the Korean Penninsula

        Most caterpillars, the larvae of butterflies and moths(Lepidoptera), are herbivorous. Sometimes they are regarded as serious agricultural pest that cause to damage agricultural produce, whereas some species such as Bombyx mori is known to be valued as sources of silk. Even though their major role in the ecosystem, details of biology are poorly known for most species. This study has been carried out to identify larvae and to acquire useful biological information of insect larvae through rearing from 1997 to 2014 in HECRI(Holoce Ecosystem Conservation Research Institution). From 2012 HECRI are carrying out ‘Larvae Identification and Specimen Security of Primary Endemic Moths in Korea Peninsula through Rearing’ supported by National Institute of Biological Resources(NIBR). Of these this study is the list about special caterpillars feeding on the foliage Willow Trees (Salix spp.) Total 143 species belonging to 13 families were collected and identified 69 species belonging to 12 families. Among them Willow Treesdependent monophagous caterpillars are 34 species belonging to 5 families, oligophagous 36 species belonging to 3 families and polyphagous 16 species belonging to 12 families. The dominant family is Geometridae(51 species), second is Noctuidae(35 species) and followed by Notodontidae(21 species).

        Regulation of Membrane-Associated Mucins in the Human Corneal Epithelial Cells by Dexamethasone

        PURPOSE:: To study the influence of dexamethasone on membrane-associated mucins produced by human corneal epithelial cells. METHODS:: Human corneal epithelial cells were cultured in medium supplemented with various concentrations of dexamethasone (ranging from 10 to 10 M). Reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis using monoclonal antibodies specific for human MUC1 (HMFG-1), MUC4 (1G8), and MUC16 (OC125) were performed to evaluate the effect of dexamethasone on membrane-associated mucin expression. The effect of glucocorticoid receptor antagonist (RU38486) on dexamethasone-induced mucin expression was estimated. RESULTS:: RT-PCR revealed that MUC1 and MUC16 gene expression were upregulated 48 hours after addition of dexamethasone and that MUC4 gene expression was downregulated in the same condition. Western blot analysis showed that MUC1 and MUC16 proteins were increased after addition of dexamethasone. However, MUC4 was not detected by anti-MUC4 monoclonal antibody (1G8) for ASGP-2 under our conditions. Treatment with RU38486 inhibited the changes of MUC1, MUC4, and MUC16 by dexamethasone; thus, the effect of dexamethasone on mucin expression is mediated by glucocorticoid receptors. CONCLUSIONS:: This study shows that MUC1, MUC4, and MUC16 are regulated differently by dexamethasone in human corneal epithelial cells. External application of dexamethasone might affect the precorneal mucin.

        Effects of Y-27632, a Rho-associated Kinase Inhibitor, on Human Corneal Endothelial Cells Cultured by Isolating Human Corneal Endothelial Progenitor Cells

        Purpose: Human corneal endothelial progenitor cells (HCEPs), which has been selectively isolated and differentiated into human corneal endothelial cells (HCECs), are crucial for repairing corneal endothelial damage. In this study, we evaluated the roles of a Rho-assisted kinase (ROCK) inhibitor, Y-27632, on the isolation and expansion of HCEPs, and assessed the in vitro effects of different concentrations of Y-27632 on the differentiated HCEPs. Methods: HCEPs were isolated and expanded in a medium with and without 10μM Y-27632, and then differentiated into HCECs in a medium with fetal bovine serum. The characteristics of HCEPs and differentiated HCEPs were confirmed by immunofluorescence staining. The proliferation, viability, morphology, and wound-healing ability of differentiated HCEPs were assessed in the presence of different concentrations of Y-27632. Results: Y-27632 enabled the isolation and expansion of HCEPs from the corneal endothelium. The differentiated HCEPs showed an optimal increase in proliferation and survival in the presence of 10μM Y-27632. As the concentration of Y-27632 increased, differentiated HCEPs became elongated, and actin filaments were redistributed to the periphery of cells. Y-27632 also caused a concentration-dependent enhancement in the wound-healing ability of differentiated HCEPs. Conclusions: Y-27632 enabled the isolation and expansion of HCEPs. It also enhanced the proliferation, viability, and migration of differentiated HCEPs.

        불국사 상업지역의 정체성확보를 위한 간판 개선방안

        간판은 도시의 이미지 형성과 정보전달에 큰 역할을 한다. 하지만 우리나라의 간판은 정체성과 미학적 특질이 부재하고 정보전달기능이 미약하기 때문에 경관을 훼손하는 요소이다. 이러한 현상은 역사문화지역에서 더 심각한 실정이다. 이는 역사도시에서 더욱 큰 영향을 초래하고 있다. 최근 많은 지자체는 간판개선을 실시하고 있으나 지역과 어울리지 않은 획일적인 디자인과 관련주체의 참여부족으로 인하여 그 효과는 미진한 것으로 나타나고 있다1)2)3)4). 본 연구는 불국사 상업지역을 대상으로 지역정체성 확보를 위한 간판개선방안을 제시하고자 한다. 구체적으로 본 연구에서는 불국사의 상업주와 방문자, 관련 공무원을 중심으로 설문조사가 진행되었다. 설문조사에서는 상업주 97명, 방문자 130명, 관련 공무원 32명이 참여하였다. 연구 결과는 세 가지로 요약할 수 있다. 첫째. 현재 간판현황에 대한 만족도는 3주체 모두 낮게 나타났으나, 방문자의 만족도가 다른 두 주체보다 더 낮게 나타나고 있다. 둘째, 모든 응답자들은 현 간판이 정보전달기능이 낮고 지역정체성도 반영하지 않는다고 인식하고 있어, 간판 개선이 필요함을 알 수 있다. 간판개선이 특히 시급한 것으로 인식하는 요소는 디자인 요소의 배치와 색상, 문구로 나타났다. 마지막으로, 관련 공무원들은 상업주의 의식부족과 현실에 맞지 않는 법규 등이 개선대상으로 인식하고 있었다. 이와 같은 연구에 근거하면 지역정체성확보를 위한 간판 개선을 위하여 제도적 방안과 디자인적 방안이 제시될 수 있다. 우선 제도적 방안으로는 현실과 지역에 맞는 이해하기 쉬운 법령의 개선이 필요하다. 또한 실제 적용에 필요한 구체적인 지침이 마련되어야 한다. 디자인적 방안으로는 지역 정체성을 나타내는 이미지의 개발이 우선되어야 한다. 이와 함께, 간판의 기능을 극대화 시킬 수 있는 디자인 요소의 배치와, 대상지 주변 색상의 활용 및 대상지 이미지와 알맞은 문구의 도입이 필요하다. Outdoor advertizing signs of the city plays an important role in formulating the image of the city and communicating information. But outdoor advertizing signs of Korea are more likely to damage urban landscape because of the lack of the aesthetic quality, local identity, and communicating ability. This negative effect is more serious in the historic city. Recently, many local governments plan and implement the projects for the improvement of outdoor advertizing signs, which are proved to be ineffective due to the uniformed design and the lack of participation of the related stakeholders 1)2)3)4). This study aims to suggest the guidelines for the improvement of outdoor advertizing signs in Bulguksa business district in order to establish the sense of identity in the street. Specifically, questionnaire survey was performed to the three stakeholders, such as 97 storeowners, 130 visitors, and 32 related public officials. The results revealed three important things. First, although the satisfaction for the present sings of the three stakeholders were low, the satisfaction of the visitors was found lowest, compared to the other two stakehoders. Second, all the participants thought that the present signs neither successfully communicated information nor reflected the regional identity, which indicated the need of improvement. The sign factors that found to need the urgent improvement were the disposition of design elements, colors, and wording. Lastly, the public officials pointed the lack of storeowners` sense on the good signs and the unrealistic regulations as the targets of improvement. Based on these results, two kinds of guidelines for sign improvement were suggested; one for regulation and the other for design. It is important that the regulation should be realistic and correspond to the characteristics of the region. Also specific information for implementation should be prepared. For design, the first and important thing is to develop the representative regional image. Then, endeavors are needed to find the ways to lay out the design elements, to utilize the surrounding colors, and to use the appropriate wordings.

        인공눈물 점안 후 안구 표면 두께 간섭계를 이용한 눈물 지질층 두께의 변화 분석

        목적: 농도 및 보존제 유무에 따른 히알루론산 나트륨제제의 인공누액 점안 후 LipiView?? II 간섭계(TearScience Inc., Morrisville, NC, USA)를 이용하여 눈물 지질층 두께의 시간에 따른 변화를 측정, 비교 분석함으로써, 인공눈물의 사용이 LipiView?? II 검사 결과에 미치는 영향을 분석하고자 한다. 대상과 방법: 건성안 환자를 대상으로 전향적 연구를 시행하였다. 모든 대상자에게 눈물막 파괴 시간 측정, 쉬르머 검사, 각막 및 결막형광염색 검사, 마이봄샘 기능부전 평가, ocular surface disease index 설문조사를 실시하였으며, 대상자를 난수표를 이용한 무작위 추출로 인공누액 종류에 따라 세 그룹(1군: 무보존제 0.1% 히알루론산 나트륨제제 인공눈물, 2군: 무보존제 0.3% 히알루론산 나트륨 인공눈물, 3군: 0.003% 염소 벤잘코니움을 포함하는 0.1% 히알루론산 나트륨제제 인공눈물)으로 나눠 각각의 인공눈물을 점안 전, 직후, 1, 3, 6시간 후의 눈물지칠층의 두께를 LipiView?? II 간섭계로 측정 후 비교 분석하였다. 안검염의 유무에 따라 두 군으로 나누어 눈물지칠층의 두께를 추가로 분석하였다. 결과: 눈물 지질층의 두께를 측정한 결과, 인공눈물제제를 점안하고 3시간 뒤 눈물 지질층의 두께 변화가 유의하게 나타났다(p=0.03). 제1군 및 2군에서 시간에 따른 눈물 지질층 두께의 변화가 유의하게 측정되었고(p<0.01, p<0.01), 제3군에서는 눈물 지질층 두께 변화가 관찰되지 않았다(p=0.71). 환자를 안검염 그룹과 비안검염그룹으로 세분화하여 분석해 보았을 때, 비안검염 그룹에서는 인공눈물점안과 상관 없이 눈물지질층의 두께가 일정하였고(p=0.19), 안검염 그룹에서는 제1군과 2군에서 인공눈물 점안 후 시간 경과에 따라 눈물지질층 두께가 변화하였다(p<0.01, p=0.01). 안검염 그룹에서 제1군과 2군의 눈물 지질층의 두께를 비교하면, 인공눈물 점안 직후 및 1시간, 3시간 뒤까지 유의한 차이를 보였으며(p=0.04, p<0.01, p=0.02), 6시간째에는 유의한 차이가 없었다. 제1군과 3군의 경우에는 시간별 차이가 없었다(p=0.08). 결론: LipiView?? II 간섭계를 이용해 측정한 눈물 지질층 두께는 인공눈물의 농도와 보존제의 유무에 따라서도 수치의 차이를 보였으며, 안검염의 유무에 따라서도 인공눈물 점안 후 눈물 지질층의 두께 변화 양상에 차이가 있었다. 이는 LipiView?? II 간섭계로 눈물 지질층을 측정하는 경우 인공눈물 종류와 관계없이 적어도 6시간의 점안을 금해야 하며, 또한 안검염이 있는 환자군과 보존제를 포함하는 인공눈물을 사용하는 환자에서 더욱 유의해야 함을 시사한다. Purpose: The changes in tear film lipid layer thickness (LLT) after artificial tears application using LipiView??II interferometer were assessed. Methods: We performed a prospective study of patients with dry eye disease. All subjects underwent measurement of tear film break-up time, Schirmer test, ocular surface staining, meibomian gland evaluation, and subjective score assessment using the Ocular Surface Disease Index. All subjects were randomly assigned to 1 of 3 groups using table of random numbers (group 1, sodium hyaluronate [HA] 0.1% eye drops without preservatives; group 2, HA 0.3% eye drops without preservatives and group 3, HA 0.1% with benzalkonium chloride 0.003%). LLT was measured before, immediately after and 1 hr, 3 hrs, and 6 hrs after artificial tears application. Additionally, the patients were divided into 2 subgroups depending on the presence of meibomian gland dysfunction (MGD) and further evaluated. Results: Significant change in LLT was observed at 3 hrs after artificial tears instillation. LLT in groups 1 and 2 showed significant changes over time (p < 0.01 and p < 0.01, respectively). However, LLT in group 3 showed no change. LLT was unchanged in patients without MGD. Conversely, in MGD patients, a significant difference in LLT between groups 1 and 2 was observed immediately after and 1 hr and 3 hrs after instillation of artificial tears (p = 0.04, p < 0.01 and p = 0.02, respectively) but not at 6 hrs. However, no significant difference in LLT between groups 1 and 3 was observed in MGD patients. Conclusions: LLT after instillation of artificial tears measured using LipiView??II interferometer was affected by artificial tear concentration and presence of preservatives. Additionally, the presence of MGD can impact the pattern of LLT changes induced by artificial tear instillation. Therefore, LLT measurements using LipiView??II interferometer require at least a 6-hrs interval after use of eye drops, especially for patients with MGD or using artificial tears with preservatives.

        스포츠 활동프로그램이 비만여자 초등학교 4학년의 혈중지질과 비만인자에 미치는 영향

        The purpose of this study is to investigate the effects of exercise programs on the fourth grade of obese female elementary schoolers. For the purpose, this study selected the fourth grade of 35 obese female elementary schoolers, whose body fat percentage was over 22%. Among them, this study selected 10 students for an experimental group, who continued to participate in aerobic and strength exercise programs for 12 weeks, and 10 students for a control group, who stopped participating in the exercise program within a week from the beginning. This study found that among the obesity factors, levels of leptin and body fat percentages showed significant differences between the experimental group and the control group while levels of adiponectin and ghrelin did not show significant differences. This findings indicate that the fourth grade of female elementary schoolers in the early stages of growth and puberty experienced mixed effects of blood lipids and obesity factors due to various hormonal actions and genetic factors. However significantly different levels of leptin and body fat were found between the experimental and control groups, meaning that regular exercise is necessary for obese female elementary schoolers.

        CD166 promotes the cancer stem-like properties of primary epithelial ovarian cancer cells

        Cancer stem cells (CSCs) or tumor-initiating cells are thought to play critical roles in tumorigenesis, metastasis, drug resistance, and tumor recurrence. For the diagnosis and targeted therapy of CSCs, the molecular identity of biomarkers or therapeutic targets for CSCs needs to be clarified. In this study, we identified CD166 as a novel marker expressed in the sphereforming CSC population of A2780 epithelial ovarian cancer cells and primary ovarian cancer cells. The CD166+ cells isolated from A2780 cells and primary ovarian cancer cells highly expressed CSC markers, including ALDH1a1, OCT4, and SOX2, and ABC transporters, which are implicated in the drug resistance of CSCs. The CD166+ cells exhibited enhanced CSC-like properties, such as increased sphere-forming ability, cell migration and adhesion abilities, resistance to conventional anticancer drugs, and high tumorigenic potential in a xenograft mouse model. Knockdown of CD166 expression in the sphereforming ovarian CSCs abrogated their CSC-like properties. Moreover, silencing of CD166 expression in the sphere-forming CSCs suppressed the phosphorylation of focal adhesion kinase, paxillin, and SRC. These results suggest that CD166 plays a key role in the regulation of CSC-like properties and focal adhesion kinase signaling in ovarian cancer. [BMB Reports 2020; 53(12): 622-627]
