      • Enhancement of the hydrogen productivity in microbial water gas shift reaction by <i>Thermococcus onnurineus</i> NA1 using a pressurized bioreactor

        <P><B>Abstract</B></P> <P>Here, we developed a pressurized bioreactor system that increase carbon monoxide (CO) transfer efficiency in order to enhance the hydrogen productivity in the microbial water gas shift reaction by <I>Thermococcus onnurineus</I> NA1. The effects of CO pressure on the hydrogen production rate, CO consumption rate and the cell growth were investigated using small scale stainless steel bottles at various CO partial pressures. It was found that CO solubility increased by applying pressure can affect hydrogen production positively as long as the increased toxicity of CO is endurable to cells. The hydrogen productivity increased to some extent with CO pressure, but decreased drastically at the pressure higher than 4 bar. On the other hand, the effect of pressure itself on the cell's activity was not as significant as that of CO solubility increase. In the experiments at various system pressures with identical CO partial pressure of 1 bar, more than 80% of the cell activity remains even at total pressure of 10 bar. Also, it was important to determine the appropriate time to increase pressure for preventing excess CO in the reactor. Based on these results, a fermentation strategy for the pressurized system was designed and applied to a 5 L bioreactor with the continuous supply of the gas containing 60% CO. When the pressure was introduced to the bioreactor up to 4 bar at CO limitation condition, the unprecedented high productivity (360 mmol L<SUP>−1</SUP> h<SUP>−1</SUP>) could be obtained.</P> <P><B>Highlights</B></P> <P> <UL> <LI> CO gas solubility increased by pressurizing can be beneficial to H<SUB>2</SUB> production. </LI> <LI> The impact of pressure per se was marginal. </LI> <LI> CO toxicity impacts were reduced by applying pressure at mass transfer limitation condition. </LI> <LI> When the pressure was introduced to the bioreactor, the unprecedented high productivity could be obtained. </LI> </UL> </P>

        Rapid and Visual Detection of Barley Yellow Dwarf Virus by Reverse Transcription Recombinase Polymerase Amplification with Lateral Flow Strips

        Barley yellow dwarf virus (BYDV) has been a major viral pathogen causing significant losses of cereal crops including oats worldwide. It spreads naturally through aphids, and a rapid, specific, and reliable diagnostic method is imperative for disease monitoring and management. Here, we established a rapid and reliable method for isothermal reverse transcription recombinase polymerase amplification (RT-RPA) combined with a lateral flow strips (LFS) assay for the detection of BYDV-infected oat samples based on the conserved sequences of the BYDV coat protein gene. Specific primers and a probe for RT-RPA reacted and optimally incubated at 42oC for 10 min, and the end-labeled amplification products were visualized on LFS within 10 min. The RT-RPA-LFS assay showed no cross-reactivity with other major cereal viruses, including barley mild mosaic virus, barley yellow mosaic virus, and rice black streaked dwarf virus, indicating high specificity of the assay. The sensitivity of the RT-RPA-LFS assay was similar to that of reverse transcription polymerase chain reaction, and it was successfully validated to detect BYDV in oat samples from six different regions and in individual aphids. These results confirm the outstanding potential of the RT-RPA-LFS assay for rapid detection of BYDV.

      • Improving the subcritical water extraction of flavonoids narirutin and hesperidin from Citrus unshiu peel by pulsed electric fields

        Citrus fruit is important source of flavonoids, mainly flavanones which are narirutin and hesperidin. Those citrus flavonoids have been found to have health-related properties including antioxidant, anticancer, and anti-inflammatory. The main purpose of this study was to verify that the extraction of narirutin and hesperidin from Citrus peel can be more effective by combining pulsed electric field (PEF) pre-treatment and subcritical water extraction (SWE). Citrus unshiu peels were treated with PEF under conditions of electric field strength (3 kV/cm) and times (1 and 2 min). Subsequent SWE was conducted by using a Dionex Accelerated Solvent Extractor (ASE, Model 350) at extraction temperature 170°C for 10 min. The total flavonoids content was measured by using the aluminum chloride colorimetric method and the antioxidant capacity was analyzed by the Ferric reducing antioxidant power (FRAP) assay using spectrophotometer. The concentrations of narirutin and hesperidin were increased as PEF pre-treatment time increased. The highest concentrations of narirutin and hesperidin were 13.41 mg narirutin/g dry citrus peel and 141.16 mg hesperidin/g dry citrus peel at PEF pre-treatment condition of 3 kV/cm and 2 min. The total flavonoids contents of the extracts increased 105.2% and 123.1% for citrus peel PEF treated at 1 and 2 min, respectively. In addition, compared to the untreated sample, PEF pre-treatments of 1 and 2 min increased the antioxidant capacity of the extracts 109.2% and 160.8%, respectively. Therefore, the results demonstrate the potential of PEF pre-treatment to improve the SWE of flavonoids from citrus unshiu peel.

      • 여가활동 프로그램이 정신지체아동의 사회적 기술에 미치는 효과

        본 연구에서는 정신지체아동에게 놀이 중심의 여가 활동이 이들의 사회적 활동에 미치는 향요인에 대하여 알아보고자 하였다. 연구대상은 특수학교 고등부 정신지체 학생 20명을 대상으로 하였으며,사회적 기술의 도구는 Gresham Elliott(1990) 의 사회적 기술평정척도(Social Skills Rating System: SSRS) 중에서 사회적 기술척도인 협동,자기주장,자아통제 을 측정하였다. 그 결과로는 여가활동은 정신지체아동의 전반석인 사회적 기술에 영향을 치는 요인으로 나타났으며 정신지체아동의 협동을 증진시키는 영향요인으로 나타났으나,사회적 기술의 하위변인인 자기주장과 자아통제는 통계적으로 유의한 차이가 나타나지 않았다. The purpose of this study was investigating the effects of leisure activities prograrn on social skill of students with mental retardation. The subjccts of this study, were 20 high school students of special school locateel in a Kyongbuk province. The test instrument utilized in this studly was Social Skills Rating System developed by Greshum and Elliott(990). The collected data was analyzed by t-test using the SPSS/PC for windows 9.0 program.

        Reverse Transcription Recombinase Polymerase Amplification Assay for Rapid and Sensitive Detection of Barley Yellow Dwarf Virus in Oat

        Barley yellow dwarf virus (BYDV) is an economically important plant pathogen that causes stunted growth, delayed heading, leaf yellowing, and purple leaf tip, thereby reducing the yields of cereal crops worldwide. In the present study, a reverse transcription recombinase polymerase amplification (RT-RPA) assay was developed for the detection of BYDV in oat leaf samples. The RT-RPA assay involved incubation at an isothermal temperature (42°C) and could be performed rapidly in 5 min. In addition, no cross-reactivity was observed to occur with other cereal-infecting viruses, and the method was 100 times more sensitive than conventional reverse transcription polymerase chain reaction. Furthermore, the assay was validated for the detection of BYDV in both field-collected oat leaves and viruliferous aphids. Thus, the RT-RPA assay developed in the present study represents a simple, rapid, sensitive, and reliable method for detecting BYDV in oats.

      • 결장직장암 환자의 임상적 고찰 : 조선대학교 부속병원의 10년 간의 결과

        Background and Objectives: The colorectal cancer rates 4th in the total cancer prevalence in Korea. The clinical patient demographics were considered with the incidence rate of the colon and rectal cancer. The clinicopathological factors were analyzed for the 5 year survival rate. Materials Methods: Of the 282 patients who were operated from Jan. 1, 1992 to Dec. 31, 2001 and diagnosed by pathological biopsy as colo-rectal cancer, 234 patients were followed up. The age, sex, location of the cancer, TNM stage, operation method, histopathological classification, metastatic region, complications after operation and the survival rate were investigated. Results: Of the 234 patients, male and female consisted of 120 cases (51.3%) and 114 cases (46.7%), with an average age of 61.6 years. The average follow up period was 52.3 months and the peri operative mortality was 5.6%. The site of the cancer was most frequent at the rectum with 127 cases (54.3%). The patients were classified according to the TNM stages with 38 cases of stage 1 (16.2%), 98 cases of II (41.9%), 72 cases of III (30.8%) and 26 cases of IV (11.1%). The 5 year survival rate in the age group older than 50 was 63.3 % and 73.5% in the group younger than 50. In regard to the operation type, the 5 year survival rate was 65.4% for elective operation and 65.8% for emergency operation. The 5 year survival rate for well differentiated, moderately differentiated, poorly differentiated cancer, and mucinous adenocarcinoma was 73.7%, 60.2%, 58.3%, and 92.6%, respectively. The 5 year survival rate of each of the TNM stage is 81.1%, 74.8%, 57.0%, and 11.5% for Classes I, II, III, and IV, respectively. Conclusion: The clinical factors were analyzed for the 5 year survival rate with no statistical significance for the age, sex, and location of the cancer. The TNM stage and histopathological differentiation were statistically significant. The colon cancer showed better prognosis compared to the rectal cancer.

      • Comparison of the rebound tonometer (TonoVet<sup>®</sup>) with the applanation tonometer (TonoPen XL<sup>®</sup>) in normal Eurasian Eagle owls (<i>Bubo bubo</i>)

        <P>Abstract</P><P>Objective </P><P>To examine the feasibility and accuracy of a handheld rebound tonometer, TonoVet<SUP>®</SUP>, and to compare the intraocular pressure (IOP) readings of the TonoVet<SUP>®</SUP> with those of an applanation tonometer, TonoPen XL<SUP>®</SUP>, in normal Eurasian Eagle owls.</P><P>Animals studied </P><P>Ten clinically normal Eurasian Eagle owls (20 eyes).</P><P>Procedures </P><P>Complete ocular examinations, using slit-lamp biomicroscopy and indirect ophthalmoscopy, were conducted on each raptor. The IOP was measured bilaterally using a rebound tonometer followed by a topical anesthetic agent after 1 min. The TonoPen XL<SUP>®</SUP> tonometer was applied in both eyes 30 s following topical anesthesia.</P><P>Results </P><P>The mean ± SD IOP obtained by rebound tonometer was 10.45 ± 1.64 mmHg (range 7–14 mmHg), and by applanation tonometer was 9.35 ± 1.81 mmHg (range 6–12 mmHg). There was a significant difference (<I>P =</I> 0.001) in the IOP obtained from both tonometers. The linear regression equation describing the relationship between both devices was <I>y</I> = 0.669<I>x</I> + 4.194 (<I>x</I> = TonoPen XL<SUP>®</SUP> and <I>y</I> = TonoVet<SUP>®</SUP>). The determination coefficient (<I>r</I><SUP>2</SUP>) was <I>r</I><SUP>2</SUP> = 0.550.</P><P>Conclusions </P><P>The results suggest that readings from the rebound tonometer significantly overestimated those from the applanation tonometer and that the rebound tonometer was tolerated well because of the rapid and minimal stress-inducing method of tonometry in the Eurasian Eagle owls, even without topical anesthesia. Further studies comparing TonoVet<SUP>®</SUP> with manometric measurements may be necessary to employ rebound tonometer for routine clinical use in Eurasian Eagle owls.</P>

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        영구치 치수 기질세포를 이용한 연골 분화 및 분화 시기에 따른 형태학적 변화

        Objectives: The aim was to confirm the stem cell-like properties of the dental pulp stromal cells and to evaluate the morphologic changes during in vitro chondrogenesis. Materials and Methods: Stromal cells were outgrown from the dental pulp tissue of the premolars. Surface markers were investigated and cell proliferation rate was compared to other mesenchymal stem cells. Multipotency of the pulp cells was confirmed by inducing osteogenesis, adipogenesis and chondrogenesis. The morphologic changes in the chondrogenic pellet during the 21 day of induction were evaluated under light microscope and transmission electron microscope. TUNEL assay was used to evaluate apoptosis within the chondrogenic pellets. Results: Pulp cells were CD90, 105 positive and CD31, 34 negative. They showed similar proliferation rate to other stem cells. Pulp cells differentiated to osteogenic, adipogenic and chondrogenic tissues. During chondrogenesis, 3-dimensional pellet was created with multi-layers, hypertrophic chondrocyte-like cells and cartilage-like extracellular matrix. However, cell morphology became irregular and apoptotic cells were increased after 7 day of chondrogenic induction. Conclusions: Pulp cells indicated mesenchymal stem cell-like characteristics. During the in vitro chondrogenesis, cellular activity was superior during the earlier phase (within 7 day) of differentiation.
