      • Capsicum chinense, 재래종 및 저항성 고추 유전자원에서 역병 저항성 계통 선발

        2000년도에 검정에서 역병에 살아남은 개체들로부터 채종하여 육성한 Capsicum chinense 31계통에 대하여 역병 저항성을 검정한 결과 고도의 저항성을 나타내는 것은 발견되지 않았다. 2001년도의 검정에서 역병에 살아남은 개체로부터 채종한 재래종 26계통에 대하여 다시 역병 저항성 검정을 실시한 결과 KC180, KC230, KC195, KC194에서 다수의 개체가 살아남아 저항성을 나타내었다. 그러나 KC180과 KC230은 각각 AC2258과 CM334와 혼종된 것으로 관찰되었다. KC195와 KC194는 재래종의 형질을 유지하고 있는 것으로 관찰되었다. CM334의 보존 증식과정에 자연교잡이 일어난 것으로 보여 이의 순도향상을 위하여 채종년도별로 시료를 꺼내어 역병저항성 검정을 실시한 결과 가장 오래된 1992년도 채종종자에서부터 약간의 이형주가 관찰도기 시작하여 1995년부터 2001년도까지 시간이 경과함에 따라 많이 변형되어 있는 것을 알 수 있었다. 1992년도 종자에서 이형주를 제거하고 원형의 개체로부터 자식종자를 대량으로 채종하였다. 함께 공시한 AC2258은 순수한 것으로 확인되었다. 1995년도 채종 CM334 종자에서는 비록 혼종은 되었으나 측지발생이 적은 개체들이 발견되어 이들을 개체 선발하여 역병에 저항성이며 측지발생이 적은 계통으로 육성하고 있다. A total of 31 c. chinense lines selected from 2000 screening were tested again for resistance to P. capsici but resistance was not found in the lines. A total of 26 selections from Korean land races tested 2001 spring were tested again for resistance to P. capsici. KC180, KC230, KC195 and KC194 showed moderate resistance to P. capsici. However, it was apparent on the basis of hortivultural characteristics that KC180 and KC230 had been naturally crossed with AC2258 and CM334, respectively. CM334 and AC2248 seed lots that were increased in different years were taken out and tested for resistance to improve their genetic purity because the resistant genetic resources have been showing some off-types in the population. Off-types began to be found in 1992 seed lots and the proportion and degree of the offs was increasing with time up to 2001. Plants true to the type in 1992 seed lots were selected and their inbred seeds were mass produced in a net cage in the greenhouse. AC2258 included in the experiment number of lateral branching at cotyledonary axil, although they were off from the original in the experiment together was uniform. In 1995 seed lost of CM334, plants with resistance to P. capsici and low or no number of lateral branching at cotyledonary axil, although they were off from the original CM334, were found and selection was applied to breed lines fixed in the characters.

      • TNF-α 유전자형과 방광암과의 관계

        연구목적: TNF-α는 일부 종양의 종양화 과정과 관련이 있는 것으로 알려져 있다. 본 연구는 TNF-α 발현에 영향을 미치는 TNF-α 촉진자 -308 부위의 유전적 다형성이 방광암과 관련이 있는지 유무를 알고자 시행하였다. 대상 및 방법: 유전자 분석을 위하여 환자 113명 및 대조군 109명으로부터 혈액을 채취하여 genomic DNA를 분리한 후 PCR-RFLP 및 direct DNA sequencing을 통하여 TNF-α유전자의 다형성을 조사하여 방광암의 발생, 병기 및 분화도와 비교 검토하였다. 결과: TNF-α 촉진자 -308 부위의 유전형은 대조군에서는 GG형이 83.5%(90례 및 GA형이 16.5%(19례)로 관찰되었으며 AA형은 없었다. 환자군에서는 GG 형이 85.4%(97례), GA형 및 AA형은 각각 13.1%(15례)및 0.8%(1례)에서 관찰되었다. 두 군 모두에서 GG형이 가장 많이 나타났으며 다음으로 GA형을 보이고 AA형은 1례의 방광암 환자에서만 관찰되었다. -308부위의 경우도 두 군 사이에 유전자형의 차이는 없었다(p=0.259) 분화도별 분포를 보면 grade I이 20례, grade II가 49례, grade Ⅲ은 34례였고 병기별로 표재성인 경우가 90례였으며 침윤성은 14례였다. 분화도가 나빠질수록 GA형이 증가하였다(p=0.04). 그러나 병기와 TNF-α promoter -308부위의 유전자형 사이에는 유의한 상관 관계가 없었다(p=0.123). 결론: 방광암 환자의 혈액에서 GA genotype이 관찰되는 경우, 분화도가 나쁠 가능성이 매우 높기 때문에 좀 더 적극적인 치료와 세밀한 추적관찰을 함으로써 방광암으로 인한 사망과 암의 진행을 예방할 수 있을 것으로 기대한다. Purpose : Tumor necrosis factor-alpha (TNF-α) is involved in tumorigenesis of several cancers as an endogenous tumor promoter. The purpose of this study was to investigate whether genetic polymorphism of TNF-α promoter region (-308) was associated with human bladder tumor. Materials and Methods: The DNA from 113 and 109 respective blood samples of bladder tumor Patients and control group was analyzed by PCR-based restriction fragment length polymorphism (RFLP) and direct DNA sequencing methods to characterize the genetic polymorphism of -308 promoter region of the TNF-α gene in bladder tumor patients. We compared the association of bladder tumor with genetic Polymorphism of TNF-α promoter region(-308) in relation to the stage, grade, recurrence and progressio. Results : Eighty-six percents(97/113) of bladder tumor patients and 83.5% (90/109) of control group showed genotype GG at -308 region of TNF-α. Difference in genetic variations of TNF-α promoter (-308) did not exist between bladder tumor patients and control group(p=0.259). Tumor grade was significantly related to the GA genotype (p=0.04). The higher is the grade in bladder tumor, the more frequent was the GA genotype. Tumor stage, recurrence and progression were not significantly associated with genetic polymorphism of TNF-α promoter region (-308). Conclusion: The GA genotype of TNF-a promoter region (-308) had a significant impact on TNF-α production and was related to higher grade tumor compared to GG genotype. TNF-α serum levels in bladder tumor patients were significantly higher than controls. These data suggested that TNF-α might involve the tumorigenesis of the bladder rather than treatment or prevention of bladder tumor.

      • 조혈모세포이식 환자의 삶의 질에 대한 연구 : 화학요법 환자 및 정상인과의 비교

        연구배경: 조혈모세포이식후 신체 및 정신적 장애의 정도가 얼마나 환자의 '삶의 질(QOL)'에 영향을 미치는지에 대하여 외국에서는 연구들이 활발히 진행되고 있다. 그러나 우리나라에서는 조혈모세포이식이 비교적 활발히 시술되고 있음에도 불구하고 이에 대한 연구가 미미한 실정이었다. 연구자 등은 우리 나라에서 조혈모세포이식을 받은 환자들의 QOL은 어떤지를 알아보고자 하는 목적으로 본 연구를 시행하였다. 방법: 본 연구에 참여한 각 기관에서 조혈모세포이식을 받고 주된 치료가 끝난 후 1999년 2월 시점에서 최소 3개월이 지나고 관해상태에서 일상생활을 영위하고 있는 재생불량성 빈혈과 급성 및 만성 백혈병 환자들을 각각 자가 및 동종 조혈모세포이식을 시행받은 조혈모세포이식군(1군)과 항림프구 글로불린, 공고요법 등으로 치료를 끝낸 화학요법군(2군)으로 분류하고 연구자 병원에 내원한 건강한 환자 보호자들로 이루어진 정상대조군(3군)을 선정하여 각각의 QOL을 비교, 분석하였다. QOL의 측정은 EORTC QLQ-C30 문항을 우리나라의 실정에 맞게 번역하여 사용하였다. 결과: 조혈모세포이식군(1군)은 53명, 화학요법군(2군)은 57명, 정상대조군(3군)은 55명 이었고, 성별, 나이, 질환별 분포, 검사시점 등에서의 유의한 차이는 없었다. 각 군간의 QLQ-C30 평균 점수를 비교한 결과, 전반적인 삶의 질에서는 1군이 73.5, 2군이 51.7, 3군이 79.3으로 2군이 1.3군보다 낮았고 역할 및 사회기능은 각각 1군이 64.3/68.9, 2군이 57.6/70.3, 3군이 85.2/85.5로 1,2군이 3군에 비하여 낮았다.(P<0.01). 또한, 오심/구토가 1군은 8.7, 2군은 4.5, 3군은 3.2로 1군이 2, 3군보다 높았으며 통증은 1군은 17.0, 2군은 19.2, 3군은 9.6으로 1,2군이 3군에 비하여 높았다(p〈0.01). 기타 신체, 감정, 인식기능 및 피로에서는 각군간에 유의한 차이를 발견할 수 없었다. 결론: 본 연구에 참여한 조혈모세포이식환자들의 전반적인 삶의 질은 화학요법을 받은 환자들보다 우월하면서 정상인들과 유사하였으나 역할기능 및 사회기증, 통증에서는 화학요법 환자들처럼 정상인들에 비하여 열악하였고 오심/구토는 화학요법 환자들 및 정상인에 비하여 심한 양상을 보였다. 그러므로 본 연구를 바탕으로 하여 국내 실정에 맞는 QOL 측정 방법을 개발하고 조혈모세포이식을 시행받는 환자들의 삶의 질에 관한 연구들이 향후 계속 진행되어야 하리라고 생각된다. Background: It is very important to endow patient with satisactory quality of life (QOL). However, little is known about QOL after hematopoietic stem cell transplantation (HSCT) in Korea. In this study we tried to measure QOL in the patients treated with HSCT and compare QOL scores with those of patients treated with usual chemotherapy and normal populations. Methods: The QOL evaluation was performed with EORTC QLQ-C30 questionnaires interpreted to Korean language. The study populations were consisted with 53 patients treated with HSCT (Group 1), 55 patients treated with usual chemotherapy (Group 2), and 55 normal populations (Group 3). The QLQ-30 scores of each group were evaluated and compaired with x² test. Results: Global quality of in group 1, 3 (score: 73.5, 79.3, respectively) were higher than that (score: 51.7) of group 2. Role and social function were lower in group 1, 2 (score: 64.3/68.9, 57.6/70.3, respectively) than those (score: 85.2/85.5) of group 3. The degree of nausea and vomiting was more severe in group 1 (score: 8.7) than those of group 2, 3 (score: 4.5, 3.2 respectively). Pain scores of group 1, 2 (score: 17.0, 19.2, respectively) were more higher than that of group 3 (score: 9.6) In other parameters of QLQ-C30, there was no significant difference between each groups. Conclusion: Our perliminary result may suggest that HSCT is not necessarily associated with deterioration of QOL. This underlines the necessity of undertaking perospective studies using reliable and well-validate methods for measuring QOL

      • 봄철 익산지역 환경대기 중 수용성 이온성분의 일별 침적량

        Deposition samples were collected in wet gauge and dry gauge containers at downtown of Iksan city, located southwest of the Korean peninsula, from April 20 to May 1, 2004. The volume of 30-50㎖ deionized water was added for wet gauge before sampling, but dry gauge was installed in the dry state with no deionized water adding. These samples were collected twice a day during daytime and nighttime and were analyzed for anions(Cl^(-), NO^(3-), SO₄^(2-)) and cations (NH₄^(+), Na^(+), K^(+), Mg^(2+), Ca^(2+)) using ion chromatography. Data quality was checked by the data obtained from re-injection of standard solution, Dionex cross check standard solutions, and random several deposition samples, and measured data was estimated to be reliable. Considering the deposition sample volume, the sampling time, the surface area of sampling container, and the ion concentration measured, the deposition amounts were calculated in mg/㎡/day. The sum of ion dry deposition amounts for dry gauge and wet gauge was 9.1±6.7 mg/㎡/day and 26.5±9.1 mg/㎡/day, respectively. A significant increase in deposition amount during rainfall days and Asian yellow dust period was observed for both wet gauge and dry gauge, especially during rainfall days having no difference of deposition amount between in wet gauge and in dry gauge. The mean deposition of all ions measured in this study were higher in wet gauge than in dry gauge because of the surface difference of the sampling container. The mean deposition amount of NH₄^(+), SO₄^(2-), and NO^(3-) in wet gauge were found to be about 27.6 times, 5.5 times, and 3.4 times higher. than that in dry gauge, while the rest of the chemical species were equal or a little higher in wet gauge than in dry gauge. Dominant species in dry gauge were SO₄^(2-) and Ca^(2+), accounting for 14.7% and 47.3% of the total ion dry deposition, whereas those in wet gauge were SO₄^(2-) and NH₄^(+), accounting for 27.7% and 27.1% of the total ion dry deposition, respectively.

        Kidney Toxicity Induced by 13 Weeks Exposure to the Fruiting Body of Paecilomyces sinclairii in Rats

        Paecilomyces sinclairiis (PS) is known as a functional food or human health supplement. However concerns have been raised about its kidney toxicity. This study was performed to investigate the kidney toxicity of PS by 13 week-oral administration to rats. Blood urea nitrogen (BUN), serum creatinine, and kidney damage biomarkers including beta-2-microglobulin (${\beta}2m$), glutathione S-transferase alpha (GST-${\alpha}$), kidney injury molecule 1 (KIM-1), tissue inhibitor of matrix metalloproteinase 1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, cystatin C, neutrophil gelatinase-associated lipocalin (NGAL) and osteopontin were measured during or after the treatment of PS. BUN, creatinine and kidney damage biomarkers in serum were not changed by PS. However, kidney cell karyomegaly and tubular hypertrophy were observed dose-dependently with higher severity in males. KIM-1, TIMP-1 and osteopontin in kidney and urine were increased dose dependently in male or at the highest dose in female rats. Increased urinary osteopontin by PS was not recovered at 2 weeks of post-exposure in both genders. Cystatin C in kidney was decreased at all treatment groups but inversely increased in urine. The changes in kidney damage biomarkers were more remarkable in male than female rats. These data indicate that the PS may provoke renal cell damage and glomerular filtration dysfunction in rats with histopathological lesions and change of kidney damage biomarkers in kidney or urine. Kidney and urinary KIM-1 and cystatin C were the most marked indicators, while kidney weight, BUN and creatinine and kidney damage biomarkers in serum were not influenced.

        교과과정의 조직은 체계적이고 뜻이 있는 순서로 학생들을 가르치기 위해 지식을 어떻게 조직화할 것인지와, 과목들 사이에서 공통된 내용은 통합시켜 중첩되는 일이 없도록 조정할 필요가 있다. 이에 현재 본 대학 간호과에서 운영되었던 97학년도의 17개 전공과목들의 교과과정의 내용을 비교, 분석하여 중복을 줄이며 순차적 지식의 습득을 도모하는 새로운 통합교과과정의 틀을 개발, 제시함으로써 보다 나은 전문직 간호교육이 이루어지도록 하고자 하였다. Most curricula are considered how to teach the student in systematic and meaningful orders. And it is necessary to coordinate the duplicate contents in major subjects in nursing. So this study was designed to set up a new comprehensive curriculum through comparing and ananlyzing each seventeen major subjects in Nursing Department of Kyung-Bok College. Therefore, the purpose of this study is to present the new comprehensive curricula for promoting the efficiency and effectiveness in diploma degree course of nursing education.

        학생체질검사제도에 대한 인식과 개선방안 연구

        Recent domestic findings indicate steep increase of geriatric diseases such as obesity, diabetes, and high boll d pressure among children and adolescents. It is, therefore, necessary to establish a new system for constitution inspection th at reflects domestic adolescents' health conditions by the introduction of feasible items and systematic methods in the inspect ion. The objective of this study is to grasp the problems and to present proper ways of improving the system. As the method of the research during the period from September 2001 to March 2002 we refer to the domestic and foreign li terature, and ask the advice of the specialists in pediatrics, family medicine, endocrine, cardiology. As on-site research we us a survey of nationwide elementary, middle, and high schools, visit representative local schools and have an interview with shoo l nurses. Through the interview we try to find practical problem of school heath care and find proper method of students physic al examination. The abstract of the problems of student examination system is as following. To the question about the extent of the resu lt of physical examination 46. 46.4%(school doctor) and 42.6%(parents and students) of the respondents answered "don't know". It means neglectfulness about examination itself. 62% of the school nurses responded "unsatisfactory" people who take charge of practical affairs have complaint about pre sent examination method. Because of the short time of the examination, and numerous students, the examination was incomplete. So veral questions, method of physical examination, content of physical examination and list of physical examination, were raised. Reform measures for physical examination system of students were presented by physician, school nurses, students and parents of student.

      • 중증 재생불량성 빈혈 환자에서 신우신염에 대한 광범위항생제치료 중에 속발한 Saccharomyces cerevisiae 진균감염 1예

        Most patients with aplastic anemia who do not respond to immunosuppressive treatment or are not candidates for bone marrow transplantation die of infection or bleeding. The neutropenia in acute leukemia, aplastic anemia, or occurring subsequently to chemotherapy and bone marrow transplantation increases susceptibility to infection. In general, the number of infectious episodes correlate with the degree and duration of neutropenia. Global immunosuppression produced by conditioning for bone marrow transplantation or graft-versus-host disease, is associated with unusual bacterial and fungal pathogens, or serious viral and protozoan infections. In addition, repeated treatment with broad-spectrum antibiotics is associated with the emergence of resistant organisms and fungal diseases because of the altered microbial microenvironment of the host. The incidence of invasive fungal infection caused by Saccharomycetes eerevisiae in immunosuppressed patients is very rare, compared with that of infection by candida or aspcrgillus species. Cases of Saccharomycetes cerevisiae fungemia occurring in the course of treatment with broad-spectrum antibiotics are reported in patients with extensive burn or with prosthetic valve endocarditis. We experienced a case of urinary tract infection by Saccharomycetes cerevisiae in a 27-year old female patient with severe aplastic anemia. We report the case with a review of relevant literatures.

        순 티탄늄 판재의 프레스 성형성 평가(제 1보)

        본 논문에서는 판형 열교환기 등에 널리 이용되고 있는 순 티타늄 판재의 프레스 성형성을 평가하기 위해 인장실험을 수행하였고 인장실험결과를 가장 잘 피팅할 수 있는 가공경화 모델을 제안하였다. 또한 항복곡면의 도출을 위해 단축변형, 평면변형, 순수전단변형 등 다양한 변형모드 하에서의 인장실험을 실시하였다. 이 제안된 모델을 Hora의 수정된 최대하중조건식에 적용하여 프레스 성형성을 평가하기 위해서 널리 사용되고 있는 성형한계선을 예측하였고 그 결과를 장출성형실험에서 구한 성형한계선과 비교하였다. 도출한 항복곡면을 잘 묘사하기 위한 이방성 항복조건식과 본 연구에서 제안된 가공경화모델은 순 티타늄 판재의 프레스 성형성의 척도인 성형한계선을 잘 예측함을 알 수 있었다. Commercially pure titanium (CP Ti) has been actively used in plate heat exchangers due to its light weight, high specific strength, and excellent corrosion resistance. However, compared with automotive steels and aluminum alloys, there has not been much research on the plastic deformation characteristics and press formability of CP Ti sheet. In this study, the mechanical properties of CP Ti sheet are clarified in relation to press formability, including anisotropic properties and the stress-strain relation. The flow curve of the true stress-true strain relation is fitted well by the Kim-Tuan hardening equation rather than the Voce and Swift models. The forming limit curve (FLC) of CP Ti sheet was experimentally evaluated as a criterion for press formability by punch stretching tests. Analytical predictions were also made via Hora's modified maximum force criterion. The predicted FLC with the Kim-Tuan hardening model and an appropriate yield function shows good correlation with the experimental results of the punch stretching test.
