        Multiplex PCR Detection for 3 Events of Genetically Modified Maize, DAS-59122-7, TC6275, and MIR604

        A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect 3 events of genetically modified (GM) maize. The event-specific primers were used to discriminate the following 3 events of GM maize (DAS-59122-7, TC6275, and MIR604) using multiplex PCR method. The zein gene was used as an endogenous maize reference gene in the multiplex PCR detection. The primer pair Zein-F/R producing a 99 bp amplicon was used to amplify the zein gene. The primer JI-Das-F1/R1 for DAS-59122-7, JI-TC6275-F3/R3 for TC6275, and JI-MIR F1/R1 for MIR604 yielded an amplicon of 130, 162, and 197 bp, respectively. The detection limit of multiplex PCR was 1% for DAS-59122-7, TC6275, and MIR604 for one reaction.

        보육교사의 전문성 발달 수준에 따른 이야기 나누기와 조형 활동의 수업 전문성 평가

        본 연구에서는 보육교사의 전문성 발달 수준에 따라 이야기 나누기와 조형 활동의 수업을 평가하여 보육교사의 수업 전문성 차이를 살펴보고자 하였다. 연구대상은 G 광역시에 소재한 보육시설에 근무하는 보육교사 84명이다. 연구도구는 백은주ㆍ조부경(2004)이 개발한 전문성 발달 수준 검사 도구와 선행연구 및 관련문헌(김희진ㆍ박은혜ㆍ이지현, 2000; 박승순, 2004; 정영희, 2005; 허미애, 2007)을 참고하여 연구자가 개발한 관찰 평가지를 사용하였다. 자료분석은 보육교사의 전문성 발달 수준 총점을 기준으로 집단(상집단과 하집단)을 구분하고, 각 집단에 따른 수업 전문성의 차이를 알아보기 위하여 독립표본 t-검증을 실시하였다. 연구결과를 제시하면 다음과 같다. 첫째, 전문성 발달 수준이 높은 보육교사는 낮은 보육교사보다 이야기 나누기 활동의 수업 전문성이 높았으며, 특히 이야기나누기의 주제 선정, 사전 준비 및 매체 활용, 소요 시간에 대한 수업 전문성이 더욱 높은 것으로 나타났다. 둘째, 전문성 발달 수준이 높은 보육교사는 낮은 보육교사보다 조형 활동의 수업 전문성이 높았으며, 조형 활동의 선정, 환경 준비, 지도 과정에 대한 수업 전문성에서 모두 높은 것으로 나타났다. 본 연구는 보육교사의 전문성 발달 수준과 수업 전문성은 밀접한 관련이 있음을 증명하는 것으로, 연구의 결과는 보육교사의 수업 전문성 발달 수준을 높이기 위한 현직 교사교육 프로그램 개발에 필요한 기초 자료로 활용될 수 있을 것이다. This study examines differences in instructional expertise among childcare teachers in relation to their professional development levels through evaluation of their story discussion and art activity classes. Employed as the subjects of this study were 84 teachers working for childcare institutions in G Metropolitan City. A questionnaire was used as a research tool after being prepared on the basis of a professional development test tool developed by Baek Eun-ju and Jo Bu-gyeong (2004) as well as related previous studies and bibliographies (Kim Hee-jin, Park Eun-hye, Lee Ji-hyeon, 2000; Park Seung-sun, 2004; Huh Mi-ae, 2007). For data analysis, the teachers were divided into two groups (the upper group and the lower group) in accordance with their professional development level scores. Independent t-tests were conducted to determine the differences between the two groups in instructional expertise. The findings of this study can be summed up as follows. First, childcare teachers with high levels of professional development displayed higher levels of instructional expertise in story discussion class than those with low levels of professional development. The difference was particularly conspicuous in topic selection, class preparations and the use of media, as well as time allotment. Second, childcare teachers with high levels of professional development displayed higher levels of instructional expertise in art activity class than those with low levels of professional development. The former group turned out to have better instructional expertise than the latter group in all the related areas of art activity selection, arrangement of learning environments, and the effectiveness of instructional processes. This study showed that childcare teachers' professional development levels are closely related to their instructional expertise. The findings may be used as basic materials in developing program models designed to enhance professional development levels of childcare teachers.

        부산·울산·경남지역 직업병 감시체계

        Objective: Occupational medicine specialists in the Busan, Ulsan and Kyung-Nam areas established an area-based occupational disease surveillance system and used this system to collected case information for the purpose of preventing occupational diseases Methods: l l hospital participated in this system. The authors selected five main diseases, which were hand-arm vibration syndrome (HAVS), work-related musculoskeletal disorder (WRMSD), occupational asthma, occupational skin disease and occupational and occupational lung cancer and established their case definitions. All cases were reported on the web, and real time analysis was conducted. Results: Between April 2001 and April 2003, 192 cases of HAVS, 118 cases of WRMSD, 33 cases of occupational asthma, 17 cases of occupational skin disease and 17 cases of occupational lung cancer (for a total of 377 cases of these five main diseases) were reported. most of the HAVS cases came from shipbuilding, and 172 of them (92.7%) were associated with grinding. Of the four main types of WRMSD, carpal tunnel syndrome (CTS) was the most prevalent with 46 cases and shipbuilding was also the main industry involved (83.9%). The ergonomic risk factors involved mainly associated with the hand. In 19 (57.6%) and 4 (12.1%) cases of occupational asthma, the agents involved. The causative agents of occupational lung cancer included Cr, welding fume, PAH, Ni, etc. 10(58.8%) of the cases were diagnosed as squamous cell carcinoma and 5(29.4%) as adenoma. Conclusion: This result showed that an area-based occupational disease surveillance system might provide an effective method of evaluating the prevalence of such diseases, however the Busan, Ulsan and Kyung-Nam provinces are too large to be treated as individual areas. Therefore, the authors suggest that each province should establish its own surveillance system.

        황색포도상구균 신속 검출용 간이 진단키트

        This study was performed to develop a rapid test kit for pathogenic Staphylococcus in various samples. The rapid detection kit has been fabricated based on nitrocellulose lateral-flow strip. Colloidal gold and Staphylococcus antibodies were used as a tag and a receptor, respectively. Manually spotted Staphylococcus antibody and anti-mouse antibody on the surface of nitrocellulose membrane were used as test and control lines, espectively. Feasibility of the rapid kit to detect Staphylococcus aureus in samples were evaluated. The intensity of the color of the tes line started to increase with the samples in which higher concentration of the cells were contained. The sensitivity of the sensor was 106 cfu/mL Staphylococcus spiked in PBS. Also, the rapid test kit could detect 105 cfu/mL of Staphylococcus in chicken meat extract.

      • 보리뿌리에서 질산 환원 효소 유도에 미치는 Ferulic acid의 효과

        Effects of ferulic acid(FA) on the induction of nitrate reductase(NR) were surveyed in Hordeum vulgare. Seedlings raised with 0.2mM CaCl_2 in the dark for 6 days were put into the buffered solution containing 0.2mM KNO_3 for 5 hrs to induce NR. The longer the induction time was, the more NR activity in vivo increased. Experimental group which was induced in the solution containing 0.5mM FA showed significantly lower NR activity than the control treated without FA. But it was found that NR was not affected directly by FA when the enzyme assay was performed both in vivo and in vitro with FA. NR induction was assayed at various concentrations of FA and pH. The higher the concentration of FA and the lower pH was, the more the induction of NR was inhibited. And nitrate contents in the barley roots were assayed at the same condition. The induction of NR was inhibited more severely, when nitrate contents decreased. Thus it is thought that FA affects the nitrate uptake and then the induction of NR.

      • 근대 초기 소설에 나타난 폭력의 재현과 근대성

        There are many violence motives in new-style fiction. The aspect of violence in that fiction is very cruel and expressed very concretely. But this tendency is changed in Lee Gwang-su's [Mu-jung], the first modern novel. The cruelity of violence is still the same but expression of violence motives becomes indirect and decrease. On 1920s' novel like Kim Dong-in and Choi Su-hae's, it is not different there are also many violence motives like murder, fire prevention, and terrible death etc. But it is important to stress on the reason of violence. In the Kim Dong-in's novel, the reason of violence is suggested the madness or hallucination of character, while in the Choi Su-hae's novel, irrationalities of the society as poverty, class discrimination. Accordingly, during the earliy modern times, the strength and concreteness of violence motives in the novel have become weaken in process of time. Which means the effect of enlightment of modernity, but instead of a decrease in privit violence, the invisual public violence increases. 전근대에서 근대로의 이행을 보여준다고 평가되는 신소설에서는 오늘날의 기준으로 보기에 지나치게 잔혹하고 엽기적인 폭력 모티프들이 자주 등장한다. 그러나 이러한 폭력 모티프는 이후 한국 소설사에 점차 약화되는 양상을 보인다. 최초의 근대적인 장편소설인 이광수의 <무정>에서는 재현적 차원에서의 폭력의 강도나 수위는 크게 달라지지 않았지만, 신소설과 비교했을 때, 표현 방법이 암시적이고 간접적이다. 1920년대 경향파를 대표하는 최서해나 김동인의 소설에서도 폭력 모티프가 자주 등장하는데, 주목할 것은 방화, 살인 같은 폭력의 잔혹함은 줄어들었다고 보기 어렵지만 이전의 소설에서 폭력이 개인적인 탐욕과 질투, 복수 등에서 기인한 것과는 달리, 빈곤과 같은 사회문제가 개인문제와 함께 작용하고 있다는 점, 폭력이 광기나 환각 상태처럼 특수한 상황에서 발생한 것으로 합리화시키고 있다는 특징이 있다. 말하자면 전반적으로 폭력의 잔혹함과 표현의 적나라함이 감소하고 있는 것이다. 근대 초기 소설에 나타난 폭력의 재현 양상이 이렇게 변하는 것은 이 시기가 근대적인 제도와 인식의 변화가 이루어지던 시기라는 점과 관련하여 고찰할 수 있다. 즉, 근대의 계몽주의적인 관점에서 폭력을 전근대적이고 반인간적인 것으로 간주하여 사적인 폭력을 억압하고 배제한 것이라고 볼 수 있다. 그러나 이 시기가 스펙터클한 폭력에 대한 금지가 이루어졌을지는 모르나 정신을 훈육하고 내면을 통제하는 사회적․제도적 폭력이 극대화된 시기였음을 간과해서는 안 된다.

      • NC 선반의 직선 사이클 평면 위치결정 정도 측정 및 분석

        It is very important t o measure linear cycle plane positioning accuracy of NC lathes because the plane positioning accuracy affects accuracy, performance, interchange-ability and quality of machine parts machined by the NC lathes in industries. Until now, linear cycle plane positioning accuracy test of NC lathes has not been carried out because the two axes of NC lathe are moving simultaneously crossing a t right angle each other. while positioning accuracy test of one axis of NC lathe is briskly carried out using a laser interferometer system in industries. So, in this study, a measuring units system to measure simultaneously two axes' plane. positioning accuracy of NC lathe was composed of two optical linear scales installed on the. z and x-axis of work coordinate system on NC lathe and a computer inserted with PC counter card enables to obtain measuring data. Here, ATC(Automatic Tool Changer) and moving part of linear scale are fixed with magnet bases in order to measure displacement of the ATC of NC lathe. And next, computer software was developed in order to measure the lathe under resolution of 0.1㎛ by the linear scales, and also computer soft measuring data could be modeled on plots and be analyzed to numerically.

      • Brucella abortus 국내 분리주의 36kDa 외막단백을 암호하는 omp2b 유전자의 염기 분석

        B. abortus 국내 분리주의 분자유전학적 성상을 규명하고 분자역학적 연구에 필요한 기초자료를 얻기 위해 36kDa OMP를 암호하는 omp2b 유전자를 PCR법으로 확인하고,그리고 omp2b 유전자의 핵염기 서열과 아미노산 서열을 결정하고 이 결과를 Brucella 표준균주 및 여러 동물에서 분리된 Brucella 분리주와 상호 비교 분석하였던 바 다음과 같은 결과를 얻었다. 1. B. abortus 분리 주 (7주)에 대해 omp2b primers를 이용하여 PCR을 수행한 바 공시한 참조균주와 분 리균주 모두에서 1,251bp의 특이한 DNA 절편이 각각 증폭되었다. 2. omp2b 유전자의 1,089bp에 대한 염기서열과 이에 대한 362개의 아미노산 서열을 결정하였다. 분리 균주의 omp2b 유전자의 염기서열을 참조균주와 비교 분석한 결과 B. abortus 2308 (biovar 1) 및 B abortus B3196 (biovar 5)과는 99.6%∼100% 및 99.5%∼99.9%의 매우 높은 상동성을 나타내었고,B. abortus 45/20 (biovar 1)과는 97.1%~97.4%의 상동성을 나타내었다. 또한 362개의 아미노산 서열에 대해 분석하였던 바,분리균주들은 B.abortustus 2308 (biovar 1) 및 B. abortus B3196 (biovar 5)과는 99.2%∼100%의 매우 높은 상동성을 보였으며,B. abortus 45/20 (biovar 1)과는 96.4%∼ 97.2%의 상동성을 보였다. 3. 본 시험에 공사된 브루셈라균주의 omp2b 유전자 서열과 여러 동물에서 분리된 브루셀라 분리주의 자료를 바탕으로 phylogenetic tree를 작성 한 바,국내 분리주는 B. abortus 2308 (biovar 1),B. abortus B3196 (biovar 5),B. abortus 45/20 (biovar 1),B. suis 1330 (biovar 1),B. melitensis 16M (biovar 1),B. canis NCTC 01854,B. neotomae 5K33과 97.3% 이상의 높은 근연성을 보이며 동일한 cluster에 소속되었으며,omp2b 유전자는 매우 잘 보존됨을 알 수 있었다. omp2b gene of Brucella abortus isolates encoding 36kDa outer membrane protein (OMP) was amplified by polymerase chain reaction (PCR) and the sequences of nucleotide and amino acid were determined and analyzed to obtain the basic data for molecular epidemiological features of brucellosis in Korea. In PCR using omp2b primers for the gene of 36kDa aMP, the specific signals at 1,251bp were demonstrated, The nucleotide sequences of omp2b gene of 1,089bp were sequenced by dye terminator cycle sequencing method, and 362 amino acid sequences were predicted. As compared the nucleotide sequences with those of B. abortus reference strains, the isolates showed the very high homology to the reference strains ranging 97.1%∼100%. The similar results were also obtained in the analysis of the amino acid sequences. In the phylogenetic tree, the isolates clustered with the reference strains and the majority of strains originated from pig, dog, wood rat and cattle.

        Sodium Salt of Bis (2,2,3,3,4,4,5,5-octafluoro-1-pentanol) Sulfosuccinate를 이용한 초임계 CO₂^(+)물 계의 마이크로에멀젼

        대부분의 촉매, 이온, 단백질같이 비휘발성이고 친수성인 물질들은 초임계 이산화탄소에서는 잘 용해되지 않는 단점을 가지고 있다. 이는 이산화탄소가 무극성이며, 극성인 물에 대해 용해력이 매우 낮기 때문이다. 본 연구에서는 이러한 한계를 극복하기 위한 방법으로, 물과 이산화탄소의 마이크로에멀젼 형성에 관한 연구를 실시하였다. 물과 이산화탄소의 에멀젼을 형성히키기 위하여 친수기와 친이산화탄소기를 동시에 지닌 hydrocarbon-fluorocarbon hybrid 계면활성제인 sodium salt of bis(2,2,3,3,4,4,5,5-octafluo-ro-1-pentanol) sulfosuccinate(F-AOT)를 합성하였다. 기초 데이터로 초임계 이산화탄소내에서 계면활성제의 용해도를 알아보기 위해 variable volume cell을 이용하여 계면활성제의 농도에 따른 cloud point를 측정하였고, 또한, 물+계면활성제+이산화탄소계의 cloud point 측정을 통하여 마이크로에멀젼이 안정적으로 형성되는 ‘마이크로에멀젼 안정 영역’을 구하였다. 초임계 CO_(2^(+)) 물 계의 마이크로에멀젼은 온도 308.34에서 368.25K까지, 압력 14.06에서 63.41 MPaRk지의 조건에서 안정적으로 형성되었다. Non-volatile hydrophilic substances such as ions, proteins, and catalysts have disadvantage that they are insoluble in liquid or supercritical CO₂ because of its very low dielectric constant and polarizability per volume. To overcome this disadvantage, we investigated the formation of the water-in-CO₂ microemulsion that nano size water droplets were dispersed into CO₂. In order to form this microemulsion, the fluorocarbon-hydrocarbon hybrid surfactant, that is, the sodium salt of bis (2,2,3,3,4,4,5,5-octafluoro-1-pentanol) sulfosuccinate(F-AOT) which has CO₂ philic chains and hydrophilic head group simultaneously was synthesized. To investigate the solubility of this surfactant in CO₂, the cloud point was measured with its various concentrations. And “the stability region of microemulsion” was also obtained by measuring the cloud point of water-in-CO₂ microemulsion. It was found that the water-in-CO₂ microemulsion was stably formed in the temperature range from 308.34 to 368.25 K, and in the pressure range from 14.06 to 63.41 MPa.

      • 실험동물에서의 내분비계 교란물질인 DEHP, DBP의 생체내 영향

        The major protocol features of the rodent uterotrophic assay have been evaluated using a range of reference chemicals. The protocol variables considered include the selection of the test species and route of chemical administration, the age of the test animals, the maintenance diet used, and the specificity of the assay for estrogens. The rodents were ovariectomized under general anesthesia via bilateral flank incisions and randomly assigned to groups of 5 animals. This study investigated the estrogenic effects of phthalates, which are DEHP and DBP, suspected as endocrine disruptors. It is concluded that three daily subcutaneous administration of test chemicals, which are di-ethylhexyl phthalate(DEHP) and di-n-butyl phthalate(DBP), to 9 week-old rodents. The results revealed that DEHP & DBP show similar uterotrophic activity in the rat and the mouse, using subcutaneous administration. DEHP and DBP failed to affect any of these parameters, which are body weight, mammary gland, vaginal and uterus histopathology. but, DBP increased uterus weight to only one rat of 1.0 g/kg group. The results conform the need for rigorous attention to experimental design and criteria for assessing estrogenic activity.
