DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

<P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

찰옥수수 이삭 形質의 變異와 雜種强勢

This study was conducted to examine the variations and heterosis of ear characters in waxy corn(Zea mays L.). Total of 13 inbreds and 24 hybrids were tested at the Crop Experiment Station in 1991. The average number of days to tasselling of inbreds and hybrids were 67 and 78 days after planting, respectively, while the everage number of days to silking were later about 10 days than those to tasselling date for both inbreds and hybrids. Silking of both indeds and hybrids was occurred about 3 days after tasselling in general. Growth of hybrids was more vigorous than inbreds for all plant characters, e. i., stem height was 55cm taller and ear height was 35cm higher. KW2 and KW5 were early and short inbreds, while M401WX, W8013 and W8113 were late and tall inbreds. Coefficients of variation of the characters studied were 8-29% for hybrids and those for inbreds were 9-35%. Coefficients of variation of unhusked ear length and ear diameter were small with the range of 8-10%, those of ear weight, unmber of kernels per row and filled ear length were large with the range of 19-35%, and those of filled ear length and row number ranged 11-16%. Hybrids were classified into 6 groups(I- VI) based on ear length and weight. Among 24 hybrids tested, KW1/KW4, KW1/KW2/KW3, KW3/W8013, KW3/W8113 and W8113/KW4 were classified as grade I. Correlations among husked ear length, filled ear length and number of kernels per row were highly significant in both inbreds and hybrids. Ear weight showed the greatest heterosis with 113% in average among 24 hybrids, and number of kernels per row (73%), filled ear length(42%) and husked ear length(24%) followed in that order.

TALP-35 purified from human term placenta is known to increase microtubule polymerization and stabilize the polymerized microtubule. To examine the effect of TALP-35 on immune system this study was performed. MTT assay was performed to investigate the effect of TALP-35 on the proliferation of RAW264.7 cells. TALP-35 dose dependently suppress the proliferation of RAW264.7 cells at high concentration (above 1 μM) in unstimulated cells, in case of 10 μM TALP-35 treated cells the suppression was 25% but in stimulated cells it was only 15%. Cosedimentation assay was carried out to investigate whether TALP-35 can bind to tubulin of RAW264.7, monocyte/macrophage lineage of mouse, and polymerize it. TALP-35 polymerize the tubulin of RAW264.7 cells and sedimented in dose-dependent manner. To investigate the effect of TALP-35 on the expression of iNOS protein western blotting was performed. The expression level of iNOS was decreased dose dependently in high concentration of TALP-35 treatment. To examine the activity of iNOS, secreted NO was determined by method based on Griess reaction. Interferon-γ and LPS-stimulated production of NO from RAW264.7 cells was decreased dose dependently above 0.1 μM concentration of TALP-35 and 50% is decreased at 10μM of it. This study shows TALP-35 can control cytokine induced-iNOS expression therefore it might control inflammatory diseases.

In a case of driving beginnners, it is very difficult to park a car perfectly because driver's knowledge makes more errors than intelligent mechanism when they are performed repeatedly. However skilled drivers be of experienced knowledge are able to park a car quickly. In this paper, we analyzed driving patterns of skilled drivers, designed the fuzzy controller to park based on it, and perform auto-parking simulation of a car using that controller.

We evaluated the efficacy of the liquid form of enrofloxacin against poultry respiratory and digestive infections, the major problems in the poultry industry, from diverse diagnosis viewpoints. Results obtained through this study were as follows: 1. The liquid form of enrofloxacin showed good acceptability to poultry owing to no abhorrence to the drug by chickens, which indicated its suitability for mixing in drinking water. 2. The application of the enrofloxacin formulation improved the productivity of the chicken farm to a great extent by lowering the death toll of chicken raised in the farm. 3. The use of the drug shortened the days to the market, and increased the body weight at the time of marketing. 4. Preventive and therapeutic effects of the liquid enrofloxacin were observed higher than ampicillin, a major antibiotic used in poultry farming. From above results, the liquid form of enrofloxacin would find its use for the prevention and treatment of digestive and respiratory infections, let alone the growth promotion and feed efficiency, especially in the chicken farming practice.

In this study the antimicrobial spectrum and sensitivity of a liquid formulation of enrofloxacin against the major six pathogenic bacteria isolated from the diseased poultry in Korea were evaluated in comparison with ciprfloxacin (CFX), gentamicin (GM) and penicillin (PC). A liquid form of enrofloxacin, a test material and ciprofloxacin showed at least 83% antibacterial activity against all of six test pathogenic microorganisms at the fixed concentration of 10 ㎍/㎖. Gentamicin and penicillin revealed much lower antibacterial activity at the same condition. We evaluated the liquid form of enrofloxacin as having widespread antibacterial spectrum against poultry infections. The liquid form of enrofloxacin showed potent antibacterial activity, with the MIC range being 0.12∼2 ㎍/㎖ against Gram positives, 0.06∼2 ㎍/㎖ against Gram negatives and 0.5∼2 ㎍/㎖ against Mycoplasma spp. These values were equivalent to or somewhat higher than those of ciprofloxacin, but much lower than those of gentamicn and penicillin. This new liquid form of enrofloxacin would find its way in application of mixing with drinking water for the prevention and treatment of diverse infections in chickens.

The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the third year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. Distribution of the Fairy Pitta (Pitta nympha) in the South Korea: A focus on protected areas

국내 팔색조(Pitta nympha) 분포 현황을 알아보고자 전국 국립공원 등 보호지역 중심으로 총 19개 지역을 조사하였고, 전문가 네트워크를 통해 자료를 보강하였다. 전국 19개 지역을 조사한 결과 북한산, 지리산 등 내륙지역 7개 국립공원에서는 팔색조가 확인되지 않았고, 경남 거제, 남해, 해남, 장흥 고흥 등 남해안과 인접한 12개 지역에서 팔색조 서식이 확인되었다. 또한 전문가 네트워크를 통해 수집한 팔색조 출현 위치 정보 82건을 분석한 결과, 전국 8개 광역지자체 모두 팔색조 서식이 확인되었고, 내륙지역(26.8%)보다 해안지역(73.2%)에서 더 많이 출현하였다. 연구 결과를 종합해보면, 우리나라의 팔색조는 주로 제주도와 남해안 지역을 중심으로 많이 분포하며, 그 보다 적은 개체수가 내륙 지역에 넓게 분포하고 있는 것으로 나타났다. The present study investigated the current distribution of Fairy Pitta (Pitta nympha) in South Korea. A total of 19 study areas, including protected areas such as national and provincial parks in South Korea were surveyed, and additional data obtained from an expert network. According to the results, Fairy Pitta was absent in seven inland national parks, including Mt. Bukhan and Chiri parks. Fairy Pitta inhabited 12 areas near the southern coast of South Korea, including Geoje, Namhae, Haenam, Jangheung, Goheung, and Gyeongnam Province. The analysis results of 82 datasets obtained from experts with regard to the locations where Fairy Pitta have been spotted showed that the bird inhabits all the eight metropolitan regions in South Korea, and is more frequently spotted in coastal areas (73.2%) than in inland areas (26.8%). The findings demonstrate that Fairy Pitta is mainly distributed in Jeju Island and the southern coast of South Korea, and is distributed in inland areas at relatively lower numbers.
