        The Ascidian Numb Gene Involves in the Formation of Neural Tissues

        Notch signaling plays fundamental roles in various animal development. It has been suggested that Hr-Notch,a Notch homologue in the ascidian Halocynthia roretzi, is involved in the formation of peripheral neurons by suppressing the neural fates and promoting the epidermal differentiation. However, roles of Notch signaling remain controversial in the formation of nervous system in ascidian embryos. To precisely investigate functions of Notch signaling, we have isolated and characterized Hr-Numb, a Numb homologue which is a negative regulator of Notch signaling, in H. roretzi. Maternal expression of Hr-Numb mRNAs was detected in egg cytoplasm and the transcripts were inherited by the animal blastomeres. Its zygotic expression became evident by the early neurula stage and the transcripts were detected in dorsal neural precursor cells. Suppression of Hr-Numb function by an antisense morpholino oligonucleotide resulted in larvae with defect in brain vesicle and palps formation. Similar results have been obtained by overexpression of the constitutively activated Hr-Notch forms. Therefore, these results suggest that Hr-Numb is involved in Notch signaling during ascidian embryogenesis.

        Hydroxycinnamic Acids in <i>Crepidiastrum denticulatum</i> Protect Oxidative Stress-Induced Retinal Damage

        <P>We investigated the effects of an ethanol extract of <I>C. denticulatum</I> (EECD) in a mouse model of glaucoma established by optic nerve crush (ONC), and found that EECD significantly protected against retinal ganglion cell (RGC) death caused by ONC. Furthermore, EECD effectively protected against <I>N</I>-methyl-<SMALL>d</SMALL>-aspartate-induced damage to the rat retinas. <I>In vitro</I>, EECD attenuated transformed retinal ganglion cell (RGC-5) death and significantly blunted the up-regulation of apoptotic proteins and mRNA level induced by 1-buthionine-(<I>S</I>,<I>R</I>)-sulfoximine combined with glutamate, reduced reactive oxygen species production by radical species, and inhibited lipid peroxidation. The major EECD components were found to be chicoric acid and 3,5-dicaffeoylquinic acid (3,5-DCQA) that have shown beneficial effects on retinal degeneration both <I>in vitro</I> and <I>in vivo</I> studies. Thus, EECD could be used as a natural neuroprotective agent for glaucoma, and chicoric acid and 3,5-DCQA as mark compounds for the development of functional food.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jafcau/2014/jafcau.2014.62.issue-6/jf4046232/production/images/medium/jf-2013-046232_0012.gif'></P>

        Objective: Occupational medicine specialists in the Busan, Ulsan and Kyung-Nam areas established an area-based occupational disease surveillance system and used this system to collected case information for the purpose of preventing occupational diseases Methods: l l hospital participated in this system. The authors selected five main diseases, which were hand-arm vibration syndrome (HAVS), work-related musculoskeletal disorder (WRMSD), occupational asthma, occupational skin disease and occupational and occupational lung cancer and established their case definitions. All cases were reported on the web, and real time analysis was conducted. Results: Between April 2001 and April 2003, 192 cases of HAVS, 118 cases of WRMSD, 33 cases of occupational asthma, 17 cases of occupational skin disease and 17 cases of occupational lung cancer (for a total of 377 cases of these five main diseases) were reported. most of the HAVS cases came from shipbuilding, and 172 of them (92.7%) were associated with grinding. Of the four main types of WRMSD, carpal tunnel syndrome (CTS) was the most prevalent with 46 cases and shipbuilding was also the main industry involved (83.9%). The ergonomic risk factors involved mainly associated with the hand. In 19 (57.6%) and 4 (12.1%) cases of occupational asthma, the agents involved. The causative agents of occupational lung cancer included Cr, welding fume, PAH, Ni, etc. 10(58.8%) of the cases were diagnosed as squamous cell carcinoma and 5(29.4%) as adenoma. Conclusion: This result showed that an area-based occupational disease surveillance system might provide an effective method of evaluating the prevalence of such diseases, however the Busan, Ulsan and Kyung-Nam provinces are too large to be treated as individual areas. Therefore, the authors suggest that each province should establish its own surveillance system.

      • 일 정신병원에 입원한 정신분열병 환자에 대한 사회인구학적 및 임상적특징

        Objectives and Methods: The purpose of this study was to evaluate the sociodemographic characteristics, clinical characteristics, season of birth and consequence of these characteristics in recently admitted schizophrenics and to compare these results with earlier studies. The author retrospectively reviewed the hospital records of the 103 schizophrenics admitted to Naju National Mental Hospital from January to December 1997. The data analysis was performed by the chi-square test. Results: 1)Age distribution was most frequent in the 20s for both sexes(47.6%), and the sex ratio of male: female was 1: 0.9 2)Unmarried(92.6%), non-religion(66.7%), unemployed(83.3%) was more frequent in male patients and the mean age of onset was earlier(24.4) in male patients. 3)Married or cohabiting(42.9%), Christian(65.3%), employed(57.1%) was more frequent in female patients and mean age of onset was later(27.4) in female patients. 4)Education level, subtype and age of onset were most frequent in high school(52.4%), paranoid type(68.0%) and in the 20s(57.3%) for both sexes respectively. 5)ln the seasonal distribution of birth, there was no significant difference among spring, summer, autumn and winter and no significant differences according to age of onset and subtypes. 6)Subtypes had no significant difference according to Age of onset. Conclusion: This study suggests that the mean age of onset is similar in both sexes of recent times. The catatonic and disorganized type has a descending tendency in incidence according to the progression of time. There was no correlation between the season of birth and age of onset, subtypes. There was no correlation between age of onset and subtype.

        Case Report : Non-infected and Infected Bronchogenic Cyst: The Correlation of Image Findings with Cyst Content

        We hereby report a case on bronchogenic cyst which is initially non-infected, then becomes infected after bronchoscopic ultrasound (US)-guided transesophageal fine-needle aspiration (FNA). The non-infected bronchogenic cyst appears to be filled with relatively echogenic materials on US, and the aspirate is a whitish jelly-like fluid. Upon contrast-enhanced MRI of the infected bronchogenic cyst, a T1-weighted image shows low signal intensity and a T2-weighted image shows high signal intensity, with no enhancements of the cyst contents, but enhancements of the thickened cystic wall. The patient then undergo video-assisted thoracic surgery 14 days after the FNA. The cystic mass is known to be completely removed, and the aspirate is yellowish and purulent. To understand the image findings that pertain to the gross appearance of the cyst contents will help to diagnose bronchogenic cysts in the future.

        Background : In recent years gonorrhoea has been panendemic and remains one of the most common Sexually Transmitted Diseases in the world, especially in developing countries. Objective & Method : For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria Gonorrhoeae(PPNG), Ive have been trying to study the patients who have visited the VD Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results : In 1994, 168 strains of N. gonorrhoeae were isolated, among which 109 (64,9% ) were PPNG. Conclustion : Our results suggests that the prevalence of PPNG in Seoul is still increasing.

        Quantification and Distribution Evaluation of Quercetin and Kaempferol in Rat Eyes after Oral Administration of EEDK Using Ultra-Performance Liquid Chromatography–Tandem Mass Spectrometry

        Although recent pharmacological studies have demonstrated the beneficial effects of the ethanol extract of Diospyros kaki (EEDK) against eye diseases in vitro and in vivo, they could not conclude whether two strong antioxidants, quercetin and kaempferol, in the EEDK were bioavailable in eye tissue after oral administration. A method based on ultra-performance liquid chromatography coupled with triple quadrupole tandem mass spectrometry (UPLC-MS/MS) was herein developed for the highly selective and sensitive quantification of quercetin and kaempferol in rat eye tissue. The method was applied to investigate the distribution of quercetin and kaempferol in rat eyes after oral administration of EEDK at 0, 5, and 35?min after it was successfully validated for specificity, linearity, a low limit of quantification, precision, accuracy, extraction recovery, and matrix effect. The distribution of quercetin and kaempferol in rat eyes at 35?min after oral administration of the EEDK was 0.0099% and 0.0041%, respectively. These results will be useful for the precise determination of optimized in vivo EEDK dose and time for the effective prevention and treatment of eye diseases.

      • Chlorogenic Acid and Coffee Prevent Hypoxia-Induced Retinal Degeneration

        <P>This study explored whether chlorogenic acid (CGA) and coffee have protective effects against retinal degeneration. Under hypoxic conditions, the viability of transformed retinal ganglion (RGC-5) cells was significantly reduced by treatment with the nitric oxide (NO) donor <I>S</I>-nitroso-<I>N</I>-acetylpenicillamine (SNAP). However, pretreatment with CGA attenuated cell death in a concentration-dependent manner. In addition, CGA prevented the up-regulation of apoptotic proteins such as Bad and cleaved caspase-3. Similar beneficial effects of both CGA and coffee extracts were observed in mice that had undergone an optic nerve crush (ONC) procedure. CGA and coffee extract reduced cell death by preventing the down-regulation of Thy-1. Our <I>in vitro</I> and <I>in vivo</I> studies demonstrated that coffee and its major component, CGA, significantly reduce apoptosis of retinal cells induced by hypoxia and NO, and that coffee consumption may help in preventing retinal degeneration.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jafcau/2014/jafcau.2014.62.issue-1/jf404285v/production/images/medium/jf-2013-04285v_0008.gif'></P>
