        Effect of Body Condition Score (BCS) on In Vivo Embryo Production and Pregnancy Rate Following Superovulation in Hanwoo

        Body condition score (BCS) is a useful management tool for distinguishing differences in nutritional needs of cows in the herd. Although it is not always possible to quantify the nutrient content of the feed supplied to the donor cow, the nutritional status can be determined by the BCS. The objective of this study was to evaluate in vivo embryo production, return to estrous of donor and pregnancy rate of recipients following BCS in Hanwoo superovulation. Sixty nine Hanwoo donor cows were flushed on day 7 of estrus cycle with same FSH and artificial insemination by the same technicians. Embryos were recovered on 7 days after the third insemination by flushing the uterus with Embryo Collection Medium. The results obtained were as follows: No differences were observed in the efficiency of superovulation rates regardless of BCS (≤2.0, 2.5 to 3.0, and ≥3.5). The mean number of total embryos was each 5.20±0.86, 11.56±1.04, and 6.23±1.07. The mean number of transferable embryo from ≤2.0, 2.5 to 3.0, and ≥3.5 of BCS was 2.60±0.87, 7.94±0.89, and 4.75±1.32, respectively (p<0.05). Return to estrous regardless of donor BCS was no difference. The pregnancy rates of recipient were BCS ≤2.0 11.76%, 2.5 to 3.0 40.79%, and ≥3.5 11.11%, following transfer of fresh embryos produced in vivo, respectively. These results indicate that if the Hanwoo with BCS 2.5 to 3.3 are used for donor and recipient, the embryo production and the conception rate will be greater.

        열공형과 비열공형 피질하 혈관성 치매에서 위험인자의 차이에 관한 비교 연구

        Backgrounds and Objectives: Vascular dementia is a group of dementing disoders arising from various stroke syndrome. Among these. subcortical ischemic vascular dementia (SIVD) is regarded as a relatively distinct clinical entity. However, MRI patterns of SIVD are not homogenous. In some patients, lacunes are dominant, and in others, subcortical white matter changes are. This study was designed to compare risk factor profiles between SIVD with and without multiple lacunes. Methods: We divided 47 subjects (22 males, mean age. 68 years) recruited from VADAPET (Multicenter Trial For Evaluation Of The Changes In the PET Images Of Subcortical Vascular Dementia Patient) study into two groups one with more than 5 lacunes in deep gray matter (lacune group) and the other with 5 or less(non-lacune group) Clinical characteristics and laboratory findings of two groups were compared. Results: Nineteen of 47 patients (40%) belonged to the lacune group. The lacune and non-lacune groups d d not differ in the following variables: age, hypertension, diabetes mellitus, hyperlipidemia heart disease, history of stroke or TIA, history of trauma or major surgery, family history of hypertension stroke, or dementia, age at diagnosis of dementia, body mass index, white blood cell count, ESR, CRP, fibrinogen, hemoglobin A1C, total cholesterol. LDL cholesterol creatinine, proteinuria, glucosuria, and microhematuria. However, male sex, smoking alcohol. hemoglobin, and HDL cholesterol were possibly associated more with lacune group SIVD than with non-lacune group (p<0 1) Multivariate analyses revealed that smoking, hemoglobin, and HDL cholesterol were independent predictors of SIVD with multiple lacunes Conclusion: Our study suggests that SIVD with multiple lacunes may be significantly different in smoking habits hemoglobin, and HDL cholesterol from SIVD without multiple lacunes.

      • 편도 절제술 후 출혈에 관한 임상적 분석

        Background and Objectives : Hemorrhage is one of the most common complication after tonsillectomy. Despite of many various efforts, it still remains unsolved problem to many otolaryngologist. Because so many factors are related to postoperative bleeding after tonsillectomy, so we tried to see the postoperative hemorrhage rate in relation to various parameters. Material and Methods : A retrospective study on patients who had underwent tonsillectomy in Wallace Memorial Baptist Hospital for 6 years from January 2002 to December 2007. Parameters such as sex, seasons of year, age distribution, tonsil size, previous tonsillitis history and anesthesia type were considered. Results : Overall hemorrhage rate was 2.6%. Postoperative hemorrhage was common in adult males with incidence of 4.5% and most of them was secondary hemorrhage. In age distribution, the incidence was high in the group of 16~30 year. Bleeding was common in autumn, grade II tonsil and increased when the history of tonsillitis was frequent, and local tonsillectomy was better in terms of bleeding compared with tonsillectomy under general anesthesia. but there was no statistical significance. Conclusion : To reduce the incidence of postoperative hemorrhage, perioperative education and complete bleeding control during operation is important for high risk patients.

      • 묏대추나무(Zizyphus jujuba Miller)의 器官分化에 미치는 培地造成 및 生長調節物質의 影響

        This experimet was carried out to establish regeneration systems of Zizyphus jujuba Miller, which will be a fundamental tool for breeding of new cultivar of Zizyphus spp. through biotechnological techniques and for in vitro physiological researches. Explants such as cotyledon segments, hypocotyl segments, epicotyl segments, internodal segments and leaves were excised from in vitro grown sterile seedlings and cultured on Woody Plant Medium(WPM) and half strength Murashige Skoog Medium(1/2MS) containing 6 kinds of growth regulators BAP, kinetin, 2iP, zeatin 2,4-D and NAA at 5 levels of 0, 0.2, 0.5, 1.0, 2.0 mg/l respectively. After 6 weeks in culture, formation and growth of shoot, root and callus were investigated. All the five kinds of explants tested showed organogenetic potentials(both shoots and roots) and callus formation by the media and growth regulator regulations, but the regeneration rate and growth were varied with the factors above. The best results in shoot regeneration were obtained when epicotyl segments were cultured on 1/2MS containing 0.2 mg/l 2,4-D(40%). and better result on WPM containing 0.2 mg/l zeatin(30%). Roots were easily formed and grown when excized leaves were cultured on 1/2MS medium containing 1.0 mg/1 NAA, where regeneration rate was 90% average root number, 5.55. Of the basal media tested for shoot regeneration in case of cotyledon and epicotyl which showed better results. 1/2MS was more efficient than WPM from cotyledons, but vice versa from epicotyls. For root regeneration, WPM was better than 1/2MS from cotyledons but vice versa from leaves. Callus formation and growth was not affected by the basal media tested. Two auxins, NAA and 2.4-D supplemented to basal media, as we know, was effective for root regeneration and callus formation. But a strange result was obtained:2,4-D supplemented to 1/2MS showed better shoot regeneration rate from epicotyls than any cytokinins. NAA promoted root regeneration and growth from all the explants tested. Of the cytokinins tested BAP and zeatin were more effective than 2iP and kinetin in shoot regeneration. Zeatin was effective in shoot regeneration from cotyledons, epicotyls and internodes. From the above results, a protocol could be suggested that epicotyls are cultured on 1/2MS media containing 0.2mg/1 2,4-D for shoot regeneration and cultured on 1/2MS medium containing NAA for rooting. For callus formation explants are cultured on WPM containing 1.0mg/1 2,4-D and the calli formed could be used for shoot regeneration and for somatic embryogenesis by further culture.

        Statistical Parametric Mapping 분석을 이용한 알쯔하이머병과 혈관성 치매의 Tc-99m HMPAO SPECT 영상 소견

        Background:Functional neuroimaging of single photon emission computed tomography (SPECT) has been used to diagnose or to define the pathophysiological mechanisms of dementias. The purpose of this study was to assess the specific patterns and the differences of regional cerebral blood flow (rCBF) in the patients with Alzhermer's disease (AD) and vascular dementia (VD) using Tc-99m HMPAO SPECT. Methods:Fifty five AD, 34 VD, and 30 normal control subjects participated in this study. Patients with VD and AD were matched for age and clinical dementia rating scores. Three groups were evaluated with Tc-99m HMPAO SPECT for measuring rCBF. Statistical parametric mapping (SPM) software was used for analysis of SPECT image data. The SPECT data in the patients with AD and VD were compared with the normal control subjects and then compared with each other. Results:SPM analysis of the SPECT image showed significant perfusion deficits in the bilateral frontotemporal regions in the patients with AD and in the bilateral frontotemporoparietal regions in the patients with VD compared with normal control subjects (uncorrected p<0.001). AD patients, compared to VD patients, revealed significant hypoperfusion in the bilateral frontal areas, left middle temporal gyrus, right fusiform gyrus, right precuneus and right postcentral gyrus (uncorrected p<0.01). VD patients, compared to AD patients, revealed significant hypoperfusion in the bilateral thalami, left insula, bilateral postcentral gyri, right cuneus, right cingulate gyrus and bilateral frontal subcortical regions (uncorrected p<0.01). Conclusions:Our study shows the characterisitcs and patterns of perfusion deficits in AD and VD patients, and defines the specific areas of hypoperfusion in these two dementias.

      • 고혈압쥐에서 성장에 따른 신장 세포막 Na, K-ATPase와 Adrenergic Receptor의 변동에 관한 연구

        To investigate the developmental changes of Na, K-ATPase and adrenrgic receptors in the kidney basolateral membrane(BLM) of hypertensive rat, Na, K-ATPase activities, [^3H]ouabain, [^3H]prazosin and [^3H]dihydroalprenolol(DHA) binding sites were measured in the kideny basolateral membrane prepared from 1, 3, 5, 10 and 15 weeks old Sprague-Dawley rats(SD) and spontaneously hypertensive rats(SHR). Results are as follows ; 1. Systolic blood pressure in SHR was not elevated at 3 and 5 weeks old, but elevated significantly to 185 mmHg at 10 or 15 weeks old. 2. Na, K-ATPase activity of BLM in 3 weeks old SD was markedly increased compared to that in 1 week old and there was little change in 5, 10 or 15 weeks old rat. 3. Na, K-ATPase activity of BLM in SHR was also markedly increased at 3 weeks old, but continuously increased by 15 weeks old. Although the Na, K-ATPase activity in BLM of 1, 3 or 5 weeks old SHR was lesser, that of 10 or 15 weeks old SHR was higher than that observed in corresponding age of SD. 4. The numbers of ouabain binding sites were increased in both groups with increasing age. In the SHR, the numbers of ouabain binding; sites were lower in 1, 3 or 5 weeks old but higher in 10 or 15 weeks old, than those in corresponding age of SD. 5. In the dose-response curves of ouabain sensitive Na,K-ATPase activity and the ouabain binding experiments, Na, K-ATPase in the BLM of SD or SHR was consistent with the putative existence of low affinity site(IC_50 : 35 - 55 μM, Ed : 40 - 90 μM) 6. α1-Adrenergic receptor densities([^3H] prazosin binding sites) were markedly increased by 3 weeks after birth and further increased slightly by 5, 10 or 15 weeks old SD. 7. The numbers of prazosin binding sites in SHR were also increased with increase of age. However those were lower in 1, 3 or 5 weeks old, higher in 10 or 15 weeks old SHR than those in corresponding age of SD. 8. In the prazosin binding experiments, Ed value was 0.1 - 0.4 nM and there was no difference between in Kd values of SD and SHR. 9. β-Adrenergic receptor densities([^3H] DHA binding sites) in SD were markedly increased in 3 weeks old compared to those in 1 week old and there were no further changes of receptor densities in 5, 10 or 15 weeks old. But those in SHR were still increased until 15 weeks old. The numbers of DHA binding sites in 1, 3 or 5 weeks old SHR were slightly lower but almost same in 10 or 15 weeks old, compared to those in corresponding age of SD. 10. In the DHA binding experiments, Kd value was 1.0 - 1.8 nM and there was no difference between in Kd values of SD and SHR. From the above results, it is suggested that as increase of age, Na, K-ATPase, α1 and β-adrenergic receptors in the kidney basolateral membrane of SHR were increased with different pattern from normotensive SD ; lower in young age, but same or higher in hypertensive period of SHR than SD. These observed changes in Na,K-ATPase activity and adrenergic receptor densities of kindney basolateral membrane may partly contribute to the regulation of blood pressure in SHR.

        전기화학적 방법을 이용한 수소제거에 관한 연구

        Hydrogen embrittlement (HE) mitigation on the low alloy steel was studied using electrochemical technique. This method can be applied to reduce HE susceptibility. Hydrogen induced cracking (HIC) characteristics of the low alloy steel was investigated in aerated and deaerated borate buffer solution (0.3M H₃BO₃ + 0.074M N₂B_(4)O_(7), pH 8.4). The electrochemical properties of the low alloy steel showed passive behavior in this environment. By performing galvanostatic and potentiostatic test, hydrogen was charged and discharged to specimens respectively. By applying the potential of +630 mV_(SCE), the accumulated hydrogen was eliminated electrochemically. A specimen discharged for 24h indicated the efficiency of the hydrogen discharge up to 99.9%. Slow strain rate tests (SSRT) results showed that a specimen with higher hydrogen concentration revealed inferior mechanical properties such as shorter time to fracture, lower elongation, ultimate tensile strength and yield strength values. Furthermore, SEM fractographs revealed that the higher the hydrogen concentration is, the more probable the features of brittle fracture are. (Received July 26, 2004)

      • 순차적 크리깅기법을 이용한 미소혼합기의 최적화

        An active micro-mixer, which is composed of an oscillating micro-stirrer in the micro-channel to provide effective mixing was optimized. The effects of molecular diffusion and disturbance by the stirrer were considered with regard to two types of mixer models: the simple straight micro-channel and micro-channel with an oscillating stirrer. Two types of mixer models were studied by analyzing mixing behaviors such as their interaction after the stirrer. The mixing was calculated by Lattice Boltzmann methods using the D2Q9 model. In this study, the time-averaged mixing index formula was used to estimate the mixing performance of time-dependent flow. The mixing indices of the two models were compared. From the results, it was found that the mixer with an oscillating stirrer was much more enhanced' and stabilized. Therefore, an approximate optimization of an active micro-mixer with an oscillating stirrer was performed using Kriging method with OLHD (Optimal Latin Hypercube Design) in order to determine the optimal design variables. The design parameters were established as the frequency, the length and the angle of the stirrer. The optimal values were obtained as 1.5754, 0.803D and ±45°, respectively. It was found that the mixing index of the optimal design increased by 83.36% compared with that of the original design.
