        발아현미가 LPS로 유도된 지방세포의 염증반응에 미치는 영향

        Germinated brown rice (GBR, Orysa sartiva L.) has been reported to have anti-obesity and anti-inflammatory effects. However, the mechanisms underlying these effects in adipocytes are not fully understood. Therefore, this study was conducted to explore the anti-inflammatory mechanisms of GBR on lipopolysaccharide (LPS)-stimulated 3T3-L1 adipocytes. 3T3-L1 adipocytes were pretreated with GBR extracts (0-20 mg/mL) 1 h before LPS stimulation. The mRNA expression of adipokines and Toll-like receptor 4 (TLR4) were measured by RT-PCR. The protein expressions of TLR4- related molecules were detected by western blotting and nuclear factor-κB (NF-κB) activation was measured. Our results showed that GBR extract dose-dependently inhibited mRNA expression of LPS-induced tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1). GBR extract was found to inhibit LPS-induced mRNA expression of TLR4 and protein expression of both myeloid differentiation factor 88 (MyD88) and TNF receptorassociated factor 6 (TRAF6). Furthermore, GBR extract significantly inhibited extracellular receptor-activated kinase (ERK) phosphorylation and NF-κB activation. These results suggest that GBR extract has the anti-inflammatory effects on LPSinduced inflammation via inhibition of TLR4 signaling, includingthe ERK and NF-κB signaling pathways, in adipocytes.

        생명공학분야의 연구데이터 공유 사례에 관한 연구

        In this study, major steps of the research data management plan were derived through the research data management guidelines of major countries (NISO DMP in the US, UK DMP in UK archives, etc.). The results obtained are support for research data policy & planning, research data technical support, research data sharing support, research data legal mechanism support, and research data education support. In this study, we analyzed seven cases of data sharing among 7 domestic and foreign biotechnology. Shared use case countries are limited to the United Kingdom and the United States, which play a leading role in the management of research data. In Korea, shared cases were analyzed for the Korean Bio Information Center and related systems, which is a research and performance management and distribution agency designated by the Ministry of Science and Technology. 연구데이터 관리․공유의 연구 패러다임 변화로 미국 및 유럽은 연구데이터 관리 의무화를 법제도화 하고 있으나 국내 연구현장은 인식부족과 인프라 미비로 연구데이터 관리지침 및 관리계획 등이 부재한 상황이다. 본 연구에서는 주요국가의 연구데이터 관리지침(미국 NISO DMP, 영국 UK 아카이브 DMP 등)을 조사하여 연구데이터 관리계획의 주요 단계를 도출하였다. 도출한 결과는 연구데이터 정책 계획 지원, 연구데이터 기술적 지원, 연구데이터 공유 활용 지원, 연구데이터 법적 매커니즘 지원, 연구데이터 교육 지원이다. 데이터 활용도가 가장 높은 생명공학분야 국내외 7개 기관의 연구데이터 공유 활용 사례를 연구데이터 관리 구성요소, 연구데이터 공유방법에 따라 비교 분석하였다. 유럽생물정보학연구소와 미국 국립생명공학정보센터는 연구데이터 공유 활용을 하고자 연구데이터 관리계획, 문서화, 데이터포맷, 데이터 저장, 공유 및 접근, 보존 등 관리를 위한 각 단계를 시행하고 있으나 국내의 경우 연구성과물 중 생물자원에 대한 제출만 이루어지고 있다. 연구데이터 관리계획(DMP)지원, 기술적 지원, 공유 활용 체계 지원, 법적 저작권 지원 등에 대한 가이드 및 매뉴얼 제작 배포로 국내 연구데이터 관리 및 공유 활용 대책을 연구데이터 관리계획 단계별로 마련해야 한다.

        Chemopreventive Action of Anthocyanin-rich Black Soybean Fraction in APCMin/+ Intestinal Polyposis Model

        Background: Anthocyanins have been shown to inhibit cancer cell growth by suppressing oxidative stress and inflammatory responses. The purpose of this study was to investigate the effects of an anthocyanin-rich extract (AE) from black soybean coat on intestinal carcinogenesis. Methods: ApcMin/+ mice were fed a diet of 0.2% or 0.5% AE for 7 weeks. We analyzed the number of intestinal tumors, oxidative stress and inflammatory markers associated with β-catenin and cytosolic phospholipase A2 (cPLA2) signals. The number of intestinal tumors, and cellular expression of β-catenin were determined. Results: The number of intestinal tumors was significantly lower in mice fed a 0.5% AE diet compared to those of the other groups. Cytosolic β-catenin expression was significantly decreased in the AE supplemented groups compared to that of the control animals. In addition, mucosa expression of cyclooxygenase-2 and cPLA2 were also significantly decreased in the 0.5% AE group, by 32% and 62%, respectively, compared to the control group. Conclusions: These results suggest that dietary AE reduced the development of intestinal tumors, possibly through the ability to suppress oxidative stresses, decreasing inflammatory responses mediated by β-catenin associated signals.

        다양한 열처리 조건에 따른 우엉뿌리의 항산화 활성

        This study examined the changes in antioxidant activity and contents of phenolic compounds inblanched, steamed, and autoclaved burdock root (BR). The total polyphenolic and flavonoids contents of raw and cooked BR were determined spectrophotometrically. The antioxidant activity of BR was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2- azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and oxygen radical absorbance capacity (ORAC) assays. The main phenolic compounds in BR were quantified by HPLC (high performance liquid chromatography). Both blanching and steaming treatments significantly increased the antioxidant activities of BR in all groups (5 min, 15 min, and 30 min), whereas in autoclaving treatment, the 30 min treatment only showed an increase in the antioxidant activities of BR. The 30 min blanched BR exhibited the strongest DPPH and ABTS radical scavenging activities and possessed the highest total polyphenol and flavonoid phenolic contents. The 15 min-steamed BR showed the highest ORAC value. The main phenolic compound of the 15 min-steamed BR was CGA (chlorogenic acid). These results suggest that heat cooking methods, such as blanching and steaming, improve the antioxidant activity of BR by increasing the concentration of phenolic compounds.

        검사의 증인사전면담의 필요성과 한계에 대한 연구

        형사재판에서 조서재판의 문제점이 부각되면서 증인신문의 중요성은 점차 강조되고 있다. 법정에서 증인신문이 효과적이고 원활하게 이루어지기 위해서는 검사의 증인사전면담이 허용되어야 할 것이나, 이는 증인에 대한 회유나 압박 등으로 증언이 왜곡될 가능성이 늘 존재하므로 구체적인 한계를 미리 설정하는 법 규정 마련이 필요하다. 검사의 증인사전면담의 한계를 구체적인 규정으로 마련하는 것은 피고인에게 불의의 타격을 주지 않기 위한 목적도 있지만, 한편으로는 검사가 그 규정을 잘 지켜서 면담을 한 경우에는 증언의 신빙성을 인정받아 실체적 진실발견에 한걸음 더 나아갈 수 있기 때문이다. 그동안 우리 형사실무에서 이루어진 검사의 증인사전면담 사례는 대부분 피고인에게 유리한 증언을 한 증인의 증언을 번복하기 위함이거나 증인의 증언 번복을 방지하기 위한 목적에서 이루어진 경우가 대부분이었는데, 검사의 증인사전면담의 구체적인 기준을 마련함으로써 암암리에 이루어지던 검사의 증인사전면담을 형사재판의 준비단계로 끌어올려 제도화할 때가 되었다. 이 논문에서는 그와 관련하여 해외 각국의 실무와 국제형사재판 실무를 검토하고, 국내 학계의 견해 및 검사의 증인사전면담과 관련한 대법원 판례 등을 정리한 후 검사의 증인사전면담과 관련한 구체적 기준을 제시하였다. As the problem of the written trial is highlighted in the criminal trial, the importance of the witness interrogation is gradually emphasized. In order for witness interrogation to take place effectively and smoothly, prosecutors should be allowed to conduct pre-trial witness interviews, but there is always a possibility that testimony will be distorted due to conciliation or pressure on witnesses.The limitation of the prosecutor's pre-trial witness interviews is set by certain regulations so as not to hurt the defendant unfairly, but if the prosecutor complies with the regulations, the witness's credibility can be recognized and take a step forward in discovering the substantive truth.In the meantime, most of the cases of prosecutor's pre-trial witness interviews in our criminal trials have been made to reverse the testimony of the witness who testified in favor of the defendant or to prevent the witness's testimony from reversing. It is time to establish specific standards for the prosecutor's pre-trial witness interviews and institutionalize the prosecutor's pre-trial witness interviews as a preparation stage for criminal trials.This paper reviewed the practice of overseas countries and international criminal trials, summarized the opinions of domestic academia and the Supreme Court's precedents related to pre-trial witness interviews, and presented specific standards for pre-trial witness interviews by prosecutors.

        초등 예비교사와 현직교사의 수학 평가문항 개발사례 연구

        The purpose of this study is to investigate the assessment expertise of Mathematics' teachers, focusing on the competence in designing assessment item development. In this present research, I analysed how the teachers' competence appears in designing assessment items development when they generated problems the given problem into a new one. To examine this assumption, the following research questions were posed and investigated in the present study : How do Pre-service and In-service Teachers in primary schools develop the assessment item when generating problems the given problems into new problems? The result from the case study of metamorphosing the given problem into a new problem teachers used similar patterns switching numbers or changing units in order to develop new problems. Also, teachers in primary schools tend to develop problems as commonly as in the mathematics workbooks. In-service teachers tend to have better skills developing assessment items, but there were quite much of variability between individuals.

        High-fat Diet Accelerates Intestinal Tumorigenesis Through Disrupting Intestinal Cell Membrane Integrity

        Background: Excess energy supply induces chronic low-grade inflammation in association with oxidative stress in various tissues including intestinal epithelium. The objective of this study was to investigate the effect of high-fat diet (HFD) on intestinal cell membrane integrity and intestinal tumorigenesis in ApcMin/+ mice. Methods: Mice were fed with either normal diet (ND) or HFD for 12 weeks. The number of intestinal tumors were counted and biomarkers of endotoxemia, oxidative stress, and inflammation were determined. Changes in intestinal integrity was measured by fluorescein isothiocyanate (FITC)-dextran penetration and membrane gap junction protein expression. Results: HFD group had significantly higher number of tumors compared to ND group (P < 0.05). Blood total antioxidant capacity was lower in HFD group, while colonic 8-hydroxy-2’-deoxyguanosine level, a marker of oxidative damage, was higher in HFD group compared to that of ND group (P < 0.05). The penetration of FITC-dextran was substantially increased in HFD group (P < 0.05) while the expressions of membrane gap junction proteins including zonula occludens-1, claudin-1, and occludin were lower in HFD group (P < 0.05) compared to those in ND group. Serum concentration of lipopolysaccharide (LPS) receptor (CD14) and colonic toll-like receptor 4 (a LPS receptor) mRNA expression were significantly higher in HFD group than in ND group (P < 0.05), suggesting that significant endotoxemia may occur in HFD group due to the increased membrane permeability. Serum interleukin-6 concentration and myeloperoxidase activity were also higher in HFD group compared to those of ND group (P < 0.05). Conclusions: HFD increases oxidative stress disrupting intestinal gap junction proteins, thereby accelerating membrane permeability endotoxemia, inflammation, and intestinal tumorigenesis.
