        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

      • The Antioxidant Effect of the Rhaponticum uniflorum Extracts on the Oxidative Stress in H₂O₂ induced Human Keratinocytes

        In this study, To investigate the antioxidant activities of Rhaponticum uniflorum extracts (RUE), we assessed scavenging activities on DPPH, ABTS radicals, and reducing power. The DPPH and ABTS radical scavenging activities (SC50) of RUE were 96.97±1.19 μg/mL and 31.26±0.13 μ g/mL, respectively, and the reducing power significantly increased dose-dependent manner. To study the antioxidant effecters of RUE in H₂O₂ induced human keratinocytes, expression of Nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were determined by Western blot and qRT-PCR. RUE induced the expression of HO-1, a critical target enzyme of Nrf2, in a concentration-dependent manner. The expression of HO- 1 mRNA was also induced in H₂O₂-treated HaCaT cells but no significant difference in the level of HO-1 mRNA was observed. Taken together, these results suggest that RUE may be useful as a potential ingredient for anti-aging products.

      • 콩나물의 asparagine 함량에 미치는 요인 구명

        알코올의 숙취해독에 뛰어난 효과가 있는 것으로 알려진 sparagine은 콩나물의 뿌리에 함량이 가장 많은 것으로 알려져 있다. 본 연구논문에서는 콩나물 뿌리 발생에 미치는 재배실의 온도, 일당 관수횟수, 재배일수 및 동일용기에 사용한 콩 량에 대한 영향을 조사하여 얻어진 연구결과를 요약하면 다음과 같다. 1. 아가통과 풍산나물콩의 수율은 재배일수에 따라 경시적으로 증가하였으며, 재배실 온도가 20℃에서 보다 30℃에서 높았으며, 콩나물의 수율은 5회 관수, 8일 재배시 가장 높았다. 2. 아가콩과 풍산나물콩의 콩나물 부위별 건물중의 비율은 자엽이 가장 많았고 뿌리가 가장 낮았으며, 재배실 온도가 20℃에서 보다 30℃에서 비율이 더 높았다. 3. 아가콩의 sparagine 함량은 재배실 온도 30℃에서 5회 관수 8일 재배한 하배축에서 18.9%로 가장 높게 나타났다. 4. 풍산나물콩의 sparagine 함량은 재배일수가 길수록, 재배온도가 높은 경우 높았으며, 하배축에서 가장 높았고, 30℃에서 2회 관수 8일 재배시 하배축에서 18.8%로 가장 높게 나타났다. 5. 아가콩은 300g, 풍산나물콩은 500g 치상시 수율이 가장 높았으며, 잔뿌리의 수는 아가콩과 풍산나물콩 모두 치상량이 증가함에 따라 증가하는 경향이었다. It is known that the content of asparagine, an excellent detoxifying substance of alcohols in human body, is the highest in the roots of soy-sprout. At the same time, it is inferred that soy-sprouts producing more roots are better for detoxifying. In this experiment, the effects of room temperatures on number of watering per day, and duration of soy-sprout culture were carefully investigated. Some of the results obtained are as follows; 1. The yield rate of soy-sprouts for Agakong and Pungsannamulkong was continuously increased up to 9 days. It was higher in room temperature of 30℃ than in 20℃ and was the highest at 8th day of culture with 5 times of watering per day. 2. The asparagine content in soy-sprouts of Agakong and Pungsannamulkong was the highest in cotyledon and the lowest in roots. This rate was higher in the room temperature of 20℃ than in 30℃. 3. The highest asparagine content of soy-sprout of Agakong was 18.9%, obtained in the room temperature of 30℃, cultivated for 8 days with 5 times of watering per day. 4. The highest asparagine content of Pungsannamulkong was 18.8%, obtained in hypocotyl cultivated in the room temperature of 30℃ for 8 days with the number of 2 times watering per day. 5. When an cultivation apparatus of 5 liters volume was used, the optimum seed amount for the highest yield rate was 300g for Agakong and 500g for Pungsannamulkong. At the same time, the number of lateral roots showed increasing tendency with more amounts of soybean seeds used.

        This study was conducted to investigated the major characteristics of genetically modified rice of "Milyang 204" originated from Dongjinbyeo compared to a non-transgenic rice varieties Dongjinbyeo and Junambyeo. Basta resistant transgenic rice lines carrying bar gene produced by the Yeongnam Agricultural Research Institute were evaluated for their agronomic characters. The transgenic Japonica rice of "Milyang 204" showed inferior phenotypic traits compared to a non-transgenic rice variety Dongjinbyeo and Junambyeo. On the basis of UPOV (Union Internationale Pour la Protaection des Obtentions Vegetables) and NSMO(National Seed Management Office) the transgenic "Milyang 204" showed difference in some traits out of some agronomic traits, such as leaf color, angle of flag leaf, number of spikelets, culm length, white core and white belly compared to the nontransgenic varieties rice.

        Quantitative determination of diarylheptanoids (oregonin and hirsutanonol), which were characteristic components of Alnus spp. has been conducted by High Performance Liquid Chromatography. The HPLC analysis was carried out using an YMC J'sphere ODS-H80 column [150×4.6mm, S-4μm. 80A(particle)] and a combination of acetonitrile and H2O was used as mobile phase in gradient solvent system. The results showed differnce in the species (Alnus japonica, A hirsuta and A. hirsuta var. sibirica)

        불완전 전뇌 허혈후 소생의 백서 모델을 이용한 재관류 뇌혈류량과 허혈성 뇌조직 손상도와의 관계

        Background: Experimental data indicate that low-flow reperfusion following prolonged cardiocirculatory arrest may aggravate early cerebral microcirculatory reperfusion disorders. We investigated the influence of cerebral reperfusion flow change to the ischemic histopathologic damage of brain tissue after incomplete forebrain ischemia in rats. Materials and method: Anesthetized Sprague-Dawley rats were undergone ligation of both internal carotid artery by microvascular clamp for 10 minutes. After release of the clamp, reperfusion was started with several different flow levels (0, 10, 20, 30, 50, and 100%) of internal carotid artery comparing to pre-clamping phase using flowmeter. After 15minutes of reperfusion, rat brains were prepared by perfusion-fixation with 3% formaldehyde. Under light microscopic examination of Hematoxylin-Eosin stained tissue slide, histopathologic damage was examined at cortex, putamen, and hippocampus regions. Categorical hisotopathologic damage scores were derived in each regions by manual counts of ischemic neurons. Result: The histopathologic damage scores were 0, 10.2±0.5, 7.6±1.5, 5.9±1.4, 5.0±2.8, 3.5±0.7, and 1.0±0.0 in control, 0, 10,20, 30, 50, and 100% reperfusion groups, respectively(p<0.05). Conclusion: Our results showed significant increment of brain histopathologic damage scores along with decreasing amount of cerebral reperfusion flow after incomplete forebrain ischemia. We believe restoration of reperfusion flow to pre-ischemic level would be a critical component in attenuation of brain ischemic damage.

      • 태음인 체질의 비만아동에 대한 운동처방

        본 연구는 비만해소 방안과 효과의 검토 및 자료를 제시하기 위하여, 비만도 20% 이상 초등학교 5·6학년 남학생 중 체질진단결과 태음인체질의 10명을 대상으로 10주 동안 주4회, 50∼70% HRmax의 강도로 운동처방프로그램을 실시하여 운동처방 전·후의 신장, 체중, 혈압, 심박수, 체지방량의 신체적 변화와 특수 인성검사A의 심리적 변화를 측정하였으며, 자료분석은 SPSS통계프로그램의 비모수 검정의 Wilcoxon 부호순위 검정을 이용하였다. 신체적 변화에서는 신장은 근사 유의확률이 P < .005, 혈압은 수축기와 이완기에서 P < .011과 P < .026, 체지방량은 P < .028로 유의한 것으로 나타났으나, 신장은 운동처방에 의한 영향이라고 볼 수 없으므로 유의한 차이가 없는 것이 타당하다. 체중과 심박수에서는 유의한 차이가 나타나지 않았다. 심리적 변화에서는 타당도 척도(Validity Scales), 일반 성격척도(General Personality Scales), 특수적응척도(Special Adaptation Scales)에서는 유의한 차이가 나타나지 않았다. 아동비만을 사상의학의 바탕인 체질의학에서 체질적 특징과 비만에 대한 신체적·심리적 특징을 파악하여 체질에 맞는 10주간의 짧은 기간이지만 규칙적으로 꾸준히 운동을 실시한다면 비만을 억제하거나 치료할 수 있고, 나아가 성인비만으로 이환될 가능성을 예방할 수 있을 것이라 생각된다. The purpose of this study was to investigate the effective anti-obesity measures and its effects. The research was conducted to ten boys(the 5th and 6th graders in elementary school) whose obesity degree is over 20 percent. The subjects with Taeeum-In constitution are exposed to exercise programs with 50 to 70 HRmax for 10 weeks. The exercise prescription program was implemented 4 times a week. The physical and psychological changes was measured by the pre & post-program. The analysis of data was relied on Wilcoxon Signed Ranks Test among nonparametric tests(significant level P<.05). The results of this research are as follows: 1) in respect of the physical changes, the blood pressure is significant(P<.011 and P<.026) in the contraction period and the relaxation period respectively. The amount of body fat is significant(P<.028). But heights is not significant(in spite of P<.05) because it was not influenced by the exercise prescription. Weights and heart beats are also not significant; 2) in respect of the psychological changes, the desirability of validity scales and General personality scales and special adaptation scales are not significant. This study suggests that regular exercise in accordance with human constitution may treat obesity and prevent transferability for adult obesity.

      • 주기억장치 DBMS Tachyon을 위한 동시성 제어

        In this paper, we deal with concurrency control in the Tachyon, a main memory DBMS(MMDBMS). Since an MMDBMS, unlike a disk-based DBMS, performs all of data update or retrieval operations by accessing main memory only, the portion of the cost for concurrency control in the total cost for a data update or retrieval is fairly high. Thus, the development of an efficient concurrency control manager highly accelerates the performance of the entire system. The concurrency control in the Tachyon basically employs 2-phase locking protocol, and has the following characterisics. First, it adapts the partition, an allocation unit of main memory, as a locking granule, and thus, dffectively adjusts the trade-off between the system concurrency and locking cost through the analysis of applications. Second, it enjoys low locking costs by maintaining the lock information dirctly in the partition itself. Third, it provides the latch as a mechanism for physical consistency of system data. Our latch supports both of and Unix semaphore facility. Fourth, for solving the deadlock problem, it periodically examines whether a system is in a deadlock state using lock waiting information. In addition, we discuss various issues arising in development such as mutual exclusion of a transaction table, mutual exclusion of indexes and system catalogs, and realtime application supports.

      • Capsicum chinense, 재래종 및 저항성 고추 유전자원에서 역병 저항성 계통 선발

        2000년도에 검정에서 역병에 살아남은 개체들로부터 채종하여 육성한 Capsicum chinense 31계통에 대하여 역병 저항성을 검정한 결과 고도의 저항성을 나타내는 것은 발견되지 않았다. 2001년도의 검정에서 역병에 살아남은 개체로부터 채종한 재래종 26계통에 대하여 다시 역병 저항성 검정을 실시한 결과 KC180, KC230, KC195, KC194에서 다수의 개체가 살아남아 저항성을 나타내었다. 그러나 KC180과 KC230은 각각 AC2258과 CM334와 혼종된 것으로 관찰되었다. KC195와 KC194는 재래종의 형질을 유지하고 있는 것으로 관찰되었다. CM334의 보존 증식과정에 자연교잡이 일어난 것으로 보여 이의 순도향상을 위하여 채종년도별로 시료를 꺼내어 역병저항성 검정을 실시한 결과 가장 오래된 1992년도 채종종자에서부터 약간의 이형주가 관찰도기 시작하여 1995년부터 2001년도까지 시간이 경과함에 따라 많이 변형되어 있는 것을 알 수 있었다. 1992년도 종자에서 이형주를 제거하고 원형의 개체로부터 자식종자를 대량으로 채종하였다. 함께 공시한 AC2258은 순수한 것으로 확인되었다. 1995년도 채종 CM334 종자에서는 비록 혼종은 되었으나 측지발생이 적은 개체들이 발견되어 이들을 개체 선발하여 역병에 저항성이며 측지발생이 적은 계통으로 육성하고 있다. A total of 31 c. chinense lines selected from 2000 screening were tested again for resistance to P. capsici but resistance was not found in the lines. A total of 26 selections from Korean land races tested 2001 spring were tested again for resistance to P. capsici. KC180, KC230, KC195 and KC194 showed moderate resistance to P. capsici. However, it was apparent on the basis of hortivultural characteristics that KC180 and KC230 had been naturally crossed with AC2258 and CM334, respectively. CM334 and AC2248 seed lots that were increased in different years were taken out and tested for resistance to improve their genetic purity because the resistant genetic resources have been showing some off-types in the population. Off-types began to be found in 1992 seed lots and the proportion and degree of the offs was increasing with time up to 2001. Plants true to the type in 1992 seed lots were selected and their inbred seeds were mass produced in a net cage in the greenhouse. AC2258 included in the experiment number of lateral branching at cotyledonary axil, although they were off from the original in the experiment together was uniform. In 1995 seed lost of CM334, plants with resistance to P. capsici and low or no number of lateral branching at cotyledonary axil, although they were off from the original CM334, were found and selection was applied to breed lines fixed in the characters.

      • 西海 沿岸의 養殖場 環境 調査 : 2. 高亭理 海域 김養殖場의 水質環境

        To understand the effects of oceanographic condition on the harvest of laver and establish the basic environmental data for cultivation farm of laver, a series of survey was carried out in Gojeong-ri sea area, west coast of Korea from January to September, 1987. Variation of water temperature in Gojeong-ri sea area was 3.8-25.8℃, pH was 7.8-8.4, and transparency was 1.3-3.1m. Salinity was changed from 19.57 to 31.99% and lower remarkably in July and August owing to a heavy rain. The concentration of DO in surface water wart varied from 9.4-10.1㎎/ℓ and in lower layer was 8.0-9.99㎎/ℓ . COD was 0-4.28㎎/ℓ by alkalic modification of potassium permanganate method. Distribution of total suspended solid was 13.7-488.7㎎/ℓ and the rate of light penetration in 1m water column was 7.2-15.5%. The concentration of nutrients were changed in 0.48-23.63㎍-at./ℓ for ammonia nitrogen, 0-4.45㎍-at./ℓ for nitrite nitrogen, 0.39-3.13㎍-at./ℓ for nitrate nitrogen, 0-21.42㎍-at./ℓ for phosphate, and 0.84-157.32㎍-at./ℓ for silicate. It was showed high concentration in summertime, raining season in general. The concentration of chlorophyll a was 0.39-20.62㎎/㎥. Total 57 species of phytoplankton and 21 groups of zooplankton were identified from collected samples. Dominant species were Coscinodiscus radiatus, Eucampia zodiacus as a phytoplankton and Noctiluca scinitillans, Cladocera, Copepods sp. as a zooplankton. The total standing crop of phytoplankton was ranged of 5,500-1,524,400cells/ℓ and that of zooplankton was 3,000-43,000 individuals/㎥.
