Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

<P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression. Determination of Limiting Nutrient for Algal Growth by Algal Bioassay. Oh, Hee-Mock, Seog June Lee, Seong-Bin Kim, Mi- Kyung Park, Byung-Dae Yoon and Do-Han Kim (Environmental Microbiology Research Unit, Korea Research Institute of Bioscience and Biotechnology, Taejon 305-333, Korea and Water Resources Research Institute, Korea Water Resources Corporation, Taejon 305-390, Korea) Water quaiity and algal growth potential (AGP) using Microcystis aeruginosa were investigated from April to September 1997 in Daechung Reservoir. Secchi disk transparency was 1.1 m in August. Chlorophyll-a concentrations were higher than 10㎍/l at all samples. Total phosphorus concentration was in a range of 0.020∼0.027mg/l which is at the approximate value of 0.03 mg/l, the general critical level of eutrophication. These results indicate that Daechung Reservoir is in an eutrophic state except some temporary times and restricted sites. Phosphorus uptake rate of M. aeruginosa was higher in the water from the epilimnion than in the hypolimnion and also higher under light condition than under dark condition. Therefore, phosphorus uptake rate appears to depend on cellular phosphorous content and light condition, a meaning of endergonic process. Phosphorus was the most frequently limiting nutrient: phosphorus was limiting algal gowth in April and May, nitrogen, phosphorus and trace elements were the limiting factors for algae in August, and trace elements in September. If a fast growing alga, M, aeruginosa, is used as a test organism for assaying AGP, in vivo fluorescence instead of dry weight can be measured for algal biomass and incubation period can be reduced from traditional 14 days to 7 days.

대량생산을 위한 김치의 산업화에 있어서 가장 큰 문제점인 유통시의 산패현장과 포장팽대 및 파손을 지연시켜 김치의 저장기간을 연장하고자 이산화탄소 흡수능력을 가진 물질을 가스흡수제로 처리하여 저장중 김치의 품질을 검토하였다. 품질평가를 위한 측정항목은 pH, 산도, 색상, 이산화탄소 함량 및 젖산균수 그리고 총균수를 측정하였다. 수산화칼륨, 수산화칼슘, 수산화나트륨과 같은 알칼리물질과 연화마그네슘, 염화칼슘과 같은 염화합물을 가스흡수제로 사용한 결과, 수산화칼륨과 수산화나트륨처리구가 타처리구에 비해 pH의 감소가 적었으며, 또한 산도의 증가가 다소 낮았다. 수산화칼륨과 수산화나트륨을 각각 0.25%, 0.5%, 0.75% 세단계의 농도로 처리한 결과 pH와 산도에 있어서 수산화칼륨처리구가 수산화나트륨처리구에 비해 높은 pH와 낮은 산도를 나타내었다. 또한 각 처리구의 농도에 따라 큰 차이가 나타나지 않아 간편성과 경제적인 면, 그리고 시각적인 면을 고려해 볼 때 0.25% 수산화칼륨처리구가 탄산가스흡수에 의해 김치의 저장기간을 연장하는데 다소 효과를 보였다. PE bag으로 포장된 수산화칼슘과 PE flim(0.02㎜)으로 포장한 0.25% 수산화칼륨을 가스흡수제로 사용하여 무처리구와 비교하여 저장중 김치의 품질을 검토하였다. 저장중 0.25% 수산화칼륨처리구와 시판용 김치의 가스흡수제인 수산화칼슘처리구를 비교할 때, 수산화칼륨처리구가 무처리구에 비해 pH 및 산도의 차이가 컸으며, 김치의 조직과 국물속에 용해되어 있는 이산화탄소의 함량도 적었다. 그리고 총균수에 있어서 타처리구에 비해 수산화칼륨처리구가 높은 값을 나타내었다. When kimchi was manufactured and circulated, largest question is rancidity phenomenon and packing swelling. This study was conducted to prevent these phenomenon and prolong the shelf-life of kimchi. KOH, NaOH, Ca(OH)_2, MgCl_(2), and CaCl_(2) were used to gas-absorbent. The pH of kimchi used KOH, NaOH was higher than the others. The acidity of kimchi used KOH, NaOH was lower than the others. The concentration of KOH, NaOH was 0.25%, 0.50%, 0.75%. The decrease of pH and increase of acidity that kimchi dealt with KOH were lower than kimchi used NaOH. There in no significant difference in concentration of gas-absorbent. Comparision of kimchi used 0.25% KOH and Ca(OH)_2. Ca(OH)_2 packaged PE bag and 0.25% KOH packaged PE film(0.02㎜) were used to gas-absorbent. On comparision to control, kimchi dealt with 0.25% KOH was higher than kimchi used Ca(OH)_2 at pH and acidity. The CO_(2) contents of kimchi dealt with 0.25% KOH was lowest. The number of total microbe kimchi dealt with 0.25% KOH was highest.
수행 평가 도구를 이용한 심화·보충형 수준별 교수 전략 개발 : 과학 3의 물질의 구성 Composition of Matter Section in Science 3

제7차 교육 과정의 과학 교과에 대한 심화·보충형 수준별 수업 전략을 개발하여 현장 과학 교사들에게 구체적인 도움을 주고자 하는 것이 본 연구의 목적이다. 수업 운영 형태는 동일 학급 내 교과 수준별 분단 편성이며, 연구된 구체적인 단원은 중학교 과학 3의 ‘물질의 구성’ 단원으로 총 17차시가 소요된다. 먼저 현실적으로 중등 과학 교육 현장에서 실현 가능한 과학과 수준별 교육 과정에 관련된 이론적 배경을 고찰하였다. 둘째, ‘물질의 구성’ 단원에 대하여 차시 수업의 목표로 사용 가능한 구체적인 성취 기준을 지식, 탐구, 태도 영역으로 개발하였다. 셋째, 앞에서 개발한 성취 수준의 도달 정도를 판별하는 평가 기준을 개발하고, 이 성취 수준과 평가 기준을 토대로 하여 동일 학급 내 과학 교과 수준별 분단 편성을 위한 진단 평가 도구들을 개발하였다. 넷째, 이 진단 평가 도구에 의하여 학생들을 수준별 소집단 분단으로 분류하는 구체적인 방법을 제시하였다. 마지막으로, 각 동일 학급 내 수준별 분단에 사용할 수 있는 보충 활동지를 개발하였으며 사용하는 방법을 제시하였다. The purpose of this study is to development of instructional strategy for differentiated curricular by enrichment and supplementary instructions about junior high school science under the 7th National Curricular Revision. The ability grouping for differentiated curricular in this study was within class ability grouping in same year. The composition of matter in junior high school science 3 was selected for this study and analyzed to develope the detailed achievement standards and assessment standards of school class level for science teacher. Based on those detailed achievement standards and assessment standards of school class level, the concept clusters of the composition of matter section was classified. The diagnostic evaluation which was the performance assessment tools according to the concept clusters was developed to classify for enrichment and supplementary groups. Also the method of classification about enrichment and supplementary groups in science class was suggested in details. Finally, teaching and learning materials for supplementary groups according to the concept clusters were developed about the composition of matter section in science 3 at junior high school.

Measurement of lysosomal enzyme activity in the rabbit eye was effected using acid phosphatase as enzyme markers. Acid phosphatase in the retinal pigment epithelium of the rabbit eye was studied using standard enzymological techniques. The enzyme activity in the lysosomal extract of the pigment epithelium was 54.9±23.0 King-Armstrong Unit/dl.
