        Safety Assessment of Chromium by Exposure from Cosmetic Products

        Low level impurities often reside in cosmetic products. The aim of the present study was to estimate the human exposure to chromium from cosmetic products purchased at a local market in South Korea, and to assess the risk on public health. Hexavalent chromium is an impurity substance that contaminates cosmetic products during manufacture. The potential for chromium to induce and elicit allergic contact dermatitis, as well as the degree of chromium exposure from cosmetic products, were assessed. Chromium exposure was estimated using the chromium concentrations found in cosmetic samples taken from the local market along with the expected user pattern data that was taken from the literature. Of the cosmetics we tested and available for purchase on the Korean market, seven had chromium contents above the detection limit of 0.1 ppm (0.1 μg/mL), ranging from 0.2 to 3.15 ppm. In risk assessment, scientifically defensible dose-response relationships must be established for the end points of concern. In the case of chromium contaminated cosmetic products, this includes conducting dose-response assessments for allergic contact dermatitis following dermal exposure. This dose-response information can then be integrated with site-specific exposure assessments to regulate consumer safety by use of these products. We found that dermal exposure to chromium concentrations ranging from 0.0002 to 0.003 μg/㎠ does not appear to cause concern for eliciting allergic contact dermatitis.

      • 아리스톨로크산 함유 생약제에 대한 안전성평가연구 : 3개월간 반복투여독성시험을 통한 신장독성평가

        본 실험에서는 아리스톨로크산을 함운하고 있는 생약제중 하나인 마두령(.4risfoforfioe JTurruf)의 90일 반복투여독성시헌을 실f,」하였다. 마두령에서 아리스톨로크산을 정량분석한 결과 마두령 단일 건조븐말의 경운 2.112mg/g, 잉상에서 처방되고 있는 마든령복방 (마두령외 9가지 생약제 복합처방) 건조분말꼭 경무에는 0.066m9/5으로 각각 검출되었다. 본 실험에서 용량설정은 마두령 처리군 (저용량 군; 21.3m9/k9/day (임상용량), 중용량군; 2t3mgfg/day, 고용량군; 2430mg딘g b.w./day), 마두령복방 처리군 (427mg/kg b.w./da?; 임상용량) 및 positive control (아리스톨로크산) 처리군 (저용량군;0.05mgag/da17, 중용량군; 0.Smg/kg/day, 고용량군; 5mg./kg/dal·1으료 하여 랫드에게 경구투여하였다. 마두령단방 처궈근 및 마두령복방 처리군에서 저용량군과 중응량군에서는 잉상관찰 및 생화학적 분석에서 패조군과 유의적인 차이가 없었다. 그러나 마두령단방 고용량근에서는 간, 신장의 상대적인 무계가 증가되었고, 신장의 유두이행상피증식 및 암종이 관찰되었으며, 위에서는 편평세포암종이 관찰되었다. 하자만 간손상이나 간암은 유발하지는 않는 것으로 판단되었다. 결론끌으로 본 실험의 결과에서는 아리스톨로크산 함유 생약제인 마두령이 임상용량인 저응량 처리군에서는 독성을 나타내지 않았으나, 고용쏭으로 장기 복용시켰을 깅우 설치류의 전위부위 및 신장에서 독성이 있음을 알 수 있었다. Chines herbs nephropathy (CHN) has been described in young women who had taken a slimming pills containing some chines herbs. Aristolochic acid (AA), suspected substance as the causal factor of CHN, is known a carcinogen. The Aristolochiae fructus (fruit of Aristolochia contorta) was used in Korean Traditional Medicine consists of appropriate amounts of mixed natural products. Subchronic toxicity of A. furctus containing aristolochic acid was investigated in SD rats. The body weight and clinical signs were observed after orally administration of A. futctus at doses of 21.2, 213, and 2130 mg/kg/day; mixture (including 9 other herbs) at dose of 427 mg/kg/day; aristolochic aicd (Ⅰ+Ⅱ) at doses of 0.05, 0.5, and 5 mg/kg/day for 3 months. At the end of the treatment, high dose treated animals revealed a deficit in final body weight about 25% compared with that of control. Organ weights of kidney, liver, testis, or ovary were increased with dose dependent manner. No changes considered to be due to the administration of A. furctus, mixture or aristolochic acid were in hematological and clinical study. However, significant changes at histopathological study of kidney and stomach were boserved in high-dose treated groups. In conclusion, for human health safety it needs appropriate regulatory actions regarding the use of natural herbal medicines known or suspected of containing aristolochic acid.

      • Nicotine 및 Tobacco-specific nitrosamine이 발암과정에 미치는 영향

        Nicotine은 그동안 계암발생파정에 관여되어 있는 것으로 추정되어지고 있으나 현재 이에 대한 기전.은 밝혀진 것이 거의 없다.본 연구에서는 폐암발생과정에서 있어서 nicotine의 역할을 조사하기 위해 첫번째 실험군으로 nicotine을 Sprague-Dawley 랫드에 연속적으로 10ㅇ리간 투여한 후 암유전자 및 암억제유전자의 발현변화를 단백질과 mRNA레벨에서 검토하엿다. 그 결과 5종류의 암유전자인 ras,raf,myc,fos, jun,및 2종류의 암억제유전자인 p53,Rb 단백질의 발현변화는 거의 없는것으로 나타났다.그리고 2종류의 암유전자인 myc,fos및 암억제유전자 p53 mRNA발현변화 역시 거의 없어 단백질 발현 실험결과와 일치하는 것으로 나타났다. 두번째 실험군으로 nicotine 및 sodium nitrite그리고 NNK를 Fischer344 랫드에 투여하여 발암물질인 NNK의 생성여부를 검토한 결과 NNK를 단독투여한 실험군의 경우 8-OHdG 레벨이 1.8배에서 2.3배까진 현저하게 증가하였으나 nicotine 및 sodium nitrite를 단독 혹은 병용 투여한 경우 8-OHdG 레벨의 유의한 차이가 관찰되지 않아 NNK 생성을 확인할수 없었다. Nicotine has been implicated as a potential factor in the pathogenesis of human lung cancer, however its mechanism of action in the development of lllng cancer remains largely unknown.To explore the role of Bicotine in the developmene of lung cancer, lue first investigated the effects of nic-otine on the expression of tumor associated genes by treating Spragve-Barley rats with nirotine (10 mf/kg) by gavage once daily for 10 days. We determined the expression of proteins and rnRNAs of the raf,raf, mrt JHn, foa oncogenes and p53, ab tumor suppressor genes by Western and Northern blotting,respectively. We did not detect any changes on t31e levels of prote·ins aBO mRNAs of these tumor associ-ated genes in the lung of Sprague-Dawley rats from 3 days to 12 ·weeks after the last treatment of nico-tine, indicating that nicotine appears to have no effect on the exE)ression of these oncogenes aBd turnersuppressor genes at an early stage in multistage chemical carcinoifenesis. In a second experiment, we in-vestigated the possibility that 4-(methylnitrosamino)- t- (3-p!rridyl)-1-butaaone (NNK) could beformed eBdogenously bf treating with nicotine and sodium nitrite. We treated groups of Fischer 344 ratswith nicetiBe (60 rrnol/kg) and sodium nitrite (180 rmol/kg), nicotiae, sodiu:n-nitrite and NNK (120 #mol/kg) flfne by gavage once daily to, 7 dafs, .espectively and dete,mined the 8-hydroxy-deoByguanosine (8-OHdG), as an indicator of NNK formation, in the lungs of rats 24 hours and 48hours after the last treatmeilt by HPLC/ECD method. We detect 3ncreased level of 8-OHdG in the tungsof rats treated with NNK, but in the case of nicotine plus sodiLlra nitrite, nicotine and sodium nitritealone we could not detected any changes of 8-OHdG, respectively.

        Association between Urinary Bisphenol A and Waist Circumference in Korean Adults

        Bisphenol A (BPA) is widely used in the production of polycarbonate plastics, epoxy resins, and food and beverage containers. In the present study, we aimed to investigate the relationship between urinary concentrations of BPA and waist circumference in Korean adults. A total of 1,030 Korean adults (mean age, 44.3 ± 14.6 years) were enrolled in the study on the integrated exposure to hazardous materials for safety control, conducted by the Ministry of Food and Drug Safety from 2010 to 2012. Abdominal obesity was defined as having a waist circumference of at least 90 cm and 85 cm for men and women, respectively. The participants were divided into 4 groups according to the urinary BPA concentration quartile. Waist circumference was significantly higher among subjects with a urinary BPA concentration in the highest quartile relative to those in the lowest quartile (p = 0.0071). Linear regression analysis revealed a significantpositive association between urinary BPA concentrations and body mass index, body fat, after adjusting for potential confounders. Moreover, subjects with urinary BPA concentrations in the fourth quartile were more likely to be obese compared to those with urinary BPA concentrations in the first quartile (odds ratio, 1.938; 95% CI: 1.314~2.857; p for trend = 0.0106). These findings provide evidence for a positive associationbetween urinary BPA concentration and waist circumference in Korean adults.

        아태지역 식품안전관리 협력과 역량강화를 위한 APEC의 활동

        Persistent accidents related to food safety and expanded international trades have urged the world to be aware of the gravity of the accidents. Accordingly many countries have tried to come up with various laws, regulations,measures, support networks and educational programs for the agenda, particularly focusing on harmonizing food safety technologies among nations and fostering professionals. Also, APEC newly organized Food Safety Cooperation Forum (FSCF) in 2007 to exert multi-dimensional efforts to improve food safety for Asia Pacific nations. Up to now, 35 activities have been promoted since 2007 and additional 20 activities are waiting for their turns for action on the list of APEC project agenda. FSCF has the objective that it helps the stakeholders in food supply chain develop their own competence in that area, thus increase international trade among nations and maintain the globe healthy by applying the highest standards and best practices for the management of food safety ranging from production to consumption. To achieve this strategic objective, APEC subsequently formed Partnership Training Institute Network (PTIN) to build up the multilateral networks of specialists from governmental agencies engaging in food safety management,industries, academia and international organizations in Asia-Pacific region. This attempt made it possible for the world to exchange their scientific and technological information concerning food safety and strengthen related education and training. Today, international cooperation is essential for food safety management. Therefore, we need to participate actively in the activities of APEC FSCF to contribute to improving food safety technologies for the member countries of APEC. We also need to connect the domestic support programs with theirs.
