        Stain improvement in the white button mushroom ‘Seolgang’ and its varietal characteristics in Agaricus bisporus

        The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, ‘Seolgang’ was developed by crossing two monokaryons ‘CM020913-27’ and ‘SSU423-31’. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of ‘Seolgang’ on CDA was better at 20℃ and 25℃ when it was compared with that of ‘505 Ho’. The mature cap shape of new strain ‘Seolgang’ is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain ‘505 Ho’, the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of ‘505 Ho’. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of ‘505 Ho’. Genetic analysis of the new strain ‘Seolgang’ showed different profiles compared to ‘505 Ho’, CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.

        전통주 주박의 항혈전 활성 평가

        전통주 주박을 이용한 고부가가치 식품소재 개발을 위해, 상업적 시설에서 생산된 3종 약주(J-B, J-S, J-Y) 및 2종 탁주(J-H, J-W) 주박의 ethanol 추출물 및 열수 추출물을 조제하고 이들의 혈액응고 저해활성, 혈소판 응집저해 활성 및 인간 적혈구 용혈활성을 평가하였다. 5종 주박의 pH는 3.90~4.29로 유사하였으나, brix는 5.0~27.0으로 다양하게 나타났으며, 수분 및 알코올 함량에서도 시료에 따라 1.8배의 차이를 나타내었다. 주박의 색차와 성분은 첨가된 부재료 및 사용누룩에 좌우되었으며, J-W 주박의 경우 수분함량이 80.3%, brix 13, 알코올 함량 1.8%를 함유하여 다른 주박에 비해 다양한 식품제조에 용이하게 이용 가능하리라 판단되었다. Ethanol 추출효율은 J-H, J-W, J-B, J-S, J-Y의 순, 열수 추출효율은 J-S, J-B, J-W, J-H, J-Y의 순으로 높았으며, 총폴리페놀 및 총플라보노이드함량은 ethanol 추출물 중에서는 J-H, 열수 추출물 중에서는 J-Y 주박에서 가장 높았다. 5종 주박의 10종 추출물은 모두 5 mg/ml 농도까지 인간 적혈구에 대한 용혈활성이 나타나지 않았으며, J-B, J-S, J-Y의 약주 주박의 ethanol 추출물에서 유의적인 혈액응고저해 활성이 나타났으며, J-W 탁주 주박의 열수 추출물에서 thrombin 저해 활성과 J-B, J-S 및 J-H 주박 열수 추출물에서 혈액 응고인자 저해활성을 확인하였다. 혈소판 응집저해 활성평가의 경우 J-W 탁주 주박의 ethanol 및 열수 추출물에서만 아스피린에 필적하는 우수한 활성이 확인되었다. 본 연구결과는 다양한 약주 및 탁주 주박이 항혈전 활성을 가지고 있으며, 주박으로부터 항혈전제 개발이 가능함을 제시하고 있다. In this study, ethanol and hot water extracts of lees from Korean traditional wine (J-B, J-S, J-Y, J-H, and J-W) were prepared, and their effects on blood coagulation, platelet aggregation, and hemolysis of human red blood cells (hRBCs) were investigated to develop functional food ingredients from lees. The pH and brix of the lees ranged from 3.90 to 4.29 and 5.0 to 27.0o, respectively, and there was a huge difference in the water and ethanol content among the lees. The nuruk and additives used affected the color and physicochemical properties of lees. The J-W takju made from only rice and traditional nuruk, which has 13o brix and 1.8% of alcohol, has potential as functional food ingredient. With regard to the extraction yields of lees, higher yields were obtained from J-H, which contains different medicinal plants, in ethanol, followed by J-W, J-B, J-S, and J-Y. Higher extraction yields of lees were obtained from J-S in hot water, followed by J-B, J-W, J-H, and J-Y, respectively. The ethanol extract of J-H and the hot water extract of J-Y had the highest contents of total polyphenol and total flavonoids among the lees extracts. The 10 lees extracts did not show hemolysis activity against hRBCs up to 5 mg/ml. In an anticoagulation activity assay, the ethanol extracts of three yakju lees (J-B, J-S, and J-Y) and the hot water extract of J-W inhibited thrombin activity, whereas the hot water extract of J-B, J-S, and J-H inhibited blood coagulation factors. In an antiplatelet aggregation activity assay, only the J-W takju lees showed significant inhibition activity. Our results suggest that lees from traditional wine had high potential as a novel antithrombosis agent.

      • Hepatoprotective and Antioxidative Activities of <i>Cornus officinalis</i> against Acetaminophen-Induced Hepatotoxicity in Mice

        <P>The fruit of <I>Cornus officinalis </I>Sieb. et Zucc. is commonly prescribed in Asian countries as a tonic formula. In this study, the hepatoprotective effect of ethanolic extracts of the fruit of <I>C. officinalis</I> (ECO) was investigated in a mouse model of acetaminophen- (APAP-) induced liver injury. Pretreatment of mice with ECO (100, 250, and 500 mg/kg for 7 days) significantly prevented the APAP (200 mg/kg) induced hepatic damage as indicated by the serum marker enzymes (AST, ALT, and LDH). Parallel to these changes, ECO treatment also prevented APAP-induced oxidative stress in the mice liver by inhibiting lipid peroxidation (MDA) and restoring the levels of antioxidant enzymes (SOD, CAT, and HO-1) and glutathione. Liver injury and collagen accumulation were assessed using histological studies by hematoxylin and eosin staining. Our results indicate that ECO can prevent hepatic injuries associated with APAP-induced hepatotoxicity by preventing or alleviating oxidative stress.</P>

        KAERI 부지 주변의 환경선량 측정을 위한 온라인 감마선량 감시시스템

        환경감마선량 변동을 감시하기 위해 한국원자력연구소 부지 주변에 온라인 감마선량 감시망을 구성하였다. 모니터링 포스트에 설치된 전리함 검출기에 의해 측정된 자료는 무선전송방식으로 중앙통제소에 전송되어 실시간으로 감시된다. 무선전송 방식은 자료의 전송, 처리 및 자자에 신뢰도가 높고 경제적이었다. 감시시스템은 급격한 선량증가시 조기경보체제와 연결할 수 있게 하였다. On-line gamma monitoring system around KSERI-site was set up to monitor the radiation fluctuations in environment. Data on gamma exposure rates measured by the ionication chamber in the monitoring posts are transmitted to a computer of central control station with radio telemetry transmission and reliable on handing and storing of data. This monitroing system can triger an early warning system in the event of abnormal radiation levels.

      • 矮化劑 處理가 바나나의 生育 및 收量에 미치는 影響

        바나나에 알맞은 矮化劑를 選拔하고자 몇종의 矮化劑를 Dwarf Cavendish에 처리하였던 결과를 요약하면 다음과 같다. 1. 주당 80mg 이하의 paclobutrazol 土壤灌注 처리는 草長, 莖葉特性, 果實收量, 果實特性 등에 영향을 주지 않았다. 200mg 이상 처리구에서는 草長과 假莖長이 단축되는 경향을 보였고 果梗長은 유의성 있게 단축되었으나 다른 莖葉特性이나 果實收量, 果實特性은 무처리구와 큰 차이가 없었다. 2. 주당 20mg의 ancymidol 土壤灌注에 의해서 草長과 假莖長이 단축되는 경향이나 다른 莖葉特性이나 果實收量, 果實特性은 무처리구와 큰 차이가 없었다. 3. 5000ppm의 chlormequat 수용액 2회 葉面撤布區에서는 草長, 莖葉特性, 果實收量, 果實特性 등이 무처리구와 비슷하였다. 4. 草長과 假莖을 현저히 단축시키는 藥量의 矮化劑를 처리할 때 果梗이 假莖에서 빠져나오지 못할 것으로 보여 바나나에 있어서 矮化劑의 실용성은 없을 것 같다. This study was conducted to select growth retardants for bananas grown in a plastic greenhouse. The various rates of paclobutrazol(40, 80, 200 and 400mg/plant) and 20mg/plant of ancymidol were soil drenched and 5000ppm solution of chlormequat was foliarly sprayed twice. Application of less than 80mg of paclobutrazol did not affect plant height, pseudostem and leaf characteristics fruit yield and fruit characteristics. Application of 200 or 400mg of paclobutrazol tended to reduce plant height and pseudostem length and significantly shortened fruit stalk but did not affect the other characteristics. Application of 20mg of ancymidol tended to reduce plant height and pseudostem length but did not influence the other characteristics. Foliar spray of 5000ppm solution of chlormequat did not influence any measured plant traits. Application of growth retardants to bananas grown in a plastic greenhouse may not be beneficial because growth retardant rates to reduce plant height and pseudostem length significantly appear to prevent fruit stalks from fully emerging.

      • Fluoxetine이 Schedule-Induced Polydipsia가 유발된 백서 뇌에서 Tyrosine Hydroxylase 발현에 미치는 영향

        연구목적: Fluoxetine은 serotonin을 매개하여 간접적으로 dopamine 신경전달기능을 억제한다고 추정되고 있다. 또한 운동장애에서 운동기능의 악화를 유발한다고 알려져 있다. 그러나 신경세포체에서 fluoxetine이 dopamine에 어떠한 영향을 주는지는 아직까지 확실치 않다. 저자들은 schedule-induced polydipsia를 유발시킨 백서 뇌의 흑질, 복부피개영역, 미상핵에서 tyrosine hydroxylase(TH) 발현이 저하됨을 발견하였다. 이를 통해서 fluoxetine이 백서 뇌의 dopamine 기능에 긍정적인지 혹은 부정적인지를 규명하고자 하였다. 방법: 4주간의 schedule-induced polydipsia 과정을 거친 백서에서 면역죄치화학적인 방법으로 흑질, 복부피개영역, 미상핵의 tyrosine hydroxylase 발현이 저하됨을 확인한 후, 실험동물들에게 fluoxetine 10mg/kg를 3주간 복강내 주사하였다. 실험백서들을 희생시켜 뇌 조직을 적출하여, TH 면역조직화학 염색법을 이용하여 흑질, 복부피개영역, 그리고 미상핵의 TH 면역반응세포를 관찰하고 이를 정상백서와 비교하였다. 결과: 1) 다갈증이 유발된 백서의 흑질, 복부피개영역, 미상핵에서 tyrosine hydroxylase 발현이 정상백서 보다 저하됨을 관찰하였다. 2) 3주간에 걸친 fluoxetine 투여후 흑질, 복부피개영역, 미상핵의 tyrosin hydroxylase 발현이 다시 증가하는 소견을 보였다. 결론: Fluoxetine 만성투여가 흑질, 복부피개영역 그리고 미상핵의 tyrosin hydroxylase를 증가시키는 소견을 얻었다. 이러한 결과는 임상에서 dopamine 결핍과 연관된 질환들에서 fluoxetine을 만성투여하면 운동기능을 포함한 증상들의 개선을 가져올 수도 있다고 추정된다. Objective: It has been suggested that fluoxetine inhibits the dopaminergic neurotransmission by serotonergic mediation. And also, it has been shown to inhibit synthesis of DOPA in dopamine-rich areas of the rat forebrain. These dopamine-antagonistic capacity of fluoxetine is only supported by anecdotal report that the increased amount of motor disability in patients with idiopathic Parkinson's disease after exposure to fluoxetine. However, there is still no evidence of the direct effect of fluoxetine on dopaminergic neuronal cell body in the substantia nigra, VTA, caudate & putamen. This study was designed to evaluate the effects of fluoxetine in rat brain which showed decreased numbers of dopaminergic neuronal cell body induced by schedule-induced polydipsia(SIP). Method: We incidentally found that 4 weeks of schedule-induced polydipsic rats revealed the suppression of tyrosine hydroxylase expression in the substantia nigra, VTA, caudate & putamen with the immunohistochemistric measures. After 3 weeks of intraperitoneal injection of 10mg/kg of fluoxetine to the schedule induced polydipsic rats, the tyrosine hydroxylase expression was also measured with immunohistochemistry. We compared the tyrosine hydroxylase expression among the normal control, the polydipsic rats, and the rats with fluoxetine treatment. Results: 1) By contrast with the control, the polydipsic rats revealed the evidence of decreased tyrosine hydroxylase expression in the substantia nigra, VTA, caudate & putamen. 2)After daily injection of fluoxetine for 3 weeks, the polydipsic rats showed increment of tyrosine hydroxyase expression in those areas. Conclusions: In previous studies, a great deal of results suggest that fluoxetine negatively influence the dopaminergic systems indirectly via serotonergic activation such as inhibition of dopamine synthesis or transport system. Although our results are obtained from rodents, we suggest that fluoxetine directly and positively enhance the dopamine system in the substantia nigra, VTA, caudate & putamen. The chronic adminstration of fluoxetine may be helpful to dopamine-depleted condition in clinical situations. We anticipate the replication studies of our findings and well-controlled clinical trial.

        충남대학교 학술림 산책로 훼손실태 및 관리방안에 관한 연구

        This study concentrates an the trail of the CNU(Chungnam National University) forest. The amount of use and the pattern of damage are considered, as well as the extent of environmental damage. Finally, some managerial strategies are proposed in connection with sustainable maintenance. The trail of the university forest is composed of three sections; one is the neighborhood park and the Institute of Information Technology/Communications, the second is the Institute of Information Technology/Communications and the Dormitory, and the third section is the Dormitory and the Foreign prof's Apt. The total length of the trail is 2.6km. The daily amount of use is 1,269 visitors per day. The amount of using increased three times the amount of that in 1998. This study shows the pattern of damage of the trail in the order of root-exposure, widening, rock-exposure, deepening and the divergence in the frequency of occurrence of all these factors. Environmental damage is also graded thus: grade 3 is 27.4m, grade 4 is 0.8m, grade 5 is 1.5m, and grade 6 is 0.2m. Accordingly, some managerial strategies were proposed in connection with the sustainable trail maintenance.

        Sarcomatoid Carcinoma Arising from Mature Cystic Teratoma

        Malignant transformation of mature cystic teratoma (MCT) is rare. Sarcomatoid carcinoma is a neoplasm comprising malignant mesenchymal cells and a conventional carcinomatous area. Here, we report on a case of sarcomatoid carcinoma arising from an MCT in the left ovary of a 45-year-old female. A unilocular cyst consistent with MCT was observed; however, a nodule within the cyst was confirmed from the resected ovary. Microscopically, the nodule showed both squamous cell carcinoma and pleomorphic sarcomatous components admixing with each other. Lining epithelial cells at the periphery of the main tumor showed squamous metaplasia. When a sarcomatous component is observed in the ovary tumor, it is important to find a squamous cell component, either benign or malignant.

      • 봉독약침이 류마티스 관절염 환자의 관절 통증, 종창 및 급성 염증 반응에 미치는 영향

        Objective In order to study the effects of bee venom(BV) on the pain, edema, and acute inflammatory reactant of rheumatoid arthritis(RA) patients. Methods Patients with RA who met the ACR(American College of Rheumatology) 1987 revised criteria for the diagnosis of RA were treated with the BV therapy twice a week for 3 months. Tender Joint counts, swollen joint counts, Visual analog scale(VAS), morning stiffness, ESR. C-reactive protein(CRP) were analyzed before and after BV therapy. Results The results as follows. 1. Tender joint counts in patients after BV therapy were significantly lower than those before BV therapy(9.0±7.9 vs 15± 11.4, p=0.002). 2. Swollen joint counts of the patients after BV therapy were significantly lower than those before BV therapy(50±61 vs 15±23, p=0.001). 3. VAS in patients after BV therapy was significantly lower than those before BV therapy(608± 17.6 vs 380± 159, p=0.000). 4. Duration of morning stiffness in patients after BV therapy was significantly reduced compared with that before BV therapy(119.1± 112.6 min vs 59.0±89.7 min, p=0.009). 5. ESR and CRP were not significantly changed before and after BV therapy, suggesting BV itself could make inflammatory reaction as well as therapeutic effect. Conclusions BV therapy improved tender joint counts, swollen joint counts and duration of morning stiffness in this study, and further study is needed on long-term effect of BV therapy.
