        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        수술환자의 수술장 내 대기시간 단축방안

        문제: 환자가 수술장내에서 수술을 기다리면서 느끼는 불안감을 최소화할수있도록해야하지만수술실의 효율성 및 의료진 편의성 위주로 운영되고 있어 수술실 내 대기시간이 연장되고 있다. 목적: 수술장내 대기시간을 단축시키기 위해 수술환자의 이동경로에 따른 지연요인과 문제점을파악하고 개선하여 환자가 수술을기다리면서 느끼는 불안감을 최소화하고자 한다. 의료기관: 부산시에 소재한 481병상의 종합병원 수술실 질 향상 활동: 수술환자의 대기시간 지연요인과 문제점을 파악하고 개선을 통해 질 향상을 도모하였다. 개선효과: 대기시간 수행율에서 수술장 도착까지의 수행율이 개선전 95%에서 개선후98%로, 수술방입실까지의 수행율이 개선전 88%에서 개선후 94%로, 마취시작 까지의 수행율이 개선전 93%에서 개선후 96%로 수행율이높아졌다.

        부산·울산·경남지역 직업병 감시체계

        Objective: Occupational medicine specialists in the Busan, Ulsan and Kyung-Nam areas established an area-based occupational disease surveillance system and used this system to collected case information for the purpose of preventing occupational diseases Methods: l l hospital participated in this system. The authors selected five main diseases, which were hand-arm vibration syndrome (HAVS), work-related musculoskeletal disorder (WRMSD), occupational asthma, occupational skin disease and occupational and occupational lung cancer and established their case definitions. All cases were reported on the web, and real time analysis was conducted. Results: Between April 2001 and April 2003, 192 cases of HAVS, 118 cases of WRMSD, 33 cases of occupational asthma, 17 cases of occupational skin disease and 17 cases of occupational lung cancer (for a total of 377 cases of these five main diseases) were reported. most of the HAVS cases came from shipbuilding, and 172 of them (92.7%) were associated with grinding. Of the four main types of WRMSD, carpal tunnel syndrome (CTS) was the most prevalent with 46 cases and shipbuilding was also the main industry involved (83.9%). The ergonomic risk factors involved mainly associated with the hand. In 19 (57.6%) and 4 (12.1%) cases of occupational asthma, the agents involved. The causative agents of occupational lung cancer included Cr, welding fume, PAH, Ni, etc. 10(58.8%) of the cases were diagnosed as squamous cell carcinoma and 5(29.4%) as adenoma. Conclusion: This result showed that an area-based occupational disease surveillance system might provide an effective method of evaluating the prevalence of such diseases, however the Busan, Ulsan and Kyung-Nam provinces are too large to be treated as individual areas. Therefore, the authors suggest that each province should establish its own surveillance system.

        The measurement of unsteady flow rate is importance to clarify and improve the dynamic characteristics in pipeline, hydraulic components and systems. There is also demand for a real time flow at the sensor in order to measure unsteady flow rate with high accuray and fast response to realize feedback control of flow rate in fluid power systems therefore We propose an approch for estimating unsteady flow rate through a pipeline and dynamic characteristics of pulsting flow rate components under high pressure condition. Unsteady flow rate is estimated by using semiconductor type pressure transducers and the measured pressure values at two distant points along the pipeline.

        유압관로의 주파수변화 따른 압력전파특성

        In this paper, an oil hydraulic pipeline is terminated by both rotary sinusoidal flow generator at upstream and oriffice at down stream. The pulsating pressure wave form, generated by the rotary sinusoidal flow generator, is measured by pressure sensor. In the analysis of this paper, a component of the fundamental frequency is obtained by using Laplace transformation. The experimental and analytical results make clear that (1) viscosity is significant role in hydraulic pipe. (2) When pulsating frequency is matched with the natural frequency, resonance frequency occurred periodically. According to the study proposed here, dynamic pressure in a circular oil pipe is expressed in propagation of pressure wave with respect to frequency and Bessel function. The resonance frequency is 108Hz at oil temperature 20℃ in this study. The abrupt change of gain value id due to effect of resonance frequency. The results of experiment are compared with the calculated results, and agreement of both results is fairly good.

        플륨 모니터링에 의한 SM45C 레이저 용접특성 평가

        With the increased use of lasers in industrial welding applications, techniques for monitoring and controlling these processes become increasingly important. It is very important that we understand the dynamic behaviors of the laser induced plume in welding because the laser induced plume has considerable effects on welding efficiency and the quality of materials. As the plume fluctuation was associated with keyhole instability, unstable vapor plume indicated the process was unstable and would result in poor welds. An Infrared Themal-vision Camera can be utilized compensate for incurracies encountered in real-time monitoring during laser welding. We have results that instabilities of plume are closely dated with hot cracking and defect of laser welding.

        Poly(p-phenylenesulfide)와 poly(p-phenylene)으로부터 제조된 탄소를 리튬이온 2차전지용 음극재료로 사용하여 전기화학적특성을 연구하였다. 이들 고분자들을 질소 분위기하에서 승온속도 2℃/min로 1000℃까지 상승시킨 후, 1시간 동안 탄화시켜 탄소를 제조하였다. Poly(p-phenylene)으로부터 제조된 탄소가 보다 더 규칙적인 구조를 가지기 때문에 Poly(p-phenylenesulfide)으로부터 제조된 탄소보다 더 많은 용량과 충방전 효율을 나타내었다. 수산화리튬과 염화주석(Ⅱ)을 첨가하여 용량의 증가와 충방전 효율의 증가를 가져올 수 있었다. 이중에서 poly(p-phenylene)에 염화주석(Ⅱ)을 첨가시켜 제조된 탄소가 가장 큰 충방전 용량과 충방전 효율의 향상을 가져왔다. Carbon inaterials manufactured from poly(p-phenylene sulfide) and poly(p-phenylene), were studied on electrochemical characteristics as anode materials for lithium ion secondary battery. These polymer precursors were heat treated for 1hr at 1000℃ with the rate 2℃/min under nitrogen atmosphere. Carbon manufactured from poly(p-phenylene) showed higher capacity and coulomb efficiency of charge/discharge than carbon from poly(p-phenylene sulfide) because the former has better ordered structure. Carbon manufactured from polymer precursors adding stannous chloride or lithium hydroxide showed higher capacity and better efficiency of charge/discharge. Also, carbon manufactured by adding stannous chloride to poly(p-phenylene) showed the highest capacity and efficiency of charge/discharge.

        In this paper, we attempt to help alleviate this difficulty by pointing out that an encompassing perspective on Taquchi's Philosphy can be provided by statistical decision analysis. The subject deals with decision making in the face of partial or no imformation, and prescribes that an optimum decision is one that maximizes expected utility. The role of experimental design is to obtain partial information about the unknown quanties in an efficient manner. The objective of this paper was to argue that it is design by decision analysis rather than design by design that better encapsulates Taguchi's approach to offline quality control.
