        Toxicity of the recombinant human hyaluronidase ALT-BC4 on embryonic development

        Cumulus-oocyte complexes (COCs), which contain immature oocytes, are matured in vitro for in vitro embryo production. Oocyte and cumulus cells are then separated using hyaluronidase. To date, there have only been a few reported cases of the toxic effects of hyaluronidase on porcine oocytes. The aim of this study was to compare the effects of bovine testis-derived hyaluronidase and recombinant human hyaluronidase on oocyte denudation and quality. Porcine COCs were matured for 44 h and denuded using different hyaluronidase concentrations and exposure times. Then, oocytes were activated by electrical parthenogenesis. In experiment 1, COCs were denuded using bovine-derived, ovine-derived (Hirax), and human recombinant (ALT-BC4) hyaluronidases for 10 and 20 min. In experiment 2, bovine-derived and human recombinant (ALT-BC4 and ICSI Cumulase<sup>®</sup>) hyaluronidases were used to denude the COCs for 2 and 20 min. In both experiments the oocytes were all completely denuded, and there was no degeneration. Rate of embryo development was significantly increased in group treated ALT-BC4 for 2 min and not significantly different in other treatment groups. In general it slightly decreased with longer exposure times. These results have confirmed that different sources of hyaluronidase do not have detrimental effects on the quality of porcine oocytes and suggest that the human recombinant hyaluronidase ALT-BC4 is suitable for oocyte denudation with an increased blastocyst rate.

      • 록인 증폭기에 의한 L(C,R) 회로 공진점 결정의 한 방법

        L(C,R)회로의 공진점을 결정하기 위해 록인 증폭기에 의한 sequential spectrum analysis(SSA)법을 적용해 보았다. 시료는 변압기였으며, 단일 소인방식과 왕복 소인방식을 비교해 보았다. 또한 실험 결과의 신뢰도를 확인해보기 위해 실험결과를 변압기의 제작사 자료와 Q 미터(HP,4277A USA)에 의한 측정 결과와 비교하였다. 제작사 자료에 의하면 시료의 공진점은 두 권선의 각각은 5kHz(10kHz/2)였으나, 왕복소인에 의한 측정에서 시료의 공진점은 5.1kHz, 실험 결과로부터 계산된 Q 값은 4.3으로 주어졌고, 단일 소인법에 의해서는 공진점이 5.78KHz로 주어졌으며, Q 미터에 의한 Q 값은 4.9(9.8/2)였다. 실험 결과들과 기준 Q 미터에 의한 측정결과를 비교해본 결과, 양방향 소인법이 단일 방향소인법 보다 더 신뢰할 수 있음을 알 수 있었으며, SSA법에 의한 측정이 기존의 상용 전문기기에 의한 측정에 못지 않음과 전문기기의 측정 하한 문제를 극복하는 좋은 대안이 될 수 있음을 알 수 있었다. An accurate method named the sequential spectrum analysis(SSA) using a lock-in amplifier(LIA) of the heterodyne type was investigated to determine the resonance frequency of a LC(C,R) circuit : A heavy winding transformer was used as an inductor in the circuit. In this experiment, two types of the sweep method-one way and round trip method were examined. The resonance frequency of each method were compared with the manufacturer's data and those by the reference Q meter(HP,4277A, USA)for the experimental confidence: Measured resonance frequency by the round-trip method was 5.1 kHz, Q value at this frequency was about 4.3, while those by the single-sweep method were given by 5.78 kHz for the resonance frequency, 2.3 for the Q. On the other hand, the resonance frequency of manufacturer's data was 5 kHz and Q value by the reference Q meter was 4.9(9.8/2)at that frequency. From the comparison, it might be concluded that the SSA by the round-trip method was more reliable and accurate than the one-way method and relatively inexpensive, good alternative with respect to the professional instrument.

        수중의 Pyrene, Chrysene 및 Benzo[a]pyrene의 광분해(I)

        The photodegradation of pyrene, chrysene and benzo[a]pyrene, that were similar in structure among polycyclic aromatic hydrocarbons (PAHs), were investigated in water irradiated with a low-pressure mercury lamp (wavelength of 253.7㎚ and UV output of 1.35×10^-3J/s). The effects of several factors (t-BuOH, HCO_3^- and pH) on photodegradation of above three PAHs were also examined. The photodegradation rates of PAHs decreased with increasing the concentration of t-BuOH, but decreased little with increasing the concentration of HCO_3^- under the concentrations used in this study. The photodegradation rates of PAHs decreased with increasing pH, but their change were greater in case of pH increase from acid to neutral and were little in case of pH increase from neutral to base. The photogradation rates of PAHs fitted a first-order kinetic model and their photogradation rates decreased in the following sequences: pyrene>chrysene>benzo[a]pyrene among the PAHs used.

        상아질 접착제의 용매 증발이 접착 효율에 미치는 영향

        연구목적: 본 연구는 상아질 접착제의 잔류 용매가 접착 효율에 미치는 영향을 평가하고자 하였다. 연구 재료 및 방법: 본 연구에서는 5세대 2단계 산부식형 접착제와 7세대 단일과정 자가부식형 접착제를 사용하였다. 상아질 접착제의 증발률과 전환률, 적용 후 용매의 공기건조 방법에 따른 미세인장결합강도를 측정하였으며 접착 계면을 FE-SEM을 이용하여 관찰하였다. 결과: 1. 시간에 따라 접착제의 증발률은 증가하나 접착제의 종류에 따라 증가 양상이 서로 달랐다. 2. 대부분 전환률은 증발률에 비례하는 경향을 나타내었다. 3. 공기건조 방법에 따라 결합강도는 대부분 under군, control군, over군 순으로 증가하는 양상을 나타내었다. 4. FE-SEM에서 아세톤을 용매로 사용하는 접착제는 공기건조 방법에 따라 droplet이나 gap이 관찰되었다. 결론: 상아질 접착제의 잔류 용매는 접착 효율에 부정적인 영향을 미치기 때문에, 상아질 접착제의 선택과 사용 시 용매의 종류와 특징에 대한 적절한 이해가 필요할 것이다. Objectives: The purpose of this study is to evaluate bonding efficacy by means of measuring the effect of remained solvent on Degree of conversion(DC) and μTBS and FE-SEM examination. Materials and Methods: Two 2-step total etching adhesives and two single-step self etching adhesives were used in this study. First, volume weight loss of 4 dentin adhesives were measured using weighting machine in process of time in normal conditions and calculate degree of evaporation (DE). Reaction/reference intensity ratio were measured using micro-Raman spectroscopy and calculate DC according to DE. Then 2 experimental groups were prepared according to air-drying methods (under, over) and control group was prepared to manufacturer's instruction. Total 12 groups were evaluated by means of micro tensile bond strength and FE-SEM examination. Results: Degree of evaporation (DE) was increased as time elapsed but different features were observed according to the kind of solvents. Acetone based adhesive showed higher DE than ethanol and butanol based adhesive. Degree of conversion (DC) was increased according to DE except for S³ bond. In μTBS evaluation, bond strength was increased by additional air-drying. Large gaps and droplets were observed in acetone based adhesives by FE-SEM pictures. Conclusions: Additional air-drying is recommended for single-step self etching adhesive but careful consideration is required for 2-step total etching adhesive because of oxygen inhibition layer. Evaporation method is carefully chose and applied according to the solvent type.

        측두하악장애 환자 중 하악과두의 퇴행성 골 변화를 보이는 환자의 분포 및 임상적 특징

        측두하악장애 환자 중 하악과두의 퇴행성 골 변화를 보이는 환자의 분포와 임상적 특징을 규명하고자 측두하악장애로 진단받은 6,070명의 환자들을 대상으로 연구를 시행하였다. 환자들의 초진 시 파노라마와 TMJ 파노라마 사진을 관찰하여 하악과두의 퇴행성 골 변화 유무를 기준으로 환자들을 퇴행성 골 변화를 보이는 군(DJD군)과 골 변화를 보이지 않는 군(non-DJD군)으로 나누고, 환자들의 초진 의무기록을 바탕으로 두 군 간의 특징을 비교, 분석하였으며, 각종 변수들이 DJD 발병에 미치는 상대적 위험성을 측정하기 위하여 로지스틱 회귀분석을 시행하였다. 연구 결과 전체 환자 중 DJD군에 속한 환자는 31.7%였고, non-DJD군에 속하는 환자는 68.3%였다. DJD군은 성별에 따라 다른 분포를 보여 남자보다 여자의 이환율이 높았고, 10대와 20대에서 가장 높은 분포를 보였다. 환자의 내원 주소 중에서는 전치부 개방교합, 턱의 후퇴감, 안모 비대칭, 개구장애를 주소로 내원한 환자가 다른 주소로 내원한 환자들에 비해 DJD를 보일 위험성이 높았으며, 교정치료 경력이나 중심위-중심교합위 변위 그리고 염발음을 보이는 환자도 DJD군에 속할 위험성이 큰 것으로 나타났다. 따라서 측두하악장애를 가진 환자 중 30세 이하의 여성이 위와 같은 임상적 특징을 보이는 경우, 교정치료를 계획할 때 DJD에 이환 되었을 가능성에 대하여 충분히 주의를 기울여야 할 것으로 생각된다. (대치교정지 2006;36(6):402-11) Objective: This study was carried out to evaluate the distribution and clinical features of patients with degenerative change of the mandibular condyle. Methods: Six thousand and seventy patients with TMD (temporomandibular disorder) were selected for this study, who had complete initial clinical records and radiographs. Panorama and TMJ panorama radiographs were used to screen the degenerative change in the condyle, and the patients were divided into DJD (degenerative joint disease) and non-DJD groups. Results: The distribution and clinical features of the two groups were compared. Out of the total number of patients, 31.7% were in the DJD group, and 68.3% were in the non-DJD group. The portion of females was larger in the DJD group (80.8%) than in the non-DJD group (67.5%), and the DJD group had high prevalence in the second and third decades. Lack of incisal contact, retrusive chin, facial asymmetry, and mouth opening limitation were the chief complaints of the patients who had positive relation to DJD. Conclusion: Patients with an orthodontic treatment history, CO-CR discrepancy and crepitation were at possible risk of having DJD. (Korean J Orthod 2006;36(6):402-11)

        상아질 접착제의 미세누출에 관한 연구

        본 연구는 여러가지 상아질 접착제들의 법랑질 변연과 상아질 변연에서의 미세누출을 비교 평가하기 위해 이루어졌다. 발거된 건전한 영구치의 협면과 설명에 5급 와동을 형성하고 형성된 50개의 와동을 무작위로 5개의 군으로 나누어 각군에 Adper™ Scotchbond™ Multi-purpose Plus Adhesive (SM; 3M ESPE. USA), Adper™ Single bond 2 (SB; 3M ESPE, USA), Clearfil™ SE Bond (SE; Kuraray Medical Inc., Japan), Adper™ Prompt™ L-Pop™ (PL; 3M ESPE, USA), G-Bond™ (GB; GC Co., Japan)를 적용하고 Filtec™ Z350 A3 (3M ESPE. USA)로 충전하였다. 시편을 37℃ 증류수에 24시간 보관한 후 5℃와 55℃에서 10초씩 1000회 열순환하고 2% methylene blue 용액을 24시간 넣어 염색한 뒤, 주수 하에 치아를 협설로 절단하여 색소의 침투 정도를 화상분석현미경을 이용하여 침투깊이를 측정한 뒤 각 군간의 미세누출 정도를 비교하여 침투깊이에 따라 0~3점으로 분류하여 다음과 같은 결과를 얻었다. 1. 범랑질 변연에서의 미세누출은 PL(0.85), SB(0.55), GB(0.50), SM(0.35), SE(0.25) 순이었으며 PL은 SM 및 SE와 통계학적으로 유의한 차이를 나타냈다(p<0.05). 2. 상아질 변연에서의 미세누출은 GB(2.10), SE(1.45), PL(1.40), SB(1.05), SM(0.70) 순이었으며 GB는 SB 및 SM과 통계학적으로 유의한 차이를 나타냈다(p<0.05). 3. 각 실험재료 군내에서 법랑질 변연에 비해 상아질 변연에서 미세누출이 더 크게 나타났으며, SE, PL, GB에서 통계학적으로 유의한 차이를 나타냈다(p<0.05). The objective of this study was to compare the microleakage of five adhesive systems in the enamel and dentin of permanent teeth. Class V cavity preparations with occlusal margins in enamel and gingival margins in dentin were prepared on both buccal and lingual surfaces of 25 extracted human molar teeth. The tested adhesives were: Adper Scotchbond Multi-purpose Plus Adhesive (SM), Adper Single bond 2 (SB), Clearfil SE Bond (SE), Adper Prompt L-Pop (pL) and G-Bond (GB). The results were as follows: 1. At the enamel margins, PL showed the highest leakage value(0.85), and others showed values of SB(0.55), GB(O.50), SM(O.35) and SE(0.25) in decreasing order. There were statistically significant differences in PL vs. SM and PL vs. SE(p<O.05). 2. At the dentin margins, GB showed the highest leakage value(2.10), and others showed values of SE(1.45), PL(1.40), SB(1.05), SM(0.70) in decreasing order. There were statistically significant differences in GB vs. SB and GB vs. SM(p<O.05). 3. Dentin margins showed high dye penetration rate than enamel margins in all material tested groups and there were statistically significant differences for SE, PL and GB.

        기억력 저하를 호소하는 노인들을 대상으로 INM176를 3개월간 임상시험한 후의 치료성적 및 부작용 평가 보고

        Objectives : We examined the effects of INM 176 (K-l107) compared with placebo on the cognitive functions of 92 old aged subjects with cognitive impairment. Methods : This was a prospective, 12 week, double-blind, Placebo-controlled clinical trial. The elderly who achieved a score of less than 25 points on the K-MMSE or showed a high risk of Alzheimer's disease from the 7-Minute Neurocognitive Screening Battery were considered to have objective impairment and were selected as subjects for this study. The subjects were randomized to placebo or INM 176 group. The outcome measures were from the Alzheimer's Disease Assessment Scale-cognitive subscale (ADAS-Cog), the Instrumental Activities ofDaily Living (IADL) and the Korean Geriatric Depression Scale (KGDS) and two kinds of computerized priming tests. After setting the total error score in the Alzheimer's Disease Assessment Scale-Cognitive section (ADAS-cog) as the repeated measurement factor, an analysis of variance of the combined factor design was done between the placebo and INM 176 group. Results : The interaction effect of time (pre- and post-trial) and group (placebo and INM 176 group) was significant in the analysis of the ADAS-cog's total error score. The INM 176 group's total error score in the ADAS-cog decreased significantly (p<0.01), whereas the placebo group showed a slight increase. The mean changes in IADL and GDS from baseline scores favored in the INM 176 group than in the placebo group. Outcome changes ofADAS-cog, IADL, KGDS scores during the 12 week clinical trail ofINM 176 and placebo demonstrated favorable responses in the INM176 administered group. Conclusions : This is a preliminary clinical trial result of INM176 as a memory pill. Based on these results, INM176 may be a candidate molecule for the improvement of cognitive functions, including memory, Further clinical trial should demonstrate its efficacy.

        하수 2차 처리수 재이용을 위한 저압 및 중압 고도산화시스템의 성능평가

        In the present study, the feasibility of UV/H₂O₂ systems was investigated using low and medium-pres sure lamps on biologically treated wastewater effluents for secondary effluent reclamation. Two types of UV lamps were used as the light sources (a 33W lowpressure mercury lamp and a 350-W mediumpressure mercury lamp). The results from these UV systems showed that the removal of organic compounds could be achieved in the contact time of longer than 30min (i.e., low UV doses). Efficiencies of color removal and disinfection were far better than those of organic matters measured as TOC, DOC and TCOD_(cr). In the low pressure lamp UV system, it has been found that DOC and color removals were 60.9 and 86.2% with 50㎎/L of H₂O₂ and contact times of 30 minute, respectively. Whereas, with the mediumpressure lamp UV system, TOC, DOC and color removal were 27.1, 5.6 and 95% with 14.3㎎/L of H₂O₂ and 14 minute of contact times, respectively. Both systems could be applied for the reclamation of secondary effluent treated with biological treatment processes.
