        Ethanol Extract of Ganoderma lucidum Augments Cellular Anti-oxidant Defense through Activation of Nrf2/HO-1

        Objectives: The mushroom Ganoderma lucidum has been widely used as a traditional herbal medicine for many years. Although several studies have focused on the anti-oxidative activity of this mushroom, the molecular mechanisms underlying its activity have not yet been clearly established. The present study investigated the cytoprotective effect of ethanol extract of Ganoderma lucidum (EGL) against oxidative stress (hydrogen peroxide, $H_2O_2$) and elucidated the underlying mechanisms in a C2C12 myoblast cell line. Methods: Oxidative stress markers were determined by using the comet assay to measure reactive oxygen species (ROS) generation and deoxyribonucleic acid (DNA) damage. Cell viability and Western blotting analyses were employed to evaluate the cellular response to EGL and $H_2O_2$ in C2C12 cells. Transfection with nuclear factor erythroid 2-related factor 2 (Nrf2)-specific small interfering ribonucleic acid (siRNA) was conducted to understand the relationship between Nrf2 expression and $H_2O_2$-induced growth inhibition. Results: The results showed that EGL effectively inhibited $H_2O_2$-induced growth and the generation of ROS. EGL markedly suppressed $H_2O_2$-induced comet-like DNA formation and phosphorylation of histone H2AX at serine 139 ($p-{\gamma}H2AX$), a widely used marker of DNA damage, suggesting that EGL prevented $H_2O_2$-induced DNA damage. Furthermore, the EGL treatment effectively induced the expression of Nrf2, as well as heme oxygenase-1 (HO-1), with parallel phosphorylation and nuclear translocation of Nrf2 in the C2C12 myoblasts. However, zinc protoporphyrin IX, a HO-1 inhibitor, significantly abolished the protective effects of EGL against $H_2O_2$-induced accumulation of ROS and reduced cell growth. Notably, transient transfection with Nrf2-specific siRNA attenuated the cytoprotective effects and HO-1 induction by EGL, indicating that EGL induced the expression of HO-1 in an Nrf2-dependent manner. Conclusion: Collectively, these results demonstrate that EGL augments the cellular anti-oxidant defense capacity through activation of Nrf2/HO-1, thereby protecting C2C12 myoblasts from $H_2O_2$-induced oxidative cytotoxicity.

      • 뇌손상 후 운동신경기능 회복 기전 : 기능적 뇌 자기공명영상의 유용성 The Usefulness of fMRI Study

        목적: 본 연구는 뇌손상환자에서 기능적 뇌 자기공명영상(functional magnetic resonance imaging, fMRI)을 이용한 대뇌운동피질 뇌지도화를 시행하여, 운동신경기능 회복 기전에 대한 기능적 뇌 자기공명영상의 유용성에 대하여 연구하였다. 방법: 신경학적 증상과 병력이 없는 성인 8명과 뇌손상환자 5인을 대상으로 하였다. 뇌손상환자 중 2명은 뇌종양환자이었으며 1명은 뇌출혈, 1명은 뇌경색, 1명은 뇌낭미충증환자이었다. 기능적 자기공명영상은 1.5T MR scanner로 Blood Oxygen Level-Dependent(BOLD) 기법을 적용하였다. 운동과제는 손가락을 1-2Hz의 주기로 쥐었다 펴기를 반복하도록 하였다. 결과: fMRI를 촬영한 결과 정상인 8인, 즉 총 16개의 손중 15개의 손과 환자 5인의 이환되지 않은 손운동시 반대측 일차 운동피질 꼭지(knob)를 중심으로 일차 감각운동피질(SM1)이 활성화되었다. 정상인 중 1인의 한 손에서 반대측 SM1과 동측 일차 운동피질(primary motor cortex, M1) 및 보조운동영역이 동시에 활성화되었다. 뇌손상환자의 이환측 손운동시 환자 1에서는 일차감각피질(primary sensory cortex, S1), 환자 2와 환자 3에서는 양측 SM1, 환자 4와 환자 5에서는 반대측 SM1이 활성화되었다. 환자 1에서는 손상된 M1의 운동신경기능이 S1으로 이전된 것으로 추정되며, 환자 4와 5에서는 손의 운동신경기능이 피질척수로에 의하여 유지되고 있는 것으로 추정되었다. 그러나 환자 2와 환자 3에서 발견된 동측 운동피질 활성화는 본래 정상적으로 존재하던 운동신경 경로인지 뇌손상 후에 발생한 운동신경기능 회복 경로인지 구분할 수는 없었다. 결론: fMRI을 통한 대뇌 운동피질 지도화는 뇌손상 후 병변 주위로의 재조직되는 회복 기전과 피질척수로에 의해 운동신경기능이 유지되고 있음을 입증하는데에 유용할 것으로 생각된다. Objective: This study investigated the usefulness of functional magnetic resonance imaging (fMRI) in the research of motor recovery mechanisms in brain injured patients. Method: Eight control subjects and five brain injured patients were investigated. Out of five patients, two were with brain tumor, one cerebral hemorrhage, one cerebral infarction, and one neurocysticercosis. fMRI was performed using the blood oxygen level-dependent (BOLD) technique at 1.5 TMR scanner with a standard head coil. The motor activation task consisted of finger flexion-extension exercises in 1-2 Hz cycles. Results: fMRI showed activation of the contralateral primary sensori-motor cortex(SM1) by the hand movements of all control subjects and by the unaffected hand movements of all brain-injured patients. Activation of the supplementary motor area(SMA) by one hand movement was also seen in one control subject. Hand movements of the affected hand of the brain injured patients resulted in activation of the contralateral primary sensory cortex(S1) in patient 1, both SM1 in patients 2 and 3, and the contralateral SM1 in patients 4 and 5. Conclusions: It is concluded that fMRI is useful for the research of perilesional reorganization and sparing of the corticospinal tract during the motor recovery mechanisms after brain injury.

        In this researching area, Iron ore deposit of skarn type is distributed in the line with the part of contact with pungchon limestone of great limestone group in relative rock of the cretaceous Samwha granite. The Granite has a high SiO₂, K₂O, ,Na₂O, contents on the contrary A1₂O CaO MgO TiO₂ low and they are of a uniform composition generally. when we investigate the interrelation of trace elements versus D.I, high differentiation have come out. And we found that Zr, th, Rb have a tendency to increase but Ba, Sr is decrease and Nb, Y, Rb has not systematic change and Rb/Sr versus D.I has a positive relation according as differentiation index increase. At the end, the age dating of this granite is not certain through whole rock isochron because of insufficiencey and alteration sample but I infered it was made in Mesozoic cretaceous.

      • 청정지역과 도시지역의 오존농도 특성 연구

        A study has been performed on the characteristics of rural and urban surface ozone concentration for the period of March 1992 to February 1993. The monitoring station of rural ozone is located at Kosan, Cheju and other urban monitoring stations are located at Seoul, Pusan and Kwangju. Rural's and urban's ozone data exhibit a distinct features in many ways. First, annual mean of rural ozone concentration is very high(42 ppbv) but urban's are very low(10~15 ppbv). Second, rural ozone data shows a seasonal variation with it's maximum in spring, and minimum in summer, but urban's show a seasonal variation with it's maximum in spring, and minimum in winter. Third, diurnal variation of rural data is very small but that of urban's are very large. Fourth, monthly mean of daily minimum of rural data is not low and shows a large seasonal variation, but those of urban's data are extremely low(<3 ppbv) and have no seasonal variations.

      • 제주도 고산에서 수용성 에어로졸의 화학적 성분 분석

        Atmospheric aerosols were collected by a High Volume Tape Sampler from March 1992 to December 1993 at Kosan, Cheju, korea. The water soluble ion concentrations in aerosol were analyzed. The concentrations of cations(Na+, K+, Ca²+, Mg²+, NH₄+) were determined by an Inductively Coupled Plazma(ICP) or an Atomic Absorption Spectrometer(AAS), and those of anions(Cl-, NO₃-, SO₄²-) were analayzed by the capillary electrophoresis method. The Ca²+, SO₄²-and NO₃- concentrations in spring were higher than those in other seasons. The lowest concentrations of these elements were found in summer, largely due to scavenging by frequent rains. Especially the ca²+ concentration on April was three to four times higher than the annual mean concentration. The enrichment factor(E.F.) of each element was calculated. The annual mean E.F. values of the Ca²+, Mg²+ and Cl- in 1992 were the same as those in 1993 except K+ and SO₄²-. The correlation formula between all cations and anions for the whole period was Anions=0.759xCations+0.066.

        월동 볏짚의 미생물 분포 및 청국장 발효효과

        월등한 볏짚을 1998년 2월 말부터 3월 초순 사이에 주로 대전을 중심으로 하는 충청지역에서 회수하고 이를 부위별로 구분하여 부착 세균류 및 진균류의 분포를 조사한 결과 도시 근교지역보다 농촌지역의 볏집에서 일반 중온성 세균수가 많고 동시에 bacilli도 많았으나 진균류는 오히려 적은 경향을 보였다. 고온성 세균과 방선균의 분포는 낮았으며, 대장균도 일부 지역에서 검출되었다. 볏짚의 부위별 분포를 보면 대체적으로 가운데 부위에서 미생물수가 높게 나타났으며 bacilli의 분포를 고려한다면 청국장 제조에는 하루보다 중부 및 상부가 접종원으로서 바람하다고 할수있다. 청국장 제조를 위한 볏짚시료로서는 어느 지역의 것이라도 무방하겠으나 하수 및 축산폐수의 오명이 없는 것을 선택할 필요가 있겠다. 볏짚의 중간부위를 미생물 접종원으로 사용하여 청국장을 발효시킨 다음 그 품질을 평가한 결과, 볏짚을 사용한 경우 그대로 발효시킨 대조구에 비하여 외관, 점성도, 아미노태 질소함량, 단백질 분해력에 있어서 현저하게 양호한 결과를 보였다. 볏짚의 사용 방법간에, 즉 실험구 사이에는 발효후 성분상의 차이가 별로 없었으므로 사용방법에 따른 영향은 적었다. The populations of microorganisms adhered on rice straws which had been ricked in the fields around Chungchong areas during the winter season from February to March in 1998 were investigated. The number of mesophilic bacteria including bacilli was generally high in the middle part of straw. And it appeared to be higher in the samples from rural areas than those from suburb. Thermophilic bacteria and actinomycetes were rarely to be detected in most samples. Coliform bacteria were detected in a few samples which collected from Kwanpyong-dong of Taejon-city, Jangpyong-meun and Jungsan-meun of Chongyang-koon, indicating that these areas were contaminated by sewage and livestock wastes. Following the fermentation of Chungkukgang employed the middle parts of straws from Mok-meun of Chongyang-koon as a source of microbial inoculum, qualities of the fermented products such as amino-nitrogen content, viscosity and protease activity, were examined: the application of rice straws resulted in better qualities of the fermented products when compared to the control, however the methods of application appeared to have little or no effect on the quality.
