      • RAW264.7 세포에서 interferon-r 및 LPS에 의해 유도되는 NO생성에 미치는 TALT-35의 영향

        TALP-35 purified from human term placenta is known to increase microtubule polymerization and stabilize the polymerized microtubule. To examine the effect of TALP-35 on immune system this study was performed. MTT assay was performed to investigate the effect of TALP-35 on the proliferation of RAW264.7 cells. TALP-35 dose dependently suppress the proliferation of RAW264.7 cells at high concentration (above 1 μM) in unstimulated cells, in case of 10 μM TALP-35 treated cells the suppression was 25% but in stimulated cells it was only 15%. Cosedimentation assay was carried out to investigate whether TALP-35 can bind to tubulin of RAW264.7, monocyte/macrophage lineage of mouse, and polymerize it. TALP-35 polymerize the tubulin of RAW264.7 cells and sedimented in dose-dependent manner. To investigate the effect of TALP-35 on the expression of iNOS protein western blotting was performed. The expression level of iNOS was decreased dose dependently in high concentration of TALP-35 treatment. To examine the activity of iNOS, secreted NO was determined by method based on Griess reaction. Interferon-γ and LPS-stimulated production of NO from RAW264.7 cells was decreased dose dependently above 0.1 μM concentration of TALP-35 and 50% is decreased at 10μM of it. This study shows TALP-35 can control cytokine induced-iNOS expression therefore it might control inflammatory diseases.

Analgesic Effect of Manual Acupuncture and Electroacupuncture combined with Twirling Method. Proceeding of International Symposium on East-West Medicine Seoul. 187-200, 1999.-Twirling the needle (TN) is one of several needing methods frequently used for acupuncture in the oriental medicine. The present study was conducted to see if TN enhanced the antinociceptive effect produced by plain-or electroacupuncture in the rat tail flick test. Rats (Sprague-Dawley, 200-300g)lightly anesthetized with thiopental sodium(40mg/kg, i.p.) were used in the tail-flick test. The basal reaction time for tail-flick was 2 sec. For "plain" acupuncture(PA), a needle was inserted for 20 min into a Chock-samni (ST36) acupuncture point which (is located at the anterior tibial muscle and about 10mm below the knee joint). For electroacupuncture(EA), train-pulses(3 Hz,0.3-ms pulse width, 0.2-0.3mA) were applied to the inserted needle for the period of insertion. For combining TN with either PA or EA, TN was performed during the each acupuncture period. Two methods for TN were used; twirling 3 times for 1 second persisted for 1 min in every 5 mins(long-duration and long-interval;LDLI) and twirling 3 times for 1 second persisted for 10 sec in every 1 min(short-duration and short-interval;SDSI). PA and EA produced an increase in tail flick latency (TFL); peak increases were 21.3±5.1% and 42.1±17.3% of the pre-acupunture control, respectively, and occurred immediately after cessation of 20min-acupuncture. Performing TN combined with PA increased TFL more than PA itself by showing a greater peak increase in TFL when performed with a SDSI-TN than with a LDLI-TN(60.6±12.5% and 42.7±22.7% of the pre-acupuncture control, respectively). Performing TN combined with EA also increased TFL more than EA itself and a greater peak increase in TFL was observed with a SDSI-TN as compared to a LDLI-TN(77.5±13.8% and 67.3±14.0% of the pre-acupuncture control, respectively). These results indicate that TN enhances both PA-and EA-produced antinociception where an greater enhanced antinociception than a LDLI-TN. It is suggested that performing a SDSI-TN combined with EA seems to be one of the most effective ways to produce analgesia.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the second year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. The systems were as follows; - differential diagnosis according to condition of body fluid, differentiation of syndromes according to the state of qi and blood, differential diagnosis according to relative excessiveness or deficiency of yin and yang(氣血陰陽津液辨證) - differentiation of diseases according to pathological changes of the viscera and their interrelation - analyzing and differentiating of febrile diseases in accordance with the theory of the six channels(傷寒辨證) The individual diagnosis pattern was arranged by the diagnostic requirements in the following order : another name(異名), notion of diagnosis pattern, index of differentiation of symptoms and signs(辨證指標), the main point of diagnosis, analysis of diagnosis pattern(證候分析), discrimination of diagnosis pattern(證候鑑別), a way of curing a diseases(治法), prescription(處方), herb in common use(常用藥物), diseases appearing the diagnosis pattern(常見疾病), documents(文獻調査).         Adipose sirtuin 6 drives macrophage polarization toward M2 through IL-4 production and maintains systemic insulin sensitivity in mice and humans

        <▼1><P>Adipose tissue inflammation is a reproducible feature of obesity and obesity-linked insulin resistance. Although sirtuin 6 (Sirt6) deficiency has previously been implicated in diet-induced obesity and systemic insulin resistance, the adipocyte-specific role of Sirt6 in the regulation of adipose tissue inflammation and systemic metabolic dysfunction in mice fed normal chow and in humans remains elusive. Here, using <I>Adipoq-Cre</I>-mediated adipocyte-specific Sirt6 knockout (aS6KO) mice, we explored whether adipocyte Sirt6 inhibits adipose tissue inflammation and its underlying mechanism. aS6KO mice fed normal chow gained more body weight and fat mass than wild-type mice and exhibited glucose intolerance and systemic insulin resistance. Measurement of plasma and tissue cytokines and flow cytometric analysis of adipose stromal vascular cells indicated a decrease in alternatively activated M2 macrophages in the adipose tissue of aS6KO mice. Mechanistically, Sirt6 regulated the expression of the canonical type 2 cytokine IL-4 by adipocytes in a cell autonomous manner, which in turn affects M2 macrophage polarization. Consistent with animal experimental data, the degree of obesity and insulin resistance demonstrated by the body mass index, fasting blood glucose and HbA1c correlated negatively with the expression of Sirt6 in human visceral fat tissues. Collectively, these results suggest that adipocyte Sirt6 regulates body weight gain and insulin sensitivity independent of diet, and the increased IL-4 production by Sirt6 and resultant M2 polarization of adipose tissue macrophages may attenuate proinflammatory responses in adipose tissue.</P></▼1><▼2><P><B>Obesity and diabetes: A control protein in fat</B></P><P>A protein in adipose tissue (composed of fat cells) helps protect against inflammation and the development of resistance to insulin that develops in obesity and can lead to type 2 diabetes. Researchers in South Korea, led by Eun Ju Bae at Woosuk University, Wanju, and Byung-Hyun Park at Chonbuk National University, Jeonju, investigated the role of Sirt6 in mice and in human adipose tissue. Deleting the mouse gene that codes for Sirt6 in adipocytes promoted the impaired response to insulin and associated increase in blood glucose levels that are two key aspects of diabetes. Changes in biochemical signaling pathways controlling immune cells called macrophages were implicated in these effects and suggest an anti-inflammatory role for Sirt6. Analysis of human adipose tissue supported these findings. The research will help understand how obesity promotes type 2 diabetes.</P></▼2>

      • 염화콜린 처리가 고추 및 토마토묘의 생육 및 몇가지 원예작물 삽목묘의 발근과 생육에 미치는 영향

        Effects of choline chlorides of different formulations and their analogues on pot-grown hot pepper and tomato seedlings were investigated. Choline chloride solutions of different concentrations were sprayed or applied to the pot-soil approximately one month before the expected transplanting and the quality of seedlings was evaluated shortly before the transplanting. The effects of choline chloride on the rooting and subsequent growth of cuttings of several horticultural crops were also investigated. Major results can be summarized as follows. 1. Spray treatment of 250-1,000ppm choline chloride to hot pepper seedlings were effective in improving the seedling vigor by promoting the seedling growth and increasing the fresh weight of roots of several cultivars tested. 2. Applications of choline chloride solutions directly to the pot at the rate of l0ml per 12-cm diam pot were effective in improving the seedling vigor prior to transplanting in 'Dabok' hot peppers. 3. Spray of 250-500ppm choline chloride to tomato seedlings promoted the growth though decrease in the number of flowers in the first flower cluster was observed in the seedlings sprayed with solutions of higher concentrations. 4. Significant promotion of rooting was obtained by dipping the sweetpotato cuttings in choline chloride solution and rooted a t cool, shady place during the early spring whereas no such promotion was found in the cuttings rooted under favorable environments. 5. Cucumber cuttings dipped in 3 ppm choline chloride solution prior to rooting in vermiculite medium showed a marked promotion of rooting as comparable to that with IBA treatment whereas the watermelon cuttings treated in the same way showed only slight promotion of rooting. 6. The effect of choline chloride treatment was not quite apparent when treated to the easy-to-root floricultural plants such a s devil's ivy and English ivy.

        We have used molecular biology technique to label the side chains and backbone of HBV preS1(1-56) with stable isotope (??) during E. coli over-expression and purified the protein to near homogeneity. The expression level of ?? labelled preS1(1-56) seems to be ∼15% of the total protein. to characterize the secondary structure of the labelled protein, circular dichroism was used. In different buffer conditions including pH 3.3 and pH 7.4 the protein seems to be devoid of any stable secondary structure probably suggesting that the N-terminal portion (preS1(1-56)) of HBV preS1 might be unstructured before binding to its target cells.

      • Gastric Calcifying Fibrous Tumor : A Rare Cause of Gastric Subepithelial Lesion

        석회화 섬유종양은 원인 미상의 흔하지 않은 양성 중앙이다. 이 종양은 복강이나 복막에서 기원하는 경우가 많으나 위에서 기원한 석회화 섬유종양은 매우 드물 다. 건강검진시 위내시경에서 우연히 발견된 위 점막하 종양 평가를 위해 28세 여성이 본원으로 전원되었다. 본원에서 시행한 위 내시경에서 위 중간체부에 1.2cm의 평탄한 표면의 위 점막하 종양이 관찰되었다. 초음파 내시경을 시행하였고 점막하층에 위치한 불균질한 저에코성의 병변과 함께 종양 내부에는 음향 음영을 동반한 다수의 고 에코성의 부위가 관찰되었다. 위 점막하 중앙은 악성종양을 완전히 배제할 수 없기 때문에 내시경적 점막하 절제술을 시행하였다. 조직 검사결과 종양은 석회화률 동반하는 호산구성 콜라겐 섬유가 풍부한 위 석회화 섬유 종양으로 진단되었다. 이에 위 점막하 중앙에 관한 다른 문헌을 고찰하면서 본 증례를 보고하는 바이다.

      • 반건식탈황법을 이용한 SOx/NOx의 제거에 관한 연구

        This study has been carried out to investigate the variation of removal efficiencies with SOx/NOx concentrations and the improvement of removal efficiencies with additives addition. The results are summarized as follows ; 1. For the SOx cases, the removal efficiencies of 95%, 95% and 90% were achieved at 300ppm, 1000ppm and 1500pm respectively with 15 Ca/S stoichiometric ratio in SDA-FF Process 2. For the NOx cases, the removal efficiencies of SDA were obtained extremely low levels. But the removal efficiencies of SDA-FF were achieved approximately 50% regardless of Ca/S stoichiometric rations and concentration loadings. 3. For the cases in variation of NOx removal efficiencies with additives addition, the improvement of removal efficiencies were obtained 5%, 10% by NaOH, NH₄Br respectively with 1.0 Ca/S stoichiometric ratio in SDA-FF.

      • DCT 영역에서 분류 벡터 양자화를 이용한 블럭 예측 부호화

        In this paper, an effective block predictive coding was presented. This method makes use of the basic properties of image and the analysis of the correlation within spatial domain and transform domain. For this block predicive coding, first, the correlations between spatial domain and transform domain are analyzed, and then with these properties a very effective classifier is designed for obtaining minmized errors. After setting delegate blocks out of each class passed through the classifier, the errors between input blocks and delegate blocks are classified-vector quantized. Simulation shows that significantly improved results are obtained compared to Aizawa method[4], that is, in terms of the quality of reconstructed image, 3∼5 dB at the same bit rate, and the blocking effect is reduced even for low bit rates.
