        흰쥐 해마박편에서 veratrine과 고농도 포타슘자극시 칼슘이온이 gamma-aminobutyric acid 유리에 미치는 영향에 관한 연구 : A role of calcium

        Present study was performed to clarify the effect of calcium on the release of gamma-aminobutyric acid (GABA) employing hippocampal slices. Hippocampal slices(300-400㎛ thick) were prepared by the method of Kim et al(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. In case of veratrine-induced GABA release, pre-equilibrated slices were incubated in fresh KBM and then veratrine (25μM)-containing KBM for 10min period in the presence or absence of 2.5mM Ca^2+. In case of potassium-induced GABA relaese, pre-equilibrated slices were incubated in fresh KBM and then potassium(50mM)-containing KBM for 5min period in the presence or absence of 2.5mM Ca^2+. Basal and veratrine and potassium-induced release of GABA was determined from recovered medium by HPLC. The observed results were as follows: 1. The release of GABA induced by the 10min-exposure of 25μM veratrine and 5min-exposure of 50mM potassium in the presence of 2.5mM Ca^2+ was 228.9±11.2 nmol and 100.1±8.9nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 6.8 and 4.6-fold increase respectively. 2. The release of GABA induced by the 10min-exposure of 25μM veratrine and 5min-exposure of 50mM potassium in the absence of Ca^2+ was 381.4±30.2 nmol and 55.1±4.1 nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 11.3 and 2.4-fold increase respectively.

        흰쥐 해마박편에서 acetylcholine이 gamma-aminobutyric acid 유리에 미치는 영향에 관한 연구

        Present study was performed to clarify the effect of acetylcholine on the release of gamma-aminobutyric acid(GABA) employing hippocampal slices. Hippocampal slices (300∼400㎛ thick) were prepared by the method of Kim et al(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. Pre-equilibrated slices were incubated in fresh KBM and then potassium(50mM)-containing KBM for 5 min period. Basal and potassium-induced release of GABA were determined from recovered medium by HPLC. After 30min resting period, in the presence of physostigmine(20μM) slices were reincubated in acetylcholine-containing KBM and acetylcholine plus potassium-containing medium consecutively for 5min period each to investigate the effect of acetylcholine on basal or potassium-induced GABA release from hippocampal slices. The observed results were as follows: 1. The release of GABA induced by the first and second 5 min-exposure of 50mM potassium was 107.3±8.2 nmol and 90.6±3.2nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 4.6 and 4.6-fold increase respectively. 2. Physostigmine(20μM) had no significant effect on the spontaneous release of GABA. 3. Acetylcholine(10-1000μM) increased spontaneous and potassium-induced GABA release in a dose-dependent manner.

        하악골에 발생한 골막 골육종

        Periosteal osteosarcoma is a distinct entity of malignant bone tumor with characteristic clinical, morphological, and histological features within the group of juxtacortical osteosarcoma. Periosteal osteosarcoma is predominantly located in the tubular long bones, and extremely rarely involving the jaws. A case of periosteal osteosarcoma of the right mandible is presented. A 27-year-old woman complained of the gingival swelling and bleeding tendency of the right posterior mandible. Clinical examination revealed a reddish brown strawberry-like swelling on the affected mucosa, which measured 1.5㎝×1.5㎝. The tumor was located on the lingual cortex of the mandible and extended into the surrounding soft tissues. Microscopically, the tumor consisted exclusively of atypical chondroblastic cells with a small osteoblastic area. A minimal bone marrow involvement was noted and the adjacent cortex was free of tumor. These findings suggested that the tumor originated from the periosteal cambium layer, which lies between the periosteal fibrous layer and the cortex of mandible.

        Lidocaine이 흰쥐 해마박편에서 Gamma-Aminobutyric Acid 유리에 미치는 영향에 관한 연구

        Present study was conducted to clarify the role of GABA in lidocaine-induced convulsion employing hippocampal slice. Hippocampal slices(300-400㎛) were prepared from hippocampal tissue blocks using a glass guide while the tissue was continuously moistened with oxygenated (95% O_2-5% CO_2) Krebs-bicarbonate media (KB) at 37℃. After preparation, the slices were equilibrated for 1hr before biochemical testing in KB. Individual equilibrated slices were transferred to tissue holder, incubated for 5min in fresh oxygenated KB, and then exposed for 5min to 50mM K^+ KB and transferred again to fresh KB for 30min rest period. Each slice served as its own control by exposing it a second time to 50mM K^+ KB for 5min after the 30min rest and a second 5min incubation in fresh KB. After the 30min rest and a second 5min incubation in fresh KB, slices were transferred again to fresh KB containing 1, 2, 3, or 4mM lidocaine, incubated for 10min and then exposed to 50mM K^+ KB+lidocaine solution for 5min. The results were as follows : 1. The release of GABA induced by the first and second 5min exposure of 50mM K^+ was 95.9±17.28nmol and 80.6±13.04nmol, respectively. When compared with released amounts of GABA during the corresponding spontaneous periods, these were 4.7 and 4.9-fold increase respectively and revealed that consecutive K^+─exposure increased GABA release with similar fashion. 2. The concentration of lidocaine necessary for 50 percent inhibition(IC_50) of potassium─induced release of GABA was 1.82mM.

      • Stage II B형 골육종에 대한 술전 항암화학요법 및 수술적 치료 결과

        Recent advances in imaging techniques, surgery and combination anti-cancer chemotherapy have brought high survial rates in osteosarcoma. To investigate the survival rate, local recurrence and complications in treatment, we analysed 25 osteosarcoma cases who had been treated with preoperative neo-adjuvant chemotherapy, surgery and post operative chemotherapy at Department of Orthopedic Surgery, Catholic University. From May 1988 to April 1995, 42 cases of stage IIB osteosarcoma were admitted in Department of Orthopedic surgery. Among them, 17 cases who didn't follow our treatment guidance were excluded in this study. The average age were 19 years. There were 21 males and 4 females. The involved sites were 4 humerus, 10 femur, 10 tibia and 1 talus. Eleven cases had received intra-arterial cisplatin and intravenous adriamycin chemotherapy, and 7 T-10 protocol and 7 intravenous ifosfamide, ADR, methotrexate, cisplatin. Among twenty-three cases were treated with limb salvage surgery, and 2 amputation. The average follow-up was 35 months(3-82). There were 14 cases of continuous disease free, 9 cases of died of desease, 1 case of alive with disease, and 1 case of no evidence of disease at final follow-up. There were three cases of local recurrence at 6,8 and 12 months after operation. The Estimated kaplan-Meier's 5 year survival rates for all, ADR-cisloatin group, T-10 protocol group, and ifosfamide regimen group were 66%, 73%, 44% and 72%, respectively.

      • 口眼渦斜 患者의 Gadolinium-DPTA enhanced MRI 所見에 대한 臨床的 考察

        This study is designed to evaluate the clinical implications of Gd-DPTA (Gadolinium-diethyl - enetriamine pentacetic acid) enhanced MRI(Magnetic resonance imaging) in Bell's palsy and find it's usefulness in Oriental Medicine In this study, 25 outpatients with Bell's palsy were studied that MRI was performed. To evaluate degree of facial palsy, H-B(House-Brackmann) Grade was used. In Oriental Medical therapy, Acupuncture and Herbal medicine were treated. Subjective causes was divided into exposure to chill, fatigue, stress, mixed cases. Enhanced site was compared with symptoms which were disorder of eye, hearing, taste, and facial muscle palsy. Also, Relation between time which was performed MRI and enhancement was analyzed. The enhanced lesion in MRI was divided into five segments; Internal auditory canal, Labyrinthine segment, Geniculate ganglion, Tympanic segment, Mastoid segment. In Bell's palsy, 20 of 25 patients(80%) had abnormal contrast enhancement of the facial nerve. The H-B grade and interval performed MRI from onset were directly proportionate to enhancement. That is to say, Severe facial palsy short interval show high possibility of enhancement. There was no relation between subjective causes and enhanced site of facial nerve in MRI. Also Clinical symptoms didn't coincide with MRI findings.

      • 蜂毒藥鍼刺戟이 腦幹 神經細胞와 Serotonin性 神經細胞의 活性變化에 미치는 영향

        This study was designed to evaluate the effect of the bee venom(BV) aqua-acupuncture on the neuronal activities of serotonergic(5-HT) system in the brainstem. After the BV aqua-acupuncture was applied on Chok-Samni(ST36) and the gluteal part(Blank locus) in rats, the number of Fos immunoreactive neurons was counted by using computerized image analyzing system. Also, the number of colocalization between 5-HT containing neurons Fos immunoreactive neurons were analyzed by using the double immunohistochemical technique. The results of the experiments were summarized as follows : 1.In almost every neucli, the Chok-Samni group and Blank locus group showed more increase in the number of Fos immunoreactive neurons than the control group. Especially, in Arc, DR, LC, RMg, Gi, PAG Rost and PAG LV, the Chok-Samni group showed more significant increase than the control group. Also, in PAG LV Mid and Arc, Chock-Samni group showed more significant increase than the Blank locus group. 2.In DR and PAG LV Mid, Chok-Samni group and the Blank locus group showed more significant increase in the number of colocalization between 5-HT containing neurons and Fos immunoreactive neurons than the control group after the BV aqua- acupuncture. Also, the Chok-Samni group showed more significant increase than the Blank locus group. Consequently, the BV aqua-aqupuncture increased more potent the number of Fos immunoreactive neurons and the activity of serotonergic neurons. Furthermore, the BV aqua-acupuncture was more effective on Chok-Samni than Blank locus group. These results indicate that the BV aqua-acupuncture is very effective therapy to control pain. The therapeutic effect of BV aqua-acupunture may associated with the endogenous modulatory system such as serotonin Those data from the study can be applied to establish the effective treatment of the BV for pain control in the clinical field.

        The current study provides an efficient method connecting the free-selected terminal among the divergent terminals, which may fit best in the various business operation characteristics, to existing host computer. More specifically, the review of network components has been made to control interface between the computer and the terminal of different type, and the analysis of NETWORK PROTOCOL controlling interface also has been made. For the better compatibility, the modification has been made between the host computer and the terminal of different type to operate properly in the general-purpose NETWORK PROTOCOL which differ from the existing host computer.

        當歸의 種類에 따른 四物湯이 白鼠의 胸部大動脈 血管弛緩에 미치는 影響

        We have examined the relaxational reponse to the water extract of Angelica gigas Nakai (AG), A. sinensis(Oliv.) Diels (AS), A. acutiloba Kitagawa (AA), and Four-Substance Decoctions (AG-FSD, AS-FSD, AA-FSD) in isolated thoracic aorta from sprague dawley(SD) rat in the presence and absence of endothelium. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to AG, AS, AA and AG-FSD, AS-FSD, AA-FSD were investigated in vessels precontracted with 5-hydroxytryptamine(5-HT) were compared in vasodilation effect. We found that the thoracic aorta segments responded to AG, AS, AA and AG-FSD, AS-FSD, AA-FSD with a dose-dependent vasodilation. The 5-hydroxytryptamine induced contraction at 10-4M were inhibited by 26.3%, 2.7%, 2.1% and 24.6%, 1.1%, 3.2% after addition of the 0.1 g/㎖ water extract of AG, AS, AA and AG-FSD, AS-FSD, AA-FSD. The 5-hydroxytrptamine induced contraction at 10^-4M with and without endothelium were inhibited by 24.6% and 6.9% after addition of the 0.1 g/㎖ water extract of AG-FSD. In conclusion, AG, AS, AA and AG-FSD, AS-FSD, AA-FSD-induced relaxation in the isolated rat thoracic aorta were composed of dose-dependent relaxation. AG-FSD has very potent vasodilation.

        흰쥐 해마박편에서 콜린성 수용체와 glutamate 유리와의 상호관계에 관한 연구

        Present study was performed to clarify the effect of cholinergic agents on the release of glutamic acid employing hippocampal slices. Hippocampal slices(300∼400㎛ thick) were prepared by the method of Kim et al.(1988) and pre-equilibrated in Krebs-bicarbonate medium(KBM, pH 7.4) for 1hr at 37℃. Pre-equilibrated slices were incubated in fresh KBM and then potssium(50mM)-containing KBM for 5 min period. Basal and potassium-induced release of GABA and glutamic acid were determined from recovered medium by HPLC. After 30 min resting period, slices were reincubated in cholinergic agents-containing KBM and cholinergic agent plus potassium-containing medium consecutively for 5 min period each to investigate the effect of cholinergic agent on basal or potassium-induced glutamic acid release from hippocampal slices. The observed results were as follows: 1. The release of glutamic acid induced by the first and second 5 min-exposure of 50mM potassium was 139.7±14.05 nmol and 114.5±10.01 nmol, respectively. When compared with released amounts of glutamic acid during the corresponding spontaneous periods, these were 5.3 and 5.6-fold increase respectively. 2. Acetylcholine(10-1000μM) inhibited potassium-induced glutamic acid release in dose-dependent manner. 3. The inhibition of glutamic acid release caused by acetylcholine(1mM) was antagonized by atropine(50μM) but not by mecamylamine(50μM).
