      • Novel dentin phosphoprotein frameshift mutations in dentinogenesis imperfecta type II

Novel dentin phosphoprotein frameshift mutations in dentinogenesis imperfecta type II.</P><P>The dentin sialophosphoprotein (<I>DSPP</I>) gene encodes the most abundant non‐collagenous protein in tooth dentin and DSPP protein is cleaved into several segments including the highly phosphorylated dentin phosphoprotein (DPP). Mutations in the <I>DSPP</I> gene have been solely related to non‐syndromic form of hereditary dentin defects. We recruited three Korean families with dentinogenesis imperfecta (DGI) type II and sequenced the exons and exon–intron boundaries of the <I>DSPP</I> gene based on the candidate gene approach. Direct sequencing of PCR products and allele‐specific cloning of the highly repetitive exon 5 revealed novel single base pair (bp) deletional mutations (c.2688delT and c.3560delG) introducing hydrophobic amino acids in the hydrophilic repeat domain of the DPP coding region. All affected members of the three families showed exceptionally rapid pulp chambers obliteration, even before tooth eruption. Individuals with the c.3560delG mutation showed only mild, yellowish tooth discoloration, in contrast to the affected individuals from two families with c.2688delT mutation. We believe that these results will help us to understand the molecular pathogenesis of DGI type II as well as the normal process of dentin biomineralization.</P>

        사료내 식물성 Phytase 첨가가 산란계의 생산성 및 인 이용성에 미치는 영향

        본 연구는 식물성 phytase 처리수준이 산란계의 생산성에 미치는 영향 및 인 이용률에 미치는 영향을 검토하기 위하여 실시하였다. NRC 사양표준의 유효 인 요구량을 충족시킨 대조구와 무기태인 사용량을 50%로 제한한 처리구로 나누고, 각각의 처리구에 식물성 phytase를 각각 0.05%, 0.1% 및 0.2% 첨가하였다. 58주령의 갈색 산란계(ISA-Brown) 240수를 공시하여 총 6주간 사양실험을 실시하였고 산란종계를 이용하여 인 및 질소의 이용성을 측정하였다. 그 결과를 요약하면 다음과 같다. 1. 50% 무기태인 수준에서 식물성 phytase의 첨가에 따른 일당증체량, 산란율의 저하는 나타나지 않았다. 2. 50% 무기태인 수준에서 식물성 phytase 0.05% 첨가수준에서 일시적으로 난각질 저하 현상이 나타났으나 0.1% 이상 첨가수준에서는 난각질의 변화가 관찰되지 않았다. 3. 효소의 첨가수준이 증가함에 따라 P 배설량의 감소와 P 이용률이 개선되었다(P<0.05). 4. 식물성 phytase 첨가는 산란계의 성적 저하없이 사료내 무기태인의 사용량을 줄일 수 있음이 시사되었다. Two experiments were conducted to investigate the effect of dietary supplementation of plant phytase(Phytazyme^R) in corn-soybean meal based diets on utilization of phytase-bound phosphorus in laying hens and evaluate nitrogen(N) digestibility and phosphorus(P) availability in breeders. In he experiment one, three levels of the Phytazyme^R(0.05, 0.1, and 0.2% of diet) were added to diets containing a half of control TCP level(0.96%) for 4 wks. Feed consumption, egg production rate, egg weight and eggshell quality were recorded weekly. At the end of experiment, 8 birds per treatment were sacrificed, liver weight were weighed and right tibiae were removed for determination of P content. The second experiment was conducted to evaluate the P availability and nitrogen digestibility in breeders fed same diets for 2 wks. Feed and excreta were collected to determine the P and N contents for the last three days of experiment two. Addition of Phytazyme^R resulted in no effects on feed intake, egg product rate, egg weight and egg shell quality. P excretion decreased and its availability enhanced as phytase supplementation increased in diets. Dietary supplementation of Phytayme ^R above 0.1% level in corn-soybean meal based diets did not have an adverse effect on production and decreased level of phosphorus in excreta.

      • Synthesis and characterization of ampelopsin glucosides using dextransucrase from Leuconostoc mesenteroides B-1299CB4: Glucosylation enhancing physicochemical properties

        Novel ampelopsin glucosides (AMPLS-Gs) were enzymatically synthesized and purified using a Sephadex LH-20 column. Each structure of the purified AMPLS-Gs was determined by nuclear magnetic resonance, and the ionic product of AMPLS-G1 was observed at m/z 505 (C<SUB>21</SUB>H<SUB>22</SUB>O<SUB>13</SUB>.Na)<SUP>+</SUP> using matrix-assisted laser desorption ionization time-of-flight mass spectrometry. AMPLS-G1 was identified as ampelopsin-4'-O-α-d-glucopyranoside. The optimum condition for AMPLS-G1, determined using response surface methodology, was 70mM ampelopsin, 150mM sucrose, and 1U/mL dextransucrase, which resulted in an AMPLS-G1 yield of 34g/L. The purified AMPLS-G1 displayed 89-fold increased water solubility and 14.5-fold browning resistance compared to those of AMPLS and competitive inhibition against tyrosinase with a K<SUB>i</SUB> value of 40.16μM. This value was smaller than that of AMPLS (K<SUB>i</SUB>=62.56μM) and much smaller than that of β-arbutin (K<SUB>i</SUB>=514.84μM), a commercial active ingredient of whitening cosmetics. These results indicate the potential of AMPLS and AMPLS-G1 as superior ingredients for functional cosmetics.

      • 외상성 심내 이물질의 응급 개심술 치험 : 1 case report

        A case of a 33-year-old man who sustained the penetrating wound of the heart by the iron-works accident in March, 1986, is reported. He was arrived at this emergency room for 45 minutes after the accident. The physical examination showed the small inlet in the left 3rd intercostal space and parasternal border, and drowsy mentality with a systolic pressure under 80mmHg when he arrived. The chest roentgenography revealed enlarged heart and 0.5×0.3cm sized radio-opaque metalic fragment at the mid-portion of the heart. About 500ml of dark pure blood was immediately aspirated through subxyphoid pericardiotomy. The emergency operation was done under the extracoporal circulation. The inlet of the heart injury was noted at the anterior wall of the right ventricle(0.3cm) and obstructed with the blood clot. Through the right ventriculotomy incision, the iron fragment was removed from the septum near the origin of the medial papillary muscle of the tricuspid valve. The post-operative course of the patient was smooth except complete rght bundle branch block. The patient discharged at 14th post-operative days.

      • 산란계에 대한 한방 부산물의 영양적 효과

        본 실험은 사료원료로서 한방 부산물의 영양적 가치를 평가하고 산란계 사료의 배합비를 작성하여 실제 사양실험을 통한 한방부산물의 이용 가능성 및 계란 품질변화를 구명하고자 대사 실험을 통하여 한방부산물의 진정대사에너지(TME)와 진정아미노산이용율(TAAA)를 측정한 후 90수의 산란계에게 한방부산물 및 생약을 각각 0, 2, 4%수준으로 배합하여 4주간 사양하였으며 매주 산란율, 사료섭취량, 계란품질(난각강도, 난각두께, 난황색 및 Haugh Unit)를 측정하였다. 1. CP함�c은 10.4%, TME는 1,121kcal/kg, 필수 아미노산의 이용률은 평균 47.01%로 다소 조악한 경향을 나타내었다. 2. 모든 처리구에서 한방부산물의 첨가에 따른 산란율과 사료섭취량 및 계란의 품질에 유의적인 차이는 발견되지 않았다. 3. 본 연구결과 한방부산물을 산란계 사료원료로써 4%까지의 첨가는 영양적 불균형을 초래하지 않아 사용 가능할 것으로 판단되었다. Two experiments were conducted to evaluate the feeding values of oriental medicine residue (OMR) and to investigate effects on its dietary supplementations in laying hen. True metabolizable energy(TME) and true amino acid availability(TAAA) values of OMR were determined in the first experiment. In the second experiment, a total of 90 laying hens (Isa-Brown) were divided into 5 groups and were fed diets containing 0, 2, 4% OMR and 2, 4% OM(oriental medicine) for 4 weeks. Weekly measurements were made on egg production rate, feed intake, eggshell strength, eggshell thickness, egg yolk color, and Haugh Unit. Crude protein content and TME for OMR were 10.4% and, 1,121kcal/kg respectively. The average TAAA of essential amino acids was 47.01%. There were no significant differences in egg production rate, feed intake and egg quality between the treatment. It could be concluded that OMR can be used for laying hen feeds up to 4% without any significant negative effects.

        Seventy cases of the open heart surgery were performed in the Department of Thoracic and Cardiovascular surgery of Soonchunhyang University in 1985. Of them, 52 cases were congenital anomalies which were classified as 43 acyanotic patients and 9 cyanotic patients, and 18 cases were aquired valvular diseases which contained 17 rheumatic valvular diseases and 1 mitral valve prolaps syndrome . The most common anomaly of acyanotic congenital group was VSD(22cases) and the one in cyanotic group was TOF(5 cases). In 18 cases of the acquired valvular diseases, open mitral commissuratomy was done in 2 patients, tissue valve replacement(Carpentier-Edwards Bioprosthesis) in 9, mechanical valve replacement(Medtronic-Hall) in 6, and mitral valvulo-plasty with Carpentier ring in 1 patients. The most frequent complications during postoperative course were arrhythmia and low cardiac output syndrome, Perioperative mortality rate was 5.7%.

        사이버 윤리지표 개발에 관한 실증연구 : 한국, 미국, 인도의 차이를 중심으로

        정보화 시대에서 인터넷이 주는 여러 장점이 존재하기는 하지만 그에 따른 비윤리적 행위에 대한 폐해도 점차 심각해지고 있기 때문에 사이버 상에서의 윤리지표가 필요한 시점이라 생각한다. 본 연구에서는 다음과 같은 두 가지 목적을 가지고 있다. 첫째, 사이버 공간에서 발생되는 범죄행위에 대한 문제인식을 바탕으로 본 연구에서 제시된 인터넷 사용자들의 윤리의식 지수(Self-Evaluation)를 스스로 측정해 볼 수 있는 표준화된 사이버 윤리 프레임워크를 개발하는 것이다. 둘째, 제시된 프레임워크의 타당성을 검증하기 위해 3개국(한국, 미국, 인도) 인터넷 사용자들을 대상으로 실증분석을 함으로써 각기 문화권이 다른 한국, 미국, 인도의 인터넷 사용자들의 윤리지수 차이를 문화적 관점에서 국가간 어떤 차이를 보이는지를 규명하는데 그 목적이 있다. 연구의 시사점으로는 국가별 전통문화의 차이 규명과 함께 행동의도를 분석하였다는 점과 인터넷 공간에서 사용자들이 윤리적인 부분에 있어 불안감이나 민감하게 느끼는 요인들을 분석하였다는 점에서 연구의 의의를 찾아볼 수 있다. The Internet-assisted communication transforms conventional information systems into vast human interactive networks, and offers people opportunities to experience new forms of social contacts, without any real social presence. Because of this anonymity of Internet users, they have a strong tendency to behave in an unrestricted manner. This experience is do stimulating, rewarding and reinforcing that ome user find it hard to know when to stop. These behaviors frequently go over the fun, and are developed into the crimes. To reduce these side-effects, various computer security solution and laws have been introduced. However, the most important factor in reducing the Internet side-effects is individual user’s ethics. This research proposes a Cyber Ethics Evaluation Model to measure the ethics of Internet users and compares cyber ethics of South Korea, U.S.A, and India by using the model. It conduct Confirmatory Factor Analysis (CFA) to test the validation of the model.
