        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        This study attempted to determine the degree of asbestos release from the aging slate roof by comparing the asbestos content in the slate roof rain gutter with a colored steel plate (tin plate) over the slate roof. Four slate roof houses located in Haengmok-ri, Asan-si, Chungcheongnam-do, and one house constructed with a colored steel plate on the slate roof were selected to collect the sediment of the roof rain gutter. The asbestos fiber content was calculated by a point counting method using a polarization microscope after pretreatment with conversion treatment and hydrochloric acid treatment. The average asbestos content of the four slate roof rain gutter were 1.89%. However, asbestos was not detected in the Slate covering roof rain gutter, which were constructed on the slate roof. Asbestos fiber content was the highest at 2.89% in the slate roof rain gutter installed in 1976, followed by 2.44% in 1953. From the above results, it is necessary to minimize secondary damage as asbestos fibers released from slate roof houses to the surrounding atmosphere or leaked from slate roofs as rainwater may cause soil pollution and seriously affect residents' health. Although covering with colored steel plates (tin plates) has been shown to prevent the leakage of asbestos fibers to some extent, it is believed that a policy alternative to remove the slate roof as soon as possible is needed to solve the fundamental problem.

      • 토양환경지도 자료를 이용한 콩밭 토양의 화학성 비교

        It is important to interpret the soil chemical properties in order to effectively manage the farmland. This study was conducted to investigate the soil chemical properties of upland soybean field according to the soil series at 32 upland fields located in the parts of Gangwon-do. Soil sampling sites were selected by using the National Statistics DB and KSIS DB that is a soil environment information system provided by the Rural Development Administration. Soil samples were collected from 0~15 cm of top soil before seeding or transplanting, air-dried, passed through a 2 mm sieve, and analyzed for soil pH, EC, organic matter, available phosphorus (P2O5), exchangeable potassium, exchangeable calcium and exchangeable magnesium. The average chemical properties have exceeded the each nutrient optimum range level of RDA, except the organic matter in experimental soil 2015. And despite application of conventional fertilization, each soil series nutrient contents were change respectively. The results of this study suggest that conventional fertilization should be applied based on the analysis about the amount of accumulated nutrient contents in the soil.

      • 권운과 대기의 복사수지: 열대해양에서의 권운의 모사 및 광학적 성질

        The spectral data obtained from Infrared Interferometer Spectrometer(IRIS) have been simulated in order to examine optical features of cirrus clouds in the tropical oceans. The sensitivity experiments of upwelling intensity to several optical and meteorological factors have been done using the models of the Mie and multiple scattering. The intensity in the experiments is highly sensitive to the particle size and ice-water content (or optical depth) of clouds, while it is less sensitive to their phase and altitude. From theoretical simulation of the infrared spectra the tropical cirrus clouds generally have a two-layer vertical ()tratification in particle size, with larger particles(∼12㎛)in their bottom and smaller ones(∼6㎛) aloft. The vertical structure of these clouds differs from those of the cirrus clouds in polar regions, in that the polar cirrus clouds have an upper layer of smaller particles(≤1㎛). Among 400-1,250() infrared region, the scattering effect due to cloud particles is strong in the relatively short wavelength of 1,050-1,250() portion, while the absorption effect is predominant in the other spectral region. The forward scattering due to the cloud particles for upwelling intensity is the strongest in the large cloud particle size and optical depth. In addition, the scattering effect in the forward direction of water particles generally is a little stronger than that of ice. The cirrus clouds which have been distributed globally and extensively are very persistent and play an important role in the earth's climate by influencing the heat balance and transport in the earth-atmosphere system.

        정신분열병의 임상적 고위험군의 특징 : '서울청년클리닉'의 자연 추적 관찰 예비연구

        Objectives : The aims of this study were to identify the intake and following process for subjects at high risk of transition to schizophrenia and to examine the neurocognitive, social, and emotional functions of the subjects compared with healthy controls. Methods : Symptomatic individuals judged at high risk for schizophrenia ('clinical high risk') within Youth Clinic were assessed and followed up. They performed a neurocognitive function test, a social function test and an affective function test. Twenty healthy controls were recruited in this study. Results : Among eighty-two subjects contacted through the Youth clinic, sixteen subjects were judged as the clinical high risk group. Fourteen subjects among the clinical high risk group showed deficits in several domains of neurocognitive functions, such as visual recall memory, verbal short term memory and executive function. Social and affective functions are also impaired in the Cli-nical high risk group compared with healthy controls. Two of 15 subjects (13%) developed a psychotic disorder within 6 months. Conclusion : This study illustrates the state of follow-up study for a clinical high risk group. Despite low numbers and short durations, some impairment of several functions in the clinical high risk group suggests possible predictors of psychosis.

        Gastritis cystica profunda (GCP) is a disease characterized by hyperplastic and significant extension of cystic dilatation of the gastric mucous glands, which results in a cystic lesion in gastric submucosa. It often occurs on the part of gastroenterostomy, but can be found in the stomach without any previous surgery. GCP has variable gross finding including solitary polyps, diffuse ones, submucosal tumors, and rare giant gastric mucosal fold. It is difficult to tell GCP from a cancerous lesion by gross finding that the disease demands a necessary tissue biopsy, though GCP is commonly showed as a benign in the progress. We report a case of GCP in polypoid types diagnosed by endoscopic polypectomy.

        To produce reliable and high quality software, software quality assurance is very important in relation to software evaluation process. One of the key issue to be considered in quality evaluation is how to achieve objectiveness and quantitative measurement in evaluation process. A typical evaluation algorithm relies on experts' assured. In this paper, we propose an alternative algorithm for generating weight value on evaluation criteria which represents the degree of importance among evaluation characteristics conforming to ISO9126 standards. Especially, Dempster-Shafer theory is used to enumerate the hierarchical relationship among quality characteristics, and thus generate quantitative value from qualitative information such as evaluator's judgement or knowledge.

        혈청이 마우스 간 세포주 BNL CL.2의 Nitric Oxide 생성에 미치는 영향

        마우스 간 세포주인 BNL CL.2의 시험관내 배양에서 혈청과 IFN-γ가 세포주의 nitric oxide(NO) 생성과 세포 손상에 미치는 영향을 알아보기 위한 실험을 하였다. 혈청이 공급된 배양에서 IFN-γ에 의한 세포 생존율은 거의 변동이 없었으나, 혈청을 제거한 배양에서는 약 65%의 생존율이 유지되었으며, NO 생성 억제제인 N -monomethy-L-arginine (NMA)의 첨가는 농도 의존적으로 세포의 생존율을 감소시켰다. 혈청이 제거된 BNL CL.2세포주는 IFN-γ단독 처리에서도 NO 를 생성할 수 있었으며, IFN-γ와 lipopolysaccharide (LPS)의 복합 처리는 세포주의 NO 생성을 상승적으로 증가시켰다. 또한 protein tyrosine kinase (PTK) inhibitor인 herbimycin A와 genistein에 의해서 NO 생성이 억제되어 PTK의 활성이 혈청이 고갈된 BNL CL.2 세포에서 NO의 생성에 중요한 역할을 담당하고 있기 때문으로 판단된다. IFN-γ의 독성은 혈청을 제거시킬 때 NO 생성 억제제에 상승적으로 간 세포를 손상시키며, 이때 NO가 IFN-γ에 의해 유도된 손상을 어느 정도 억제시키는 것을 알 수 있었다. Nitric oxide (NO) plays an important role in immunologic defense, and influences upon the functioning of secretory tissues and cells. It also exhibits cytotoxic/cytostatic activity as one of major operating effectors of the cellular immunity system. We investigated the effects of serum on the cell damages and NO production in the mouse liver cell line BNL CL.2 to establish the role of NO. We observed that, when BNL CL.2 cells were cultured in serum-free medium, they were induced to cell damage by the stimulation of IFN-γalone or IFN-γplus LPS. Serumstarved cells showed large amount of nitrite accumulation and NO synthase (NOS) expression in response to IFN-γalone in dose-and time-dependent manners, but serum-supplied cells did not. The production of NO was blocked by protein tyrosine kinase (PTK) inhibitors, genistein and herbimycin. These results suggest that the deprivation of serum in the BNL CL.2 cell culture medium might primed with the cells to produce NO when the cells are triggered by IFN-γ and the involvement of PTK signal transduction pathway in the expression of NOS gene in murine hepatocytes.

        치조골 복합골절의 정복고정시 탈구치아의 치근관을 통한 배농술의 효과

        Treatment of compound alveolar bone fracture consists of reduction and fixation with wound closure of soft tissue laceration. For the proper reduction and fixation, interosseous wiring or splinting of the involved teeth are applied during about 4 weeks. But wound infection around the fracture site may be occurred, because of the presence of devitalized tissues(pulp, bone and soft tissue), residual hematoma and anergy effect of trauma stress. The wound infection results in bone resorption, pulp degeneration and necrosis, wound dehiscence and soft tissue necrosis around the fracture site. Therefore, if the infection will be expected, early proper drainage should be established via the fracture site and root canal of the luxated teeth. The authors treated two cases of compound alveolar bone fracture by the rubber drainage in the fracture site and the endodontic drainage by open canal of the luxated teeth, with the usual reduction and fixation by use of arch bar and resin. The prognosis was more favorable without wound infection, bone resorption, teeth mobility and specific root resorption.
