        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        무청시래기를 첨가한 추어 떡갈비의 개발 및 품질특성

        이번 연구는 남원식 추어탕의 주재료인 추어와 무청시래기에 대한 인식개선과 이를 이용하여 젊은 연령층의 기호도에 적합한 분쇄육 제품을 개발하고자 추진되었다. 떡갈비 주원료인 돈육과 우지의 15%를 추어 페이스트로 대체한 추어 떡갈비를 대조구로 사용하였으며, 대파의 일정비율(25%, 50%, 75%, 100%)을 무청시래기로 대체하여 추어와 무청시래기가 첨가된 떡갈비 시료를 제조하였다. 무청시래기를 첨가한 떡갈비는 가열·조리 후 pH와 당도는 각각 6.07-6.28과 9.94-10.9로 시중에서 유통되는 떡갈비의 개별 특성에 적합하였다. 가열·조리에 따른 가열감량은 대파를 사용한 대조구에 비하여 대파를 무청시래기로 대체한 떡갈비 시료에서 유의적으로 낮았지만 무청시래기의 첨가량에 따른 유의적인 차이는 없었다. 가열 전과 가열·조리 후 무청시래기를 첨가한 떡갈비의 수분함량은 모두 대조구 보다 높았으며 이는 무청시래기의 식이섬유가 육가공품의 보수성을 높여 가열감량을 감소시킨 것으로 판단되었다. 대파를 무청시래기로 대체한 떡갈비 시료의 명도(44.5-47.8)는 시료 간의 유의적인 차이를 보이지 않은 반면 적색도(0.77-4.97)와 황색도(6.92-12.8)는 무청시래기의 첨가량에 따라 유의적으로 감소하였다. 떡갈비 시료에 대한 TPA 결과, 부착성은 대파를 무청시래기로 대체한 떡갈비 시료에서 대조구보다 유의적으로 낮아졌으나 경도, 응집성, 검성, 씹힘성에서는 대조구와 무청시래기를 첨가한 시료 간에 유의적인 차이를 확인할 수 없었다. 무청시래기가 첨가된 떡갈비 시료에 대한 관능적인 평가에서 색, 연도, 다즙성에서 대조구보다 낮은 점수를 받았다. 전반적인 기호도 역시 무청시래기가 첨가되지 않은 대조구에서 가장 높은 점수를 받았으나 무청시래기를 첨가한 시료에서는 대파의 50%를 대체한 시료에서 높은 점수를 받았다. 무청시래기 자체가 지닌 특유의 향미는 관능적인 평가에서 확인되지 않았으며 전자코를 이용한 향기패턴 분석에서도 낮은 분별지수(-90)로 시료 간의 향기패턴 차이를 확인하기 어려운 수준이었다. 이번 연구를 통해 떡갈비의 제조에서 대파를 대체하는 무청시래기의 함량은 25-50%가 적절하였으며 무청시래기와 추어가 지닌 특유의 맛과 냄새로 인한 영향을 받지 않는 떡갈비를 개발함으로써 향후 남원지역의 향토음식과 연계되는 새로운 메뉴개발이 가능하게 되었다. This study was performed to develop tteokgalbi suitable for young-aged individuals by using mudfish and radish greens, major ingredients of Chueo-tang. Mudfish-tteokgalbi (control), in which 15% of the meat and tallow was replaced with mudfish paste, was used while substituting 25 to 100% of the green onions with radish greens. The pHs and sugar contents of the tteokgalbi developed in this study were 6.07-6.28 and 9.94-10.9, respectively, and it was suitable for tteokgalbi sold in the market. Replacing green onions with radish greens while manufacturing tteokgalbi significantly reduced the color (a and b values) and weight loss during cooking compared to the control. Replacing green onions with increasing quantities of radish greens also significantly reduced the adhesiveness of the samples in the texture profile analysis. Although the overall acceptability of the control was the best, the ideal quantity of radish greens as a substitution for green onions appears to be 25-50%. The unique flavor of the supplemented radish greens was indistinguishable in the sensory evaluation and the principal component analysis with an electronic nose. Finally, radish greens were confirmed as a good ingredient for manufacturing tteokgalbi together with mudfish paste.

        요로결석 환자에 대한 응급실에서의 진단방법

        Nephrolithiasis is one of the most common disease seen in the Emergency Department. Among the patients who visited in the Emergency Department with acute abdominal pain, many people were suspected urolithiasis. Because of limitation of diagnostic method on Emergency Department we planned this study in order to confirm the our accuracy of diagnosis and the possibility of misdiagnosis. During 5 months from April 1996 to August 1996, the 189 patients was suspected the urolithiasis. Among this patient, 51 cases was excluded due to incomplete study. After performing P/Ex, U/A, KUB in the Emergency Department, they were followed out patients department of Urology and perfomed Intravenous Pyelogram. The sensitivity and specificity for KUB were 84.5% and 94.0%, respectively. The positive predictive value was 93.8%, the nagative predictive value was 93.8%. Diagnostic accuracy of urolithiasis in our Emergency Department is 89.1%. There was no relationship between urinary occult blood and KUB or IVP. There was no relationship between degree of occult blood and diagnosis of urolithiasis. For differential diagnosis of urolithisis, emergency ultrasound and IVP will be available. So combination of this diagnostic methods will improve accuracy and safty of diagnosis and treatment of urolithiasis.

        노인 인구에서 신경인지기능저하와 혈청 지질농도와의 상관성

        Objectives : In this study, the authors evaluated the correlation between eurocognitive impairments and serum lipids levels among Korean elderly over the age of 65. Methods : A total of 609 elderly individuals participated in this study. Screening for cognitive impairments were carried out using the Mini-Mental Status Examination-Korean version (MMSE-KC). There were 197 subjects above 1.5SD Of MMSE- KC and they were evaluated using the Korean versions of the Consortium to Establish a Registry for Alzheimer's Disease (AD) (CERAD-KC) and Geriatric Depression Scale (GDS-K). Results : There was a correlation between old age/low levels of education and AD (p<0.05, P<0.01). There was an inverse correlation between the serum level of total cholesterol and the word list recognition test scores. The serum levels of LDL cholesterol were inversely correlated with the scores on the constructional praxis test, word list recall test and word list recognition test. Inverse correlations between the serum level of triglyceride and scores on the word list recall test and word list recognition test were also observed. Conclusion : There were inverse correlations between the serum levels of lipids and language/memory function in subjects with AD.

        비아그라??복용 후 발생한 급성 심근경색증

        Erectile dysfunction(ED) is defined as the inability to achieve and/or maintain an erection which is sufficient for satisfactory sexual activity. Risk factors for ED include many of the same risk factors for cardiovascular disease: hypertension; lipid abnormalities, including low high-density lipoprotein cholesterol; diabetes; and smoking. Therefore, some patients with ED will have ischemic heart disease. Viagra□, an oral therapy for ED, is the citrate salt of sildenafil, a selective inhibitor of cyclic guanosine monophosphate(cOMP)-specific phosphodiesterase 5(PDE5). The adverse effects of sildenafil include headache, flushing, dyspepsia, nasal congestion, visual disturbance, and cardiovascular complications. A general impression exists that cardiovascular complications of sildenafil occur mainly when the drug is taken concomitantly with nitrate. We report the case of a 52-year-old man who developed an acute myocardial infarction shortly after taking one tablet of sildenafil without sexual activity. The patient had not been using nitrates. Physicians treating patients with chest pain should be aware of the potential consequences of giving nitrates to patients taking sildenafil. We suggest that before initiating therapy, the history of sildenafil ingestion be obtained from patients with suspected myocardial infarction.

      • Fractal을 이용한 多孔質 硅素의 微細孔 形成 課程에 관한 硏究

        단결정 실리콘(P형)을 양극 산화반응으로 다공질 규소를 제작하고 AFM과 이차원 DLA 모델을 사용한 컴퓨터 모의 실험으로 표면구조를 조사했다. 컴퓨터 모의 실험에는 양공의 확산거리 D를 변수로 사용했다. AFM 측정 자료와 컴퓨터 모의 실험을 비교해본 곁과 전하 밀도 lmA/cm2에 해당하는 확산거리는 약 100nm였으며 그 때 프랙털 차원은 1.85였다. 또한 반응전류 10 mA/cm2에 해당하는 확산거리는 약 150nm의 결과가 얻어졌으며 그 때의 프랙탈 차원은 1.75이다. 이러한 컴퓨터 모의 실험 결과와 AFM을 통하여 관찰한 p-PS의 표면 구조를 비교해보면 양극 반응 전류와 양공의 확산 길이는 비례 관계가 있다. Surface structures of porous silicon are investigated by AFM and a qualitative theoretical model based on 2 dimensional DLA model is used to explain their fractal structures. The diffusion length L is used as a starting parameter, which sets the distance of a hole which can reach to a tip of a pore. The best result is obtained when the diffusion length was 100 nm (200 lattice distance). In this condition, the average pore distance is about 650nm, which corresponds to lmA/cm2 current density in the experiments and its fractal dimension is 1.85. Our results verify that a diffusion length L is proportional to the current density used in an experiment.
