Reading Performance Improvement in PDF-417 2-dimensional Bar code

As life style becomes to be complicated, lots of supportive technologies were developed. The bar code technology is one of them. It was renovating approach to merchandise industry. However, data storage ability of one dimensional bar code is limited because of growing the number of merchandise.. Two-dimensional bar code was proposed to overcome one-dimension bar code. PDF-417 bar code most commonly used in standard two-dimension bar codes has good performance at data decoding and error correction area. An algorithm to improve reading performance with PDF-417 is suggested. The threshold value is computed, and bar and space are extracted according to threshold value. PDF-417 is used everyday and is printed in many different materials. The printed PDF-417 is especially influenced by various light sources. It is hard to recognize the bar and space separation under the circumstance of changing illumination. The fixed threshold value to distinguish the bar and space can not be applied. The proposed algorithm is developed to investigate the variable threshold. The variable threshold can be obtained by simple calculation.

To evaluate the effect of 6-month short term antituberculosis chemotherapy with INH, Rifampin, Ethambutol, and Pyrazinamide(2HREZ/4HRE) in the patients with pulmonary and/or extrapulmonary tuberculosis, the authors prescribed 2HREZ/4HRE regimen in 79 tuberculosis patients for 6 months with measuring the sputum staining for AFB, chest X-ray findings, recurrence rates and possible side reactions of the treatment. The result were as follow; 1. Pulmonary Tuberculosis 1) Among the 56 pulmonary tuberculosis patients who had taken 2HREZ/4HRE regimen. 32 patients showed initial positivity in sputum AFB smear stain(57.2%). Negative conversion occurred usually within 2 months after initiation of chemotherapy and the mean period of negative conversion was 1.4 months. 2) Among the 56 pulmonary tuberculosis patients, chest X-ray finding changed in 41 patients (37.21%). From these 41 patients 39 patients showed continuous improvements in chest X-ray finding, though 3 patients showed initial aggravation in spite of continuous medication. The remainder 2 cases aggravated due to the failure of treatment. 3) Treatment failure occurred in 2 patients (3.57%) during the chemotherapy among 56 patients of pulmonary tuberculosis and they were infected with secondary drug-resistant strains of M. tuberculosis. 2. Extrapulmonary tuberculosis. Among 8 patients with tuberculous pleurisy and 15 patients with superficial tuberculous lymphadenitis, there were no evidence of treatment failure after completion of antituberculosis chemotherapy for 6 months with 2HREZ/4HRE regimen. 3. Follow-up study was performed from 6 months to 50 months after completion of antituberculosis chemotherapy and the relapse was not noted in both pulmonary and extrapulmonary tuberculosis patients group during this period. 4. Serum AST/ALT elevated in 9 patients(11.4%) during the treatment and this occurred usually within 3 months after the initiation of antituberculosis chemotherapy. However treatment interruption occurred in 2 patients (2.5%) due to the development of hepatitisone due to drug-induced hapatitis and the other due to type B viral hepatitis. In conclusion, we could find this 2HREZ/4HRE 6-months short-term antituberculosis regimen is effective and could be recommanded as a promising regimen for the treatment of tuberculosis.

This study was designed to exam the aged's needs for spiritual nursing care. The purpose was to serve as a basis for the development of spiritual nursing practice. The major findings are as follows : 1. The degree of needs for spiritual nursing care as area was that needs of love and relationship mean 22.0, needs of meaning and object mean 28.2, needs of forgiving mean 13.5. Total needs for spiritual nursing care mean 63.7, which was on the upper middle level. The needs of meaning and object was rated highest. 2. Among the general characteristics of the subjects, needs of love and relatiohship wasn't significanlty different. 3. Among the general characteristics of the subjects, needs of meaning and object was significantly different according to two factors : age(F=7.260, p=0.001), religion(F=5.275, p=0.001). Higher needs of meaning and object was possessed by the older than the younger, by the one who have religion than the other. 4. Among the general characteristics of the subjects, four factors made a significantly difference to needs of forgiving : sex(t=-2.851, p=0.006), age(F=8.201, p=0.001), religion(F=6.928, p=0.000), disease(t=2.327, p=0.024). Higher needs of forgiving was possessed by man than woman, by the older than the younger, by the one who have religion than the other, by the one who have disease than the other.

FOXA2 Overexpression Promotes Hepatic Differentiation of Adipose Tissue Derived Stem Cells

Aims: We used non-viral vector gene delivery system and three dimensional culture using scaffold to increase the efficacy of hepatocyte differentiation. Methods: The adipose derived stem cells (ADSCs) were isolated from human adipose tissue. Using microporator, Foxa2 was over-expressed in ADSCs and cultured in poly-lactic-co-glycolic acid (PLGA) scaffold. Later, for hepatic differentiation ADSCs were pre-cultured in IMDM with 20 ng/mL epidermal growth factor and 10 ng/mL fibroblast growth factor-basic (bFGF). After 2 days, the cells were cultured in Step-1 differentiation medium consisting of IMDM with 20 ng/mL hepatocyte growth factor (HGF), 10 ng/mL bFGF, and nicotinamide 0.61 g/L for 7 days. Finally, the cells were cultured in Step-2 differentiation medium consisting of IMDM with 20 ng/mL oncostatin M, 1 mol/L dexamethasone, and 50 mg/mL ITS+ premix for 7 days. For in vivo experiments, Foxa2 over-expressed ADSCs cultured in IMDM with 20 ng/mL EGF and 10 ng/mL bFGF and were loaded in scaffold for 1 day and later were implanted in nude mice dorsum for 2 weeks. After 2 weeks the scaffold was retrieved from mice. After differentiation, periodic acid-Schiff (PAS) staining, and qPCR gene evaluation were performed. Results: After hepatic differentiation, Foxa2 over-expressed ADSCs increased the gene expression of hepatocyte-specific gene markers (alpha- fetoprotein [AFP], Cytokeratin 18 [CK18], Albumin[ALB]) in 2D cultures. In 3D cultures, Foxa2 over-expressed ADSCs increased the gene expression of hepatocyte-specific gene markers (AFP, CK18). In 2D, 3D cultures, Foxa2 over-expression increased glycogen storage ability in hepatic differentiated cells. In vivo experiments showed that after 2 weeks scaffold implant, the mRNA expression for AFP and CK18 increased in foxa2 over-expressed group while ALB expression remained the same as in vector group. Moreover, PAS staining was more pronounced in Foxa2 group. Conclusions: Foxa2 promotes hepatic differentiation by increasing AFP, ALB and CK18 expressions in 2D, while increasing AFP, and CK18 expressions in 3D culture and in vivo system.

The utility of a biodegradable hyaluronic acid (HyA)-based hydrogel as a tissue scaffold for intravitreal carriage of mesenchymal stem cells (MSCs) into the retina was tested. A rat model of retinal ischemia-reperfusion (IR) injury was used. DiI-labeled MSCs from three passages were loaded onto hydrogel and injected into the vitreous body of experimental and control eyeballs 1 week after IR. The neutral hydrogel was also modified to a basic pH. A control carrier, 0.01 M phosphate-buffered saline (pH 7.2; PBS) was compared. Retinal tissues were prepared 1 week and 2 weeks after MSCs application. MSCs localization and effects were evaluated on immunostained retinal preparations with confocal microscopy. MSCs localization was apparent in the retinas loaded onto hydrogels, adhering tightly to the inner limiting membrane, whereas those in PBS floated in the vitreous body. Nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) were expressed in the end feet of M?ller cells in the normal retina. NGF expression was slightly reduced 2 weeks IR, had expanded into the proximal processes 3 weeks IR, but it appeared reversely 1 week and 2 weeks after MSCs application, respectively. BDNF expression was higher in the ischemic and MSCs-treated ischemic retinas than in the normal and MSCs-treated control retinas, respectively. These findings demonstrate that HyA-based hydrogel is an efficient vehicle for intravitreal MSC transplantation into the retina, and that MSCs thus transplanted induce M?ller glial cells to produce growth factors concerned with the survival of retinal ganglion cells.
Plug-in Repressor Library for Fine-tuning of Metabolic Flux in Escherichia coli

In metabolic engineering, enhanced production of value-added chemicals requires precise flux control between growth-essential competing and production pathways. Although advances in synthetic biology have facilitated the exploitation of a number of genetic elements for precise flux control, their use requires expensive inducers, or more importantly, needs complex and time-consuming processes to design and optimize appropriate regulator components, case-by-case. To overcome this issue, we devised the plug-in repressor libraries for target-specific flux control, in which expression levels of the repressors were diversified using degenerate 5′ untranslated region (5’ UTR) sequences employing the UTR Library Designer. After we validated a wide expression range of the repressor libraries, they were applied to improve the production of lycopene from glucose and 3-hydroxypropionic acid (3-HP) from acetate in Escherichia coli via precise flux rebalancing to enlarge precursor pools. As a result, we successfully achieved optimal carbon fluxes around the precursor nodes for efficient production. The most optimized strains were observed to produce 2.59 g/L of 3-HP and 11.66 ㎎/L of lycopene, which were improved 16.5-fold and 2.82-fold, respectively, compared to those produced by the parental strains. These results indicate that carbon flux rebalancing using the plug-in library is a powerful strategy for efficient production of value-added chemicals in E. coli.
