        Multi‑tissue lipotoxicity caused by high‑fat diet feeding is attenuated by the supplementation of Korean red ginseng in mice

        Background Excessive intake of fat, one of the causes of obesity, is associated with low-grade infammation in various susceptible organs and eventually causes tissue toxicity. This study examines the multifaceted suppressive efects of Korean red ginseng extract (KRG) on high-fat diet (HFD)-induced lipotoxicity and infammatory responses in the aorta, liver, and brain. Methods Male C57BL/6 mice were fed HFD with or without KRG for 12 weeks. The improvement efect in KRG on lipotoxicity and infammatory potential was determined in the blood and the aorta, liver, and brain tissues. Results KRG signifcantly inhibited 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity by >20% in vitro. KRG supplementation suppressed HFD-associated body weight gain, lipid profle changes, and excessive fat deposition in the liver and increased leptin, insulin, and ALT levels in the blood. Infammatory markers in the aorta, liver, and brain were also signifcantly reduced by KRG treatment. In microvascular endothelial cells, the 15% cyclic stretch-mediated upregulation of ICAM-1 and vascular cell adhesion protein-1 (VCAM-1) expression was signifcantly attenuated in the presence of KRG. Conclusion KRG supplementation attenuates HFD-mediated body weight gain, lipid profle changes, and multi-tissue infammatory responses.

      • 상피성장인자가 치주인대세포 및 치은섬유아세포의 증식능에 미치는 영향

        성장인자 가운데 미분화중배엽세포의 활성을 조절하는 것으로 알려진 상피성장인자를 초기배양한 치주인대세포와 치은섬유아세포에 각기 다른 농도와 시간에 따라 주입하였을 때 두 세포의 세포증식능에 미치는 영향을 알아보고 각 조건에 따른 두 세포간의 증식능을 상호 비교해 보고자 본 실험을 실시하였다. 교정치료를 목적으로 내원한 환자의 제 1 소구치 부위의 정상치은을 절제하고, 건강한 제 1 소구치를 발거하여 치은섬유아세포와 치주인대세포를 분리, 배양하여 상피성장인자를 주입시키지 않은 군을 대조군으로 하고 상피성장인자를 각각 1, 10, 50, 100, 200ng/ml로 실험군으로 하여 24시간, 48시간, 72시간 동안 배양하였으며, 각 시간별 배양 6시간전에 10μl/200 ml 5-Bromo-2'-deoxy-uridine을 첨가하여 5-Bromo-2'-deoxy- uridine이 DNA내로 편재되는 양으로써 두세포군의 증식능을 측정하여 다음과 같은 결과를 얻었다. 치은섬유아세포에 상피성장인자를 주입후 24,48,72시간 동안 배양시 각시간별로 농도의존형으로 증가하는 경향을 보였으며 48시간까지 세포의 증식능이 증가 하였다가, 72시간 배양시에는 24,48시간 배양시에 비해 증식능이 전반적으로 감소하는 경향을 보였다. 24시간 배양시에느 1,10Πg/ml를 제외한 모든 실험군에서, 48시간 동안 배양시에는 1Πg/ml를 제외한 모든 실험군에서, 72시간 배양시에는 1, 10, 100Πg/ml를 제외한 모든 실험군에서 대조군에 비해 통계적으로 유의한 차이(P<0.01,P<0.05)를 나타내었다. 치주인대세포에 상피성장인자를 주입후 24,48,72시간 동안 배양시 24시간에서는 농도의존적으로 미약한 증가를 보이다가 200Πg/ml에서 감소하는 경향을 보였으며, 48시간에서는 농도의존적으로 증가하다가 72시간에서는 24시간과 유사한 양상을 보였다., 24, 72시간 배양에서는 48시간 배양에 비해 증식능이 감소하는 경향을 보였다. 24,48,72시간 배양시 모두에서 1Πg/ml를 제외한 모든 실험군에서 대조군에 비해 통계적으로 유의한 차이(P<0.01,P<0.05)를 나타내었다. 치주인대세포와 치은섬유아세포의 증식능을 비교해 보면, 24시간 배양시에 모든 실험군에서 치주인대세포가 치은섬유아세포보다 조금 더 낮은 경향을 나타애었고, 두 세포 상호간에도 모든 실험군에서 통계적으로 유의한 차이 (P<0.01)를 나타내었다. 48시간 배양시에는 모든 실험군에서 치주인대세포가 치은섬유아세포보다 증식능이 더 높은 경향을 나타내었으며, 두 세포 상호간에 통계적으로 유의한 차이 (P<0.01,P<0.05)는 나타내지 않았다. 72시간 배양시에는 1Πg/ml까지 치주인대세포가 더 낮은 증식율을 보이다가 10Πg/ml이상에서는 치주인대세포가 치은섬유아세포보다 그 증식능이 더 높은 경향을 나타내었으며, 두 세포 상호간에 50,200Πg/ml를 제외한 모든 실험군에서 통계적으로 유의한 차이(P<0.01,P<0.05)를 나타내었다. Epidermal growth factor(EGF) is one of polypeptide growth factors. EGF has been reported as a biological mediator which regulates activities of wound healing process including the cell proliferation, migration and metabolism. The purposes of this study is to evaluate the effects of EGF on the human periodontal ligament cells and human gingival fibroblast cells that promote regeneration of periodntal tissue. The mitogenic effects of epidermal growth factor on human periodontal ligament cells and human gingival fibroblasts were evaluated by dentermining the incorporation of 5-Bromo-2'-deoxy-uridine into DNA of the cells in a dose dependent manner. The prepared cells were the primary cultured gingival fibroblast and periodontal ligament cells from humans, the fourth or sixth subpassages were used in the experiments. Cells were seeded in DMEM containing 10% FBS. 1, 10, 50, 100, 200Πg/ml and epidermal growth factor were added to the quiescent cells for 24 hours, 48 hours and 72 hours. They were labeled with 10μl/200μl 5-Bromo-2'-deoxy-uridine for the last 6 hours of each culture. The results of the five determinants were presented as mean and S.D.. The results were as follows: The DNA synthetic activity of human gingival fibroblasts were increased dose dependently by epidermal growth factor at 24 hours, 48hours and 72 hours. The mitogenic effects were similar at the 24 and 48 hours of epidermal growth factor, but the DNA synthetic afctivity of human gingival fibroblasts generally decreased at 72 hours. The DNA synthetic activity of human periodontal ligament cells were increased dose dependently by epidermal growth factor at 24 hours but the DNA synthetic activity decreased at 200Πg/ml of each hour. Generally the maximum mitogenic effects were observed at the 48 hours application of epidermal growth factor. The DNA synthetic activtity of human periodontal ligament cells generally decreased lower at 24,72 hours than at 48 hours the application of epidermal growth factor. In the comparison of DNA synthetic activity between human gingival fibroblasts and human periodontal ligament cells, human periodontal ligament cells had slightly higher proliferation activity than human gingival fibroblasts for a longer time at the high dosage of the epidermal growth factor. In conclusion, epidermal growth factor have important roles in the stimulation of DNA synthesis in human periodontal ligament cells and human gingival fibroblasts, and thus may be useful for clinical applications in periodontal regenerative procedures.

      • Retinoic acid에 의해 유발된 흰쥐의 구개열에서 Transforming Growth Factor-beta 1, 2, 3의 발현의 변화

        Objective : Craniofacial malformations are among the most frequent congenital birth defects in humans : cleft palate, that is inadequate fusion of the palatal shelves. Retinoic acid is teratogenic in many species, producing multiple malformations, including cleft palate. Also it plays an important role in embryogenesis, by regulating morphogenesis, cell proliferation, differentiation, and deposition of the extracellular matrix. The effects of retinoic acid which leads to cleft palate vary depending on the stage of development exposed. After exposure of embryonic mice to retinoic acid on gestational day(GD) 10, abnormally small palatal shelves form. And the elevation of palatal shelves is delayed. The growth and differentiation factor, transforming growth factor-beta(TGFβ) family is thought to play important roles in multiple developmental processes, especially, the development of the embryonic palate. The expression of the TGFβ family in embryonic palate suggests a functional role for these molecules. In this study, author has determined the tissue distributions of TGFβ family in murine embryonic palate, and compared them in terms of developmental stage and locations in normal and retinoic acid-induced cleft palate. Methods and Material : Retinoic acid was administered orally at gestational day(GD) 10 to ICR mice. The pregnant mice were sacrifeced on GD 12, 13. 14, 15 to obtain the fetuses and immunohistochemistry was performed. And using the scanning electron microscope, morphology of normal palate and cleft palate was studied. Results : In the retinoic acid-treated fetuses, palatal shelves did not elevate and cleft palate was induced. On GD 13 in the palates of the retinoic acid-treated fetuses, expression of the TGFβ1, 3 decreased dramatically, but that of TGFβ2, did not change. The expression of the TGFβ1, 3 in the palates of the retinoic acid-treated fetuses, increased on GD 14, 15, Significant differences in TGFβ family expression were observed with areas of the palate. Conclusion : These results suggest that retinoic acid inhibits the expression of TGFβ 1, 3, which is critical in the developing palate, and elevation of palatal shelves is delayed and impaired, therefore cleft palate is induced. These data demonstrate a role for retinoic acid and retinoic aicd-induced TGFβ family in the regulation of palatal development and suggest a role for TGFβ in retinoic acid-induced cleft palate.

      • 정상인과 혈액질환 환자에서의 혈소판 평균용적치에 관한 연구

        Molphologic study of platelet have been done by many authors for the past years, they investigated that the heterogeneity of platelet size was shown even in normal person and various hematologic disorder. In the past the platelet size was not utilized in clinical medicine because of its technical difficulties of measuring. Recently, and automated blood cell counter, Coulter counter Model Splus Ⅱ®, was introduced and platelet size could be easily gained, which expressed by mean platelet volume(MPV). Therefore, this study was planned to evaluated the clinical significance of MPV in health person and various hematologic disease. The following results were obtained: 1. In healthy person, MPV was decreased along with increasing the platelet number. 2. In thrombocytopenic patients, MPV was decreased more than normal. 3. In various hematologic disease, MPV was decreased in acute leukemia, marrow failure and aplastic anemia, normal in iron deficiency. anemia, increased in reactive marrow, MPV was varied by platelet number in various hematologic disease. This disclosed that MPV was related with marrow function and production of megakaryocytes. From these results, MPV could be used as a valuable parameter in the evaluation of platelet together with measuring the platelet number. Particular relation-ship between MPV and various hematologic disease were not observed. We should be more study for MPV.

        GC/NPD를 이용한 다성분 잔류농약의 동시분석

        Pesticides were extracted from samples with 70% acetone and methylene chloride in order, and then cleaned up via open-column chromatography apparatus packed with fiorisil, and finally analyzed simultaneously the organophosphorus pesticides using GC/NPD, Ultra-2 and Ultra-1 fused silica capillary columns were used to separate and identify the products. Recovery of most analytes from soybean sample, taken from pesticide residues well, was greater than(80%) for all except(6) analytes. This method can simultaneously determine multiple pesticides with a high degree of accuracy and precision.

      • 응력경로에 따른 섬진강 모래의 탄성특성

        Usually, Many Engineers have used predicting the settlement and displacement using linear elastic modulus(E_(50) calculated by the conventional triaxial test results. But it is results are very diffemt from those of field, which follows deffent stress path and nonlinear behaviour. Therefore, to investigate the dependence of stress path with the variation of confining pressure and relative density on Sum-Jin River sand, local strain triaxial tests are preformed. It is showed as follows ; For the same stress path condition, the stiffness increased with increasing confining pressure and relative density, especially it is more affected by confining pressure. From the relations between Young's modulus and axial strain showed all nonlinear-elastic behaviour and independent on testing conditions, such as, confining pressure, relative density and stress path, etc. The mount of stiffness at very small strain range showed very high value and linear elastic behaviour. And compression results are rather overestimated than that of extention.

        유산소 리듬 운동프로그램이 노인의 혈압, 심박수 및 생활만족도에 미치는 효과

        The purpose of this study was to test the effects of aerobic rhythmical exercise program on blood pressure, pulse and the life satisfaction of the elderly. This quai-experimental study was designed as a non-equivalent control group pretest-posttest study. The subjects were 36 subjects(experimental group: 19 subjects, control group: 17 subjects), aged between 65 and 73 years who have normal cognition, sensory function and cerebellum function. Aerobic rhythmical exercise program was developed on the basis of aerobic exercise, Korean tradititional dance and music by the author. The blood pressure, pulse rate and life satisfaction were measured prior to and following the experimental treatment. Data were analyzed with x^2-test, t-test, mean, standard deviation, percentage of change and ANCOVA test using SAS program. The results were as follows. 1. There was no significant difference of systolic blood pressure between experimental and control group following the aerobic rhythmical exercise program(F=1.39, P=0.247). 2. The diastolic blood pressure of the experimental group was significantly lower than that of control group following the aerobic rhythmical exercise program(F=2.96, p<0.1). 3. There was no significant difference of resting heart rate between experimental and control group following the aerobic rhythmical exercise program(F=0.07, P=0.793). 4. The life satisfaction of the experimental group was significantly higher than that of control group following the aerobic rhythmical exercise program(F=9.94, p<0.01). On the basis of the above findings, aerobic rhythmical exercise program is effective in reducing diastolic blood pressure and promoting life satisfaction of the elderly. Thus, this program can be recommended as an effective nursing intervention for the elderly.
