        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        가평 전주이씨묘 출토복식 고찰

        In 1995, the costumes were restored in an unknown woman's tomb at the Jeonju Lee's family cemetery in Gapyeong, Gyeonggi-Do, and there are currently nine items remaining. In this study, we first introduced these 9 relics to academia and analyzed the morphological characteristics of the costumes. We also estimated the time and person of burial compared with costumes unearthed from other burials. Jeogories (short jacket) are all four items. One of these items was unusual in shape, and the upper part of the seop (gusset) was wrinkled and shaped. This type of Jeogori appears only till the 1520s in other tombs. There are also two skirts, one of which is characterized by a superimposed pattern. The top and bottom / middle part of the skirt were rolled up, and the skirt, which was double-rolled up in this way, was first discovered. An analysis of the shape of Jeogori and skirt indicated that the person buried in the Jeonju Lee's family tomb was believed to have survived until the early 16th century. Based on these estimated periods, as a result of looking at the genealogy of Jeonju Lee's family, it was concluded that the tomb was probably a woman named Lee Geum Myeong (李金命) born in the mid-15C and died between the late 15C and the early 16C.

      • 남산공원 이용객의 시간대 및 요일에 따른 이용행태조사

        The purpose of this research was to analyze pattern of utilization as a time slot and a day of week in Nam-san park visitors. To achieve the purpose of this study, 500 persons in Nam-san park visitors. Total of 436 subjects were utilized for data analysis Primary statistical methodology for data analysis were frequency test and crosstabulation. The results of this as follows; First, 50.2% of visitors answered' satisfaction' about the whole satisfaction. Seond, 25.6% of visitors answered sightseeing about Nam-san park's importance. Third, there are significant differences depending on time table. On the morining and night times, there are increase in use of health improvement facilities. On the morning and night times, there are increases in use of health improvement facilities. Forth, there are significant distinctions about facilities preferences depending on days of the week, also. During the weekday user are preferred health improvement facilities and during th weekend, they preferred sighting or nature appreciation.       • 공정 Al-Si 합금의 개량처리에 따른 조직관찰 및 기계적 특성에 관한 연구

        주조용 Al 합금(AC4B)을 주단조법에 적용하기 위해 먼저 Sr과 TiB를 첨가하여 주조성 및 단조성이 우수한 주단조 소재를 개발하고 그 기계적 특성을 평가하였다. 먼저, 중력주조시 조대하게 정출하는 침상의 공정 Si를 개량처리하기 위해 공정 Si 개량 화제인 Sr 모합금을 0.025~0.1wt.%로 변화시켜 첨가한 결과 침상의 공정 Si이 미세한 섬유상으로 변화한 것을 관찰할 수 있었으며, 0.075wt.%일 때 가장 우수한 개량화 효과를 나타내었다. 그러나, Sr을 첨가하지 않은 상태에서 균일하게 관찰되던 α-Al상이 Sr 첨가로 인해 조대한 수지상으로 형태가 변화한 것을 관찰할 수 있었다. 따라서, α-Al 고용체상을 미세화하기 위하여 Sr이 0.075w1.% 용해된 알루미늄 합금 용탕에 AlTi_{5}B 모합금을 0.05~0.5wt.%로 변화시키면서 첨가한 결과 α-Al 고용체상이 미세화됨을 관찰할 수 있었으며, 0.1wt.%일 때 가장 우수한 미세화 효과를 나타내었다. 한편, 개량화 및 미세화 처리한 시편의 기계적 특성 평가를 위해 인장시험과 내마모시험을 실시한 결과 인장강도, 신율 및 내마모성이 주방상태에 비해 크게 향상되는 것으로 나타났다. Recently, many studies have been carried out to process on the purpose of lightness in a transport parts because of the saving energy, the environmental problem. The cast-forging process can be expected to lower costs without decreasing the mechanical properties. So, the finest microstructure is needed to get for applying the cast-forging process with Al-Si alloy because the microstructure affects to the cast-forging process. For refinement treatment of eutectic Si and a-Al solid-solution phase, Sr and TiB were added in A-Si alloys. The finest microstructure could be observed when 0.075wt.%Sr and 0.lwt.%TiB were added respectively. In this case, tensile strength and elongation much more increased than as casting.

      • 토양환경지도 자료를 이용한 콩밭 토양의 화학성 비교

        It is important to interpret the soil chemical properties in order to effectively manage the farmland. This study was conducted to investigate the soil chemical properties of upland soybean field according to the soil series at 32 upland fields located in the parts of Gangwon-do. Soil sampling sites were selected by using the National Statistics DB and KSIS DB that is a soil environment information system provided by the Rural Development Administration. Soil samples were collected from 0~15 cm of top soil before seeding or transplanting, air-dried, passed through a 2 mm sieve, and analyzed for soil pH, EC, organic matter, available phosphorus (P2O5), exchangeable potassium, exchangeable calcium and exchangeable magnesium. The average chemical properties have exceeded the each nutrient optimum range level of RDA, except the organic matter in experimental soil 2015. And despite application of conventional fertilization, each soil series nutrient contents were change respectively. The results of this study suggest that conventional fertilization should be applied based on the analysis about the amount of accumulated nutrient contents in the soil.

        골수구성 백혈병 환자에게 발생한 결핵성 림프절염

        혈액질환 가운데 결핵 발생이 증가한다고 알려진 것은 호지킨 림프종을 포함한 림프증식성질환 그리고 모양세포성 백혈병 등이 있다. 또한 동종 골수이식을 받은 환자에서도 결핵은 증가한다고 알려져있다. Kaplan 등은 골수구성 백혈병 환자에서 결핵 발생이 증가한다고 보고하였다. 그러나 골수구성 백혈병 환자에서 결핵의 발생이 증가하는 이유에 대해서는 아직까지 정확하게 밝혀지지 않았다. 저자들이 2년 동안 후향적으로 조사한 180명의 성인 골수구성 백혈병 환자들 가운데 결핵이 발생한 환자는 4명이었고, 발생 부위는 모두 림프절이었다. 결핵은 골수구성 백혈병의 특정 아형에만 국한되어 발생하지 않았지만, 50%에서 FAB 분류에 의한 M4였다. 림프절이 종대된 골수구성 백혈병 환자에서 특히, 결핵의 유병율이 높은 나라에서는 결핵성 림프절염도 감별진단에 포함시켜야 한다. During the neutropenic phase, leukemia patients receiving chemotherapy are prone to bacterial and, fungal infections; occasionally mycobacterial, viral and protozoal organisms may also cause infections. Mycobacterium tuberculosis infection was reported very rarely in these patients. This report describes four patients with M. tuberculosis infection identified from 185 adult patients who were diagnosed myelogenous leukemia between January 2003, and December 2004. There was no patient with M. tuberculosis infection from 44 lymphoid leukemia and 11 acute biphenotypic leukemia patients. Sites of infection were all lymph nodes. Three among four patients were presented with lymphadenopathy at initial diagnosis of leukemia, and the other one presented with lymphadenopathy after induction chemotherapy. There was no patient presented with lymphadenopathy during the neutropenic phase. Tuberculous lymphadenitis was presented in a patient with three acute myelogenous leukemia (FAB class 2 M4, 1 M2) and a chronic myelogenous leukemia, accelerated phase. An acute myelogenous leukemia patient had a leukemic cell and tubercle bacilli in the same lymph node. Tuberculosis should also be included as a differential diagnosis in myelogenous leukemia patient with lymphadenopathy, especially in the countries in which the disease is endemic.

      • 초등 실과 「재봉틀로 용품 만들기」단원의 프로젝트 수업의 적용 효과

        The purpose of this study is to search instruction necessity of the section of 「making an article with a sewing machine」in Elementary Practical Art and discuss the application effects of Project Learning Model selected as teaching-learning method to raise educational effect. For griping instruction necessity of the section of 「making an article with a sewing machine」, this searcher saw over educational contents of terminal curriculum related to clothing living and searched educational meaning of a sewing machine instruction in elementary school. Then, he researched educational condition of a sewing machine instruction in Busan's school and experience and ability of teacher, parents, students' using a sawing machine. And then, he inspected effect of Project Learning applied to two class, deemed as characteristically similar, out of 6th grade at D elementary school in Busan. In the result, First, the section of 「making an article with a sewing machine」had instruction necessity as meaningful education activity to raise an ability of making article needed in the living with creative planning by learning skills and knowledge of handling a sewing machine. So it set up to improve own aptitude as students have various practical experiences through this course and to have sound attitude about a work and job related to clothing living. In conclusion, this section have to instruct as meaningful education activity in elementary school, because the purpose of this section is to raise creative attitude and skills which is needed to deal with the future's change and the real living. second, in the result of application of Project Learning to 「making an article with a sewing machine」, experimental group's average were upper than comparative group's average. This indicated that the former group with the learning - based on Project Learning had better academic accomplishments related to practical cognition than the other with not. But for there was not meaningful difference in the test of affective and functional level, application effects of Project Learning was not effectful in this section. Because of regarding as this is due to limitations and conditions of practical environment, a profound study about this present situation will have to be later on.

      • 濟州道內 高等學校 物理敎育 正常化에 關한 硏究

        In this stuay we investigated the status of physics education at high schools in Jeju-do and measured the experimental experiences of students graduated from high schools. We collected the data from 20 science or physics teachers and 383 freshmen in natural science classes in Jeju national college. The results of the investigations are as follows. (1) Most students and school administrators thought physics as an unimportant subject, and none of the students selected physics as an optional subject among those offered as natural sciences. (2) Experimental apparatus was not sufficiently availabie in each school and the students hardly made any physical experiments in the high schools. (3) The mean teaching hours assigned to physics teachers were nearly 21 hours per week. Many teachers were charged two or more subjectsin addition to physics. (4) Most teachers thought that an effective physics education was possible only through experimental study, but the lack of experimental apparatus made it impossible. Given these data we suggested that : (1) The necessity of physics education should be explained to the students and school administrators. (2) The teaching methods should be changed from those traditional ones to methods based on experiments and observations. (3) The physics teachers and school administrators should make an effort to improve all of the experimental equipment. (4) The physics teachers must be rid of their overload, and henceforth teach only their own major subjects.

        보육교사의 아동학대에 대한 인식 및 발견경험에 대한 연구

        The purpose of this study was to find the preschool teacher's awareness on child abuse which is defined by physical abuse, neglect, emotional abuse and sexual abuse. Also, it was to find the preschool teacher's awareness on child abuse report. The results of this study were as follows: First of all, the preschool teachers' awareness on child abuse showed a resonable low-points to all abuse patterns. Especially, the preschool teachers' awareness is lower the behavior syndrom than the physical syndrom on abuse. The next, for one year ago, experience which found out abused child was 56 of 284 teachers. And preschool teachers' awareness on the needs of the report center and report system showed mostly(97.1%) that need. For the last time, the awareness on the education experience of child abuse showed that 50% of the preschool teachers educated properly. Many of the preschool teachers(96.8%) showed that they had a great deal of education experience that they could find out child abuse during teaching. Also, many of the teachers(94.8%) showed that participate in education.
