      • 김치 유산균인 Lactobacillus sp. JJ-2001이 생산하는 Bacteriocin에 관한 연구

        The microorganisms producing bacteriocin from kimch were screened, and among them,a strain showing the highest bacteriocin activity and stability was isolated. Based on the taxonomic characteristics of its morphological, cultural and physiological properties, the isolatef strain was characterized as Lactobacilius sp..To identify optimal conditions for the growth and the production of bacteriocin from the isolated strain,nutrient source, initial pH, and culture temperature were evaluated. Physiochemical properties of bacteriocin was also investigated. The potimal condition for the production of bacteriocin and the cell growth was obtained at 2% glucose, 1% beef extract as organic nitrogen source,1%(NH_4)_2SO_4 as inorganic nitrogen source,and 0.1% Tween 80. The isolated strain showed the maximum growth at 25℃. The inhibitory range of its antimicrobial substance aginst gram positive bacteria was marrow and its inhibitory activity was maintained between pH3.0 and pH8.0. The antimicrobial activity of bacteriocin from the isolated strain was stable at 50℃.

      • 간세포암종에 대한 간동맥 화학색전술 후 발생한 급성호흡곤란증후군

        Transcatheter arterial chemoembolization (TACE) has become an effective alternative treatment strategy for patients with inoperable hepatocellular carcinoma (HCC). Although TACE is relatively safe, acute respiratory distress syndrome associated with pulmonary lipiodol embolism is a rare and potentially fatal complication. We report a rare case of acute respiratory distress syndrome after TACE for inoperable HCC. A 75-year-old man, with huge HCC in right lobe, was treated by TACE for the first time. Seven hours after uneventful TACE procedure, he felt dyspneic and his oxygen saturation recorded by pulse oximetry (SpO2) fell to 80% despite of applying non-rebreathing mask. He underwent mechanical ventilation with a protective ventilatory strategy. We experienced a case of acute respiratory distress syndrome after TACE for HCC.

      • 무산소성 운동이 혈청 지질 농도 및 면역세포 반응에 미치는 영향

        The purpose of this study was to determine serum lipids and immune responses on the anaerobic exercise. Serum total cholesterol, HDL-C, LDL-C and plasma triglyceride, percentages of WBC and their subsets, natural killer cells were evaluated between before and after the anaerobic power. All subjects were high trained male athletes (19-24 years of age). Results were expressed as mean and standard deviations. One-way ANOVA repeated measure was used to compare the all parameters between before and after the anaerobic power test. All significant differences were set at α=.05. In this study, comparison of anaerobic power(Nm/%BW) was higher in soccer players than ssirum and judo players (p<.05). The percentage of WBC, monocyte, eosinophil and basophil were no significant differences between before and after anaerobic exercise. There were increased on neutrophil, lymphocyte after anaerobic exercise in three groups. (p<.01, p<.001, respectively). Also, there were significant differences on T cell, B cell, NK cell between before and after anaerobic exercise (p<.001).

      • 구조물의 비접촉 비파괴 검사를 위한 레이저 초음파법 적용

        The defects evaluation of the interior and the surface would be considered as vital characteristics in predicting the total life span of the steel structure. More importantly, the understandings in the interior composite of welding zone and the notifications in the presence, the formation, and the positioning of the non-metallic inclusion are necessary as well, since there were signs of relatively high defect frequency presented in the welding zone. The ultrasonic testing is a highly recommended technique chosen from among other techniques because of variety of advantages in conducting the non-destructive testing for the welding zone. However, the ultrasonic testing had technical disadvantages referred as followings; the problems due to the couplant between the PZT and the specimen, the formations that were miniature and complex, the moving subject, and the high temperature surrounding the specimen. This research was conducted to resolve the technical disadvantages of the contact ultrasonic testing by studying the non-contact ultrasonic testing where the ultrasonic waves were transferred by the laser, and revealing the specimen defects at its interior part and its surface part. The ultimate goal of this research was to develop a non-destructive evaluation applying the laser manipulated ultrasonic method for the steel structure

        자유학기제 연구학교의 수업과 교육활동의 특징 분석

        본 연구에서는 2013년도에 실시한 자유학기제 연구학교 42개교의 운영 보고서를 분석함으로써 시험이 없는 교육의양상을 살펴보았다. 연구결과 자유학기제는 첫째, 공통과정 기본교과 수업방식의 대부분이 학생주도적인 활동으로이루어지고 있었다. 둘째, 진로탐색 활동이 다양화되고 심화되었다. 셋째, 자율과정은 주입식 교육에서 벗어나 다양한 활동을 제공하고 있었다. 넷째, 학교는 지역사회와의 인적․물적 자원과 연계 지도 인프라 구축을 통해 연계 지도를 하고 있었다. 반면, 자유학기제의 여러 가지 문제점도 발견할 수 있었다. 우선, 자유학기제의 차별화된 교육활동은 중학교 1학년내, 그리고 학년 간의 교육계획에 있어서 혼란을 야기하고 있었다. 또한, 자유학기제 해당 학생들에게만 편중된 교육을 제공하여 타 학년과의 위화감과 소외감도 조성하고 있었다. 그리고 진로탐색 활동의 경우 지적인 탐색과 진로 검사와상담 ,체험을 통한 체득 등 탐색하고 체득하는 활동에만 편중되어 있었다. 끝으로, 자율과정의선택 프로그램 활동, 동아리 활동등 다양한 범주들이 중복되고 비체계적이라는 문제가 발견되었다. 이러한 문제점을해결하고자 본 연구에서는 첫째, 교육과정 계획 단계부터 운영에 이르기까지 교사들이 공동으로 연구하여 자유학기제를 운영하는 학기와 일반 학기의 격차를 줄여야 한다. 둘째, 자유학기제 시행으로 인한 긍정적인 효과들을 타 학년에도적용할 필요가 있다. 셋째, 진로윤리교육의 확대를 통해 균형 잡힌 진로탐색 활동 전개를 주문하였다. 넷째, 자율과정 중 성격과 내용 면에서 서로 중복된 범주들을 체계적으로 정비하여야 할 필요가 있음을 강조하였다 This research analyzed the reports on Free Semester System of 42 pilot schools, which do not have any assessment, in order to inspect the new ways of instructions and educational activities. The results are as follows; First, most of the basic subjects instructions on common courses were student-centered activities. Second, career exploration activities were diversified and enhanced. Third, optional courses provided various activities which fitted students’ interests and needs. Fourth, all of the educational necessities were provided for the pilot schools by the local community. However, Free Semester System’s new curriculum causes some confusion in the operation of educational plans, not only for first grade students but also second and third grade students. nbalanced new educational experiences among Free Semester System students and other grade students cause a sense of incongruity and alienation. In addition, career exploration activities put too much emphasis on intellectual exploration, career-test and career-counselling, and career related experience. Finally, optional courses programs were often redundant and required organization. uggestions to solve these problems are as follows; First, School staffs should build the school curriculum together from planning to management in order to decrease the gap between Free Semester System and general semester system. Second, the positive effect of Free Semester System should be applied to other grades to solve the sense of incongruity and to increase the quality of education. Third, career exploration activities need to be balanced by increasing the learning of vocational ethics. Fourth, optional courses have to be re-arranged to be more systematic. If additional research is conducted in accordance with the suggestions made by this research, Free Semester System will regain the authentic educational role of middle school, which is different from that of junior high school, and will play a role as the bridge of lementary school’s basic education and high school’s career-based education.

      • 초음파 비파괴 검사기법에 의한 용접결함 분류성능 비교

        In this study, we made a comparative study of backpropagation neural network and probabilistic neural network and bayesian classifier and perceptron as shape recognition algorithm of welding flaws. For this purpose, variables are applied the same to four algorithms. Here, feature variable is composed of time domain signal itself and frequency domain signal itself. Through this process, we comfirmed advantages/disadvantages of four algorithms and identified application methods of four algorithms.

      • Chitosan을 이용한 초지공장의 폐수처리

        파지를 이용하여 판지·골판지 등을 제조하는 공장의 폐수는 대부분 콜로이드상으로 분산되어 있고 그 양이 대단히 많아 수질오염의 원인이 되므로 화학적 처리에 의한 현탁입자의 제거가 요구된다. 오징어의 뼈로부터 얻은 Chitosan(β-D-Glucosamine)을 이용하여 초지공장폐수의 응집효과를 실험하였다. 정제·건조된 Chitosan 0.5g을 0.5%의 CH₃COOH수용액에 녹여 응집제로 사용하였다. 응집시험은 Jar Test에 의해 최적 응집조건(온도, pH, 응집제의 양)을 결정했다. 최적조건을 결정하는 데 탁도, SS, 침강시간, COD, 탈수시간, Floc의 크기를 관찰하였다. 응집시헙은 22℃∼25℃에서 행하였으며 시험결과 Chitosan은 pH가 6일 때 탁도, SS, 침강시간, 탈수시간, Floc의 크기가 양호했으며, Chitosan의 양이 30ppm일 때 가장 적합했다. Chitosan과 Acrylamide계 응집제를 pH 6에서 공용했을 때는 응집제의 양이 40ppm에서 적합했다. 천연고분자인 Chitosan은 기존의 Acrylamide계 응집제와 응집비교한 결과 응집효율이 좋았으며, 무독, 무해하여 수질 환경개선에 기대된다. The wastewater from manufacturing industry which produce paperboards by using remnants of paper is discharged in a great quantity, in the dispersal colloidal state, and required chemical treatment to remove suspended solids which can cause water contamination. Flocculation experiments were done by using chitosan(β-D-Glucosamine) obtained from bones of squids. Refined and dried chitosan of 0.5g was dissolved in 0.5%CH₃COOH and used as a flocculant. Optimum condition of flocculation(temperature, pH, content of flocculant) was determined through jar test. Turbidity, content of SS(suspended solids), settling time. COD, dewatering time, and size of floc were investigated to find optimum condition. Experimental temperature was 22℃∼25℃. As a result turbidity, content of SS, settling time, size of floc, and dewatering time were improved in the process of flocculation when pH=6. It showed optimum dosage when the content of chitosan was 30ppm. When chitosan and acrylamide were used together as a flocculant. it showed optimun condition when the content of flocculant was 40ppm at pH 6. Natural polymer, chitosan was more effective than acrylamide in the process of flocculation, because it is nonpoisonous and harmless, it is expected to improve water pollution.

      • 결핵군 55 kDa 항원의 면역학적 분석

        Identification and characterization of mycobacterial antigens are critical for evaluation of their role in diagnosis, vaccination, and pathogenesis of mycobacterial diseases. An attempt has been made to immunologic characterize the 55 kDa protein antigen of Mycobacterium tuberculosis H37Rv which attracted our interest because it is present in high concentration in 50-80% ammonium sulfate fraction of the culture filtrate. Two monoclonal antibodies (MAbs) directed to 55 kDa antigen were produced. MAbs MT55-1 and MT55-2 reacted with a single 55 kDa protein band. On examination of degree for cross-reactivity with other mycobacterial species by enzymelinked immunosorbent assay (ELISA) and immunoblotting, these antibodies reacted strongly with M. tuberculosis and M. bovis BCG, and reacted weakly with M. marinum and M. smegmatis. To investigated the subcellular distribution of MAbs defined epitopes in the 55 kDa antigen within the mycobacterium, we isolated three major subcellular factions of M. tuberculosis, namely, cell wall, cytoplasmic membrane, and cytosol, by a simple fractionation procedure. MAb MT55-1 reactive cpitope was found in the cytosol when tested by immunoblotting. A sandwich ELISA was initially developed for detecting 55 kDa antigen using MT55-1 MAb in mycobacterial culture filtrate before detecting it in clincal specimens. The minimal detectable concentration was 1.0 ㎍/m1 for M. tuberculosis culture filtrate and 100 ㎍/ml for sonic extracts of M. bovis BCG and M. marinum, respectively. But the 55 kDa antigen was not detected in sonic extracts of other mycobacterial species examined. Although further evaluations are required, this study suggests that the 55 kDa antigen may be of interest as potential diagnostic reagent.
