      • Curcumin protects retinal pigment epithelial cells against oxidative stress via induction of heme oxygenase-1 expression and reduction of reactive oxygen

        <P><B>Purpose</B></P><P>To determine whether curcumin induces expression of the defensive enzyme heme oxygenase-1 (HO-1) and protects cells against oxidative stress in cultured human retinal pigment epithelial cells.</P><P><B>Methods</B></P><P>Effective concentrations and toxicities of curcumin were determined after 3 h of curcumin treatment with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Confluent human retinal pigment epithelium cell lines (ARPE-19) were preincubated with curcumin and oxidatively challenged with H<SUB>2</SUB>O<SUB>2</SUB>. HO-1 expression was determined with western blot analysis. To confirm the protective role of HO-1 in oxidative stress, small interfering RNA (siRNA) against HO-1 or inhibitor of HO-1 was treated with curcumin in retinal pigment epithelium cells. Intracellular levels of reactive oxygen species (ROS) were measured with flow cytometry and confocal microscopy. Apoptosis was evaluated with Annexin V-fluoroscein isothiocyanate staining.</P><P><B>Results</B></P><P>Curcumin had little cytotoxicity at concentrations less than 30 μM, and HO-1 expression was the highest at the 15 μM concentration. At this concentration, curcumin also increased the cytoprotective effect against the oxidative stress of H<SUB>2</SUB>O<SUB>2</SUB> through the reduction of ROS levels in human retinal pigment epithelial cells. Curcumin’s effect on the reduction of ROS was mediated by the increase in HO-1 expression.</P><P><B>Conclusions</B></P><P>Curcumin upregulated the oxidative stress defense enzyme HO-1 and may protect human retinal pigment epithelial cells against oxidative stress by reducing ROS levels.</P>

        청국장에 관한 연구(Ⅰ) : 청국장 제조과정에 있어서 콩단백질의 변화에 대하여 On the changes of soy - bean protein in manufacturing Chung - Kook - Jang

        As a series on the soy-bean protein and their related substances 9 samples were collected from 9 places such as straws (Rice) to obtain bacterial strains which produce protease. From these samples total of 23 strains were isolated by the use of dilution pour plate method. For all isolated strains primary screening of productivity of protease was performed and useful straines with regard to protease productivities were identified. Optimum conditions for enzyme action of protease from isolates D_9, F_(20) strains were pH 7.5 and 40℃. Chung-Kook-Jang is one of the characteristic foods in Korea made from soy-bean by fermentation. The chief bacterium is Bacillus subtilis and the chief change which takes place in soy-bean during fermentation is degradation of protein. Three kinds of Chung-Kook-Jang were prepared using three different strains of Bacillus natto, D_9 and F_(20) from isolated. Water soluble-N, TCA soluble-N, amino-N and peptide-N were measured about the steamed soybean, Chung-Kook-Jang prepared with three strains of bacteria. Water soluble-N decreased very largely in steamed soybean, but in Chung-Kook-Jang it increased to 85% of raw soy-bean.

        신규 방사성 항암제 DW-166HC 의 소핵시험

        DW-166HC (^(166)Holmium (^(166)Ho)-Chitosan complex) is a new radiopharmaceutic anticancer agent with a broad anti-tumorigenic spectrum, especially against human hepatic cancer. DW-166HC was evaluated for the appearance of micronucleus in polychromatic erythrocytes (PCEs) of mouse bone marrow cells after subcutaneous arid intravenous single administration. Bone marrow cells were prepared at 24 hr and 48 hr after DW-166HC-I (^(165)Ho-Chitosan complex : cold compound) administration and at 24 hr, 72 hr and 2 weeks after DW-166HC (^(166)Ho-Chitosan complex : hot compound) administration. The results showed there was no statistically significant increase of the numbers of PCEs with micronucleus in all DW-166HC-I administered groups compared with a negative control group but there was statistically significant increase of the numbers of PCEs with micronucleus at 24 hr arid 72 hr in all DW-166HC administered groups, which was recovered after 2 weeks from the drug administration. The results also showed the ratio of normochromatic erythrocytes (NCEs) to PCEs of all DW-166HC-I administered groups was not significantly different from that of a negative controi group but there was significant difference of this ratio at 24hr and 72 hr in all DW-166HC administered groups compared with that of negative group, which was also recovered after two weeks from the drug administration. These results suggested that DW-166HC-I may not cause any chromosomal damage but DW-166HC has in vivo mutagenic potential because of its radioactivity.

      • 백서에서 신 허혈성 손상에 미치는 칼슘 길항제의 효과

        Renal ischemia is one of the most common causes of acute renal failure. Four factors related to the pathogenesis of acute renal failure are vasoconstriction, decreased glomerular filtration rate, tubular back leak of filtrate, and intratubular obstruction. The cellular response to renal ischemic insults include decreased content of adenosine trihosphate, lipid peroxidation induced membrane degradation, alteration in cellular pH, and calcium or phospholipase induced mitochondrial dysfunction. Much attention has been given to the role of increased cellular calcium as a pathogenetic contributor to cell injury during ischemia. Author studied the protective effects of calcium antagonists on cellular injury during renal ischemia in rat. To investigate the protective role of these agents, author measured the amount of malondialdehyde(MDA) and the enzyme activities of free radical scarvengers-superoxide dismutase(SOD), catalase and glutathione peroxidase from non-pretreated group and calcium antagonists pretreated groups in control, ischemia and reflow subgroups. The results were summerized as follows: 1) The amount of MDA in non-pretreated group was higher in the reflow compared with the control(<p<0.01). But, in all pretreated groups, there was no statistically difference in the amount of MDA. 2) The SOD activity in non-pretreated group was lower in both the ischemia and the reflow compared with the control (P<0.05). But, in both verapamil and trifluoperazine-pretreated groups, there was no statistically difference in the SOD activity. 3) Both catalase and glutathione peroxidase activities in non-pretreated group were lower in both the ischemia and the reflow compared with the control (P<0.05). But in all pretreated groups, there was no statically difference in both catalase and glutathione peroxidase activities. These results suggest that free radical mediated ischemic injury by renal artery clamp in rat can be protected by intraperitoneal pretreatment with calcium antagonists. As trifluoperazine has a protective effect in renal ischemia, the calcium activated calmodulin dependent enzyme may play a role in renal ischemic injury.

      • 비확공성 골수강내 금속정을 이용한 폐쇄성 경골 간부 골절의 치료

        Most of closed fractures of the tibial shaft can be treated successfully with medullary nails. Reamed intramedullary nailing technique has become a good alternative method in treating the close tibial fracture. Unreamed interlocking intramedullary nailing has been employed successfully in the treatment of open tibial fractures. This study was undertaken to evaluate the result of interlocking intramedullary nailing without reaming in the treatment of closed fractures of the tibial shaft. From 1995 through 1998, fifty four cases of the closed, displace tibial fractures were treated by an unreamed intramedullary nailing and the average follow-up period was 1 year and 5 months(ranged from 12 months to 27 months). The results were as follows : 1. Fifty four cases were treated with closed, unreamed nailing technique and the nail which has 1mm smaller diameter than isthmic width of unaffected tibia was choosen. 2. The average time to bony union was achieved at 17 weeks without abundant periosteal b callus formation. 3. Eight tibiae were required the additional procedure such as autogenous bone graft, skin graft or local skin flap. We also have observed the screw breakage of ten tibiae but there were no intraoperative jamming or incarceration and no significant complicaton such as bending or breakage of the nail, shortening or non-union of the fractures. In conclusion, unreamed technique is an attractive method to treat the close, displaced fractures of the tibial shaft. 경골 간부 골절의 치료에 있어서 견고한 내 고정으로 조기 관절운동 및 체중 부하를 통해 골절 유합 및 기능 회복을 도모할 수 있는 골수강내 금속정 고정술이 널리 사용되어지고 있다. 골수강내 금속정술의 장점은 술 후 조기 관절운동과 체중 부하가 가능하여 이환율(morbidity)을 감소시키며 불유합의 발생률이 낮아는 것이다. 경골 간부 골절의 치료에 있어서 비확공성 금속정 고정술은 주로 심한 연부 조직 손상을 동반한 골절 및 개방성 골절의 치료에 적용되어지고 있으며, 대부분의 폐쇄성 경골 간부 골절의 치료로는 골수강 확공 후 골수강을 충만시키는 금속정을 삽입하는 확공정 금속정 고정술이 시행되어지고 있는 경향이다. 저자들은 폐쇄성 경골 간부 골절에 대해서 비확공성 금속정 고정술의 유용성 여부를 알아보기 위해서 동국대학교 의과대학 정형외과학교실에서 치료했던 54례를 평균 1년 5개월 추시하여 골 유합 기간, 합병증의 발생에 대해서 분석하였다. 본 연구에서 비확공성 금속정 고정술시 골 유합 기간은 확공성 금속정 고정술시와 비교하여 유의한 차이가 없음을 알 수 있었으며, 확공을 하지 않음으로써 발생할 수 있는 jamming이나 가는 직경의 금속정을 선택함으로써 우려되는 금속정의 휨, 부러짐 등의 합병증은 발생하지 않았다. 결론적으로 폐쇄성 경골 간부 골절의 치료에 있어서도 비확공성 골수강내 금속정 고정술은 유용한 방법으로 사료된다.

        A 1948 paper by Robert Merton established that certain social events were, to a large extent, influenced by the expectancy effects in different social settings. It wasn’t until 20 years later that Merton’s concept of the self-fulfilling prophecy came to the attention of educational researchers (Clark, 1963; Goldberg,1963). Since the Oak Elementary School experiment, researchers have conducted well over 100 studies relative to teacher expectations. It is also important to note that impressions about a certain student may not be the same for two or more teachers. Futhermore, after an extensive review of the literature, Brophy and Good(1974) concluded that teachers, irrespective of gender, are more negative toward young males, particularly regarding their potential as behavior problems. In a follow-up study(Martinek & Karper, 1984a), it was discovered that the attractiveness cue was more influential in expectancy formation for older age groups than for younger ones. These data suggested that as children get older, attractiveness becomes increasingly related to performance expectations.

      • 151종 생약제 추출물이 B형 간염 바이러스 표면 항원(HBsAg)과 항체(HBsAb)의 결합반응 그리고 HBV DNA polymerase 활성에 미치는 효과 : 한국산 각종 생약제를 이용한 B형 간염치료제의 개발

        세계인구 43억명 중 B형 간염 바이러스 보균자는 2억 1천 5백만명이며 이중 78%인 1억 6천만명 이상이 아시아 대륙에 집중되어 있다. 한국도 B형 바이러스 간염의 침윤지대이다. 본 연구는 B형 바이러스 보균자 혹은 만성간염을 치료할 수 있는 생약제를 탐색할 목적의 일환으로 한국에서 시판하는 생약제 151종을 구입하여 증류수로 추출하고 여과한 후 냉동건조하여 HBs 항원에 대한 결합능, HBV DNA polymerase억제능, tumor necrosis factor(TNF) 유리에 미치는 영향, 그리고 galactosamine 유발 간염에 대한간장보호 작용을 조사하였다. 151종의 생약제중 0.16㎍/㎖ 농도에서 간염 vaccine 내에 함유된 HBs 항원과 결합하는 생약제는 지유, 가자, 복분자, 대황, 정향, 빈량, 산수유, 소목, 마황, 쇄양, 후박, 목과, 양강, 오약, 산사, 목단피 등 16종이었다. 이들 생약제중 대황(Rheum palmatum L. 大黃), 가자(Terminalia chebula R. 訶子), 지유(Sanguisorba officinalis L. 地楡), 복분자(Rubus coreanus M. 覆盆子)는 HBV DNA polymerase 활성도를 비교적 강하게 억제하였으며 빈량(Areca catechu L. 빈랑), 정향(Eugenia caryophyllata T. 丁香), 목과(Chaenomeles japonica L. 木瓜), 산수유(Cornus officinalis S. 山茱萸)는 약하게나마 HBV DNA polymerase 활성을 억제하였다. 또한 HBV DNA polymerase를 억제한 대황, 가자, 지유, 복분자, 빈량, 정향, 마황, 소목, 산수유를 대상으로 TNF 유리에 미치는 영향을 조사한 결과 소목을 제외한 8종의 생약제 모두가 대식세포를 자극하여 TNF를 유리시켰다. 그리고 장차 임상에 사용함을 전제로 HBV DNA polymerase를 억제한 대황, 가자, 지유, 복분자, 빈량, 정향, 마황, 소목, 산수유를 galactosamine 유발 간염(백서)에 투여하여 간기능 검사에 미치는 효과를 조사하였으나 간기능 검사상으로는 특별한 보호작용을 나타내지 못하였으며 생약제 단독 투여로서는 간기능에 대하여 나쁜 영향을 주지 않았다. Hepatitis B virus infection is major cause of acute and chrome hepatitis, cirrhosis of liver, and hepatocellular carcinoma. There are over 200 million carriers of hepatitis B virus in the world, nearly 80 % of whom live in the South East Asia, Korea is also highly endemic area of hepatitis B virus infection. This study was performed to evaluate the effect of extracts of traditional Korean herbal medicines on hepatitis B virus. 151 traditional herbs were extracted, filtered and lyophilized. Using 151 herbal extracts we tested binding capability to the HBs Ag, inhibition of HBV DNA polymerase, release of tumor necrosis factor (TNF), and protection against galactosamine induced liver damage. Among 151 herbs, 16 herbs (Sanguisorba officinalis L., Terminalia chebula R., rubus coreanus M., Rheum palmatum L., Eugenia caryophyllata T., Areca catechu L., Cornus officinalis S., Caesalpinia sappan L, Ephedra sinica S., Cynomorium songaricum R., Machilus thunbergii S., Chaenomeles japonica L., Alpinia officinarum H., Lindera strychnifolia V., Crataegus pinnatifida B., Paeonia suffruticasa A. ) had binding capability to HBs Ag which contained in the commercial hepatitis vaccine, at 16 ug/㎖ of concentratioa Among the above 16 herbs, Rheum palmatum L., Terminalia chebula R., Sanguisorba officinalis L., Rubus coreanus M. significantly inhibited the HBV DNA polymerase activity, and Areca catechu L., Eugenia caryophyllata T., Ephedra sinica S., Caesalpinia sappan L., Chaenomeles Japonica L., Cornus officinalis S. also inibited the HBV DNA polymerase activity but less effective. All of the herbs, except Caesalpinia sappan L., which inhibited the HBV DNA polymerase activity, actively stimulated the mononuclear cells to release tumor necrosis factor(TNF) in whole blood culture system. The herbs which inhibited the HBV DNA polymerase activity, did not show any protective effect in galactosamine-induced hepatitis in rats, but there was no direct toxic effect when herbs were administered alone.

      • 항진균제 알릴아민류의 합성과 생물학적 평가

        For the development of antifungal agents. modification of naftifine which exhibits significant antimycotic activity was performed by replacing the naphthalene ring of it to hetero cyclic rings such as morpholine. benzothiazole. piperidine and pyridine derivatives. The synthesized compounds were tested in vitro antifungal activity against five different fungi with naftifine as a comparative antimycotic molecule. From the biological evaluation two compounds. (E)-N-(3-phenyl-2-propenyl)-N-(4-piperidinylmethyl)amine(3d) and (E)-N-(3-phenyl-2-propenyl)-N-(3-pyndylmethyl)amine(3D) showed relatively noticeable activity(MIC=50㎍/㎖). On the other hand. the other compounds had no activity.

      • 칠절봉 신갈나무(Quercus mongolica)림의 군락구조와 토양환경

        칠절봉 신갈나무림의 군락구조와 토양환경과의 관계를 파악하기 위해 식물사회학적 조사와 토양 환경을 분석하였다. 조사지역의 신갈나무림은 신갈나무군락, 철쭉꽃하위군, 큰개별꽃하위군으로 구분되었으며, 철쭉꽃하위군과 큰개별꽃하위군에서의 방형구당 평균출현종수는 각각 30종(24∼46종), 43종(31∼52종) 이었다. 피복지수치에 의한 계층별 우점순위는 교목층에서 신갈나무(6750.0), 아교목층에서 당단풍(2364.0) 관목층에서 철쭉꽃(988.5), 초본층에서 쌀새(1040.5)가 가장 높게 나타났다. 신갈 나무림의 DBH 분포는 2∼10㎝ 범위의 소경목이 전체의 29.7%를 차지하였으며 전체적으로 정규분포를 나타내어 교란 후 재생과정에 있는 것으로 조사되었다. 또한 후계목의 발달로 인한 극상상태로의 천이가 가능할 것으로 조사되었다. 전반적인 토양환경은 두 하위군 간에 뚜렷한 차이가 없었으며, 큰개별꽃하위군이 철쭉꽃하위군에 비하여 다소 양호한 것으로 조사되었다. 환경구배분석에서 까치박달, 물푸레나무, 함박꽃나무 등은 습한 곳에 분포하고 철쭉꽃, 전나무, 국수나무 등은 건조한 곳에 분포하는 것으로 나타났다. A relationship between the community structure composition and soil environmental factors of the Quercus mongolica forest on Mt. Chiljelbong was investigated from June 1996 to August 1997. The Q. mongolica community was classified into two subcommunity of Rhododendron schlippenbachii and Pseudostellaria palibiniana. The 30(24-46) species per quadrat were appeared in R. schlippenbachii subcommunity and 43(31-52)species in P. palibiniana subcommunity. The dominant sequences on each stratum determined by the coverage index showed that Q. mongolica was the most dominant in tree-1 layer with 6750.0, Acer pseudo-sieboldianum in tree-2 layer with 2364.0, R. schlippenbachii in shrub layer with 988.5, and Melica onoei in herb layer with 1040.5 value. The value of diameter breast height(DBH) showed a normal distribution displaying a highest value in the range of 2∼10㎝(29.7%) in Q. mongolica forest and it was investigated being at restoration phase after disturbance. It also was investigated that the succession to climax state would be possible due to growth of successive tree. Overall, which the soil conditions make little difference between two subcommunities, P. palibiniana subcommunity was favorable than those of R. schlippenbachii subcommunity. It was shown that in an environmental gradient analysis, Carpinus cordata, Fraxinus rhynchophylla, and Magnolia sieboldii were distributed in a mesic area, but R. schlippenbachii, Abies holophylla, and Stepanandra incisa in a xeric area.

        응급진료센터 과밀화와 특별관찰구역의 운영

        Overcrowding of emergency departments is a growing problem. Delays in admitting patients to inpatient units have been reported as a contributing factor to overcrowding. Overcrowding in ED results in many patients not receiving needed care and impairs the ability of the ED provider to meet the needs of those patients that are treated. The effect of the critically ill patients on the overcrowded emergency department make emergency physicians frustrated. So we created the 'special unit for observation' of critically ill patients and managed them continuously and closely. The special unit for observation have 4 physical beds and monitors and 2 ventilators. One nurse and senior ED residents were asigned to the management of the patients in the special unit. 160 patients(3.4% of all ED visits) admitted to special unit during the period of March 1, 1996 to May 31, 1996. we could continue assessment and management of critically ill patients without psychologic strain or burnout generated by overcrowding. We think that special unit for critically ill patients is a very effective alternative to overcrowded ED.
