        A selective cyclin-dependent kinase 4, 6 dual inhibitor, Ribociclib (LEE011) inhibits cell proliferation and induces apoptosis in aggressive thyroid cancer

        <P><B>Abstract</B></P> <P>The RB-E2F1 pathway is an important mechanism of cell-cycle control, and deregulation of this pathway is one of the key factors contributing to tumorigenesis. Cyclin-dependent kinases (CDKs) and Cyclin D have been known to increase in aggressive thyroid cancer. However, there has been no study to investigate effects of a selective CDK 4/6 inhibitor, Ribociclib (LEE011), in thyroid cancer. Performing Western blotting, we found that RB phosphorylation and the expression of Cyclin D are significantly higher in papillary thyroid cancer (PTC) cell lines as well as anaplastic thyroid cancer (ATC) cell lines, compared with normal thyroid cell line and follicular thyroid cancer cell line. LEE011 dose-dependently inhibited RB phosphorylation and also decreased the expressions of its target genes such as <I>FOXM1, Cyclin A1,</I> and <I>Myc</I> in ATC. Furthermore, LEE011 induced cell cycle arrest in G0-G1 phase and cell apoptosis, and inhibited cell proliferation in ATC. Consistently, oral administration of LEE011 to ATC xenograft models strongly inhibited tumor growth with decreased expressions of pRB, pAKT and Ki-67, and also significantly increased tumor cell apoptosis. Taken together, our data support the rationale for clinical development of the CDK4/6 inhibitor as a therapy for patients with aggressive thyroid cancer.</P> <P><B>Highlights</B></P> <P> <UL> <LI> pRB and Cyclin D were expressed high in aggressive thyroid cancer. </LI> <LI> LEE011 suppressed pRB and also decreased the expressions of its target genes in ATC. </LI> <LI> LEE011 induced cell cycle G1 arrest and apoptosis, and inhibited cell proliferation. </LI> <LI> LEE011 inhibited in vivo tumor growth with decreased expressions of pRB and Ki-67. </LI> <LI> We could explain the anticancer effects with the RB-E2F pathway. </LI> </UL> </P>

        벼, 콩 및 채소류에 대한 Mn-54, Co-60, Zn-65, Cs-137의 토양-작물체간 전이계수

        벼, 콩, 상치, 당근, 호박의 가식부위에 대하여 몇가지 토양에 있어서 토양-작물체간 Mn-54, Co-60, Zn-65, Cs-137의 전이계수를 포트 재배에 의한 방사성 추적자 흡수실험을 통하여 조사하였다. 핵종간 전이계수는 거의 모든 경우 Zn-65>Mn-54>Cs-137>Co-60의 순이었다. 콩이 벼보다 전반적으로 한 자리 정도 높은 값을 보였고 채소류의 경우에는 대체로 상치에서 가장 높고 호박에서 가장 낮은 값을 보였다. 강산성 토양에서는 약산성 토양에서보다 전이계수가 훨씬 높았다. 본 조사결과에 입각하여 한국인의 섭식경로 피폭선량 평가에 이용하기 위한 각 핵종의 전이계수치가 작물별로 제안되었다. Soil-to plant transfer coefficients of Mn-54, Co-60, Zn-65 and Cs-137 were estimated for the edible parts of the rice, soybean, lettuce, carrot, and squash grown in different soils by radiotracer uptake experiments using pot cultures. The transfer coefficients of radionuclides were n the order of Zn-65>Mn-54>Cs-137>Co-60 in most of the cases studied. The coefficients for soybean were roughly and order of magnitude higher than those for rice. Among vegetables, lettuce mostly showed the highest value and squash, the lowest. In the strongly acidic soils. From the data obtained, crop-specific transfer coefficients of the four nuclides were proposed for the use in Korean food-radiation dose assessment.

        The Korean Neuropsychiatic Association (KNPA) was founded in 1945 and has now grown to over 2000 members as of 2003. The Association's academic meeting has also been increasing in size every year. To further develop the academic meeting of the KNPA, the task force team conducted this project. We suggest several points based on analysing the abstracts of the meeting that has conducted during the last 5 years, the survey results performed at the 2002 annual meeting, the annual meeting of the American Psychiatric Association, and the academic activities of a Korean medical assoction, the Korean Society of Otolaryngology.

        TDI계 수분산성 폴리우레탄 접착제의 특성에 Diisocyanate의 구조가 미치는 영향

        수년 전부터 많은 연구자들에 의해 폴리우레탄의 수용화, 수분산화의 연구가 수행되어 왔다. 그럼에도 불구하고 폴리우레탄의 특성에 디이소시아네이트가 구조에 미치는 영향에 대한 연구는 많이 진행되지 않았고 또한 미흡한 상태이다. 따라서 번 연구에서는 디이소시아네이트의 형태와 함량 변화에 따른 폴리우레탄의 특성을 연구하였다. 폴리우레1탄 수분산체(PUD)는 poly(tetramethylent ether) glycol (PTMG), isophorone diisocyanate (IPDI), toluene 2,4-diisocyanate (TDI), dimethylol propionic acid (DMPA), trethylamine (TEA) 등을 사용하여 제조하였다. 제조한 폴리우레탄 수분산체(PUD)의 구조를 FT-IR을 이용하여 확인하였다. 폴리우레탄 수분산체의 특성은 DSC, gel permeation chromatography (GPC), particle analyzer, universal testing machine (UTM) 등을 사용하여 측정하였다. 폴리우레탄 수분산체(PHD)에서 입자크기는 IPDI 함량이 증가할수록 작아지는 경향을 나타내었으며, 유리전이온도(Tg)는 IPDI 함량이 증가할수록 조금씩 낮아지는 경향을 나타내었다. 또한, 폴리우레탄 수분산체(PUD)의 접착세기 변화에서는 [NCO]/[OH] 몰비를 1.2로 유지되도록 하면서 합성에 관여하는 [NCO] 양에 해당하는 [TDI]/[IPDI] 몰비를 변화시켜 얻은 폴리우레탄 수분산체의 접착세기는 [TDI][IPDI] 몰비가 50/50일때 좋은 집착세기를 나타내었다. 또한 [NCO]/[OH] 몰비를 1.6으로 일정하게 유지되도록 하고 [TDI]/[IPDI] 몰비를 변화시켜 얻은 폴리우레탄 수분산체의 접착세기는 [TDI]/[IPDI] 몰비가 75/25일 때 좋은 접착세기를 나타내었다. Aqueous polyurethane dispersions (PUD) has been investigated by many researchers for a long time. However, relatively little systematic work has been reported, in detail, on the effect of structure of the diisocyanate on the properties of polyurethane. This work describes the properties of polyurethane in relation to the variation of diisocyanate type and contents. Aqueous polyurethane dispersions (PUD) were prepared from the poly(tetramethylene ether) glycol (PTMG), toluene 2,4-diisocyanate (TDI), isophorone diisocyanate (IPDI), dimethyol propionic acid (DMPA), and triethylamine (TEA). Differential scanning calorimetry (DSC), FT-IR, GPC, universal testing machine (UTM), and particle analyzer were utilized to characterize the physical and mechanical properties of PUD according to diisocyanate type and contents. As the content of IPDI in PUD increased, the particle size decreased. The glass transition temperatures (Tg) of PUT were shifted to a lower temperature as IPDI conent increased. When [NCO]/[OH] mole ratio was 1.2, the adhesion force of PUD shown the best result at [TDI]/[IPDI] mole ratio of 50/50. And, at [NCO]/[OH] mole ratio of 1.2, the adhesion force of PUD was the best when [TDIl/[IPDI] mole ratio was 75/25.

        Abstract - In this thesis, we propose a QoS supporting method on wireless ad hoc-based active network for improving reliability. On the loss of route path, the existing DSR routing protocol has several problems with wireless network communication because of decease of data reception rate and increase of network load. The proposed method improves reliability of data transfer by using functions of active network. That is, by using functions of active nodes and active packets, the proposed method supports QoS with reservation transfer method that saves data packets on transfer at intermediate nodes with lost path and then re-transmit the packets after route reconfiguration.

      • 親和性 크로마토그라피에 의한 大豆β-amylase의 精製 및 特性

        Soybean β-amyase 〔β-1.4-glucan maltohydrolase EC 3.2.1.2 〕has been purified from defatted soybean meal by fractional precipitation with ammonium sulfate, ion ezchange chromatography on CM-Toyopearl 650M,affinity chromatography on γ-CD-sepharose column,and some physicochemical properties of the enzyme were investigated. The purified enzyme showed 50.5 folds of purification , 1,136unit/mg protein of specific activity and 36% of yield,respectively,compared to the curde extract. The enzyme showed a single band on Sodium Dodecyl Sulfate polyacrylamide gel electrophoresis, and its molecular was estimated to be 58,000 daltons by SDS-PAGE. The prified enzyme was identified as homegenous by HPLC analysis and maltose activity assay. The optimum pH and temperature was 5.6and 55℃ ,respentively. The enzyme was stable below 60℃,but was very labile over 65℃ The enzyme was most stable at pH 5.0~6.0, and was more stable in the acidic region than in neutral region. From the Hanes-Woolf plot,the Km value for soluble starch was calculates as 0.05% and Vmax value was 1.3×10²μmol maltose. min??.mg??

      • 도시근교 관광지에서의 쓰레기 배출특성에 관한 조사연구 : The Gu-Gok Falls in Summer Season 하절기 구곡폭포를 중심으로

        This investigation was carried out ot study on the generation property, management and separate collection rate of solid waste in the Gu-Gok Falls. Generation rate of solid waste was 80.785kg/day and unit waste generation rates was 65.55g/dayㆍtourist one person. Physical component of solid waste were garbage 46%, papers 20%, empty bottles 10%, respectively. A plan is required to decrease generation rate of garbage waste, As result to investigate separated garbage can, papers and can were separated more than 60%. But empty bottles and scrap iron were separated less than 50%. P.E.T, steel can, aluminum can and empty bottles were 20% of total generation rate, and profit of reuse is ₩700,800/year.

      • 마찰력과 열처리에 따른 아크용사 및 아크용접된 Fe-Cr-B 합금 피복층의 변화

        Variations of arc thermal sprayed and arc welded Fe-Cr-B alloy coating with frictional loading and heat treatment were investigated by means of X-ray diffraction, optical microscopy, scanning electron microscopy, electron probe microanalysis and hardness test. The as-coated layers were shown to be crystalline state but peaks with the frictional loading largely disappeared in X-ray diffraction profiles. The splat is relatively wavy with the boundary in arc thermal spray coating. After heat treatment, the amount of porosity was found to be decreased and the intersplat distance was increased. As annealing temperature increased, interface layer for arc welding coating increased by the diffusion of Cr atoms. The wear type observed in arc thermal spray coatings is splat delamination. The hardness of the arc thermal spray coation layer was superior to that of the arc welding layer. Hardness of coated layer is reduced and microstructures become to be uniformed by the heat treatment.

        액체-고체 순환유동층에서 액상의 반경 방향 혼합 및 액체-고체 Chaotic 흐름 거동

        직경이 0.102m이고 높이가 3.5m인 액체-고체 순환유동층의 상승관에서 압력요동 및 액체의 반경 방향 혼합 특성을 고찰하였다. 상승관에서 얻은 압력요동을 해석하기 위하여 chaos 이론을 도입하였다. 액체 유속, 유동입자의 크기 그리고 고체 유동입자의 순환속도가 연속액상의 반경방향 분산계수 및 압력요동의 위상공간투영과 상관차원에 미치는 영향을 결정하였다. 본 연구의 결과 액상의 반경방향 분산계수는 유동입자의 크기와 순환속도가 증가함에 따라 증가하였으며 액상의 유속이 증가함에 따라 감소하였다. 상승관에서 압력요동의 위상공간투영은 유동입자의 크기가 고체 유동입자의 순환속도가 증가함에 따라 점점 분산되고 복잡하게 되었으나, 액체유속이 증가함에 따라서는 위상공간에서 궤적의 분산이 다소 감소하는 경향을 나타내었다. 압력요동의 상관차원은 유동입자의 크기와 고체 순환속도가 증가함에 따라 증가하였으나, 액체의 유속이 증가함에 따라서는 큰 변화가 없었다. 본 연구의 결과는, 상승관에서 액상의 반경 방향 혼합 특성 압력요동 측정함으로써 실시간으로 예측하는 것을 가능하게 하여 실제공정에 응용함으로써 그 실용적 측면이 크다고 하겠다. Characteristics of pressure fluctuations and liquid dispersion in the radial direction were investigated in the riser of a liquid-solid circulating fluidized bed whose diameter was 0.102m and 3.5m in height. The concept of chaos theory was employed to analyze the pressure fluctuations obtained in the riser. Effects of liquid velocity, particle size, and solid circulation rate on the liquid radial dispersion coefficient and phase space portraits and correlation dimension of pressure fluctuations were determined. It was found that the radial dispersion coefficient of liquid phase increased with increasing particle size or solid circulation rate, but decreased with increasing liquid velocity. The attractor in the phase space portraits became more scattered and complicated with increasing particle size or solid circulation rate, but it became somewhat less scattered with increasing liquid velocity. The correlation dimension of pressure fluctuations increased with increasing particle size or solid circulation rate, but it did not change considerably with increasing liquid velocity. The results of this study enable us to predict the characteristics of liquid radial dispersion by means of the pressure fluctuations for the practical applications.
