        A study has been performed on the characteristics of rural and urban surface ozone concentration for the period of March 1992 to February 1993. The monitoring station of rural ozone is located at Kosan, Cheju and other urban monitoring stations are located at Seoul, Pusan and Kwangju. Rural's and urban's ozone data exhibit a distinct features in many ways. First, annual mean of rural ozone concentration is very high(42 ppbv) but urban's are very low(10~15 ppbv). Second, rural ozone data shows a seasonal variation with it's maximum in spring, and minimum in summer, but urban's show a seasonal variation with it's maximum in spring, and minimum in winter. Third, diurnal variation of rural data is very small but that of urban's are very large. Fourth, monthly mean of daily minimum of rural data is not low and shows a large seasonal variation, but those of urban's data are extremely low(<3 ppbv) and have no seasonal variations.

      • 제주도 고산에서 수용성 에어로졸의 화학적 성분 분석

        Atmospheric aerosols were collected by a High Volume Tape Sampler from March 1992 to December 1993 at Kosan, Cheju, korea. The water soluble ion concentrations in aerosol were analyzed. The concentrations of cations(Na+, K+, Ca²+, Mg²+, NH₄+) were determined by an Inductively Coupled Plazma(ICP) or an Atomic Absorption Spectrometer(AAS), and those of anions(Cl-, NO₃-, SO₄²-) were analayzed by the capillary electrophoresis method. The Ca²+, SO₄²-and NO₃- concentrations in spring were higher than those in other seasons. The lowest concentrations of these elements were found in summer, largely due to scavenging by frequent rains. Especially the ca²+ concentration on April was three to four times higher than the annual mean concentration. The enrichment factor(E.F.) of each element was calculated. The annual mean E.F. values of the Ca²+, Mg²+ and Cl- in 1992 were the same as those in 1993 except K+ and SO₄²-. The correlation formula between all cations and anions for the whole period was Anions=0.759xCations+0.066.

      • 적혈구침강속도와 급성기물질의 혈중변화에 따른 상관성과 임상적평가

        The authors determined erythrocyte sedimentation rate(ESR) and the level of acute phase reactants such as ceruloplasmin, α1-antitrypsin, haptoglobin and C-reactive protein and evaluated their correlation in 101 patients with active inflammation, chronic infection, collagen disease and neoplastic disease from Jan. 1990 to Aug. 1990 in Chosun university hospital, Shortly after the patients had been diagnosed, the tests was performed using their blood. ESR was measured by standardized Win trobe method. Ceruloplasmin, α1-antitrypsin, and haptoglobin were measured by radial immunodiffusion method. C-reactive protein was measured by turbidimethc immunoassay. The correlation of ESR and the levels of ceruloplasmin, α1-antitrypsin, haptoglobin and C-reactive protein was analysed in the control group and the disease group. The results were summarized as follows. 1. The ESR rose with each decade increase of age in most cases of the disease group. 2. The author judged that the ESR was increased when the ESR was upper than 20mm / hr. The 45 of 101 cases of the disease group showed increase of ESR. So the sensitivity of ESR to detect disease is 44.55%. 3. The difference of the level of ceruloplasmin and α1-antitrypsin of control group and that of the disease group showing normal ESR value was not statistically significant(P>0.05). 4. The difference of the level of C-reactive protein and haptoglobin of the control group and that of the disease group showing normal ESR value was statistically significant(P<0.05). 5. The difference of the level of ceruloplasmin, α1-antitrypsin and haptoglobin of the disease group showing normal ESR value and that of disease group showing high ESR value was not statistically significant (P>0.05). 6. The difference of the level of C-reactive protein of the disease group showing normal ESR value and that of the disease group showing high ESR values was statistically significant (P<0.05). 7. C-reactive protein played a major role in increasing ESR and haptoglobin played some influence also. But α1-antitrypsin and ceruloplasmin played little influence.

      • Southern Blot Hybridization법을 이용한 악성 림프종의 면역유전형에 관한 연구

        Immunogenotyping using gene rearrangement analysis has emerged as a precise laboratory aid in the diagnosis and classification of malignant Iymphoid neoplasms. The lineage and clonality of the malignant Iymphoid neoplasms can be identified by the demonstration of rearrangements of antigen receptor genes of the immunoglobulin and T-cell receptor genes. The analysis of the gene rearrangements on the malignant Iymphoid neoplasms are also useful as a sensitive unique clonal markers to detect early recurrence in patients with malignant Iymphoid neoplasms after treatment. To analyze the sensitivity and specificity of gene rearrangements in the diagnosis of malignant Iymphoid neoplasms. 24 cases of malignat Iymphoma were examined by Southern blot hybridization using CTβ-T cell receptor β chain gene-DNA probe and JH-immunoglobulin heavy chain gene-DNA probe. The results of the immunogenotypings using Southern blot hybridization disclosed high correlation between the immunophenotyping using immunohistochemical stain with monoclonal antibodies (B-cell Iymphoma 84.2%. T-cell Iymphoma 75% ). The analysis of the gene rearrangement of the angioimmunoblastic lymphadenopathy(AILD) and unclassifiable Iymphoma using immunohistochemical stain could resolve the monoclonality and lineage . Rearranged bands to the CTβDNA probe were observed in one case out of 2 cases of AILD. One case of unclassifiable Iymphoma showed rearranged bands to the CTβ DNA probe. There were no rearrangements in reactive follicular Iymphoid hyperplasia and paracortical Iymphoid hyperplasia. In conclusion, DNA gene rearrangement study should be applied to differentiate the clonality and cell lineage in the malignant Iymphoma with indistinctive immunophenotype.

        방사선 조사 마우스에서 소장움세포의 Apoptosis 발생에 미치는 생약의 효과

        Apoptosis의 유발을 억제하는 한약제를 파악하여 apoptosis와 관련된 정상적 또는 질병에 대한 연구에 자료를 제공하고자 방사선에 의해 유도된 apoptosis를 지표로 한의학적 처방에서 많이 사용되는 대표적 한약제(24종)의 효과를 검증하였다. 용안육, 산조인, 원지, 인삼, 복령, 목향, 천궁, 백작약, 승마, 시호 및 눈꽃동충하초 투여군에서 apoptosis는 감소되었으며, 이들 생약제는 apoptosis와 관련된 질병의 예방 및 치료에 적용할 수 있을 것이다. 추 후 이들 생약의 작용 기전에 대한 연구가 요구된다. This study was performed to determine the effect of several herbs on radiation-induced apoptosis in jejunal crypt cells. Longyanrou(Euphoris longana), Suanzaoren(Zizyphus vulgaris), Yuanzhi(Polygala tenuifolia), Rensan(Panax ginseng), Fuling(Poria cocos), Muxiang(Saussurea lappa), Chuanxiong(Cnidium officinale), Baishaoyao(Paeonia lactifolia), Shengma(Cimicifuga heracleifolia), Chaihu(Bupleurum falcatum) and Dongchongxiacao(Paecilomyces japonica) reduced the frequency of radiation-induced apoptosis(p<0.05). Although the mechanisms of this effect remain to be elucidated, these results indicated that Longyanrou, Suanzaoren, Yuanzhi, Rensan, Fuling, Muxiang, Chuanxiong, Baishaoyao, Shengma, Chaihu and Dongchongxiacao might be useful inhibitors of apoptosis, especially since these are relative nontoxic natural products.

      • Emulsion형 化粧品의 Escherichia coli 汚染防止에 대한 防腐制效果

        Most of cosmetics are emulsion products that contain the source of nutrition-vegetable oil, mineral oil and carbohydrate etc. -so there is much possibility of contamination by microbials. The purpose of this study is to get the data necessary not only to prevent dermatopathia occurred by Escherichia coli but to sustain the quality and the wear of cosmetics as the result of finding optimum growth condition and the growth condition of Escherichia coli for enviro-factor, utilizing antiseptic so as to prevent contamination by observing it in emulsion type cosmetic. The results are as follows : 1. The optimum growth condition of Escherichia coli : pH7.0 and 37.5℃ The mixing volume ratio of water / oil was 70/30 in this experiment. 2. Adding antiseptics into emulsion type cosmetic, the number of Escherichia coli was decreased from 10^8(cell/ml) to 8 x 10^3(cell/ml) 3. Antibacterial effect against Escherichia coli was better as follows, p-hydroxy benzoic acid propyl ester + phosphoric acid buffer solution > p-hydroxy benzoic acid buthyl ester + acetic acid buffer solution > p-hydroxy benzoic methyl ester + phosphoric acid buffer solution > p-hydroxy benzoic acid methyl ester + p-hydroxy benzoic acid buthyl ester >p-hydroxy

        급성 일산화탄소 폭로가 백서 뇌조직의 immediate early gene, c-fos의 발현에 미치는 영향

        The c-fos, an immediate early gene homologous to viral oncogene v-fos, controls protein expression of cells as a DNA transcription factor. Carbon monoxide(CO) may induce tissue hypoxia and neurotoxicity and play an important role in cell signal transduction as a retrograde messenger. However, little is known about the effects of CO on expressions of immediate early genes. This study was performed to evaluate the effects of CO on c-fos expression, and to compare the effects to those of hypoxia in rat brain. Ninety male Sprague-Dawley rats were divided into 3 groups (CO, hypoxic, and ambient air groups), and then exposed to 4,000 ppm CO, 10% O₂-90% N₂gas, or ambient air for 30 minutes, respectively. Immediately, 15 minutes, 30 minutes, 45 minutes, 60 minutes, and 120 minutes after exposure, rat brains were excised and cerebral cortexes, corpus striata, hippocampuses, and cerebellums were separated. Northern blots for c-fos were performed, and GAPDH expression levels were checked as an internal control of total mRNA expression. The c-fos band densities increased till 30 or 45 minutes after exposure. and thereafter decreased. The expression levels of c-fos did not increase in hippocampuses and carebellums, but in cerebral cortexes and corpus striata of hypoxic and CO-exposed groups. In corpus striata of the CO-exposed group, further increase of c-fos expression levels was observed in comparison with the hypoxic group. These findings suggest that the effects of hypoxia and CO are different by the anatomical sites in brain, and CO is involved in signal transduction in corpus striatum, but not in cerebral cortex, hippocampus, and cerebellum within 2 hours after exposure to CO.

      • 기능성 고등어 Fillet 제조 및 저장 중 품질 변화

        고등어 fillet 제품의 저장 중 항산화의 기능성을 부여하고 고등어 특유의 비린내를 제거하기 위해 식염 외에 녹차, dillweed, 키토산, 올리고당, 생강을 첨가하여 진공포장한 후 5℃, 0℃, -20℃에 80일간 저장하면서 제품 특성을 조사하였다. 5℃에 저장한 고등어 fillet의 휘발성 염기질소 측정 결과는 이들 첨가물을 단독 또는 복합적으로 첨가한 시제품에서 2주 이내에 초기부패에 도달했고, 0℃에서는 4∼7주, -20℃에서는 저장 80일간 모든 시제품에서 신선도를 유지할 수 있었다. 이들 첨가물 가운데서도 키토산과 올리고당을 첨가한 시제품이 가장 신선도가 양호함을 알 수 있었다. 항산화성은 초기 과산화물가가 5.4∼7.8 meq/kg 이었던 것이 5℃ 저장시 35일째 15.7∼20.4 meq.kg, 0℃저장시 49일째 13.9∼18.4 meq/kg, -20℃저장시 80일째 11.6∼20.2 meq/kg으로 저온 일수록 산화속도가 늦었고 각각의 저장온도에서도 녹차, dillweed, 생강, 키토산, 올리고당을 첨가한 시제품에서 가장 산화속도가 늦었다. 친유성 갈색도는 -20℃에서 80일간 저장기간동안 거의 변화가 보이지 않았고 5℃와 0℃에서는 저장기간이 연장됨에 따라 갈색도는 증가하는 경향을 나타내었다. 저장 중 생균수 변화는 5℃와 0℃에서는 저장 80일째까지 생균수 변화는 보이지 않아 신선도 유지가 가능하였다. 관능검사 결과는 5℃저장시 14∼21일, 0℃저장시 28∼49일 점질물이나 산패취가 발생하여 상품적 가치를 상실하였고, -20℃에서는 80일간 저장기간 동안에 이들 변화는 나타나지 않았다. In oredr to endow mackerel fillet with antioxidant effect, functionality and remove fishy smell, chitosan, oligosaccaide, extracts of green tea, dill weed and ginger was used as soaking solution additives. Quality characteristics of soaked mackerel fillet(SMF) was investigated during storage temperature and time after vacuum packing with polyvinyl chloride film. From the results of VBN measurement, initial spoilage was showed within 2 or 3 weeks in SMF soaked with brine including extracts of green tea, herb and ginger at 5℃. In case of adding chitosan and oligosaccarides to the solution mentioned above, initial spoilage was found from 4 to 7 weeks, oxidation was reduced remarkably and SMF stored at -20℃ maintained the freshness during 80 days. Viable cell counts were reached to 108 for 3 weeks storage at 5 and 0℃. But no change of viable cell counts was founded at -20℃. The shelf-lifes of SMF according to 5, 0 and -20℃ storage temperatures were 2 to 3, 4 to 7 weeks and about 3 months, respectively.

      • 한국인 여성의 자궁경부암에서 PCR을 이용한 Human papillomavirus 아형의 검출에 관한 연구

        The polymerase chain reaction (PCR) was used for detection of human papillomavirus (HPV) in paraffin-embedded tissue. Tissues stored as paraffin blocks were a potential source of DNA for retrospective clinicogenetic analysis. Sections in 6 ㎛ (three or five) thick cut from paraffin blocks of 29 uterine cervix dysplasia, 34 uterine cervix carcinoma in situ, 42 uterine cervix squamous cell carcinoma and 10 uterine cervix metaplasia samples were deparaffinized and incubated at 37℃ for 48hrs. in the lysis solution. Following organic extraction and alcohol precipitation, and extracted DNAs were identified by gel electrophoresis. These DNAs carried out PCR. After repeated cycles of specific oligonucleotideprimed extension of viral DNA with thermophilic DNA polymerase. the type of HPV present was then determined on the basis of the size of the ethidium-bromide stained band visible after agarose gel electrophoresis : for HPV 6 is 263 bp. for HPV 11 is 144 bp, for HPV 16 is 601 bp. and for HPV 18 is 360 bp. This study detected HPV DNA in four cases. which broke down to HPV 16 in two case of uterine cervix carcinoma in situ and two case of uterine cervix squamous cell carcinoma, no HPV type in the remainder. Histologically, HPV types 6, 11 are mainly detected in benign genital warts and lowgrade cervical dysplasias, squamous cell carcinoma frequently contained HPV 16, whereas, HPV 18 was present in adenocarcinoma, adenosquamous cell carcinoma and small cell carcinoma of the cervix. These results indicate that cervical cancers might be clinically influenced by the differences in sutible of the infecting HPV.
