Novel signaling axis for ROS generation during K-Ras-induced cellular transformation

Reactive oxygen species (ROS) are well known to be involved in oncogene-mediated cellular transformation. However, the regulatory mechanisms underlying ROS generation in oncogene-transformed cells are unclear. In the present study, we found that oncogenic K-Ras induces ROS generation through activation of NADPH oxidase 1 (NOX1), which is a critical regulator for the K-Ras-induced cellular transformation. NOX1 was activated by K-Ras-dependent translocation of p47<SUP>phox</SUP>, a subunit of NOX1 to plasma membrane. Of note, PKCδ, when it was activated by PDPK1, directly bound to the SH3-N domain of p47<SUP>phox</SUP> and catalyzed the phosphorylation on Ser348 and Ser473 residues of p47<SUP>phox</SUP> C-terminal in a K-Ras-dependent manner, finally leading to its membrane translocation. Notably, oncogenic K-Ras activated all MAPKs (JNK, ERK and p38); however, only p38 was involved in p47<SUP>phox</SUP>-NOX1-dependent ROS generation and consequent transformation. Importantly, K-Ras-induced activation of p38 led to an activation of PDPK1, which then signals through PKCδ, p47<SUP>phox</SUP> and NOX1. In agreement with the mechanism, inhibition of p38, PDPK1, PKCδ, p47<SUP>phox</SUP> or NOX1 effectively blocked K-Ras-induced ROS generation, anchorage-independent colony formation and tumor formation. Taken together, our findings demonstrated that oncogenic K-Ras activates the signaling cascade p38/PDPK1/PKCδ/p47<SUP>phox</SUP>/NOX1 for ROS generation and consequent malignant cellular transformation.

In this paper, a method for constructing and verifying datasets used in deep learning technology is proposed to prevent safety accidents in the construction environment. The virtual construction simulator was developed to implement various construction environments, and custom data in the virtual environment was set to diversify the location and parameters of the sensors. A database for real-time deep learning models was built and detected at the pixel level to detect uneven roads and objects in the construction site. When deep learning-based detection is performed, it may interfere with the accurate judgment due to occlusion areas hidden by other objects. Therefore, the database for developing the occlusion area detection algorithm in virtual environments was constructed and verified.
Repair of Pd-based composite membrane by polishing treatment

In this study, polishing treatment was introduced to repair Pd-based composite membrane. Some pinholes remained after sputtering with palladium followed by heat-treatment at 723 K for 2 h. The pinholes drastically decreased the hydrogen perm-selectivity of the Pd-based composite membrane. In order to remove the pinholes, we introduced polishing treatment after the palladium sputtering. Scanning election microscopy (SEM) analysis and helium leak tests confirmed the effectiveness of the polishing treatment in removing the pinholes. The helium leaks were drastically decreased by the polishing treatment. Hydrogen permeation and helium leak tests showed that the polishing treatment increased the hydrogen perm-selectivity from 52 to >40,000 with a hydrogen permeation flux of 1.8 x 10<SUP>-1</SUP> mol m<SUP>-2</SUP> s<SUP>-1</SUP> at a pressure difference of 100 kPa and temperature of 673 K. The advantage of our membrane fabrication procedures lies in its simplicity. Our coating and polishing method offers the promise of highly reproducible Pd-based composite membrane.

Autoimmune hypoglycemia is characterized by hyperinsulinemia, fasting hypoglycemia, and presence of insulin autoantibody without previous exposure to exogenous insulin. Though this conditions were reported exclusively in Japan, we recently experienced the first case of autoimmune hypoglycemia in Korea and present it with a literature review. Purpose: The purpose of this study was to identify the relationship between satisfaction toward appearance and the level of mental health among female middle school students. Methods: The total study sample was 432 consisting of 3rd year female students who attended co-educational or girls’ middle schools in Korea. Data were collected from August 8 to August 16, 2012. Statistical analyses were conducted using t-test and Pearson’s Correlation Coefficient with SPSS18.0Windows software. Results: There was correlation between appearance satisfaction and mental health, especially with personal sensitivity (r= -.447, p= .000) and depression (r= -.350, p= .000). The level of mental health significantly differed according to type of school (t= -2.668, p= .008). However, appearance satisfaction did not differ according to type of school. Conclusion: The results of this study implied the importance of appropriate education and nursing intervention which would improve satisfaction toward the appearance of female middle school students.
HVPE 방법에 의해 r-plane 사파이어 기판 위의 선택 성장된 GaN/AlGaN 이종 접합구조의 특성

본 논문에서는 혼합소스(mixed-source) HVPE(hydride vapor phase epitaxy)방법으로 선택성장(SAC: selective area growth) GaN/AlGaN 이종접합구조의 발광다이오드를 r-plane 사파이어 기판 위에 제작하였다. SAG-GaN/AlGaN DH(double heterostructure)는 고온 GaN 버퍼층, Te 도핑된 AlGaN n-클래딩층. Gan 활성층. Mg 도핑된 AlGaN p-클래딩층. Mg 도핑된 GaN p-캡층으로 구성되어있다. GaN/AlGaN 이종접합구조의 발광다이오드의 특성을 알아보기 위해 SEM을 통한 구조적 분석과 전류-전압 측정(I-V: current-voltage measurement), 전류-광출력(EL: electroluminescence) 측정을 통하여 전기적, 광학적 특성을 평가하였다. In this paper, a selective area growth (SAG) of a GaN/AlGaN double heterostructure (DH) has been performed on r-plane sapphire substrate by using the mixed-source hydride vapor phase epitaxy (HVPE) with multi-sliding boat system. The SAG-GaN/AlGaN DH consists of GaN buffer layer, Te-doped AlGaN n-cladding layer, GaN active layer, Mg-doped AlGaN p-cladding layer, and Mg-doped GaN p-capping layer. The electroluminescence (EL) characteristics show an emission peak of wavelength, 439 nm with a full width at half maximum (FWHM) of approximately 0.64 eV at 20 mA. The I-V measurements show that the turn-on voltage of the SAG-GaN/AlGaN DH is 3.4 V at room temperature. We found that the mixed-source HVPE method with a multi-sliding boat system was one of promising growth methods for III-Nitride LEDs.

We have fabricated and tested a simple circuit of the rapid single-flux-quantum(RSFQ) four-stage shift register using a single layer high-$T_c$ superconducting (HTS) $YBa_2Cu_3O_{7-x}$ (YBCO) thin film structure with 9 step-edge Josephson junctions. The circuit includes two read superconducting quantum interference devices(SQUID) and four stages. To establish a robust HTS RSFQ device fabrication process, we have focussed on the reproducible process of sharp and straight step-edge formation as well as the ratio of film thickness to step height, t/h. The spread of step-edge junction parameters was measured from each 13 junctions with t/h=1/3, 1/2, and 2/3 at various temperatures. We have demonstrated the simplified operation of the shift register at 65 K.
Long-term CO<sub>2</sub> capture tests of Pd-based composite membranes with module configuration

In this study, we investigate the configuration of a Pd-Au composite membrane on a porous nickel support and membrane modules for withstanding the capture of CO<SUB>2</SUB> from a coal gasifier for a long time. The hydrogen permeation flux, recovery and CO<SUB>2</SUB> capture were experimentally evaluated using two different modules and two conditions. As in our study, the CO<SUB>2</SUB> capturing and durability tests were performed with a 40% CO<SUB>2</SUB>/60% H<SUB>2</SUB> feed gas mixture in stainless steel (SS) 316L and 310S membrane modules. As a result, it is achieved the durability tests for more than 1150, 1100 (SS 316L module) and 3150 h (SS 310S module) with pressure cycles from 100 to 2000 kPa at 673 K. The durability of the membranes and membrane modules was demonstrated under pressure cycles from 100 to 2000 kPa at 673 K and the SS 310S module was very stable after 3150 h. The durability test for more than 3000 h demonstrated that there was no significant intermetallic diffusion between the PNS and Pd-Au layer. The CO<SUB>2</SUB> capturing test performed using a 40% CO<SUB>2</SUB>/60% H<SUB>2</SUB> mixture confirmed that the CO<SUB>2</SUB> capturing capacity of the membrane and membrane module was 2.0 L/min for a CO<SUB>2</SUB> concentration in the retentate stream of 92.3% and that the hydrogen recovery ratio increased with increasing pressure and reached 93.4%. Furthermore, we suggest that the SS 310S module configuration, CO<SUB>2</SUB> capturing test using Pd-Au/ZrO<SUB>2</SUB>/PNS membrane and membrane module is very suitable for application as an Integrated Gasification Combined Cycle (IGCC) system due to very simple numbering-up stackable module design was successful.
