      • Efficacy of Seo Dong-Il's Technique as a Method of Improving Voice Quality in Patients with Phonasthenia and Vocal Nodules

        The purpose of this study was to invesigate the effects of Seo Dong-Il's technique on voice quality in patients with vocal nodules and phonasthenia (vlcal fatigue). The patients (4 nodules and 6 vocal fatigue) participated in teh study. Each subject was assessed acoustically (Fo, Jitter, Shimmer, NNE) in the first and last session. Dr. Speech (version 3.4, Tiger-DRS) was used to compae acoustic parameters of pre- and post-treatment. Seo Dong-Il's technique consisted of breating exercise, relaxation exercise, and phona-tion exercise The results were as follows: First, Seo Dong-Il's technique tended to be effective on improving voice quality in patients with phonasthenia and vocal nodules. Secon, the nature of improvements wee as follows: there was a significant difference between pre- and post-treatment in shimmer (p < .01) and NNE (p < .001), while there was no significant difference between pre- and post-treatment in Fo and Jitter. Finally, given the fact that the number of subjects was only 10, the jitter might have shown a significant difference if more subjects partcipated in teh experiment.

      • 몇 가지 항균제가 시험관내에서 내독소와 TNF-α, IL-6 분비에 미치는 영향

        To evaluate antibiotic-induced endotoxin release(AIER) and its correlation with some cytokines, we measured endotoxin level and tumor necrosis factor alpha(TNF-α) and interleukin6(IL-6) production in mononuclear cells in vitro after exposure of Pseudomonas aeruginosa to antibiotics belonging to different class with two extreme concentrations. The tested concetration of antibiotics were set up according to peak serum level. The low concetration of ceftazidirne and low concentration of imiperiem increased AIER, but high concentration of ceftazideme, high concentration of ciprofloxacin, high concentration of cefoperazone/sulbactam, high concentration of amikacin, and high concentration of meropenem reduced AIER.Interestingly, combined treatment of these antibiotics markedly reduced AIER, But the major cyotkines, TNF-α and IL-6 were not affect by type and concettration of antibiotics, combined treatment of antibiotics, and level of endotoxin released by antiboitics. In this study, we observed AIER was different according to type of antibiotics, concentration of antibiotics, and combination of antibiotics, But AIER had poor correlation with TNF-α and IL-6 in Pseudomonas aeruginosa. It suggests that cytokine release is not solely dependent to endotoxin, but more complex cascade is needed. More invesfigations, such as endotoxin induced cytokine mRNA expression, relationship with penicillin-binding proteins and endotoxin-neutralizing effect of antibiotic itself, must be performed.

        반복적 관절내 출혈이 관절 활막과 연골 세포의 변화에 미치는 영향

        목적: 혈액의 관절강내 반복주사 동물 모델을 이용하여, 혈우병성 관절염에서 활막의 변화와 연골 파괴에 대한 병리 기전과 경과 과정을 연구하였다. 대상 및 방법: 뉴질랜드 백색 토끼 수컷 20마리에 18주간 1주 3회씩 귀에서 자가 혈액을 채취하여 우측 슬관절에 1 ㎖를 주사하고, 좌측 슬관절에 생리식염수 1㎖를(대조군) 주사하였다. 11주 후와 18주 후에 양측 슬관절의 X선 촬영을 시행하였고, 활액막과 연골을 채취하여 조직검사를 시행하였다. 활막 세포 배양에서 RT PCR를 이용한 cytokine의 변화를 관찰 하였고, 연골 세포를 추출 배양하여 GAG 및 PGE₂, MMP-1,3 생성을 측정하였다. 결과: 11주째 육안적 변화는 없었으나, 우측 슬관절의 활액막은 조직학적으로 경한 증식반응과 형질세포 및 단핵세포의 활액막내 침착을 보이며 약간의 염증 소견을 나타냈다. 단순 방사선 소견상 특이 소견은 발견되지 않았다. 18주 후에는 육안적으로 우측 슬관절이 좌측에 비하여 부어 있었고, 단순 방사선학적 소견에서는 관절의 퇴행성 변화가 관찰 되었다. 병리학적 소견은 활막의 심한 증식과 염증세포의 침윤이 관찰되었다. 활막의 real-time RT PCR 시행결과 TNF-alpha, IL-1, MMP-1 mRNA 발현이 증가되었다. 연골세포 배양에서 대조군과 비교시 GAG 생성은 감소하고 PGE₂는 증가하였으며 MMP-1과 MMP-3는 변화가 없었다. 결론: 반복적인 관절 내 출혈은 활막세포를 자극하여 활막의 심한 증식과, proinflammatory cytokines의 생성을 증가시키며, 이는 연골 세포의 재생을 억제하고 연골세포의 염증을 증가시켜 연골의 대사기능이 억제되면서 퇴행성 변화를 촉진하여 점차적으로 관절염으로 이행되는 것으로 사료된다. Purpose: We designed this study to demonstrate the pathophysiology of hemophilic arthropathy (HA) by creating an animal model for determining the effect of repeated intraarticular bleeding in the synovium and articular cartilage. Materials and Methods: 20 normal male New Zealand white rabbits were used for this study. We injected 1 ㎖ of autologous blood from the ear vein of the rabbits into the right knee joint three timeds a week for 18 weeks, and we injected 1 ㎖ of normal saline into the left knee joint three times a week for 18 weeks as a control group. We examined the pathologic changes by microscopy and plain X-ray, and we determined the mRNA expression of proinflammatory cytokines in the synovium of the HA by performing real time RT-PCR at the 11<SUP>th</SUP> week and 18<SUP>th</SUP> week after starting blood-injection. We also examined the GAG and the PGE2 production in cultured chondrocytes that were extracted from the HA knees. Results: At the 11<SUP>th</SUP> week, after blood injection there were no remarkable gross changes in the HA knees and the control knee joints. At the 18<SUP>th</SUP> weeks, the experimental knee joints (HA knees) showed grossly swelling and degenerative changes by X-ray. The infiltration of inflammatory cells and the synovial proliferation in the HA knee joints were compared with that in the control knee joints by microscopic examination. The expressions of the mRNA of TNF-alpha, IL-1, MMP-1 and MMP-3 in the HA synovium were increased, as determined by real time RT-PCR, as compared with that in the control knee. In the cultured chondrocytes, the GAG production was decreased and the PGE2 was increased, but the MMP-1 and MMP-3 were not changed, as determined by ELISA. Conclusion: Our results showed that the GAG production of chondrocytes of the HA knees was decreased and there was increased PGE2, so that the cartilage degeneration by intra-articular bleeding was caused by the decreased metabolism of chondrocytes rather than by increased catabolism of the chondrocytes. We suggest that HA was associated with synovitis and cartilage degeneration, but decreased cartilage metabolism was the major mechanism of HA.

      • Therapeutic effect of a novel histone deacetylase 6 inhibitor, CKD-L, on collagen-induced arthritis in vivo and regulatory T cells in rheumatoid arthritis in vitro

        <P><B>Background</B></P><P>Histone deacetylase (HDAC) inhibitor has recently been reported to have a therapeutic effect as an anti-inflammatory agent in collagen-induced arthritis (CIA). We investigated the therapeutic effect of a new selective HDAC6 inhibitor, CKD-L, compared to ITF 2357 or Tubastatin A on CIA and regulatory T (Treg) cells in patients with rheumatoid arthritis (RA).</P><P><B>Methods</B></P><P>CIA was induced by bovine type II collagen (CII) in DBA/1 J mice. Mice were treated with HDAC inhibitor for 18 days. Arthritis score was assessed and histological analysis was performed by hematoxylin and eosin (H&E) stain. Cytotoxic T-lymphocyte associated protein (CTLA)-4 expression in induced Treg cells was analyzed and suppression assay was analyzed using Treg cells and effector T (Teff) cells isolated from naive C57BL/6 mice by flow cytometry. Cytokines were analyzed in peripheral blood mononuclear cells (PBMC) of five patients with RA by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). Tumor necrosis factor (TNF) was analyzed using PMA- activated THP-1 cells by ELISA. Suppression assay was analyzed using Treg cells and Teff cells isolated from RA patients by flow cytometry.</P><P><B>Results</B></P><P>In the CIA model, CKD-L and Tubastatin A significantly decreased the arthritis score. CKD-L increased CTLA-4 expression in Foxp3<SUP>+</SUP> T cells and inhibited the proliferation of Teff cells in the suppression assay. In RA PBMC, CKD-L significantly inhibited TNF and interleukin (IL)-1β, and increased IL-10. CKD-L and Tubastatin A inhibited TNF secretion from PMA-activated THP-1 cells. CKD-L and ITF 2357 inhibited the proliferation of Teff cells in RA patients in the suppression assay. Tubastatin A had no effect on inhibition of proliferation.</P><P><B>Conclusion</B></P><P>CKD-L decreased the arthritis score in CIA, reduced the expression of TNF and IL-1β, and increased the expression of IL-10 in PBMC from RA patients. CKD-L increased CTLA-4 expression and the suppressive function of Treg cells. These results suggest that CKD-L may have a beneficial effect in the treatment of RA.</P>

      • 전침 자극에 의한 오디 괄약근 운동성 변화에 관한 연구

        Background/Aims: This study was designed to evaluated the effects of electroacupuncture on the sphincter of Oddi (SO) motility in humans and to correlate the manometric findings with plasma cholecystokinin (CCK) levels. Methods: Eleven patients (M: F= 5: 6) who had various kinds of biliary disorders were enrolled. SO motility was monitored with a conventional low-compliant continuously perfused technique using ERCP (n=9) or percutaneous transhepatic cholangioscope (n=2). After baseline monitoring for phasic wave contractions of SO, electroacupuncture was applied at a specific acupoint, GB 34, in these 11 patients. A nonspecific acupoint 5 cm away from GB 34 was selected as a control. Manometric parameters of SO were also checked in 6 subjects during stimulation of the control acupoint. CCK plasma levels were measured during the time course of electroacupuncture stimulation. Result: All the manometric parameters including the basal pressure of SO, amplitude, frequency and duration of phasic wave contraction of SO were significantly decreased (p< 0.05) during electroacupuncture stimulation. The inhibition of SO contractility was accompanied by increased CCK plasma levels. After removal of electroacupuncture stimulation, restoration of amplitude and duration to basal condition were noted. A tendency towards the return of SO contractility was also observed in basal pressure and frequency. Stimulation of the control acupoint, however, did not affect the SO contractility and plasma CCK levels. Conclusions: Electroacupuncture stimulation of acupoint GB 34 showed reversible inhibition of SO contraction in humans. We speculate that the response of SO to electroacupuncture stimulation may be mediated by some neurohormonal mechanisms including CCK release.         삼백초(三白草)의 소염작용(消炎作用)에 대(對)한 실험적(實驗的) 연구(硏究)

        Purpose: The purpose of this research was to investigate the effects of Saurui Herba Seu Rhizoma(SHSR) on Anti-inflammatory properties in Raw264.7 cell line and murine models of inflammation. Methods: To investigate the effects of Saurui Herba Seu Rhizoma(SHSR) on anti-inflammation, we study cytotoxicity effects of SHSR on Mouse Lung Fibroblast Cells and Peritoneal Macrophages, Inhibitory effects of SHSR on the nitric oxide (NO) release, the ROS production, and the interleukin-6 production. Results: The cytotoxicity of SHSR on mouse lung fibroblast Cells and Raw264.7 cell line was not observed. SHSR in RAW264.7 cell line inhibited IL-1β, IL-6 mRNA gene expression depending upon the concentrations of extract and inhibited IL-18 mRNA gene expression at 100 ㎍/㎖ of extract. SHSR in RAW264.7 cell line inhibit COX-2 mRNA gene expression at 100, 10 ㎍/㎖ of extract. SHSR in RAW264.7 cell line inhibited NOS-Ⅱ mRNA gene expression depending upon the concentrations of extract. SHSR in RAW264.7 cell line didn``t inhibit TNF-α mRNA gene expression. SHSR in RAW264.7 cell line decreased IL-6 production depending upon the concentrations of extract. SHSR in RAW264.7 cell line decreased ITNF-α production according to the concentrations of extract. SHSR in RAW264.7 cell line inhibited NO release specially SHSR 100, 10 ㎍/㎖ concentrations of extract. SHSR inhibit ROS production depending upon the concentrations of extract. Conclusion: These results suggest that SHSR can be used treating a lot of women disease caused by inflammation.

      • 가상가공기법을 이용한 볼엔드밀과 플랫엔드밀의 절삭력 해석 시스템 개발

        본 연구에서는 볼엔드밀과 플랫앤드밀에 의해서 야기되는 절삭력을 가상적으로 평가할 수 있는 기반을 구축하기 위한 노력으로써 가상가공기법을 이용한 절삭력 해석 시스템을 구현하였다. 공구의 기하학적 제원과 공작기계(밀링머신, 머시닝센터)의 운전 조건에 의해서 변화되는 절삭력은 공작물과 공구의 접촉영역 내에 위치한 미소 절삭날에 적용하는 미소 절삭력들을 중첩시키는 방법을 통해서 해석하였다. 특히 구조변형을 고려한 절삭력은 절삭력과 구조변형의 평형조건을 이용해서 결정하였다. 아울러서 볼엔드밀과 플랫엔드밀의 절삭력에 대한 실험결과와 해석결과의 비교를 통해서 본 연구에서 개발한 절삭력 해석 시스템의 유용성을 입증하였다. In this study, a cutting force analysis system for ball and flat end-mills is realized based on the virtual manufacturing technologies. Cutting force which depends on the specification of cutting tool and operating conditions of machine tool(milling machine, machining center), is predicted by superposing the infinitesimal cutting forces on infinitesimal cutting edges existing in the contact area between cutting tool and workpiece. Especially, the equilibrium condition between cutting force and structural deformation of cutting mechanism is introduced to determine the cutting force considering the influence of structural deformation. And comparison between the predicted and measured cutting forces for ball and flat end-mills shows the effectiveness of the cutting force analysis system developed in this study.

      • AHP와 QFD를 이용한 계층별 성과평가의 측정지표 개발과 이익효과에 관한 연구

        Today, companies try to adopt new management technologies to survive in changing business environment. But it results in a few successful cases. The reason for it is that they do not realize the exact problems of the company and they apply just the techniques without proper modification, and also they just analyze only financial factor for performance evaluation of their corporate by use of only the consequence indexes but the process measurement indexes. This paper analyzes these problem and proposes the method of improvement. That is, this paper proposes new efficient methodology and model which can evaluate not only the factor of management but also the process of the management. A company organization can be divided for evaluation index into three, those are, organization level, process level, and team/individual level. AHP and QFD methods are applied to find the critical performance measurement indexes which meet the basic selecting conditions. The critical measurement indexes would work as essential factors of evaluation system in the priority order chosen by the goal of a company. This paper proposes 5 phases of new evaluation system model. And the process of team evaluation indexes are developed and case studies are represented based on the above method. When the evauation indexes are selected, corporate organization can be divided into three levels: 1) organization level 2) process level 3) team/personal level. The index type is composed with three categories: 1) cost 2) quality 3) time. It must satisfy the customer, employee and quality systems requirement. In conclusion, the five phases for the new evaluation system are proposed in this study and also the processes to develop the system evaluating model especially for the team level and sample case studies are represented.

        We have measured the absorption spectrum of CuBr quantum dots at 8.5K. Z1.2 exciton peak moves toward the shorter wavelength relative to the bulk CuBr. After a selective excitation by the dye laser, we could find the spectral hole in the absoprption spectrum. The depth and width of spectral hole depends on the optical fluence of dye laser. There are two process in the hole burning of quantum dots. The rising times of hole depth are 1.2 sec and 250 sec respectively. We think that the fast process is the result of the reduction of quantum dots which have the specific mean diameter, the second process is caused by the variation of a environment of matrix.
