        Ethanol Extract of Ganoderma lucidum Augments Cellular Anti-oxidant Defense through Activation of Nrf2/HO-1

        Objectives: The mushroom Ganoderma lucidum has been widely used as a traditional herbal medicine for many years. Although several studies have focused on the anti-oxidative activity of this mushroom, the molecular mechanisms underlying its activity have not yet been clearly established. The present study investigated the cytoprotective effect of ethanol extract of Ganoderma lucidum (EGL) against oxidative stress (hydrogen peroxide, $H_2O_2$) and elucidated the underlying mechanisms in a C2C12 myoblast cell line. Methods: Oxidative stress markers were determined by using the comet assay to measure reactive oxygen species (ROS) generation and deoxyribonucleic acid (DNA) damage. Cell viability and Western blotting analyses were employed to evaluate the cellular response to EGL and $H_2O_2$ in C2C12 cells. Transfection with nuclear factor erythroid 2-related factor 2 (Nrf2)-specific small interfering ribonucleic acid (siRNA) was conducted to understand the relationship between Nrf2 expression and $H_2O_2$-induced growth inhibition. Results: The results showed that EGL effectively inhibited $H_2O_2$-induced growth and the generation of ROS. EGL markedly suppressed $H_2O_2$-induced comet-like DNA formation and phosphorylation of histone H2AX at serine 139 ($p-{\gamma}H2AX$), a widely used marker of DNA damage, suggesting that EGL prevented $H_2O_2$-induced DNA damage. Furthermore, the EGL treatment effectively induced the expression of Nrf2, as well as heme oxygenase-1 (HO-1), with parallel phosphorylation and nuclear translocation of Nrf2 in the C2C12 myoblasts. However, zinc protoporphyrin IX, a HO-1 inhibitor, significantly abolished the protective effects of EGL against $H_2O_2$-induced accumulation of ROS and reduced cell growth. Notably, transient transfection with Nrf2-specific siRNA attenuated the cytoprotective effects and HO-1 induction by EGL, indicating that EGL induced the expression of HO-1 in an Nrf2-dependent manner. Conclusion: Collectively, these results demonstrate that EGL augments the cellular anti-oxidant defense capacity through activation of Nrf2/HO-1, thereby protecting C2C12 myoblasts from $H_2O_2$-induced oxidative cytotoxicity.

        Queueing analysis, in general., has the intrinsic problem of large state space dimension and complex computation. In fact, the petri net is a graphical and mathematical model. It has been used for the modeling of various applications, in particular, manufacturing systems. It can deal with parallelism, concurrence, deadlock avoidance, and asynchronism. Currently it has been applied to the performance of computer networks and protocol verifications. This paper presents a technique for modeling and analyzing queueing model using stochastic activity networks (SANs). And then some examples are showed for the verification of the technique presented in this paper.

        A study on the Byungtae Ron(病能論) of the So Moon(素問), one of the classic book of oriental medicine, was carried out analytically for the right comments. The principal ideas obtained summarized as follows; Neng(能) is similar to tae(熊) in the pronunciation and the meaning of a chinese character. Byungtae(病能) means the state of the symptoms and the pulse condition of the diseases, etc. It deals with ethological factor, mechanism of disease, the condition of an illness, method of diagnosis, rule of treatment, technique of treatment of seven diseases and taking this seven diseases as a example, mainly discuss the technique and significance of clinical observation and the pathogenesis and condition of an illness, so It named 「Byungtaeron(病能論)」 In this chapter it shows we can grasp condition of illness well only if we classify the pathogenesis and condition of an illness in details. this viewpoint provides the foundation of development in oriental diagnostics. It places emphasis on pulse feeling in this chapter, so it describes pulse condition of various kinds of diseases in details. It also presents how to compare pulse feeling like comparison between Inyoungmaek(人迎脈, arterial carotid communes, the pulsation of which is easily felt beside Adam's apple) and Ch'onkumaek(寸口脈, the radial artery on the wrist where the pulse is routinely taken for diagnosis), between right and left Ch'onkumaek(寸口脈), between Yangmyungsangdong (陽明常動) and Taeyangbudong(太陽不動), Soyangbudong(少陽不動), etc. In this chapter it explains pulse feeling occupies a high position in the diagnosis of diseases in 『Huangdi's internal Classic(黃帝內經)』 period and the technique of pulse feeling is already plentiful and minute. In this chapter it presents 「Tongbyungyich(同病異治, to apply different methods of treatment to the same kind of disease)」 theory, taking tangible syndromes as examples. this is prominent viewpoint of determination of treatment based on the differentiation of symptoms and signs and much influence development of later ages' medicine. On the ground of oriental medical theory, it may bring about different symptoms in the same disease according to stage of disease, the seasons, the regions, the constitution of patient, therefore surely adopt different technique of treatment, this differentiation of symptoms and signs is an important character in oriental medicine.

        Tufted Angioma와 유사한 소견을 보인 화농성 육아종

        Tufted angiomas are rare slowly progressive vascular lesions found typically in children and young adults, usually involving the trunk and neck. Microscopically, groups of capillary tufts, many uncanalized, were found dispersed at various levels in the dermis. Some authors regarded tufted angioma as a member of lobular capillary hemangiomas. We describe a pyogenic granuloma mimicking acquired tufted angioma on the lip in a 59-year-old woman. We suggest that a term lobular capillary hemangioma$quot; may be preferable in this case.

        자궁경부암에서 치료 전 FDG PET을 이용한 예후 예측

        목적: 자궁경부암에서 예후를 예측하는 것은 임상적으로 중요하다. 이 연구에서는 치료 전 FDG PET에서 관찰된 림프절 전이 유무와 원발종양의 FEG 섭취정도가 자궁경부암 환자에서 치료 후 예후를 다른 예후인자와 비교하여 독립적으로 예측할 수 있는지 알아보았다. 대상 및 방법: 자궁경부암이 새로 진단된 74명의 환자를 대상으로 하였다(50±12세). FDG를 정맥주사하고 전신 PET 영상을 얻었다. 자궁경부의 원발종양에서 최대 표준섭취지수를 측정하였고, 두 명의 핵의학과 의사가 판독하였다. 모든 환자에 대하여 추적 관찰하여 재발 여부를 알아보았다. Kaplan-Meier 법으로 재발없는 생존기간을 분석하였고, Logrank test로 비교하였으며 Cox 회귀모형으로 독립적인 예측인자를 찾아보았다. 결과: 13명의 환자에서 재발이 있었다. 단변량분석에서는 FIGO병기, MRI 또는 CT에서 측정한 종양의 최대지름, MRI 또는 CT에서 림프절 전이 유무, PET상 림프절 전이의 유무가 재발없는 생존기간에 유의한 차이를 보였고, 나이나 병리학적 암세포의 종류, 원발종양의 최대 표준섭취계수에 따른 환자군 사이에 재발없는 생존기간의 유의한 차이는 없었다. Cox 회귀모형 분석에서는 PET상 림프절 전이의 유무(Exp(β)=6.416, p=0.002)와 MRI 또는 CT에서 림프절 전이 유무(Exp(β)=5.711, p=0.026)만이 독립적으로 재발없는 생존기간과 유의한 관계를 보였으며, PET상 림프절 전이의 유무가 재발없는 생존기간 예측 모형에 가장 크게 기여하였다. 결론: 자궁경부암에서 치료 전 FDG PET의 림프절 전이 유무는 치료 후 예후를 예측할 수 있는 가장 좋은 독립적 예후인자인 반면, 종양의 최대표준섭취계수는 유의한 예후인자가 아니었다. Purpose: In uterine cervical cancer, it is clinically important to predict prognosis on initial staging. We investigated whether abnormally increased FDG uptake of lymph nodes and peak SUV of primary tumor on initial FDG PET can predict prognosis after treatment in cervical cancer patients. Materials and MEthods: Subjects were 74 newly diagnosed cervical cancer patients (50±12yr). Whole body PEt was performed after an injection of F-18. Peak SUV was measured in the primary cervical tumor. Images were interpreted by two nuclear medicine physiciana. Follow-up for the disease-free survival was done in all patients. Survival analysis was done by using the Kaplan-Meier method, and a Longrank test was used to compare survival. Cox regression analysis was followed to find independent predictive factors. Results: Recurrence occurred in 13 patients. In univariate analysis, advanced FIGO stage, large size of primary tumor, positive lymph node on MRI or CT, and positive lymph node on PET were significant prognostic factors to predict recurrence. Peak SUV and cell type of tumor failed to show any significant correlation with disease-free survival. In multivanate analysis, positive lymph node on PET (Exp(β)=6.416, p=0.002) and on CT or MRI (Exp(β)-5.711, p=0.026) were independent prognostic factors for predicting recurrence. Conclusion: In uterine cervical cancer, lymph node metastasis of pretreatment FDG PET is the best independent prognostic factor for predicting recurrence. But, peak SUV was not significant prognostic factor.

      • TiNi 金屬間化合物의 高溫酸化擧動에 관한 硏究

        The oxidation behavior and mechanism of intermetallic compounds TiNi has been investigated in air over the temperature range from 923K to 1073K. The phases of oxidation products and matrix have been examined by scanning electron microscopy, x-ray diffractometer and optical microscopy. The rate of oxidation in intermetallic compound TiNi follows the parabolic rate law in an initial stage of the oxidation process but it does not follow the parabolic rate law in the later stage. Oxidation products formed on the matrix consist of a layer structure of TiO_2(rutile), TiO_2 phase containing TiNiO_3 particles, TiO_2, Ni phase containing Ti, TiN_i3 phase and TiNi matrix in sequence from the outer side. The rate determining process in the overall oxidation of TiNi is the inward diffusion of oxygen through the oxide layer and the outward diffusion of Ti through the Ni phase.

      • 녹동균 세포외막 단백질 백신 CFC-1-101의 안정성 및 면역원성 검토 : 임상 제 Ⅰ/Ⅱa상 시험

        목적 : 제일제당에서는 녹농균의 세포외막 단밸질을 유효성분으로 하는 백신인 CFC-101을 개발하였으며, 동물시험에서 이 백신의 안전성과 유효성을 입증하였다. 본 연구에서는 이 녹농균 백신의 인체에 대한 안전성과 면역원성을 평가하는 동시에 인체 접종시의 최적 투여 용량을 결정하기 위하여 제 I/Ⅱa상 임상시험을 수행하였다. 방법 : 건강한 성인 남자를 피험자로 선별하여 각 용량군에 백신투여자 6명, 위약투여자 2명을 배정하였다. 백신 투여군은 0.25mg, 0.5mg 또는 1.0mg 용량의 녹농균 백신을 7일 간격으로 3회에 걸쳐 근육주사 하였으며, 위약 투여군에게는 세포외막 단백질을 제외한 동일한 성분을 투여하였다. 백신접종 후 국소적 또는 전신적인 반응의 발생여부를 관찰하고, 혈액시료를 체취하여 백신의 역가와 유효성을 검정하였다. 결과 : 녹농균 백신 CFC-101은 모든 접종자에서 양호한 내약성을 보였다. 또한 0.5mg 과 1.0mg 백신 투여군에서는 100%의 항체양전율을 나타내었다. 생성된 항체는 녹농균 세포외막단백질에 특이성을 보였고, 녹농균 감염에 대해 방어효능이 있었다. 결론 : 이와같은 결과로부터 이 녹농균 백신은 인체에 안전하게 투여할 수 있으며, 높은 항체 생성능으로 감염방어 효능을 보이고 0.5mg과 1.0mg이 최적용량인 것으로 판단되었다. Background : We developed a Pseudomonas aeruginosa outer membrane protein(OMP) vaccine CFC-101, and the prophylactic efficacy of which has been demonstrated in animal models. In order to evaluate the safety and immunogenicity of the P. aeruginosa vaccine, we carried out a phase I/Ⅱa clinical trial in healthy male volunteers. Methods : Groups of eight volunteers, including two placebo subjects, were vaccinated intramuscularly with three doses of 0.25, 0.5 or 1.0 mg of the vaccine at one week intervals. Sings of systemic and local reactions observed after vaccination were recorded for each vaccinee for 5 days. Physical examinations were performed on days 0, 1, 7, 8, 14, 15, 21, and 42, and clinical laboratory tests were done on days 0, 3, and 21. Blood samples for assay of serum antibody levels were obtained up to 42 days after the first vaccination. Results : The vaccine was generally well tolerated by all vaccinees, showing no significant side effects. In the three dosage groups, all vaccinees, except one receiving the 0.25 mg dose, showed significant elevation in serum IgG antibody titers against the vaccine proteins, indicating 100% seroconversion in 0.5 and 1.0 mg groups. The human antibodies induced by the vaccine were specific for P. aeruginosa OMPs, as confirmed by western blot analysis and immunoprecipitation assays. The capacity of the human antisera to enhance opsonophagocytic killing activity by polymorphonuclear leukocytes and to confer protection against P. aeruginosa infections indicates that the antibodies elicited by the vaccine have protective efficacy. Conclusion : We conclude that the P. aeruginosa OMP vaccine is safe and effective for human use and its optimal dose to be 0.5 or 1.0 mg.

        The oxidation behavior and mechanism of pure Ti has been investigated in air over the temperature range from 973K to 1273K. The phases of oxidation products and matrix have been examined by scanning electron microscopy, x-ray diffractometer and optical microscopy. The rate of oxidation of pure Ti follows a parabolic rate law in an initial stage of oxidation process but a linear rate law in the later stage. The transition from the parabolic to linear rate law take places when the thickness of oxide layer reaches a certain value, and the oxide layer formed thereafter contains a lot of defects which have been considered to form due to the stresses present at the matrix-oxide interface. The rate determining process in the overall oxidation of pure Ti is observed to be the inward diffusion of oxygen through the oxide layer.
