        The survey study was conducted to investigate the rice cake consumption pattern including the frequency of intake and the degree of preference and the area of improvement for Korean rice cake among college students. About the motive for purchases of rice cake, 'taste is good(43.3%)' was the highest, and 'eat as a snack' the next (42.9%). The reason which does not eat the rice cake frequently was uncomfortable to eat (24.1%), insipid(21.4%), not variously assorted(17.9%) in the order named. The college students surveyed were eating rice cake as a snack mainly. They wanted the rice cake to be charmed and be in pretty shape, and fusion style for gift. And they wanted to be rice cake more sweetened and cool taste like ice cream. Therefore, traditional rice cake is required for further research for the development of product which is more acceptable for the young adults.

        We experienced two case of Wilson's disease in female and male sibling. The first case was a 10 year-old female who showed jaundice. pancytopenia and cirrhotic change in liver biopsy. She died 10 days after diagnosis. The second case of 7 year-old male showed specific symptoms and revealed increased AST/ALT, low serun copper level and increased 24 hour urinary copper excretion level He is being treated with D-penicillamine and follow up examinations.

      • 제대혈 분리방법의 차이에 따른 단핵구 및 조혈모세포의 수득 효과에 관한 비교연구

        연구배경 : 최근에 보고된 변형적 Ficoll-Hypaque 방법을 이용한 제대혈 분리방법이 지금까지의 고식적 Ficoll-Hypaque 분리방법에 비하여 조혈모세포의 수득효과면에서 얼마나 차이가 있는지 또한 비교적 수득효과가 크다고 알려져 있는 3% gelatin을 이용한 분리방법에 비하여 얼마나 효과적인지를 알아보고, 제대혈 분리방법의 차이에 따라 단핵구와 조혈모세포 사이에 어떤 상관관계가 있는지 알아보기 위하여 본 연구를 시행하였다. 방법: 21명의 건강한 산모로부터 정상 질식 및 제왕절개 분만 후 제대정맥에서 해파린 처리한 주사기를 사용하여 제대혈을 채취하였다 채취한 제대혈은 동일하게 3개월 시험관에 나누어 실온에서 24시간 이내에 고식적 Ficoll-Hypaque분리법, modified Ficoll-Hypaque 분리법, 3% gelatin 분리법을 이용하여 각각 단핵구 분리를 시도하였으며 이들의 단핵구수 및 생존률, CD34 양성세포수, 집락수를 측정 비교 하였다. 또 상관분석을 이용하여 단핵구수, CD34양성세포수와 CFU-GM집락수 사이의 상관관계를 알아보았다. 결과: 1) 세포 생존률은 고식적 Ficoll-Hypaque 분리법, 변형적 Ficoll-Hypaque 분리법, 3% gelatin 분리법 사이레 통계적 차이가 없었다. 2) 단핵구수는 3% gelatin으로 분리했을 때가 4.56±1.86 (×10^(6)/mL)로 수득효과가 가장 높았으며 (P=0.0001), Ficoll-Hypaque으로 분리했을때는 각각 1.54±0.06(×10^(6)/mL), 1.36±0.67(×10^(6)/mL)로서 고식적 방법과 변형적 방법 차이에 통계적 차이가 없었다. 3) CD34 양성세포수는 3%gelatin으로 분리했을 때가 6.08±3.99(×10⁴/mL)로 수득효과가 가장 높았으며 (P=0.0001), Ficoll-Hypaque으로 분리했을 때는 각각 2.18±2.17(×10⁴/mL), 2.24±1.63(×10⁴mL)로서 고식적 방법과 변형적 방법사이에 통계적 차이가 없었다. 4) CFU-GM 집락수는 3% gelatin으로 분리했을 때가 15.72±8.93(×10³/mL)로 수득효과가 가장 높았으며 (P=0.0002), Ficoll-Hypaque으로 분리했을 때는 각각 3.12±1.45(×10³mL), 2.35±0.82(×10³/mL)로서 고식적 방법과 변형적 방법 사이에 통계적 차이가 없었다, 5) 제대혈 고식적 Ficoll-Hypaque 방법으로 분리하였던 경우는 단핵구수와 CFU-GM집락수 사이에 상관관계가 없었으며CD34 양성세포수만 CFU-GM집락수와 상관관계가 있었다. (r=0.57). 또 변형적 Ficoll-Hypaque 방법에 의하여 분리하였던 경우는 단핵구수나 CD34 양성세포수 모두가 CFU-GM 집락수와 상관관계가 없었다. 그러나 제대혈을 3% gelatin법으로 분리하였던 경우는 CFU-GM집락수 가 단핵구수 (r=0.88) 및 CD34양성세포수(r=0.86)양자 모두와 상관관계가 있었다. 결론 : 3%gelatin을 이용하여 제대혈을 분리하는 것이 단핵구 및 조세모세포의 수득효과면에서 가장 효과적이었다. 한편 변형적 Ficoll-Hypaque 분리법이 고식적 Ficoll-Hypaque 분리법보다 시간과 경비면에서 비효율적이면서 수득효과도 높지 못하였다. 또 제대혈 분리법의 차이에 따라 단핵구나 CD34 양성세포와 VFU-GM사이에 상관관계에 차이가 있었으며, 실제로 제대혈이식 후에 생착 여부를 예측할 수 있는 인자가 제대혈 분리법의 차이에 따라 달라지는지에 관해서 보다 많은 임상적 연구가 필요할 것으로 사료된다. Background : Cord blood banking for stem cell transplantation requires volume reduction and red cell depletion for cost effective storage. To determine which technique could minimize the loss of progenitor cells during cord blood processing, we compared the efficacy of cord blood separating methods which were conventional Ficoll-Hypaque method, modified Ficoll-Hypaque method, and 3% gelatin method. Methods : Twenty-one cord blood samples were obtained with heparinized syringes from the umbilical vein following delivery. Three aliquots of each 20mL cord blood were stored at room temperature and processed by 3 different techniques within 24 hours after collection. We compared mononuclear cells(MNCs) counts and their viability, CD34+ cells counts, and CFU-GMs counts and also observed their correlations among three techniques. Results : 1) The yield of MNCs or CD34+ cells was highest in the cord blood processed with 3% gelatin method than conventional or modified Ficoll-Hypaque method(P=0.0001). 2) There were no significant difference in the yield of MNCs and CD34+ cells between conventional and modified Ficoll-Hypaque method. 3) Both MNCs counts (r=0.800) and CD34+ cells counts (r=0.86) were significantly correlated with CFU-GM counts in the cord blood processed with 3% gelatin method. 4) CD34+ cells counts were significantly correlated with CFU-GM counts in the cord blood processed with conventional Ficoll-Hypaque method (r=0.57), but MNC counts were not. 5) Both MNC counts and CD34+ cell counts were not correlated with CFU-GM counts in the cord blood processed with modified Ficoll-Hypaque method. Conclusion : Cord blood separation with 3% gelatin method could provide better yield of progenitor cells than with FICOLL_Hypaque methods. And between two Ficoll-Hypaque methods, modified method is more laborsome but not better in terms of clonogenic potential than conventional method.

      • 콩아이스크림의 상품화를 위한 고정화 효소의 이용

        The study was to investigate the functional properties of soy protein isolate hydrolyzed by immobilized α-chymotrypsin and the quality characteristics of soy ice cream made by the soy protein isolate treated with immobilized α-chymotrypsin. Up to the hydrolysis rate of 22.9%, treatement by immobilized α-chymotrypsin costed more time than that of α-chymotrypsin. Foam and emulsion capacity were decreased by the number of repetitive usage. Apparent viscosity and overrun of soy ice cream treated with immobilized α-chymotrypsin and α-chymotrypsin were shown similar tendency. In the sensory characteristics, soy ice cream by enzyme treatment was evaluated as much acceptable than untreacted soy ice cream in the item of beany flavor, icy feel and overall quality. For efficient use of immobilized α-chymotrypsin industrially, α-chymotrypsin immobilized on glass beads can be reused for about 11 times more than α-chymotrypsin.

      • 혈청 Fructosamine에 대한 임상적 유용성에 관한 연구

        Diabetes mellitus is a universal health problem and may occur at any age. The traditional biochemical measurements for initially detection patients with diabetes mellitus are random estimations of blood and urine glucose concentrations. Despite their common use both tests are fairly non-specific, being influenced by wide variety of condition. Measurement of the HbA_(1c) in a random blood specimen is a more simple screening test for diabetes, and widely used as an index of diabetic control. But unfortunately, it has difficulties with methodology. Fructosamine has been described as a new parameter for the estimation of glycosylated protein, It is based on the reduction potential of ketoamines in an alkaline medium and measures colorimethcally the concentration of all glycosylated semm proteins belonging to the class of fructosamine. It reflects the time-integrated glucose concentration over the previous 1 to 3 weeks and can be used for the follow up of diabetes mellitus. We measured serum fructosamine concentration and compared with any other items. The results are summehzed as follows : 1. Within-run and between-run imprecision were assessed by 20 analysis of 3 differential fructosamine concentration in control sera. The respective coefficients of variation were 2.9%, 6.6% in low concentration, 2.2%, 5.1% in medium concentration and 3.3%, 4.7% in high concentration. 2. The serum fructosamine concentration were 2.26±0.21 mmol/L in normal control subjects(n=29) and 3.35±0.65 mmol/L (n=62) in patients with diabetes mellitus (p<0.001). 3. In diabetic patients (n=62), the serum fructosamine concentrations correlated less significantly with glucose (Y(fructosamine) = 0.01×(glucose)+2.52, r=0.54, P<0.001) and more significantly with HbA_(1c) (Y(fructosamine) = 0.22×(HbA_(1c))+1.71 , r=0.63, P<0.001). 4. The concentrations of fructosamine in serum and EDTA plasma were not significently diferent (P<0.01). So, Plasma is as well suited to fructosamine determinations as serum(Y(serum) = 0.95×(Plasma)+0.25, r=0.96). 5. No interference was observed with lipemic sera (cholesterol) in detection of serum fructosamine(P=0.393).

        일부 부인과 양성 질환자의 지방조직 및 혈청의 PCBs와 DDE 농도

        목적 : 본 연구는 지방조직과 혈청의 PCBs와 DDE 농도를 측정하고, 이에 영향을 미치는 것으로 알려진 요인들과의 관련성을 평가하고자 하였다. 방법 : 유방암, 자궁내막증 등 유기염소계 화합물의 영향이 있는 것으로 알려진 호르몬 관련 질병을 제외한 부인과 환자 52명을 대상으로 지방조직과 혈청을 분석하였으며, 설문조사를 통하여 연령, 교육수준, 직업, 비만도, 흡연과 음주, 출산력, 수유력등을 조사하였다. 지방조직과 혈청의 PCBs와 DDE 농도는 가스크로마토그래피를 사용하여 측정하였다. 결과 : 지방조직의 경우, 13개 PCBs congeners 농도의 합은 중앙값이 48.29 ng/g, 기하평균값이 56.78 ng/g이었고, DDE 농도는 중앙값이 142.89ng/g, 기하평균값이 117.06 ng/g이었으며, 혈청의 경우는 PCBs congeners 농도의 합은 중앙값이 4.67 ㎍/L, 기하평균값이 4.85 ㎍/L이었고, DDE 농도는 중앙값이 1.75 ㎍/L, 기하평균값이 2.09 ㎍/L로 나타났다. 지방조직과 혈청의 DDE 농도 사이는 높은 상관성을 보였으나(r²=0.310, p=0,0002), PCBs는 상관성이 약한 것으로 나타났다(r²=0.029, p=0.2582). 영향요인을 범주화한 후 로그변환 시킨 지방조직과 혈청의 PCBs, DDE 평균의 차이를 분석한 결과, 지방조직의 DDE만이 출산자녀수(p=0.015), 초산연령 (p=0.014), 비만지수(p=0.035)와 연관성이 발견되었다 다중회귀분석의 결과 지방조직의 DDE는 출산자녀수가 적을수록, 초산연령이 늦을수록 증가하는 경향을 보였다. 결론 : 출산자녀수와 초산연령, 비만도가 지방조직의 DDE와 관련성을 보였으며, 또한 유기염소계 화합물의 장기간의 노출에 의한 영향을 평가하는데 있어서 혈청보다 지방조직이 더 유용한 생물지표임을 알 수 있었다. Objectives : This study was purposed to determine the levels of PCBs and DDE in adipose tissue and serum and to evaluate the relations with factors affecting these levels. Methods : We analyzed adipose tissues and sera from 52 gynecologic benign disease patients aged 27-78 years, except hormonal diseases such as breast cancer and endometriosis. We also surveyed age, education, occupation, body mass index (BMI), smoking, drinking, number of children, and duration of breastfeeding by questionnaires. Tissue and serum levels of PCBs and DDE were measured using gas chromatography. Results : The median and geometric mean levels of PCBs were 48.29 ng/g, 56.78 ng/g for adipose tissue and 4.67 ㎍/L, 4.85 ㎍/L for serum, and those of DDE were 142.89 ng/g, 117.06 ng/g for adipose tissue and 1.75 ㎍/L, 2.09 ㎍/L for serum, respectively. Adipose tissue and serum levels for DDE showed high correlation (r²=0.310, p=0.0002), but those of PCBs didn't (r²=0.029, p=0.2582). In analyses of the differences of the means of log transformed adipose tissue and serum PCBs and DDE levels for groups of potential covariates, only adipose tissue DDE levels were significantly associated with number of children (p=0.015), age at first birth (p=0.074) and BMI (p=0.035). In multiple regression analysis, adipose tissue DDE levels were significantly increased with decreasing number of children and increasing age at first birth. Conclusions : This study suggested that parity and adiposity were associated with levels of DDE in adipose tissue, and adipose tissue is a better biomarker than serum for evaluating the long-term exposure of organochlorines.

        Effect of Sulfur Enriched Young Radish Kimchi on the Induction of Apoptosis in AGS Human Gastric Adenocarcinoma Cells

        The effects of young radish (YR, yeolmu in Korean) on the induction of apoptosis were examined in AGS human gastric adenocarcinoma cells. The young radish kimchi (YRK) were made of YR cultivated in the soil without (Control YR kimchi: C-YRK) and with 1,818 g/m3 sulfur (Sulfur YR kimchi: S-YRK), respectively. Methanol extracts from S-YRK exhibited higher inhibitory effect on the growth of AGS human gastric adenocarcinoma cells in a time dependent-manner compared to C-YRK at the same concentration. 4,6-diamidino-2-phenylindole (DAPI) staining showed that S-YRK induced apoptosis accompanied by the increased Bax but decreased Bcl-2 in mRNA expression. Moreover, S-YRK decreased the levels of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) mRNA expressions. The results suggested that S-YRK cultivated in the presence of sulfur elicited stronger anticancer activity than C-YRK in vitro. Dietary intakes of S-YRK may be beneficial to decrease the risk of cancer.

        문헌에 나타난 열무김치 및 열무물김치 제조 방법의 표준화

        조리서 및 문헌을 통해 열무김치 및 열무물김치의 재료배합비 및 제조방법을 표준화하였다. 열무김치의 표준화과정에서 열무 이외에 사용빈도수가 50%이상인 부재료는 파와 홍고추였으며, 양념류로는 고춧가루, 마늘, 생강, 젓갈이 이에 속했다. 또한 재료 및 양념의 배합비는 절인 열무 100 g에 대해 파 8.0±3.8, 마늘 2.9±1.3, 생강 1.6±0.7, 홍고추 7.0±1.7, 고춧가루 4.2±1.2, 젓갈 3.7±0.5로 나타났다. 열무물김치의 표준화과정의 경우, 사용빈도수가 50%이상인 부재료로는 풋고추, 홍고추, 파가, 양념류로는 마늘, 생강, 전분이 이에 속했으며, 열무물김치의 재료배합비의 표준화는 첨가된 물의 양을 기준으로 하였다. 즉, 물 100 mL에 대해 절인 열무 50.6±10.8, 파 3.3±1.3, 풋고추 3.3±1.9, 홍고추 2.4±1.3, 마늘 3.0±0.7, 생강 1.5±0, 전분 1.5±0.6의 비율로 나타났다. Manufacturing method and ingredient ratio for young radish kimchi (yulmoo kimchi) and young radish watery kimchi (yulmoo mool-kimchi) were standardized from literatures. Ingredients having frequency of use greater than 50% were only used in the standardization process. Green onion, red pepper, red pepper powder, garlic, ginger, and anchovy juice were included in young radish kimchi. Green pepper, red pepper, garlic, ginger, and starch were included in young radish watery kimchi. The standardized ingredients ratio of young radish kimchi (yulmoo kimchi) on young radish 100 g was as follows: green onion 8.0±3.8, crushed garlic 2.9±1.3, crushed ginger 1.6±0.7, red pepper 7.0±1.7, red pepper powder 4.2±1.2, and anchovy juice 3.7±0.5. The standardized ingredients ratio of young radish watery kimchi (yulmoo mool-kimchi) on added water 100 mL was as follows: young radish 50.6±10.8, crushed garlic 3.0±0.7, crushed ginger 1.5±0, green onion 3.3±1.3, green pepper 3.3±1.9, red pepper 2.4±1.3, and starch 1.5±0.6.
