        임신 및 각종 갑상선질환에서 갑상선 기능 판정에 관한 연구 : 혈청유리 T4의 진단적 의의에 관한 고찰 The diagnostic value of free thyroxine by RIA

        To evaluate the diagnostic accuracy of the measurement of free thyroxine(FT4) by radioimmunoassay, we measured free T4 and T4, T3, T3RU, TSH and TBG serum levels by radioimmunoassay in 18 healthy persons and 52 patients with various thyroid diseases and 11 normal pregnant women. The results are as follows. 1) In 19 cases of overt hyperthyroidism, T3, free T4 and FTI, T4/TBG ratio reflect hyperfunction in all cases. T4 is increased in 94%(18/19) and TBG and TSH are decreased in 79%(15/19). 2) In 8 patients with overt hypothyroidism, TSH is increased in all cases and free T4 and FTI is decreased in all cases. T4 is decreased in 87.5%(7/8), T3 is decreased in 75%(6/8) and T4/TBG ratio is decreased in 62.5%(5/8). 3) In 5 patients who are clinically in euthyroid state after treatment of hyperthyroidism, T4 free T4, FTI and TSH are in the normal range in all cases and T3 is normal in 60%(3/5) and slightly increased in 40%(2/5). 4) In 10 patients who showed clinically borderline hypothyroidism after treatment of hyperthyroidism, TSH is increased in all cases and free T4 and FTI are decreased in all cases, but T4 and T3, T4/TBG ratio are in the normal limit in all cases. So after treatment of hyperthyroidism, TSH, free T4 or FTI are recommended as optimal function test. 5) In normal pregnancy, free T4, FTI and T4/TBG ratio reflect normal function, but the other parameters unreliable due to the influence of increased TBG. Also TBG and TSH level in pregnancy is increased significantly compared with normal healthy control group. 6) The coefficients of correlation between T4 and FTI were 0.862(p〈0.001) and 0.685(p〈0.001) between free T4 and T4/TBG ratio. In most patients, diagnostic value of free T4 was comparable and even superior to FTI, so free T4 measurement can be used routinely with thyrotropin assay in the diagnosis of hypothyrodism or with T3 for the diagnosis of hyperthyroidism.

        CIE L 기준에 의한 PSE 육과 정상육의 육질 및 도체특성에 관한 연구

        Two hundreds and twenty two pork carcasses from a commercial breed were investigated for meat quality and carcass traits of PSE and normal pork classified by CIE L value. pH and conductances were taken on 1(pH₁, C₁), 3(pH₃, C₃), 6(pH_6, C_6) and 24 hr postmortem(pHu, C_u). Significant differences between PSE and normal pork were observed in pH₁, pH_u (p $lt; 0.001) and pH₃(p $lt; 0.05). But, no significant difference was found in pH_6. Although, there was no significant difference in C₁, significant differences are found in C₃(p $lt; 0.05), C_6 (p $lt; 0.01) and C_u(p$lt;0.001). The water holding capacity(WHC) were significantly lower(p$lt;0.01) in the PSE compared to the normal pork. The cooking loss were significantly higher(p $lt; 0.05) in the PSE compared to the normal pork. The CIE L value was highly related with NPPC color scale(R²=0.77), moderately related with pH_u (R²=0.45), C_u(R²=0.57), WHC(R²=0.49), cooking loss(R²=0.45) and juiciness(R²=0.43), and poorly related with pH₃(R²=0.27), pH_6(R²=0.27) and conductances(R²=0.05-0.15). Consequently, the results suggest that pork quality classification by CIE L value expresses obvious differences in most of important pork quality parameters like pH₁, pH_u, C_u visual color and WHC.

        정상 임신에서 임신주기에 따른 갑상선기능의 변화

        It is well known that normal pregnancy is accompanied by a rise in serum concentrations of thyroxine-binding globulin(TBG) and human chorionic gonadotropin (hCG). Alterations of biochemical parameters of thyroid function are recognized during gestation and sensitive tests to evaluate the alterations easily are required. Therefore, a cross-sectional study was undertaken in 140 healthy pregnant women to evaluate the efficacy of free T_4 measured by 2-step RIA compared to other thyroid function tests and to confirm the changes of thyroid function according to the stages of normal pregnancy. The sensitivities of free T_4 index, free T_4(by 2-step RIA), T_3 and TSH were realtively high(99.3%, 93.6%, 92.9%, 83.6%, respectively) compared to those of T_4 and T_3 bead upgake(49.3%, 21.4%) during all stages of pregnancy. There were positive correlations between free T_4 index and free T_4 or total T_4(r=0.68, r=0.72; p$lt;0.001). The values of free T_4 index sharply decreased from 3.22+-0.10(meam +-SEM) during 6th-12th week to an plateau after 16th-20th week of gestation(p$lt;0.01). The serum concentrations free T_4 and T_3 bead uptake also significantly decreased from 1.65+-0.05 ng/dl, 24.7+- 0.7% during 6th-12th week to an plateau after 16th-20th week of gestation, respectively(p$lt;0.001), No differences were found in the changes of serum concentrations of T_3, T_4 and TSH according to the stages of pregnancy. In conclusion, it is adequate to measure some tests including free T_4 index and free T_4 to evaluate thyroid function during pregnancy. The thyroid physiology and changes of thyroid function according to the stages of pregnancy should be considered in the interpretation of thyroid function status during pregnancy(J Kor Soc Endocrinol 9: 183-189, 1994).

      • 돈 분뇨 액비에 미생물 첨가가 배추의 발아지수에 미치는 영향

        본 연구는 돈 분뇨 액비에 미생물 첨가가 배추의 발아지수에 미치는 효과를 구명하기 위해서 수행하였다. 돈 분뇨 액비는 107일 동안 폭기와 무폭기 상태로 액상비료를 제조하여 공시하였고 미생물 첨가는 $0.05\%,\;0.1\%,\;0.2\%,\;0.3\%$ 수준으로 하였다. $0.05\%$ 미생물 접종 처리구에서 식물 독성이 낮고 발아지수가 높게 나타났다. 첨가수준별 돈분 액비 이용시험에서 초기 돈 슬러리의 액비성상은 T-N 2,873 ppm, T-P 753 ppm, $NH_4-N$ 1,441.6 ppm, $NO_3-N$ 16.48ppm으로 50일이 되면서 폭기조건에서 T-N 3,672 ppm, T-P 164 ppm, $NH_4-N$ 183.87 ppm, $NO_3-N$ 21.97 ppm로 변화하였으며, 무폭기 조건에서 T-N 1,261 ppm, T-P 68 ppm, $NH_4-N$ 161 ppm, $NO_3-N$ 16.87 ppm로 나타났다. 미생물을 처리하지 않고 107일동안 폭기조건과 무폭기 조건으로 액비화한 시료의 발아지수는 각각 83과 40.4이였다. 돈 분뇨 액비를 무산소 제조시 $40\%$ 이상, 호기제조시 $50\%$ 이상의 발아지수가 개선되었다. 이와 같은 결과는 액비제조 처리가 무산소 제조보다는 호기제조 처리가 작물에 양호한 것으로 판단되어지며 이는 호기처리시 총질소의 함량을 높게 유지할 수 있도록 하는 결과인 것으로 사료된다. This study was carried out to investigate the effect of Synechocystis sp. KACC 91007 when added to a pig slurry or pig liquid fertilizer (PLF) on germination index (GI) of Chinese cabbage. The preliminary experiment involved the screening of inoculant levels which were; 0.05, 0.1, 0.2, and $0.3\%$, respectively. The $0.05\%$ level of inoculant was selected based on low phytotoxicity and high GI.. The PLF underwent a 107 day aerobic and anoxic processing conditions. The T-N, T-P, $NH_4$, and $NO_3-N$ concentrations of the untreated pig slurry were; 2,873, 753, 1,441.6, and 16.48 ppm, respectively. Using aerobic processing treatment, the fertilizer value of the PLF was 3,672, 164, 183.87, and 21.97 ppm, respectively. In contrast, the fertilizer value of the PLF processed under anoxic condition was reduced to 1,261, 68, 161, and 16.87 ppm. The GI value of the untreated PLF under aerobic and anoxic processing condition was 83 and $40.4^{*}\%$, respectively. With the addition of the $0.05\%$ microbial inoculant, the GI improved by more than 40 and $50\%$ respectively, when the PLF was processed under anoxic and aerobic conditions. The above findings proved that the aerobic processing of PLF for 107 days was better than anoxic and yielded higher T-N which is a macro-nutrient fertilizer material. Consequently, the addition of $0.05\%$ microbial inoculant resulted to a higher GI of the Chinese cabbage specifically under aerobic processing condition.

      • Inhibition of Porcine Endogenous Retrovirus by Multi-Targeting Micro RNA Against Long Terminal Region

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>There might be much benefit in xenotransplantation, however, the risk of infections across species barriers remains, especially porcine endogenous retrovirus (PERV). To date, many attempts have been made to knock down active PERVs by inhibitory RNA (RNAi) and micro RNA (miRNA), which target different genes of PERV. There are a few studies that have explored whether targeting promoter regions of PERV could exert an inhibition effect.</P> <P><B>Methods</B></P> <P>miRNAs were automatically selected based on an online program BLOCK-iT RNAi Designer. The inhibition efficiency between miRNAs was compared based on their inhibition of different PERV genes: long terminal repeats (LTR), gag, and pol. Both relative quantitative real-time polymerase chain reaction (PCR) and C-type reverse transcriptase activity were performed.</P> <P><B>Results</B></P> <P>The results demonstrated that miRNA targeting the LTR region degraded the target sequence, and simultaneously inhibited the mRNA expression of both gag and pol genes of PERV. The LTR1, LTR2, and dual LTR1 + LTR2 miRNA inhibited 76.2%, 22%, and 76.8% of gag gene expression, respectively. Similarly, the miRNA was found to knock down the pol gene expression of 69.8%, 25.5%, and 77.7% for single targeting miRNA (LTR1 and LTR2) and multi-targeting miRNA (LTR1 + LTR2), respectively. A stable PK15 clone constitutively expressed dual LTR1 + LTR2 miRNA and exhibited higher inhibitory up to 82.8% and 92.7% of the expressions of the gag and pol genes, respectively. Also, the result of co-cultivation of dual LTR1 + LTR2 miRNA transfected PK15 cell with a human cell line inhibited expression of LTR, gag, and pol genes of PERV.</P> <P><B>Conclusions</B></P> <P>In conclusion, this study suggested that the LTR might be an alternative target for gene silencing of PERV, and that multi-targeting miRNA had better inhibitory effect than single- targeting miRNA.       • On the possibilities to use atmospheric reanalyses to evaluate the warming structure in the Arctic

        <P>Abstract. There has been growing interest in the vertical structure of the recent Arctic warming. We investigated temperatures at the surface, 925, 700, 500 and 300 hPa levels in the Arctic (north of 70° N) using observations and four reanalyses: ERA-Interim, CFSR, MERRA and NCEP II. For the period 1979-2011, the layers at 500 hPa and below show a warming trend in all seasons in all the chosen reanalyses and observations. Restricting the analysis to the 1998-2011 period, however, all the reanalyses show a cooling trend in the Arctic-mean 500 hPa temperature in autumn, and this also applies to both observations and the reanalyses when restricting the analysis to the locations with available IGRA radiosoundings. During this period, the surface observations mainly representing land areas surrounding the Arctic Ocean reveal no summertime trend, in contrast with the reanalyses whether restricted to the locations of the available surface observations or not. In evaluating the reanalyses with observations, we find that the reanalyses agree better with each other at the available IGRA sounding locations than for the Arctic average, perhaps because the sounding observations were assimilated into reanalyses. Conversely, using the reanalysis data only from locations matching available surface (air) temperature observations does not improve the agreement between the reanalyses. At 925 hPa, CFSR deviates from the other three reanalyses, especially in summer after 2000, and it also deviates more from the IGRA radiosoundings than the other reanalyses do. The CFSR error in summer T925 is due mainly to underestimations in the Canadian-Atlantic sector between 120° W and 0°. The other reanalyses also have negative biases in this longitude band. </P>
