Seltzer Model에서 인삼과 morphine의 공동투여가 통각행동에 미치는 영향

The aim of this study was to compare the effects of the ginseng total saponin(100㎎/㎏, i.p., GTS), morphine(3㎎/㎏, i.v.) and the co-administration of the GTS and morphine at same time after the unilateral tight ligation of about half of the sciatic nerve in rats. This tight ligation evoked a markedly different responses of the neuropathic pain in both injured left sides and non-injured right side, which is representing the mirror effect in spite of the time courses of the various stimili; mechanical, chemical and cold, but exception of spontaneous. The maximal responses of each stimulus showed neuropathic pain evoked at the seventh day, and the values of the response were used as a indicater of the nerve injury group for drugs; GTS alone, morphine alone, and the co-administration of GTS and morphine at same time. The effect of the GTS was continued for approximate 48 hours and morphine for 2 hours, respectively. The co-administration of the GTS and morphine at same time, however, showed the no considerable antinociceptive effect at first injection, but the consideralble difference after injection of 24 hours of 48 hours. And also these responses showed the mirror-like response after the drug treatments. These results suggest that partially tight ligation evoked the neuropathic pain in the various stimuli showing the mirror-like effect, and the actions between GTS and morphine has the antagonistic relationship with each other, and the action mechanism is partly involved the non-opioid and opioid mechanisms.

The quantitative analysis of heavy metals(As, Cd, Cr, Hg, Pb, Cu) in the sea water around Kaduck island was conducted during the period of March, 1998 to September, 1998. The sea water in the four regions was sampled four times and the heavy metals were analyzed. The Heavy metal concentration of the sea water around Kaduck island was As 127.7㎍/ℓ , Cd 49.28㎍/ℓ , Cr 0.462㎍/ℓ, Hg 0.111㎍/ℓ, Pb 1.713㎍/ℓ, Cu 2.716㎍/ℓ in the average value, respectively.

Pseudomembranous colitis (PMC) is a disease caused by Clostridium difficile proliferation. The causative drugs are clindamycin, third-generation cephalosporins, flouroquinolone and so on. Rifampicin has been reported as a cause of PMC in the 1980s, and the frequency of PMC is increasing because rifampicin is a first line drug for anti-tuberculosis therapy. Two patients were recently admitted to our hospital due to watery diarrhea for 1 month and they were diagnosed with PMC by sigmoidoscopy. Their onset age were 74, 72 years old and latent period of symptoms were 60, 129 days, respectively. In one case, the patient displayed coexisting hypertension and diabetes. The clinical symptoms improved after discontinuing the rifampicin and then administering oral metronidazole. We report here on two cases presumed to be rifampicin-induced PMC.

카드뮴폭로후 혈액과 요중 카드뮴의 변화양상

The changes of cadmium(Cd) in plasma, whole blood, erythrocyte and urine in Sprangue-Dawley male rats exposed to intravenous singleinjection of 0.8 ㎎ CdCl₂/kg of body weight were investigated. Blood was taken at 0.5 to 672 hours, and 24 hour-urine was collected by using metabolic cage for the same period. The plasma level of Cd was reached to peak at 0.5 hour after injection and reduced rapidly in 1 hour. The Cd level in blood was the highest in plasma and the lowest in erythrocyte at 0.5 hour after injection. However, in one hour postinjection, the levels of Cd were higher in order of erythrocyte, whole blood and plasma, up to 4 weeks. The changes of urinary volume and creatinine were not significant between Cd-treated and saline-treated groups. However, urinary protein was slightly increased with time in Cd-treated group. Urinary Cd level was higher in Cd treated group than control. These results suggest that the measurement of Cd in erythrocyte and urine is valuable for the biological index to estimate recent Cd exposure.

말초성 신경아세포종 : 두개골 발생 증례 보고 Involving Skull

The peripheral neuroblastomas are malignant neoplasms that originate from the adrenal gland and sympathetic nervous tissues. They usually occur in childhood. They exhibit such malignant features that they metastasize early to lung, liver, bone, rarely skull and other structures. The patients with peripheral neuroblastoma have short duration of symptom and short survival period. They reveal the characteristic light microscopic features that resemble other small cell carcinomas. The establishment of final diagnosis of peripheral neuroblastoma needs various special stainings for small cell carcinomas, and the electron microscopic findings are the most reliable. The authers recently experienced a case of peripheral neuroblastoma in a 26-year old man which involved right frontoparietal skull vault. The only chief complaint was a local non-tender mass at right frontoparietal scalp. Neither headache nor any neurological deficits was detected. Simple skull X-ary revealed a punched out rediolucency at right frontoparietal bone and brain CT showed a superficial elliptical high density mass that enhanced strongly. The mass was totally removed by wide craniectomy. The tumor invaded and penetrated the dura but the arachnoid membrane. at the tumor bed was not invaded by the tumor. The tumor was confirmed as peripheral neuroblastoma by various special stainings for small cell carcinomas. Following surgical resection of the mass, post-operative radiotherpy was offered(4800 rads for about 5 weeks). The patient aggrevated progressively and showed numerous metastases to such bones as lumbar vertebrae, pelvis and humerus to became paraplegic. 8 months after the operation, the patient died.

급성 담낭염에 대한 내과적 치료의 역할

In 58 patients who were admitted to department of internal medicine, ChungNam national university hospital from January 1989 to March, 1993. We evaluated for medical treatments and the following results were obtained. 1) The ratio of male and female was 1 : 1 and mean age of the patients was 57.4 years. 2) At arrival on hospital, the most common clinical symptom was pain in right upper quadrant and epigastrium(93.1%), the remainders were fever and chill(79.3%), nausea(51.7%) and vomiting(36.2 %) in order of frequency. 3) The most common physical findings was tenderness on the right upper quadrant and epaigastrium(98%), the remainders were jaundice(50%) and palpable gall bladder(13.8%) in order of freguency. 4) In laboratory examination, elevated levels of ESR(82.8%), leukocytosis(67.2%), elevated levels of AST(62.0%), ALT(50.0%), serum total bilirubin(51.7%) and alkaline phosphatase (43.1%) were noted. 5) The mean period of NPO was 3.9 days and the mean hospital period was 13.5 days. 6) The rates of combined complications in patients with acute cholecystitis were 17.2%; these are gall bladder empyema(5.2%), hydrops(5.2%), perforation(3.4%), sepsis(3.4%) and intestinal obstruction (1.7%). 7) In 96.6 percent of 58 cases, the symptoms of acute cholecystitis were improved by conservative management. 8) The mean follow-up period was 25.5 months and the recurrence rate was 10% during this period.

Characterization of Cryptopygus antarcticus endo-β-1,4-glucanase from Bombyx mori expression systems.

<P>Endo-β-1,4-glucanase (CaCel) from Antarctic springtail, Cryptopygus antarcticus, a cellulase with high activity at low temperature, shows potential industrial use. To obtain sufficient active cellulase for characterization, CaCel gene was expressed in Bombyx mori-baculovirus expression systems. Recombinant CaCel (rCaCel) has been expressed in Escherichia coli (Ec-CaCel) at temperatures below 10C, but the expression yield was low. Here, rCaCel with a silkworm secretion signal (Bm-CaCel) was successfully expressed and secreted into pupal hemolymph and purified to near 90% purity by Ni-affinity chromatography. The yield and specific activity of rCaCel purified from B. mori were estimated at 31 mg/l and 43.2 U/mg, respectively, which is significantly higher than the CaCel yield obtained from E. coli (0.46 mg/l and 35.8 U/mg). The optimal pH and temperature for the rCaCels purified from E. coli and B. mori were 3.5 and 50C. Both rCaCels were active at a broad range of pH values and temperatures, and retained more than 30% of their maximal activity at 0C. Oligosaccharide structural analysis revealed that Bm-CaCel contains elaborated N- and O-linked glycans, whereas Ec-CaCel contains putative O-linked glycans. Thermostability of Bm-CaCel from B. mori at 60C was higher than that from E. coli, probably due to glycosylation.</P>

Efficient Production of Recombinant Protein in HSPs (Heat Shock Porteins) Transgenic Silkworm

Many thousands of recombinant proteins have been successfully produced in baculovirus - infected insect cells and larvae. In this study, to improve its value and the yield of recombinant protein production, we constructed transgenic silkworm using Heat shock genes with regard to protein folding. This time, we adapted GAL4/UAS system to express at necessary time point and to carry genes for foreign protein. First, we generated two transgenic cells and silkworm lines that carried the silkworm heat shock proteins, UAS-HOP and UAS-HSC70 and UAS-HSP70 and UAS-HSP40 construct plus 3xP3-DsRED. Subsequently, to drive the GAL4 gene as activatorvector, we engineered Baculoviruses that contain the GAL4 under the P10 promoter linked to the expression cassette of interest foreign genes under the polyhedron promoter. Also, activator vector linked to the GAL4 was designed expressing 6xHis and 6xHis–GST tag. Infection of silkworm larvae with recombinant virus, His-tagged human C3d gene was more efficiently produced transgenic silkworm than that of wild-type, but not His-GST tagged. We show the possibilityin use of HSPs transgenic silkworm system by GAL4/ UAS BmNPV that can generate the efficient production of foreign protein.
