      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

      • 대호 간척기 토양의 염농도별 밭작물의 염해 평가

        This study was carried out to obtain the basic data for selecting the applicable crops in reclaimed land during desalinization period. A pot experiment was conducted with 5 different electrical conductivities of the saturated extracts (ECe 1, 3, 9, 14, and 16 dS·m^(-1)) of soils taken from the Dae-Ho reclaimed tidal lands. Eight crops (Chinese cabbage, radish, tomato, red pepper, buckwheat, soybean, sesame, and green perilla) were grown for 37days. Plant height and number of leaves were surveyed on 2 and 4 weeks after seeding, and on harvest time (5 weeks). After harvest, dry weights of harvested crops were measured and soil chemical properties were analyzed. Emergence rates of crops were comparatively high except sesame. For sesame, there was no emergence at ECe over 3 dS·m^(-1). Growth and dry weight decreased significantly as increasing ECe. The ECe which decreased 50% of dry weight index were 14.2 dS·m^(-1) for radish, 11.4 dS·m^(-1) for Chinese cabbage, 10.2 dS·m^(-1) for tomato for red pepper, 8.9 dS·m^(-1) for buckwheat and green perilla, 8.6 dS·m^(-1) for soybean, and 8.9 dS·m^(-1) for tomato. At higher ECe that start the growth inhibition, increasing 1 dS·m^(-1) in ECe, 7.7, 6.5, 5.9, 5.6, 5.2, and 4.9% of dry weight decreased for buckwheat, green perilla, Chinese cabbage, radish, soybean, and tomato (red pepper), respectively. The critical value of ECe for crop survival except sesame was 15.4~23.1 dS·m^(-1).

      • 갓김치 ether 추출물의 발효 과정상의 생리 기능성 변화

        일반 김치의 경우 17±1℃에서 2일 사이에 pH가 4.0에 이르는 것에 비해 갓김치의 경우 18℃에서 저장한 경우에 14일 경에 pH가 4.0(Fig.1)에 이르러 일반 김치 보다 저장성이 3~5배 높은 것으로 나타났으며 총균수에서도 비슷한 양상을 나타내었다. 또한 간암세포에 대한 항암성에서 IC30의 농도가 발효기간별 시료 모두 1000㎍이상으로 나타났으며 발효 0일째 시료가 1000㎍에서 50%의 간암세포 사멸성을 나타내서 가장 항암효과가 우수한 것으로 나타났고, 간정상세포에 대해서는 1000㎍ 이상에서도 28% 이하의 사멸성을 나타내서 정상세포에 대해 큰 영향을 미치지 않는 것으로 나타났다. 또한 항산화 효과에 있어서는 초기(0~2일) 발효시 수소공여능이 53%로 나타나서 다른 식품 추출물의 항산화 효과에 비해서는 비교적 높은 효과를 나타내었다. 그러나 발효 14일에는 15%로 감소되어서, ethyl ether에 의해 추출되어 항산화 효과를 나타내었던 물질들이 발효가 진행됨에 따라 감소된 것으로 사료되었다. The anticancer and antioxidative effects of Mustard Leaf Kimch ether extracts according to various fermentation period were investigated. pH of the Mustard Leaf Kimchi was reached at 4.0 on 14th day, so the period reaching well-fermentating state was up to 3~5 times longer than that of ordinary Kimchi. The anticancer effect on HepG2 by the addition of fermented Mustard Leaf Kimchi ether extracts was higher than that on CCL-13. IC30 of Mustard Leaf Kimchi in HepG2 was showed more than 1000㎍.Especially, 50% of death ratio was showed in HepG2 cells by the addition of ether extracts(0 day). the other side. 28% of death ratio was shown in CCL-13 up to 1000㎍ of ether extracts. As well as the Mustard Leaf Kimchi ether extracts(0-2 days) showed higher antioxidative effect than of others. The hydrogen donor activity was 53% at initial fermentation period, but it was decreased at 14% on 14th day. So, ether soluble antioxidative materials in Mustard Leaf Kimchi was decreased during fermentation.

        The purpose of this investigation was to develop a new carrier using chitin and glucan as the model for Aspergillus oryzae cell wall. In case of the ratio of chitin and glucan(3/1), the activity of the immobilized protease had higher than that of model(chitin/glucan=1/3) and carriers were formed gel. In case of soluble chitin and glucan treated with calcium carbonate, there were a 1.8-fold increase in activity of immobilized protease compared with untreated chitin. The scanning electron microscope image showed that a pore size of this carrier had a homogenized pore size and a smooth surface.

      • 협착관내에서 멱법칙 비뉴턴 유체와 점탄성 유체의 유동특성에 관한 연구

        비뉴턴 유체의 구성방정식 중에서 수정멱법칙모델을 이용하여 원형단단면을 갖는 직선관, 급격축소-급격확대 협착관, 그리고 점차축소-점차확대관 내에서의 유동특성을 수치모사하였다. 점차축소-점차확대관은 직선으로 연결되는 경우와 코사인곡선으로 이루어지는 경우로 구분하였다. 비뉴턴 유체중에서 저항감소현상을 나타내는 점탄성유체를 이용하여 급격축소-급격확대 협착관에서의 유동특성을 실험적으로 결정하였다. 완전히 발달된 층류유동에서 멱법칙 지수 n가 작아질수록 속도 형상은 관의 중앙부분에서 평탄해지고 압력손실은 증가한다. 급격축소-급격확대 협착관이 점차축소-점차확대협착관에서 곡선의 영향은 별로 크지 않다. 점탄성 유체의 압력손실계수는 층류영역에서 Reynolds수의 함수로 나타나며, 물보다 크게 나타난다. 협착비율과 협착길이비가 클수록 압력손실계수는 크게 나타난다. The modified power law model for a non-Newtonian fluid is employed to simulate the interanal flows of smooth straight tube, sudden contraction-sudden sxpansion tube, and gradual contraction-gradual expansion tube. Geometries of the gradual contraction-gradual expansion tubes are made of straight line or cosine curve. Experimental investigations using the drag reducing polymer solutions that are known as the viscoelastic fluids are conducted for the internal flows with sudden cotraction-sudden expansion tubes. The velocity profile in the contral region becomes flat and the pressure loss increases as the power law index n decreases in the fully developed laminar flow. The pressure loss of the sudden contraction-sudden expansion tube is larger than that of the gradual contraction-gradual expansion tube. The effect of index n on the sudden contraction-sudden expansion is greater than that on the gradual contraction-gradual expansion tube. The curvature effect of the gradual contraction-gradual expansion tube is not pronounced. The pressure loss coefficients of viscoelastic fluid is larger than that of water for the same Reynolds number. The loss coefficient of the stenosed tube increases as the percent contraction ratio and the length to diameter ratio increase.

        소아에서 이모장치가 하악골 및 측두하악관절에 미치는 영향에 관한 유한요소법적 분석

        The purpose od this study was to investigate the stress distribution and displacement in the mandible and temporomandibular joint during application of chin cap. The dry skull of child in mixed edntition was used to make a 2-dimensional finite element model. It was composed of 765 elements and 1321 nodes. An orthopedic forces, 500gm,was applied around the gnathion to condylion(A plane),the second was 20 degrees upward from the first plane(B plane),the third was 50 degrees upward from the first plane(C plane). The results were as follows: 1.When the retraction was applied to A plane,the distribution of tensile stress was greater on the middle and posterior portion of mandiular fossa, apex of condylar head, anterior portion of articular disc, distal portion of first molar, posterior portion of condylar neck and head. Particulary, it was concentrated on condylar head and neck. As the retracion was directed more upward, it was decreased totally and concentrated on the first molar. 2.When the retraction was applied to A plane, Compressive stress concentrated on the sigmoid notch and anterior portion of condylar head and neck. As the retraction was directed more upward, it moved from condylar head and neck toward anterior teeth portion and it was concentrated on second premolar. 3.When the retraction was applied to A plane, displacement was directed to posterior and superior in mandibular anterior body, mandibular posterior body, coronoid process, condylar head,middle and posterior portion of mandibular fossa. But it was directed to posterior and inferior in mandibular angle and ramus, anterior portion of condylar neck, anterior portion of mandiblar fossa. As the retraction was directed more upward, it was directed to posterior and inferior in coronoid process and posterior portion of condylar neck. The posterior movement of mandible was decreased. 4.When the retraction was applied to A plane, the stress of mandibular body and ramus was small and it was distributed on compact bone. As the retraction was directed more upward, the stress was distribeted widely on teeth and permanent teeth germs.

        There exists a major impediment to learning electricity which may illuminate why direct presentation of scientists' ideas is too aggressive a strategy. This impediment lies in thinking that electricity is used up and in envisaging the light bulb as a consumer. This view is based on a powerful idea which summarises much of our experience; it declares that 'things get used up', 'you cannot get something for nothing', and 'intuitive thinking'. To replace this view with scientific view, we made the analogy model. It is suggested that the analogy model is more effective in teaching electricity circuits than conventional teaching method.

      • 십이지장 궤양 환자와 만성 위염 환자의 염증 분포의 차이 및 H. pylori 집락도와 염증도의 상관관계

        An inflammatory difference between the gastric antrum, body and fundus was studied in 39 H.pylroi positive duodenal ulcer and 29 H. pylori chronic gastritis patients. Also we studied correlation of H. pylori colony count and inflammatory severity. Patients with newly diagnosed duodenal ulcer and chronic gastritis were endoscoped and two biopsy secimens each taken from the antrum, body and fundus. Although intestinal metaplasia in antrum and inflammatory activity in fundus were more severe in chronic gastritis patients. another inflammatory difference was equally likely to be found between two groups. There was no definite histological inflammatory difference between duodenal ulcer and chronic gastritis patients. A positive correlation was found between the number of H. pylori and the severity of inflammation.
