        The purpose of this study was to reform practically a Korean Judo's Doechigi-Bon(Forms of Counterattack, judo's Gaeshi no Kata, Judo's Uradori no Kata, Judo's Gonosen no Kata) that was established in 1955 Korea, according to changing of techniques by Judo's modernization, in order to have Judo's carefulness and systematic diffusion.       • Discovery of urinary metabolomic biomarkers for early detection of acute kidney injury

        <P>The discovery of new biomarkers for early detection of drug-induced acute kidney injury (AKI) is clinically important. In this study, sensitive metabolomic biomarkers identified in the urine of rats were used to detect cisplatin-induced AKI. Cisplatin (10 mg kg(-1), i.p.) was administered to Sprague-Dawley rats, which were subsequently euthanized after 1, 3 or 5 days. In cisplatin-treated rats, mild histopathological alterations were noted at day 1, and these changes were severe at days 3 and 5. Blood urea nitrogen (BUN) and serum creatinine (SCr) levels were significantly increased at days 3 and 5. The levels of new urinary protein-based biomarkers, including kidney injury molecule-1 (KIM-1), glutathione S-transferase-alpha (GST-alpha), tissue inhibitor of metalloproteinase-1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, neutrophil, neutrophil gelatinase-associated lipocalin (NGAL), and osteopontin, were significantly elevated at days 3 and 5. Among urinary metabolites, trigonelline and 3-indoxylsulfate (3-IS) levels were significantly decreased in urine collected from cisplatin-treated rats prior to histological kidney damage. However, carbon tetrachloride (CCl4), a hepatotoxicant, did not affect these urinary biomarkers. Trigonelline is closely associated with GSH depletion and results in insufficient antioxidant capacity against cisplatin-induced AKI. The predominant cisplatin-induced AKI marker appeared to be reduced in urinary 3-IS levels. Because 3-IS is predominantly excreted via active secretion in proximal tubules, a decrease is indicative of tubular damage. Further, urinary excretion of 3-IS levels was markedly reduced in patients with AKI compared to normal subjects. The area under the curve receiver operating characteristics (AUC-ROC) for 3-IS was higher than for SCr, BUN, lactate dehydrogenase (LDH), total protein, and glucose. Therefore, low urinary or high serum 3-IS levels may be more useful for early detection of AKI than conventional biomarkers.</P>

        진동에 의한 혈관 내 폐 보조장치의 산소전달 특성

        본 연구는 급성호흡부전환자를 치료하기 위하여 사용되고 있는 혈관 내 폐 보조장치의 기체전달을 향상시키기 위하여 중공사막에 진동기법을 사용하여 기체전달을 향상시키고자 시도하였다. 그리고 혈관 내 폐 보조장치를 정맥에 삽입하기 전, 혈관 내 폐 보조장치의 설계조건을 실험적 모델을 통하여 기체전달을 예측할 수 있는 예측식을 만들고자 하였다. 실험결과, 본 연구에서는 진동기법이 기체전달을 향상시키는데 효과적임을 확인할 수 있었다. 뿐만 아니라, 충진율과 가징 주파수의 함수관계를 이용하여 기체전달을 예측 할 수 있었다. 실험에 의하여 얻어진 결과는 예측식에 의하여 얻어진 결과와 유사한 결과를 얻었다. 그러므로 본 연구에서는 충진율과 가진 주파수의 함수를 이용하여 VIVLAD의 기체전달을 예측할 수 있었다. In this paper, we tried to improve gas exchange of the vibrating intravascular lung assist device(VIVLAD) using vibrating method in the hollow fiber membrane, for the patients suffering from acute respiratory distress syndrome(ARDS). And we tried to formularize prediction equations to make a prediction about gas transfer for designing intravenous artificial lung assist device, and designed modules under various conditions were studied through an experimental modeling before inserted the artificial lung assist device into as venous. As a result, we are convinced that vibration method is very useful for the has transfer increasing. Also, we can estimate the gas transfer as a function of the packing density and excited frequencies. The gas transfer obtained from the experiment was similar to that from the prediction equation, confirming the usefulness prediction equation. Therefore, we can estimate the gas transfer of the VIVLAD as a function of the packing density and excited frequencies.

      • 2000년 시드니 올림픽 유도경기 대비 세계 우수선수들의 개인별 기술 및 패턴에 관한 연구 : 남자선수를 중심으로

        The purpose of this study was to analyze international contest levels and the competition techniques traits of each category, individual scoring - losing techniques of world elite judokas who were awarded in the international judo tournaments (I.J.T.) (part 1: ~O.G '96, part 2 :from O.G '96 to July, 2000), and who are expected to participate in the 27th Olympic Games(O.G.) Sydney 2000 to prepare the 27th O.G. which will be held from 16. to 22. September 2000 at Exhibition (Center, Darling Harbour, in Sydney, Australia The records of the results and contents of competition were obtained out of (115 for man,) I.J.T., world elite judokas. To decide individual contest levels, groups were derided into 3(A,B,C)groups and points were graded by 3 ways. The I.J.T. that were derided into 3 groups are shown in the table 1.

      • Interleukin-2가 Mouse 자궁내막의 Acid Phosphatase 활성에 미치는 영향

        Interleukin-2, a kind of lymphokine produced by the T-lymphocyte stimulated by antigen or mitogen, has been studied intensively with the development of immuno-oncology and chemoimmunotherapy of the malignancy. Interleukin-2 is known to exert on antitumor effect by stimulating the proliferation of helper and cytotoxic T lymphocytes, augmenting the cytolytic activities of the cytotoxic T lymphocyte and differentiating a precursor cell into a lymphokine-activated killer cell in vivo. Though interleukin-2 suppresses the metastasis of the malignant tumor into the adjacent organs, it develops many side effects including fever, chills, malaise, mental confusion, diarrhea, vomiting and oliguria. Interleukin-2 also causes pulmonary, hepatic and renal toxicities and, especially, increase of capillary permeability with weight gain, pulmonary edema and ascites. In this experiment, the author studied the effect of interleukin-2 on the mouse endometrium, observing histochemically the changes in the activity of acid phosphatase following the estrous cycle. Female, DDY strain, weighing 20gm and exhibiting normal estrous cycle, were used as experimental animals. The estrous cycle was recognized by the vaginal smear. The experimental animals were treated with 2 million units of interleukin-2/0.2ml per Kg of body weight and with 0.2ml of normal saline per Kg of body weight in the control groups for 5 days. And animals were sacrificed 5th day after administration of interleukin-2. The specimens obtained from the corpus uteri were fixed in 10% neutral buffered formalin solution for 24 hours at 4℃ and sliced at 14μm thickness in a frozen cryostat. The activity of acid phosphatase was observed by the Gomori's method for histochemical study. The result obtained was as follows: The acid phosphatase activity was increased at estrus and decreased at diestrus in interleukin-2 treated group. There was, however, no marked differences in acid phosphatase activity of the proestrus and metestrus between control and interleukin-2 treated groups. Consequently, it was suggested that interleukin-2 develop the changes of the acid phosphatase of the endometrium of the mouse.

      • 만성 신부전환자의 혈청 갑상선홀몬 농도의 변화

        To investigate the thyroid hormonal changes and their mechanisms in patients with chronic renal failure (CRF), the author measured serum levels of the thyroid hormones using radioimmu-noassay and calculated each ratio of serum thyroid hormones in 17 patients with CRF. Ten patients were on hemodialysis therapy and seven patients had not undergone hemodialysis. The following results were obtained. 1) The serum T3 and T4 concentrations were significantly lower in patients with CRF(0.7±0.47㎍/1 and 5.53±2.95㎍/dl, respectively) than in normal subjects(1.21±0.18㎍/1 and 9.46±1.92㎍/dl, respectively. 2) The serum FT4(free T4) concentration was significantly lower in patients with CRF(1.20±0.50μu/dl) than in normal subjects(1.01±0.48μu/dl). 4) The serum rT3(reverse T3) concentration in patients with CRF(285.7±148.1pg/ml) tends to be greater than normal subjects(204.6±43.3pg/ml). But is was not significant statistically. 5) The calculated serum concentration ratio of T3/T4 in patients with CRF(0.17±0.04: presented as Mean±SEM) did not differ significantly from that of normal subjects(0.13±0.01). But the serum concentration ratio of rT3/T4 in patients with CRF(78.78±18.98) was significantly greater than in normal subjects(21.90±1.04). 6) There was no significant difference in thyroid functions between male and female patients with CRF, and also between patients on hemodialysis and non-dialyzed uremic patients. It was concluded that the serum T3, T4, FT4 concentration were significantly decreased in patients with CRF, while the serum TSH concentration was significantly increased. The calculated serum concentration ratio of rT3/T4 was also significantly increased in patients with CRF. Thus is appears that abnormalities found in patients with CRF may reflect both the possible intrathyroidal abnormalities and alteration of serum rT3 metabolism with increased peripheral generation of rT3 from T4.

      • Diazinon이 Mouse의 肝臟에 미치는 影響

        유기체인 화합물중 Diazinon(O,O-diethyl O-(2-isopropyl 4-methyl-6-pyrimidyl phosphorothioate)은 농작물에 대한 살충제로 널리 사용되고 있으며 인체나 동물 체내에서 쉽게 가수분해되고 축척작용도 적어 우수한 살충제로 알려져 있다. 그러나 많은 용량의 diazinon에 접촉되거나 혹은 체내에 흡수되어 중독이 되면 인체의 혈액과 조직에 있는 cholinesterase의 활성을 억제하는 반면 acetylcholine을 축적시켜 부교감신경계의 과잉자극과 같은 증후를 나타내며,두통,피로감,동공축소,현기증,복부경련,구토,설사,발한,유연과 근육의 경련을 일으킨다. Diazinon과 같은 유기인제화합물인 EPN(Ethoxy-p-nitrophenoxy-phenylphosphine sulfide)을 실험동물에 투여하면 호흡곤란과 근강직이 나타나고 간장에서는 간소엽 중심대있 중간대의 간질세포에서 종창을 일으키고 공포가 형성될 뿐만 아니라 지방침착도 야기시킨다고 보고되어 있다. 저자는 포유류에서 diazinon이 다른 유기인제 화합물과 같이 체내에서 흡수되어 간장에서 해독과정을 거칠 때에 간실질세포에 독성으로 작용할 것으로 사료되어 mouse에 diazinon을 투여하여 간장의 변화를 형태학적으로 추구하였다. 실험동물은 체중 20gm 내외의 건강한ICR계 웅성 mouse에 체중 kg당 25mg되게 olive유에 희석하여 0.2ml을 복강내 주사하고 6, 12, 24및 36시간 경과 후에 희생시켜 간장을 적출한 다음 hematoxylin 및 eosin염색을 하여 간장의 조직학적 변화를 관찰하고, 일부의 간조직은 methyl greenpyronin 염색으로 간실질세포내의 pyronin호성과립의 양적 분포를 관찰하였으며, 또한 일부의 간조직은 oil red O 염색으로 간실질세포내에 지질의 축척을 관찰하여 다음과 같은 결과를 얻었다. 1. Diazinon 투여 12시간 경과 후에는 mouse의 간장에서 간소엽 중심대와 중간대의 간실질세포에서 소수의 소포와 약간의 종창을 일으켰으며, 24시간 경과 후에는 간소엽 전역에 단핵세포가 산재해있고 간소엽 중심대의 간실질세포에서 심한 종창과 대다수의 공포가 나타났고, 36시간 경과후는 간실질세포에 약간의 세포종창과 극소수의 소포가 관찰되었다. 2. Diazinon투여 12시간 경과 후에는 mouse의 간소엽 중심대와 중간대의 간실질세포에서 경도의 양성반응을 일으킨 pyronin 호성과립이 관찰되었고 24시간 경과 후에는 간소엽 전역에 걸쳐pyronin 호성과립이 집괴(clump)를 형성하고 간소엽 중심대와 중간대의 간실질세포에서는 pyronin 호성과립이 소실되었으나 36시간 경과 후에는 간소엽 전역의 간실질세포내에서 pyronin 호성과립이 균등하게 분포되어 있고 경도의 양성반응을 나타낸 pyronin 호성과립을 관찰하였다. 3. Diazinon 투여 6, 12시간 경과 후에는 mouse 간소엽중심대의 간실질세포에서 경도의 지방침착을 나타냈으며, 24시간 경과 후에는 간소엽 중심대와 중간대의 간실질세포에서 중등도의 지방침착을 나타냈으나 36시간 경과 후, 의 간소엽 중심대에서 경도의 지방침착이 관찰되었다. 이상 소견을 종합해 보면 diazinon은 mouse의 간장에 초기에는 손상을 야기시키나 시간이 경과됨에 따라 점차 회복된다고 사료된다. Diazinon(O,O-diethyl O-[2-isopropy 4-methyl-6-pyrimidyl] phosphorothioate) is considered one of the least hazardous of the toxic organic phosphorus insecticides, and is widely available for domestic and home-garden use. It is hydrolyzed and does not accumulate in the tissue. Although its toxicity is relatively low, diazinon has caused poisoning in man when absorption is sufficient amounts. The authors have studied the effect of diazinon on the liver of the mouse abserving the morphologcal changes, the accumulation of fat droplets and the loss of pyroninophilic granules in the hepatic cells. Healthy male ICR strain mice were given 0.2ml of 25mg diluted (diazinon in olive oil) per kg of body weight by intraperitoneal injection. The control mice were given olive oil alone. Animals of the control and diazinon treated groups were killed at the intervals of 6,12,24 and 36 hours after adminstration of diazinon and olive oil, respectively. The histological preparations were stained with hematoxylin and eosin method for histological changes, methyl green-pyronin stain for the loss of pyroninophilic granules and oil red Ostain for fat accumulation in the mouse liver. The results obtained are as follows: 1. The cellular swelling accompanied with vesicular formation is revealed in the hepatic parenchymal cells of the central and intermediate zones 12 and 24 hours after administration of diazinon. 2. A marked reduction or disappearance of the pyroninophilic granules in the central and intermediate zones is observed 24 hours after adminstration of diazinon. 3. Fat deposits are observed in the hepatic parenchymal cells of central and intermediate zones 6,12 and 24 hours after adminstration of diazinon. Consequently, it is suggested that diazinon would induce the cellular swelling, the fat accumulation and loss of pyroninophilic granules of the hepatic parenchymal cells in albino mice.

      • Bleomycin이 Mouse신사구체 및 근위요세관 상피세포의 미세구조에 미치는 영향

        Bleomycin, a glycopeptide antibiotic isolated from Streptomyces Verticillus, has been used as an anticancer drug active against human squamous cell carcinoma of skin and lung. It is well known that bleomycin inhibits DNA synthesis of tumor cell by scissoring DNA molecule and inhibiting DNA polymerase activity. Although bleomycin is very effective on various human cancers, it may damage certain normal cells to cause pulmonary and skin toxicity. In this experiment, the author pursued the effect of bleomycin on the mouse kidney, observing the ultrastructural changes in the proximal convoluted tubular epithelial cells and glomeruli. Healthy male mice(ICR strain) were used as experimental animals which were divided into two groups, the control and bleomycin treated groups. The bleomycin treated groups were subdivided into group of 3 times successive bleomycin treated and group of 7 times successive bleomycin treated. All experimental animals were given food and water ad libitum. The animals of bleomycin treated group were administered bleomycin 3 times and 7 times respectively, in a dose of 20 units per kg of body weight with 3 days interval. The experimental animals were sacrified in 24 hours after final administration of bleomycin. The specimens obtained from cortical region of left kidney were stained with uranyl acetate and lead citrate by the double contrast method and were observed with JEM 100cx-Ⅱ electron microscope. The results obtained were as follows: 1. In the epithelial cells of the proximal tubule the pinocytic vesicles were increased in number and also numerous autophagic vacuoles and the multivesicular bodies were found. 2. The deformed mitochondria and the dilated cisternae of rough endoplasmic reticulum were appeared in the epithelial cells of proximal renal tubule. 3. The basal infoldings were dilated and basement membrane were thickened and irregulary arranged at the basal portion of the epithelial cells of proximal tubule. 4. In the glomeruli the lamine densa of filterating membrane were irregulary arranged and thickened and many of the pedicles of podocytes were fusal together. It is consequently suggested that bleomycin damages both proximal convoluted tubular epithelial cells and glomeruli in mice kidney.
