        백서 척수후근절로부터 슈반세포의 효과적인 체외 배양법

        Schwann cells(SCs), an important component of the peripheral nervous system, intract with nerous to mutually support growth and replication for the peripheral nerve regentation. Recently, ading SCs to the lumen of guidance channel is widely tried to improve regeneration or to make regeneration possible over otherwise irreparable gaps. however, it is not easy to isolate and multiplicate SCs as much as enough to help the axonal regeneration. For the allogeneic SCs source for tubular nerve guidance, we developed a little bit improved technique of harvesting and multiplicating SCs. by culturing dispersed dorsal root ganglia in specially designed medium with growth factors and serial processing, we repeatedlly generate relatively homogenous SC cultures. Our technique was compared with other methods of literature using immunostaining methods such as GFAP, S100, BDNF and the total SC count assessment at different time interval after primary culture.

        Apoptotic Effect of Co-treatment with Chios Gum Mastic and HS-1200 on G361 Human Melanoma Cell Line

        Chios gum mastic (CGM) is a resinous exudate obtained from the stem and the main leaves of Pistacia lenticulus tree native to Mediterranean areas. Recently it reported that CGM induce apoptosis in a few cancer cells in vitro. Bile acids and their synthetic derivatives induced apoptosis in various kinds of cancer cells and anticancer effects. It has been reported that the synthetic chenodeoxycholic acid (CDCA) derivatives showed apoptosis-inducing activity on various cancer cells in vitro. This study was undertaken to investigate the synergistic apoptotic effect of cotreatment with a natural product, CGM and a CDCA derivative, HS-1200 on G361 human melanoma cells. To investigate whether the co-treatment of CGM and HS-1200 compared with each single treatment efficiently reduced the viability of G361 cells, MTT assay was conducted. To investigate augmentation of apoptosis in G631 cells co-treated with CGM and HS-1200, DNA electrophoresis, Hoechst staining, proteasome activity assay, flow cytometry, Westen blot analyses, immunofluorescent staining and confocal microscopy were performed. In this study, G361 cells co-treated with CGM and HS-1200 showed several lines of apoptotic manifestation such as nuclear condensations, DNA fragmentation, the reduction of MMP and proteasome activity, the decrease of DNA content, the release of cytochrome c into cytosol, the translocation of AIF and DFF40 (CAD) onto nuclei, activation of caspase-9, caspase-3, PARP and DFF45 (ICAD), and up-regulation of Bax whereas each single treated G361 cells did not. Although the single treatment of 40 μg/mL CGM or 25 μM HS-1200 for 24 hrs did not induce apoptosis, the co-treatment of them induced prominently apoptosis. Therefore, combination therapy of CGM and HS-1200 could be considered, in the future, as an alternative therapeutic strategy for human melanoma.

        In this paper the efficient finite element for stress analysis of plane stress/strain problems is proposed. This element is achieved by adding the bubble-mode function to 8-node element. The stiffness matrix of the element is calculated by using modified numerical integration order to avoid spurious zero energy mode. In order to demonstrate the performance of this element numerical tests for various verification problems are carried out. The results of numerical tests show accuracy and reliability of the element presented in this paper.

        An analysis on supporting programs of procuring goods for export in Korea

        To promote the nation's exports, in Korea several government led programs are alive for procuring and manufacturing goods for export. The trade supporting programs are consisted of Trade Financing Program, Trade Tax Program, and Trade Insurance. The programs are summarized ; First, Trade Financing Program is available to exporters of procuring raw materials and finished goods for export. It is extended with exceptionally low interests from the Korean commercial banks. Second, Trade Tax Program provides whole or part of customs drawback to exporters. customs drawback is` made when exporters meet requirements of Customs for export. Third, Trade Insurance Program cover risks of uncertainty which is beyond the exporters' capacity at low insurance premium It is suggested that the exporters have deep knowledge of these three programs to their ends. Especially Trade Financing Program and Trade Insurance Program can be a combined work for the financially weak SMEs. Further, there are rooms for these programs to develop to enhance the competitiveness of SME exporters.

      • 상부소화관출혈에 있어서 내시경적 분말약제산포의 효과

        Background : Bleeding after endoscopic procedure of the stomach carries an increased risk of morbidity and mortality. In case of oozing hemorrhage, not in active hemorrhage, powdered sodium alginate may play a role in control of bleeding with convenience. Therefore, we evaluated the clinical effect and efficacy of sodium alginate powder in upper G-Ⅰ bleeding. Methods : Forty-five patients who had upper G-Ⅰ bleeding after endoscopic procedure were enrolled in this study. Twenty-five patients of them were managed by the application of powder preparation of Sodium alginate(ALTO). The others were observed without treatment. The next day, we performed endoscopy in all patients to ascertain the hemostasis. Results : Of 25 patients managed with Sodium alginate, only two patients(8%) had bleeding (hemostatic rate : 92%), On the other hand, of 20 patients who did not receive any treatment, 3 patient(15%) had bleeding (hemostatic rate : 85%). There was no statistical difference between the two groups(p>0.05). Using other endoscopic procedure, We successfully treated the patient who continued to bleed. Conclusion : We evaluated the effect of the spary of sodium alginate on oozing hemorrhage. Although there was no statistical significance in our study, it seems that the spray of sodium alginate may be a useful method for the prevention of bleeding after endoscopic procedure.

        Chlorpromazine이 백서 해마에서 전기경련 충격에 의한 MAPK 타이로신 인산화에 미치는 영향

        Object : In order to examine the interaction mechanisms of electroconvulsive shock(ECS) and antipsychotic drug at the level of molecular biology, we observed the effect of chlorpromazine pre-treatment on the activation of mitogen activated protein kinase(MAPK) induced by electroconvulsive shock(ECS) in rat hippocampus Method : Male Sprague-Dawley rats were divided into 2 groups. To the experimental group chlorpromazine(20㎎/㎏) was given intraperitoneally, and to the control distilled water was given instead. Thirty minutes later, ECS was given and the hippocampus was dissected out 2 minutes thereafter. Immunoblotting with antiphosphotyrosine antibody was carried out, and the signal intensity at 42kDa band was quantitized using densitometer. The obtained result was compared by student t-test between the experimental and the control group. The absolute amount of MAPK was measured by immunoblotting with anti-MAPK antibody. Result : The tyrosine phosphorylation of MAPK reached peak at 2 minutes after ECS. However, in the chlorpromazine pre-treated group, the peak level of MAPK tyrosine phosphorylation was significantly attenuated(t=-3.12, df=14, p=0.008) compared to the control. In contrast to this, the absolute amount of MAPK did not differ between the pretreated and the control group. Conclusion : Chlorpromazine attenuated the tyrosine phosphorylation of MAPK by ECS. This finding seems to be related to the fact that chlorpromazine pre-treatment changed the cfos expression by ECS in rat brain. Antipsychotic drug and ECS might interact at the level of MAPK signal transduction system, and this might explained the observed synergistic effect of two treatment modality.

        흰쥐에서 치수노출 후 삼차신경절의 신경절아교세포에서 GFAP-IR의 변화

        Glial fibrillary acidic protein(GFAP) are a group of intermediate filaments that are distributed in the cytoplasm of many type of glial cells. The purpose of this study was to determine change of GFAP immunoreactivity (GFAP-IR) in rat trigeminal ganglion satellite cells in response to pulp exposure. The immunohistochemistry was carried out using the avidinbiotin-peroxidase complex(ABC) method and subsequently stained with AEC(3-aminoethyl-9-carbasol). 1. Contol group; Central root astrocytes had strong GFAP-IR, but ganglion satellite cells occasionlly had GFAP-IR. This reaction patterns of ganglion satellite cells was not concenturated in any specific region of trigeminal ganglion. 2. Three day pulp exposure group' There was a highly GFAP-IR in satellite cells of trigeminal ganglion in maxillary region. GFAP-IR in neighboring mandibular and ophthalmic regions was less intense compared to maxillary region. 3. Seven day pulp exposure group; In this group, GFA-IR that was increased compared to control group was seen in the maxillary region. But GFAP-IR was less intense compared to three day pulp exposure group. These results suggest that GFAP in satellite cell increase in specific region of trigeminal ganglion after pulp exposure and offer useful tool in trigeminal pain research.

        상아질 접착제의 전단결합강도와 미세누출에 관한 연구

        New bonding agent systems have been supplied which operators can simply apply to conditioned tooth surfaces. The purpose of this study was to evaluate the shear bond strengths and the microleakages of three bonding agents and composite resins to dentin. Seventy-five extracted human maxillary and mandibular molar teeth were used in this study. For the shear bond strength test, the entire occlusal dentin surfaces of thirty teeth were exposed with Diamond Wheel Saw and smoothed with Lapping and Polishing Machine (South Bay Technology Co., U.S.A.). For the microleakage test, Class V cavities were prepared in the buccal surfaces of fourty-five teeth. They were randomly assigned into 3 groups according to dentin bonding agents (Scotchbond^TM Multi-Purpose plus, ONE-STEP^TM and Prime & Bond^TM) and composite resins (Z-100, Aelitefil^TM and TPH Spectrum^TM) to be used. Bonding agents and composite resins were bonded to exposed dentin surfaces of the tooth crown and to Class V cavities on the buccal surfaces respectively according o manufacturer's directions. The shear bond strengths were measured by universal testing machine(U^TM AGS-100, Japan). In addition, the degree of microleakage at the occlusal and gingival margin was examined by 2% methylene blue and stereomicroscope(Olymous SZH 10, Japan). The results were as follows: 1. The shear bond strength to dentin was the highest value in SBMP-Plus group(16.68±7.38MPa) and the lowest value in Prime & Bond group(11.61±5.82MPa), but there was no significant difference of shear bond strength among three groups. 2. The degree of microleakage at both occlusal and gingival margin was showed the lowest in SBMP-Plus group and the highest in ONE-STEP group. 3. At both occlusal and gingival margin, there was significant difference of microleakage between SBMP-Plus and ONE-STEP/Prime & Bond groups(p<0.05), but no significant difference of microleakage between ONE-STEP and Prim & Bond group(p>0.05).

      • 불안의 형성과정에 있어서 뇌 Cholecystokinin의 역할

        Cholesystokinin(CCK) is one of the most abundant neuropeptides and its receptors are widely distributed throughout the central nervous system, especially in hypothalamus, limbic system, basal ganglia, hippocampus and cortex. CCK is co-localized or interacts with dopaminergic, noradrenergic, GABAergic and serotonergic neurotransmitter systems. lts functional roles are not fully defined, but it appears to mediate anxiety and it may participate in satiety, nociception and drug withdrawal. CCK/dopamin neurons in ventral tegmental area project their long pathways to limbic system and striatum and their roles in the pathogenesis of dopamine-related disorders such as schizophrenia are shortly described in this article. Cholecystokinin tetrapeptide(CCK-4) is suggested to have a potent panicogenic property in human. Both clinical and animal research suggests that ?? receptors mediate the panicogenic effects of CCK-4 and their antagonists provide an important advance in the treatment of anxiety disorder. The possible involvement of CCK in the pathogenesis of panic disorder is described briefly. The antagonistic action of imipramine on CCK-4-induced panic and that of 5-HT₃ on CCK release in nucleus accumbens are discussed. As described above, there is growing speculations that CCK might be implicated in the neurobiology of anxiety and its receptor agonists could be used in the treatment of anxiety in future.
