Stauntonia hexaphylla (SH) and Vaccinium bracteatum (VB) are herbal extracts widely used in food and traditional herbal medicine, and have the ability to perform a wide range of biological activities. We aimed to investigate the effects of the SH and VB combination (SHVB) on mice models of chronic restraint stress (CRS) and pentobarbital-induced sleeping behaviors to elucidate its possible mechanisms of action. CRS-exposed mice treated with SHVB showed significantly decreased immobility time, increased swimming and climbing times in the forced swim test (FST), and increased locomotor activity in the open field test (OFT). SHVB decreased serum CORT levels, but enhanced brain monoamine neurotransmitters. SHVB significantly decreased the sleep latency and increased total sleep duration in pentobarbital-induced sleeping behavior in mice. SHVB showed inhibitory effect on 5-HT2A receptor-mediated ERK1/2 phosphorylation. These results suggest that SHVB has antidepressant and hypnotic effects by regulating the 5-HT2A receptor.
Genistein Inhibits Vibrio vulnificus Adhesion and Cytotoxicity to HeLa Cells

Vibrio vulnificus (V. vulnificus) is a useful model for bacterial septicemia as the bacterial infection generates a wide pathogenic spectrum in addition to a high mortality rate. This study was aimed at investigating the effects of genistein on the growth, cytotoxicity, adhesion, and the mouse mortality caused by V. vulnificus. The results of our study indicated that genistein (50 or 100 mg/L) effectively minimized the morphologic damages and inhibited acute death of HeLa cells by V. vulnificus. Strikingly, genistein significantly inhibited the adhesion of V. vulnificus to HeLa cells. This report confirmed that genistein showed bacteriostatic activity against V. vulnificus, but it did not exhibit any bactericidal activity. Nevertheless, genistein (20 mg/kg) effectively decreased CD-1 mice mortality caused by V. vulnificus infection.

P. tenebrifer (PT) belongs to the Diptera order and Stratiomyidae family. Recently, insect industry have been focused as food, animal feed and environmental advantages. γ-aminobutyric acid (GABA) and melatonin have been associated with regulating sleep and depression. GABA is the primary inhibitory neurotransmitter and is synthesized via biotransformation of monosodium glutamate (MSG) to GABA by lactic acid bacteria. In this study, we first used a GABA-enhanced PT extract, wherein GABA was enhanced by feeding MSG to PT. The underlying mechanisms preventing stress and insomnia were investigated in a corticosterone (CORT)-induced endoplasmic reticulum (ER) stress and chronic restraint stress (CRS)-exposed mouse model, as well as in pentobarbital (45 mg/kg)-induced sleep behaviors in mice. In the present study, the GABA peak was detected in high-performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) analysis and showed in Ptecticus tenebrifer water extract (PTW) but not in non-PTW extract. The results showed that PTW and Ptecticus tenebrifer with 70% ethanol extract (PTE) exerted neuroprotective effects by protecting against CORT-induced downregulation of phosphorylated extracellular signal-regulated kinase 1/2 (ERK1/2) and cAMP-response element binding protein (CREB) expression. In addition, PTW (300 mg/kg) significantly reduced CORT levels in CRS-exposed mice. Furthermore, PTW (100 and 300 mg/kg) significantly reduced sleep latency and increased total sleep duration in pentobarbital (45 mg/kg)-induced sleeping behaviors, which was related to serum melatonin levels. In conclusion, our results suggest that PTW exerts anti-stress and sleep-enhancing effects by regulating serum CORT and melatonin levels.

Geographical isolation may lead to the populations having different ecological characteristics. Geographic isolation have been known to cause echolocation call differences in bats. In order to look at geographic differences of echolocation calls of Great Horseshoe Bats (Rhinolophus ferrumequinum), populations of inland and Jeju island habitats were examined in Korea. The study areas were some abandoned mines in the inland and caves of Jeju island, which were known as rest sites of Great Horseshoe Bats during the active season. Recording was done in two ways: Hand-held, Free-flying. Recording pulse was analyzed into five parameters: Maximum Frequency (FMAX), Minimum Frequency (FMIN), Peak Frequency (PF), Duration (D), Inter pulse Interval (IPI). Interestingly, The present study shows that the difference in echolocation between the inland and island populations. The PF of bats inhabited inland was 69 kHz. but, the PF of bats inhabited island (Jeju) was 71 kHz. There was a difference between regions. Discriminant analyses also showed clear difference between the inland and the island populations. Especially, PF of Korea population is lower than that of Europe (82 kHz) and higher than that of Japan (65 kHz).

Vibrio vulnificus, a good model organism of bacterial septicemia, causes fatal septicemia manifesting a fulminating course and a high mortality rate within days. In order to identify new natural substances preventing V. vulnificus infection, a plant library was screened for inhibiting cytotoxicity to host cells by using Trypan blue staining and LDH assay. We found that Polygoni Cuspidati Radix potently suppressed the acute death of HeLa and RAW264.7 cells in a dose dependent manner. Further studies revealed that Polygoni Cuspidati Radix inhibited V. vulnificus growth and survival in HI broth and seawater, respectively. We confirmed that Polygoni Cuspidati Radix contained high level of emodin by thin layer chromatography (TLC). Emodin showed direct antibacterial activity against V. vulnificus. In addition, emodin prevented the morphologic damages and acute death of HeLa cells caused from V. vulnificus. The safety of Polygoni Cuspidati Radix and emodin to host cells was confirmed by MTT assay. Polygoni Cuspidati Radix and emodin protected mice from V. vulnificus infection.

Our previous results suggest that the Rosa rugosa Thunb. (family Rosaceae) alleviates endurance exercise-induced stress by decreasing oxidative stress levels. This study aimed to screen and identify the physiological antistress effects of an extract of R. rugosa (RO) on sleep deprivation–induced anxiety-like behavior and cognitive tests (in vivo) and tested for hippocampal CORT and monoamine levels (ex vivo), corticosterone (CORT)-induced injury, N-methyl-d-aspartate (NMDA) receptor, and serotonin 6 (5-hydroxytryptamine 6, 5-HT6) receptor activities (in vitro) in search of active principles and underlying mechanisms of action. We confirmed the antistress effects of RO in a sleep-deprived stress model in rat and explored the underlying mechanisms of its action. In conclusion, an R. rugosa extract showed efficacy and potential for use as an antistress therapy to treat sleep deprivation through its antagonism of the 5-HT6 receptor and resulting inhibition of cAMP activity.
Multifunctional antistress effects of standardized aqueous extracts from Hippophae rhamnoides L.

This study aims to screen and identify the multi-mechanism antistress effects of an extract of Hippophae rhamnoides L. (HR) leaves on corticosterone (CORT)-induced injury, N-methyl-daspartate (NMDA) receptor and serotonin 6 (5-hydroxytryptamine 6, 5-HT6) receptor activity tests (in vitro), electric foot shock and forced swimming tests (FSTs) (in vivo), and tests for hippocampal CORT and monoamine levels (ex vivo), in search of active principles and underlying mechanisms of action. We confirmed that the water extract of HR (HRW) and various ethanol extracts of HR confer protective effects against CORT-induced impairments in SH-SY5Y cells and antagonistic effects on NMDA receptors and the 5-HT6 receptor by using primary cultured rat hippocampal neurons and a stable 5-HT6 receptor-expressing cell line, respectively. In addition, we confirmed the antistress effects of HRW in an electric foot shock stress model in mice and explored the underlying mechanisms of its action. We observed that HRW treatment significantly reversed the reduction in immobility times and increased climbing times in FSTs induced by electric foot shocks in the stress model. The levels of CORT, dopamine, and norepinephrine were increased, and the level of serotonin in the hippocampus was decreased in the electric foot shock stress model. The standardized HRW effectively restored abnormal CORT and monoamine levels in the hippocampus that were induced by stress. The results of the present study demonstrate that the standardized HRW produces novel multifunctional antistress effects.
