      • 2차원적 전기영동에 의한 톡소포자층 당단백 항원의 분석

        Serious disease due to Toxoplasma gondii occurs in congenitally infected children and in immunosuppressed patients, particularly in the case of HIV infection. Glycoproteins modulate the physicochemical properties, and are involved in biological and immunological activities. Although there are many reports about the immune responses after T. gondii infection, there are few reports to identify the glycoproteins from T. gondii. For this reason, we purified the glycoproteins from tachyzoites and cysts of T. gondi and then evaluated the antigenic characterizations of toxoplasmic glycoproteins. Tachyzoites of T. gonii were purified by Percoll-gradient centrifugation. Glucosamine was the major monosaccharides released from the tachyzoites of T. gondii by acid hydrolysis methods. In order to define the antigenicities of both strains, we did 2-dimensional electrophoresis(2-DE). Over 40 protein spots were reproducibly separated in tachyzoites of T. gondii by 2-DE in range of the pH 5-8. Glycoproteins were isolated from tachyzoites of T gondii by Con A-Sepharose 4B affinity chromatography. The major bands of glycoproteins were approximately 22 kDa, 30 kDa and 38 kDa in tachyzoites of T. gondii.

        구강암 세포주의 이소위 누드마우스 종양 모델

        In order to make successful oral cancer treatment, we need to understand about tumor biology and effective chemotherapeutic agents. To achieve these studies, it is necessary to develope a proper in-vivo model. Therefore the author will make try to develop more improved animal model of more applicable in various method of cancer study. In this study, the author induced in-vivo tumorigenesis in nude mice by YD-10Bmod cell line used by YD-10B cell line originated from oral tongue squamous cell carcinoma and observed tumor formations and invasiveness of surrounding tissue, and found some results as follows : 1. The experimental group (YD-10Bmod, subcutaneous injection) produced tumors 13 out of 15 mice, while the control group produced none of 5 mice. 2. The inoculation of 1×106cells/mouse produced tumors 3 out of 5 mice and inoculation of 1×107cells/mouse, 2×107cells/mouse produced tumors in every 5 mice. 3. In the histopathologic studies, the inoculation of 1×106cells/mouse group showed the characteristic features of well-differentiated squamous cell carcinoma and demarcated expansile growth, while the inoculation of 1×107cells/mouse, 2×107cells/mouse group showed the expansile growth with partial central necrosis and invasive growth to surrounding fat & connective tissue. These findings suggest that atopic xenograft of YD-10Bmod cell line in nude mice has a improved productivity of tumors, produced tumors showed the characteristics feature of human tumor and invasive growth to surrounding tissue in histopathologic appearance. These atopic nude mouse model of tongue carcinoma might assist in studying oral cancer biology and effective choice of chemotherapeutic agents.

        In-line 마그네트론 스퍼터 장치를 사용하여 산소 분위기에서 제작한 Ag 박막의 특성

        A study was made to examine the electrical, compositional, structural and morphological properties of Ag thin films deposited by DC magnetron sputtering in $O_2$ atmosphere with deposition temperature from room temperature to 15$0^{\circ}C$ using in-line sputter system. The Ag films deposited at temperature above $100^{\circ}C$ in oxygen atmosphere gave a similar specific resistivity to and even lower oxygen content than those deposited using pure Ar sputter gas The Ag films deposited with pure Ar gas was mainly composed of crystallites with [111] preferred orientation, while, for those deposited in oxygen atmosphere, more than 50% of the volume was composed of crystallites with [100] orientation. The difference in the micro structure did not cause any difference in the specific resistivity of Ag films. The results showed that the transparent conducting oxide films and the Ag films could be processed sequentially in the same deposition chamber with careful control of deposition temperature, which might result in a cost reduction for constructing the large scale in-line deposition system.

        In-mold injection molding system is generally an advanced version of the injection molding system. The injection, printing and coating processes of In-mold injection molding take place simultaneously. The technology is mostly used in home appliances, electronic goods and cell phones. For this research we applied the In-mold technique for injection molding and process of 3D nurb-surfaces. We performed the computer simulation of injection molding to optimize the In-mold processes and identified the credibility of the products through performance evaluation.

      • 휘발성 유기용매의 In Vitro 대사속도 측정 장치의 개발

        Species, doses and routes extrapolation can be sucessfully carried out by using a physiologically-based pharmacokinetic (PBPK) approach. And PBPK approach to assess risk of hazardous chemicals is reasonable whatever the exposure scenarios are happened. Both partitioning coefficients of chemical between tissue and blood and enzymatic metabolic rate constants are key parameters to build up the PBPK model. In this study, we tried to estimate in vtlro metabolic rate constants using a special apparatus instead to measure the in vivo constants which are used to PBPK simulation since the in nitro tests are less expensive and more convenient than in vivo tests. For the purpose, we designed and tested the new system to measure continuously the headspace concentration of VOC. The newly designed system is composed with a diffusion chamber which generates gaseous substrate, a reaction vessel with a recirculating pump to establish a closed system, an automatic sampler from a gas phase, a gas chromatography to analyze the headspace. In addition, a cold water condenser is attached between the reaction vessel and pump to reduce the content of gaseous moisture which interferes with chemical analysis. To validate the newly developed methodology, in vitro metabolic rate constants of trichloroethylene (TCE) as a prototype VOC were estimated by simulating observed results with an ACSL program. The simulated results are consistent to those estimated by the other research groups. This finding suggests that our newly designed closed system may be a useful apparatus to estimate in vitro metabolic rate constants for VOC.

      • 17β-estradiol 처리에 따른 해산 어류의 vitellogenin 생성

        A common used endpoint in bioassays testing the estrogenicity of chemicals including endocrine disruptors is the induction of egg yolk precursor vitellogenin in male fish. Two marine fishes (Sebastes schlegeli and Paralichthys olivaceus) were exposed to the 17β-estradiol (E2) to determine the vitellogenin production. Vitellogenin was measured in fish blood using SDS-PAGE and Densimetry. Results showed that exposure to E2 caused vitellogenin in male fish. Especially, vitellogenin levels in young fish were about 4 times higher than in adult fish, which means young fish are more sensitive to E2 exposure. And plasma vitellogenin in fish increased related to E2 concentration and exposure duration.

        수리된 복합레진 수복물의 전달결합강도 연구

        This study was performed to evaluate the interfacial shear bond strength of base (direct and indirect) and repair composites with aging and surface treatment methods. Direct composites resin specimens (Charisma®, Heraeus Kulzer, Germany) were aged for 5 min, 1 hour, 24 hours, and 1 week in 37℃ distilled water before surface treatment, and then divided into five groups: Group 1, grinding; Group 2, grinding and application of bonding agent; Group 3, grinding, etching with 37% phosphoric acid for 30sec, and application of bonding agent; Group 4, grinding, etching with 37% phosphoric acid for 30sec, silane treatment, and application of bonding agent; Group 5, grinding, etching with 4% hydrofluoric acid for 30sec, silane treatment, and application of bonding agent. Indirect composite resin specimens (Artglass®, Heraeus Kulzer, Germany) were aged for 1 week in 37℃ distilled water and divided into seven groups: Group 1 - Group 5, equal to Charisma specimens; Group 6, grinding, etching 37% phosphoric acid for 60sec, silane treatment, and application of bonding agent; Group 7, grinding, etching with 4% hydrofluoric acid for 60 sec, silane treatment, and application of bonding agent. The repair material(Charisma®) was then added on the center of the surface (5mm in diameter, 5mm in height). The shear bond strength was tested and the data was analyzed using one-way ANOVA and the Student-Newman-Keuls test. The following conclusions were drawn. 1. The shear bond strength of Charisma® specimens aged for 1 hour was significantly higher in Group 2 and Group 5 than in Group 1 (p<0.05), and that of Charisma® specimens aged for 1 week was significantly higher in Group 3 and Group 5 than in Group 1 (p<0.05). No significant difference was found in the bond strength of specimens aged for 5 min and 24 hours. 2. In Group 2 of the Charisma® specimens, there was significant difference between the bond strength of 24 hours and that of 1 week (p<0.05). 3. In Group 4 of the Charisma® specimens, the shear bond strength of specimens aged for 24 hours was significantly higher than the others(p<0.05). 4. There was no significant diiference between the shear bond strength of the Artglass® specimens. 5. Most of the Charisma® specimens showed cohesive fractures. Artglass® specimens that were etched with acid(phosphoric or hydrofluoric) for 30 sec showed more cohesive fractures.

      • 어선원의 선상생활 건강관리에 대한 지식, 태도 및 실천도 분석

        Because the seamen must live on ship apart from the land on or off daty, seamen' health managenent must consider the characteristics of ship surroundings such as weather and job. The questionnaire, which was made in base of Guide Line of Nation Health, was surveyed to the subject of seamen who were consisted of social community in particular environment such as ship. The subject of survey of was 420 trainess of ship security who have Korean nationality in the department of fishery, as in-service training institute of sea fisher, where is Young Do Gu, Pusan. The period of survey was 13weeks from Oct. 1, 1998 to Dec. 30. 1998. Based on the results, the characteristics of the subject showed the highest as the age of 30's, 38.1% : the height of 165∼170cm, 38.8% : the weight of 61∼70kg, 43.4% : the religion belief of 68.8% : a school career, 45.4%(graduated from high-school) : embarkation career. 41.1%(from 11 to 20 years) and on-duty hours a week, 43.4%(above 81 hours). The higher age, religious-beliefs, married-life, duty-position and emparkation-career are, the higher knowledge-degree about health management is. And the higher age, religious-beliefs, married-life, embarkation-career are, the higher attitude-degree is. The higher age, religious-beliefs, married-life, duty position, ship-burden, ship-age and embarkation-career are, the higher practice-degree is. The knowledge and the attitude about health management were totally 4.20±0.27 and 3.42±0.34 in mean by 5-score scale, respectively. The practice-degrees about health management were cleanliness, 3.40 ; food, 3.15 ; drinking and smoking, 2.92 ; satisfaction of work, 3.89 ; public order, prevention of security accident, 4.08 ; healthy environmental creation, 3.00 in mean, by 5-score scale, respectively, and was totally 3.41 ±0.44. And the degrees about health management were that the knowledge score in mean was the highest by 4.20, the attitude was higher by 3.42, and the practice was the lowest by 3.41. The corelations among knowledge, attitude and practice-degree were significant difference and were corelated(0.31∼0.63). And the corelations among factors of practice-degree were significant difference and were corelated(0.19∼0.58). By regression analysis, R square of the knowledge and practice degree about health management were 0.36, 0.07 by cleanliness, respectively. R square of the attitude degree was 0.48 by both cleanliness and food. The above results showed that the health management of seamen in the ship life should be high the corelation among knowledge, attitudes and practice-degree. And it is considered that the education for the health management of seamen is needed periodically. additionally, the higher environmental sanitation and occupational environment are, the higher healthy life is. So the concern and service of shipowner about their seamen health should be needed more and more.

      • 어린이집 영유아의 성장ㆍ발달에 관한 종단적 연구(Ⅱ)

        The purpose of this study was to investigate longitudinal growth and development of young children and follow-up effects of the educare program in the PNU Day Care Center. Particularly, the first objective of this second report of study was the comparison the entire domains of development with age between the Oct. of 1995 and the Dec. of 1998. The second objective was to analyze the developmental .changes of 5 children who entered the center in 4 years old since Oct. 1995. The subjects of this study were 448 children(241 boys, 207 girls). The instruments of this study were the KISC and KEDl-WISC(for intelligence test), the Learning Readiness Test, the Social and Emotional Scale, Height Scale and Weight Scale( for physical growth). The tests were carried out 2 times per year (the early of the year and the late of the year). Results from comparsion across the 1st through 7th tests during 3 years indicated that the mean of the height was increased about 1-8cm for all of 2-5 years old children. The mean of the weight also was increased about 1.5-4.0kg except 2 years old children. The intelligence score (T score) by KISC was increased around 6-42 points except 6 years old boys and girls. In the intelligence score by KEDl, there was improvements of average 8 points for 7 years old children. In the learning Readiness Test of 5, 6 years old children, the scores all of children except 6 years old girls got were decreased. The scores of 6 years old girls were incresed 6 points in the Dec. of 1998. In the Social and Emotional Scale, there was big improvement of scores between in the 1st test and in the 7th test. These results were discussed in the view of sample size, developmental characteristics, test instruments and educare program of the PNU day care center.
