      • DJ-1/park7 modulates vasorelaxation and blood pressure via epigenetic modification of endothelial nitric oxide synthase

        <P><B>Aims</B></P><P>DJ-1/park7, a multifunctional protein, may play essential roles in the vascular system. However, the function of DJ-1/park7 in vascular contractility has remained unclear. The present study was designed to investigate whether the DJ-1/park7 is involved in the regulation of vascular contractility and systolic blood pressure (SBP).</P><P><B>Methods and results</B></P><P>Norepinephrine (NE) elevated contraction in endothelium-intact vessels in a dose-dependent manner, to a greater extent in DJ-1/park7 knockout (DJ-1/park7<SUP>−/−</SUP>) mice than in wild-type (DJ-1/park7<SUP>+/+</SUP>) mice. Acetylcholine inhibited NE-evoked contraction in endothelium-intact vessels, and this was markedly impaired in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP>. Nitric oxide (NO) production (82.1 ± 2.8% of control) and endothelial NO synthase (eNOS) expression (61.7 ± 8.9%) were lower, but H<SUB>2</SUB>O<SUB>2</SUB> production (126.4 ± 8.6%) was higher, in endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice than in those from DJ-1/park7<SUP>+/+</SUP> controls; these effects were reversed by DJ-1/park7-overexpressing endothelial cells from DJ-1/park7<SUP>−/−</SUP> mice. Histone deacetylase (HDAC)-1 recruitment and H3 histone acetylation at the eNOS promoter were elevated and diminished, respectively, in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls. Moreover, SBP was significantly elevated in DJ-1/park7<SUP>−/−</SUP> mice compared with DJ-1/park7<SUP>+/+</SUP> controls, but this elevation was inhibited in mice treated with valproic acid, an inhibitor of Class I HDACs including HDAC-1.</P><P><B>Conclusion</B></P><P>These results demonstrate that DJ-1/park7 protein may be implicated in the regulation of vascular contractility and blood pressure, probably by the impairment of NO production through H<SUB>2</SUB>O<SUB>2</SUB>-mediated epigenetic inhibition of eNOS expression.</P>

        해상형 국립공원 탐방수요의 요인분해

        국내관광은 국제관광의 변동성이 주는 충격을 완화하려 관광산업의 안정성을 담보하는 역할을 수행함에도 불구하고 국내관광부문은 활성화되지 못하고 있다. 이러한 점은 우리나라의 해외관광객은 2010년-2017년에 연평균 11.3%라는 높은 증가를 보인데 비해 우리나라 국립공원의 탐방객은 연평균 1.5% 성장이라는 초라한 실적을 드러내고 있다는 데서 알 수 있다. 더욱 산악형 국립공원에 대해서는 비교적 많은 연구가 이루어지고 있는데 비해 해상형 국립공원에 대해서는 상대적으로 연구와 관심이 빈약한 편이다. 이에 본고는 변이할당분석과 BCG분석을 이용하여 해상형 국립공원 탐방객의 변동요인을 식별한다. 먼저 변이할당분석을 적용하여 4개 해상형 국립공원 탐방객의 변동을 국립공원 전체에 대한 탐방수요가 개별 국립공원탐방수요에 미치는 국립공원효과, 국립공원 전체에 대한 매력과 해상형 국립공원에 대한 매력의 차이가 개별 국립공원 탐방수요에 미치는 해상형 국립공원효과, 그리고 개별 국립공원과 해상형 국립공원의 탐방수요가 미치는 소위 경쟁력효과와 같은 3개 요인으로 분해한다. 또한 변이할당분석을 정태적 분석과 동태적 분석으로 구분하여 정태적 분석과 동태적 분석의 3개효과가 부호와 크기에서 상당한 차이를 노정한다는 것도 밝힌다. 구체적으로 우리나라 4개 해상형 국립공원의 탐방객 변동을 요인별로 분해하여, 정태적 분석에서는 국가효과와 구성효과가 양의 값으로 4개 해상형 국립공원 탐방객을 증가시키나, 동태적 분석에서는 구성효과의 경우 2013년부터, 국가효과의 경우 2014년부터 음의 부호를 갖는다는 것을 밝힌다. 탐방객이 증가한 오동도, 다도해해상국립공원, 태안해안국립공원은 물론 탐방객이 감소한 변산반도국립공원에서도 개별 국립공원의 경쟁력을 의미하는 지역효과가 가장 큰 영향을 미친다는 것도 보인다. BCG 매트릭스를 적용하여 오동도와 다도해해상국립공원은 star 영역으로 상승한 후star 위상을 유지하며, 변산반도국립공원은 star에서 cash cow와 child로 위상이 변화하여 탐방객 증가가 평균 이상을 유지하고 있는데 비해 태안해안국립공원은 child에서 dog로 추락함을 보인다. 이러한 분석을 통해 4개의 해상형 국립공원 모두에서 지역효과가 탐방객 변동에 가장 큰 영향을 미친다는 것과, 오동도와 다도해해상국립공원의 경쟁력은 우수하나 변산반도 국립공원의 경쟁력은 정체상태에 있으며, 태안해안국립공원의 경쟁력은 하락하고 있음을 밝힌다. BCG분석에서는 오동도와 다도해해상국립공원이 탐방객에서 경쟁력이 있다는 것과 태안해안국립공원에서 가장 큰 경쟁력 향상이 이루어져야 한다는 것도 밝힌다. Tourism is widely considered one of the world's largest and rapidly growing industries. Domestic tourism, however, remains an under-researched theme in many countries involving Korea partly due to the difficulty to track domestic tourism and with governments and policy-makers placing emphasis on the attractive higher expenditures of foreign visitors. It is stated that a vibrant domestic tourism sector can cushion the industry from fluctuations of the international tourism market and bring stability and predictability in the industry. Nature-based tourism to national parks gives us broad benefits such as improved physical and mental health, stress reduction and increased social cohesion. Employment is one of tourism’s main development advantages and its role in promoting economic opportunities for communities adjacent to national parks has long been appreciated. But tourism performance in Korean national parks continues to dwindle and stagger. This paper introduces a shift-share decomposition to break down the tourism to the marine national parks into three components: one measuring national effect, and two other components due to changes in compositional effect and regional effect respectively. While the static shift-share analysis shows that the growth in marine national parks’tourism seems to be due to positive national park effect as well as marine national park effect, the dynamic regional effect appears to have a negative effect on visitors. This paper also employs the positioning analysis on national parks using Boston Consulting Group matrix over the period 2011-2016. The BCG analysis shows that Odongdo and Dadohaehaesang succeed in rising to a star area, Byeonsanbando ranges over a cash cow to a child, but Taeanhaean degrades from a child to a dog area.

        논문 : 국립공원 탐방로 내 위험 급경사지 유지관리 방안 연구

        국립공원은 주로 산악 지형 내 조성되어 있으므로 산사태에 의한 피해 가능성을 항상 내포하고 있다. 본 연구는 산사태 연구의 일환으로 국립공원 탐방로 내 위험 급경사지를 체계적으로 유지관리하기 위한 방안을 마련하는데 목적이 있다. 국립공원에 특화된 자체 체크리스트를 제작하였으며, 전국 19개 국립공원 183개소 급경사지에 대한 실태조사가 이루어졌다. 실태조사의 최종 목적은 위험 급경사지에 대한 안정화 대책을 마련하는 것으로 현장조사와 안정해석의 과정을 거친다. 일부 급경사지에서는 위험성 판단을 위해 지상라이다 등 특수 장비를 이용한 조사를 시도하였다. 중장기 정비계획을 위한 투자우선순위 공식을 개발하였으며, 현장자료를 이용한 순위 및 위험등급을 산출하였다. 최종적으로 위험 급경사지의 향후 체계적인 관리를 위해 “국립공원 급경사지 유지관리시스템”을 개발하였으며, 모든 현장자료들은 시스템 내 DB화 되었다. National parks, which are located mainly in mountainous areas, are always at risk of damage by landslides. The goal of this study is to establish a method for systematically maintaining hazardous steep slopes along trails in national parks. We produced a checklist suitable for each of the 19 national parks nationwide and investigated 183 slopes. The aim of these investigations is to recommend appropriate slope-stability countermeasures, including field investigations and stability analysis. We made preliminary investigations at specific sites, evaluating the slope hazard using specialized equipment such as terrestrial LiDAR. An investment priority formula was developed, and ranking and hazardous grades were calculated as part of a long-term maintenance plan. Finally, to systematically manage dangerous slopes and to house all the field data within one system, we developed the “Slope Maintenance System in National Parks” based on web server that can show various information for slopes.

        대전시 도시숲의 식생 및 토양특성에 관한 연구

        This study was conducted to suggest appropriate methods for management of urban forest after investigating the present condition and problems of urban forests by analyzing vegetation and soil properties in urban forests in the Daejeon. On the basis of our research, Pinus rigida dominate Gyejoksan and Bomunsan. Pinus densiflora dominate Wolpyeong park and Quercus acutissima dominate Namsun park. On the basis of our result of analysis of soil chemical properties, all investigated areas have low pHs, available phosphates and exchangeable cations. They indicate that the soil of those areas have been acidifying progressively. Soil hardness measurements were conducted to know the conditions of trampled soils and the results of them show that soil hardness in Namsun park was higher than the others. This indicates that human interference affect the health of the urban forest.

      • 국소 광적응 기능을 가지는 윤곽검출용 32×32 방사형 CMOS 시각칩의 설계 및 제조

        국소 광적응 기능을 가지는 윤곽검출용 시각칩을 픽셀수 32x32의 방사형 구조로 CMOS 공정기술을 이용하여 설계 및 제조하였다. 생체의 망막은 넓은 범위의 입력 광강도에 대해서 물체의 윤곽을 검출할 수 있다. 본 연구에서는 시세포, 수평세포, 쌍극세포로 이루어진 망막의 윤곽검출 기능을 모델링하여 윤곽검출용 인공시각칩을 설계하였다. 국소 광적응을 위해, 입력 광강도에 따라 수용야의 크기를 국소적으로 바뀌게 하였다. 아울러 단위셀을 방사형으로 배치함으로써 영상데이터의 양을 감소시킴과 동시에 칩의 중심부분으로 갈수록 해상도가 높아지도록 설계하였다. 설계된 칩은 0.6 ㎛ double-poly triple-metal 표준 CMOS 공정기술을 이용하여 제조되었으며. HSPICE 시뮬레이션으로 성능을 최적화 시켰다. A 32x32 pixels foveated (linear-polar) stucture retina chip with the function of local-light adaptation for edge detection has been designed and fabricated using CMOS technology. Human retina can detect a wide range of light intensity. In this study, we use the biologically-inspired visual signal processing mechanism that consists of photoreceptors, horizontal cells, and bipolar cells in order to implement the function of edge detection in the retina chip. For a local-light adaptive function, the size of receptive field is changed locally according to the input light intensity. The spatial distribution of sensing pixels in the foveated retina chip has the advantages of selective reduction of image data and good resolution in central part to carry out the elaborate image processing with still enough resolution in the outer parts. The designed chip has been fabricated using standard 0.6 ㎛ double-poly triple-metal CMOS technology and optimized using HSPICE simulator.

      • Baculovirus를 利用한 Canine Parvovirus VP2蛋自質의 發現

        국내에서 分離된 개파보바이러스주(V20주)의 VP2 遺傳子를 baculovirus system을 이용하여 발현시켜 다음의 結果를 얻었다. 1. 개파보바이러스의 VP2 유전자를 PCR에 의해 增幅하여 1755bp의 VP2遺傳子를 pUC19에 클로닝하여, 클로닝된 遺傳子를 polyhedrin promoter를 가지는 baculovirus expression vector인 pVL1393에 옮겨 VP2 발현벡터인 pVL1393-VP2를 얻을 수 있었다. 2. pVL1393-VP2 plasmid와 baculovirus DNA와의 homologous recombination에 의해 재조합바이러스인 VP2-BV를 얻을 수 있었으며, 그 발현효율은 2.000-5.000 HAU/0.05 ml이었다. 3. 免疫沈澱法에 의해 발현된 단백질은 개파보바이러스의 VP2단백질과 유사한 64 kb에 달하는 것이었으며, 血球凝集能을 지니고 있었다 4. 血球凝集能을 지닌 蛋白質이 여러 陽性血淸에 있어서 抗體수준을 測定할 수 있는지 개파보바이러스항원과의 相關性을 比較한 바 0.94 (n=125. p<0.01)의 相關係數를 보였다 5. 발현 VP2白은 virus-like particles를 形成하였으며, 그 크기로는 개파보바이러스와 비슷한 25 ㎚의 크기를 갖았다. Canine parvovirus(CPV) is a member of autonomous replicating parvoviruses and is aetiologically associated with enteritis and mycoarditis in puppies. The capsids of CPV are composed of three structural proteins: VP1, VP2 and VP3. The VP2 protein is the major component of capsid. The VP2 gene of a canine parvovirus. V20 strain isolated in Korea was cloned into baculovirus expression vector, and subsequently the VP2 protein was expressed by a recombinant baculovirus under the control of polyhedrin promoter. The recombinant VP2 protein expressed in Spodoptera frugiperda 9(Sf9) cells was detected by haemagglutination(HA) test and immunofluorescent antibody assay. Molecular weight of the recombinant VP2 protein expressed was estimated as 64Kd when tested by immunoprecipitation test using anti-CPV monoclonal antibody. In haemagglutination inhibition(Ⅲ) test. 8 HA units of the recombinant VP2 protein antigen was successfully utilized to determine a level of antibody against CPV in various positive sera. The recombinant VP2 protein showed also the capability to form virus like particles similar in size and appearance to the CPV virions.

      • 면역 부전 환자에서 병발한 급성 황색 포도상 구균에 의한 세균성 식도염 1예

        Bacterial esophagitis is an uncommon disease and has not been well characterized. Bacterial infection of the esophagus is usually presented as a superimposed infection upon a preexisting viral or fungal esophagitis and most patients are immunocompromised hosts. A 67-year-old man was admitted for retrosternal pain and hematemesis, who had a past history of long-standing diabetes mellitus and end stage renal disease, also had a history of steroid medication. Extensive esophageal ulcerations of the mucosa were visualized by endoscopy. Staphylococcus aureus grew in blood culture. After the 2 weeks of antibiotics treatment, he was successfully recovered without any sequelae. Due to its rarity, this case is herein reported with a review of the corresponding literature.       • 국내에서 분리된 canine parvovirus의 구조유전자 cloning과 염기서열 분석

        In this study gene encoding structural proteins of a CPV isolate was cloned and saquenced to elucidate the molecular genetical properties of the canine parvoviruses isolated from the field. Six recombinant plasmids of pEP3, p1471, p2070, pEP069, pEP338 and p14711p were constructed from the map positions 22 to 98 of RF DNA to clone the VP1 and VP2 genes of CPV-V20. Sequentialy the gene comprising 3780 nucleotides were sequenced by dideoxy chain termination method. When nucleotide sequence of gene encoding the structural proteins of CPV-V20 was compared with those of other strains, CPV-N, CPV-d and CPV-780929 published previously. DNA homologies to CPV-V20 were 99.87% with CPV-NM, 99.73% with CPV-d, 96.85% with CPV-780929 AND 98.4% with FPLV-Carl, respectively. The DNA sequence data of CPV-V20 showed seven point mutations and also deletion of 135 nucleotides from the nucleotide position 4745 to 4879 located in the 3-noncoding region of CPV-N.

        In order to change the crystallinity of low density polyethylene film, the sample cooling with air, ice and liquid nitrogen was studied for the temperature dependence of dielectric breakdown strength at the temperature range of l0℃∼120℃ As a result, breakdown strength was influenced that increase of crystallenety was due to the decrease of dielectric loss under the glass transition temperature and to the decrease of effect for space charge with Micro Brown Ion motion over the glass transition temperature.         The purpose of this study is to investigate the trend of aging of population within Busan. For this study, 2 population structure indexes and 3 aging indexes of each Dong were analysed and cluster analysis were used for finding out the regional trend and distribution characteristics of aging of population. Also they were analyzed by considering relationship of location with both the period of urbanization and topography. Some of the findings are summarized as follows. (1) the trend of aging of population in area being closer to CBD is more serious than other areas. (2) Aging of population of 5 groups has been serious or going to be rise sharply. Especially, Dongrae in northern CBD axis is expected even rapid depopulation (3) Areas, which are close by mountainous districts are immediately needed to bring regional improvement for aged population
