      • Biochemical analysis of recombinant CYP4A11 allelic variant enzymes: W126R, K276T and S353G

        Human CYP4A11 is the major ω-hydroxylase of fatty acids in the liver and kidneys. It produces 20-hydroxyeicosatetraenoic acid as well as hydroxylates fatty acids. In this study, we investigated the biochemical properties of three alleles of CYP4A11: W126R, K276T, and S353G. Site-directed mutagenesis of the wild type CYP4A11 was performed, to construct the W126R, K276T, and S353G variant clones. The CYP4A11 wild type and variant constructs were heterologously expressed in Escherichia coli. CO-binding spectra showed the expression of the wild type, K276T and S353G variants, indicating the functional P450 holoenzyme. The W126R variant was not expressed in E. coli. Binding affinities of lauric acid in K276T and S353G variants were stronger than that of wild type. Steady-state kinetics in the hydroxylation reaction of fatty acids were studied. The catalytic efficiencies (k<SUB>cat</SUB>/K<SUB>m</SUB>) of K276T and S353G variants in the reactions without cytochrome b<SUB>5</SUB> were approximately 2- and 4-fold higher, respectively, than that of wild type, and in the reactions with cytochrome b<SUB>5</SUB> they were approximately 2- and 3-fold higher, respectively. These results suggest that individuals carrying the alleles, K276T and S353G, might exhibit higher catalysis of CYP4A11, which may affect the endogenous metabolic products associated with regulation of blood pressure.

        비만에 따른 요통환자의 체중분포와 요부 근력차이에 관한 비교분석

        이 연구의 목적은 비만에 따른 요통환자의 체중분포와 요부 근기능 및 안정화 비율의 차이를 비교·분석하는 데 있다. 서울 강남에 소재한 J병원에 내원한 남성 만성요통환자 60명 중 체질량지수(Body Mass Index: BMI)가 25kg/m² 이상인 G1그룹 30명과 이하인 G2그룹 30명을 대상으로 Tetrax와 MedX를 이용하여 분석한 결과 다음과 같은 결론을 얻었다. 체중분포에서 G1그룹은 왼쪽 발뒤꿈치 1.97%(p>.05), 오른쪽 발뒤꿈치 2.37%에서 높게 나타났고(p>.05), G2그룹에서는 왼쪽 발앞꿈치 2.75%(p>.05), 오른쪽 발앞꿈치 1.97%(p>.05)에서 더 높은 체중이 실리는 것으로 나타났다. 요부 각도별 신전근력에서 G1그룹이 G2그룹에 비해 모든 각도에서 높은 근력을 나타냈고, 0°(p>.05), 12°(p>.05)를 제외한 24°(p<.05), 36°(p<.01), 48°(p<.001), 60°(p<.001), 72°(p<.001)에서 통계적으로 유의한 차이를 나타냈다. 요부 안정화 비율에서는 G1그룹의 경우 2.75+_1.31, G2그룹 2.48+_2.23로 나타났다(p>.05). 결론적으로 요부 각도별 신전근력에서 G1그룹이 G2그룹에 비해 모든 각도에서 근력이 높게 나타냈고. 발의 체중분포에서 G1그룹은 비만에 따른 요추의 전만을 증가시켜 힘의 중심을 더 후방으로 유지하려는 경향을 보이고 있는데, 이는 요부 신전근력의 약화로 이어져 안정화 비율에도 영향을 미치는 것으로 사료된다. This study was aimed to determine the effect for the weight distribution and lumbar extension strength associated with obesity index in male patients of Chronic Low Back Pain. Sixty subjects(obesity group(BMI:25Kg/m2): n=30, non obesity group: n=30) participated on this study. Both group were tested on lumbar extension using by MedX machine. Front heels and back heels were measured twice using by the Tetrax Portable Multiple System. Independent sample t-test was used to analyze the difference between the two groups. Study results showed that the obese back pain group(G1) had more weight loaded on both heels than the normal back pain group(G2), but there was no significant difference between the groups (p>.05). As for the lumbar extension strength at each angle, the obese back pain group appeared higher muscular strength than the normal group at 0°, but was stronger at angles 12°, 24°, 36°, 48°, 60°, and 72°. The ratio of lumbar flexion 72° and 0° angle showed 2.75:1 in G1 group and 2.48:1 in G2 group. Overall, the back pain group of obesity(G1) according to weight distribution tended to maintain their center of force more backwards by increasing the lordosis degree with abdominal distension.

        Effect of Cellular Zinc on the Regulation of C2-ceramide Induced Apoptosis in Mammary Epithelial and Macrophage Cell Lines

        Zinc is a trace element that is associated with a stimulation of immune function and regulation of ion balance for livestock production. In this study, the effect of zinc as inhibitor to apoptosis-induced cells was examined in vitro using mammary epithelial cell line, HC11 and macrophage cell line, NCTC3749. Cell viability, measured by MTT assay, indicated that 10 g/ml of zinc had a negative impact on cellular activity and 50 ng/ml was chosen for further testing. Apoptosis was induced in cells treated with C2-ceramide in serum-free media. DNA fragmentation and gene expression of acidic sphingomyelinase (a gene responsible for the progress of apoptosis) were distinctively low in zinc treated cells compared with those in non-treated controls. In conclusion, zinc is involved in the regulation of cell proliferation and apoptosis in mammary epithelial cells and macrophages.

        Castleman 씨 병에 동반된 신유전분증

        Castlemam's disease is an asymptomatic and benign lymph node hyperplasia which is frequently manifested as mediastinal mass in over 70% of cases. Amyloidosis is rarely associated with Castleman's disease. Recently we experienced a patient with nephrotic syndrome due to renal amyloidosis who had been previously diagnosed as Castleman's disease of retroperitoneal lymph node enlargement. A 47-year old woman was admitted because of generalized edema and chest tightness. Five months ago she diagnosed as Castlman's disease-plasma cell type, of retroperitoneal lymph node enlargement. The patient was followed up through outpatient department without any specific medication. Physical examination showed pale conjunctivae, palpable mass on epigastric area and pretibial pitting edema. The hemoglobin, leukocyte count and erythrocyte sedimentation rate were 96 g/L, 5.9×109/L and 127 mm/hour, respectively. The blood urea nitogen was 3.6 mmol/L and creatinine was 88 umol/L. The urinalysis showed 3 positive for albumin. Twentyfour hour urine contained 16.6 grams of protein. A computerized tomographic (CT) scan of the abdomen disclosed multiple lymph node enlargement at mesentery. Percutaneous needle biopsy of kidney showed massive infiltration of pale pinkish amorphous material in the glomerular tuft forming large nodules. Ultrastructural examination exhibited heavy deposition of amyloid fibrils in the mesangium and perpendicular to glomerular basement membrane. Polarizing microscopy with Congo-red stain revealed prominent yellow green birefringence in glomeruli, tubules and interstitium.

        S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

        S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

        PCR 기법을 이용한 한국재래산양의 α s2-casein 유전자의 특성

        This study was performed to provide the basic data for preservation and improvement of genetic resources according to finding genetic construction obtained from the analysis of genetic characteristics of α_(S2-) casein gene in Korean Native goat using PCR-RFLP. This study confirmed the amplified product of 1.3kb fragment obtained from the amplification of a α_(S2-)casein loci by PCR. The frequency of a α_(S2-)casein genotype in Korean Native goat was 100% for AA type and the frequencies of a α_(S2-)casein genotype in Saanen were 80 and 20% for AA and AB genotypes. The frequency of a α_(S2-)casein A allele was 1.000 in Korean Native goat, and the frequencies of a α_(S2-)casein A and B alleles were 0.900 and 0.100 in Saanen, respectively. The nucleotide sequence of a α_(S2-)casein gene of Korean Native goat was 99.0% homology with 12 nucleotide sequences difference of that of goat reported in GeneBank(AJ238965.1, AJ238967.1) and was 95.0% homology with 57 nucleotide sequences difference of that of Holstein reported in Gene Bank(M94327). Therefore, this study of molecular genetic characteristics by the analysis of genetic polymorphisms and sequences for a α_(S2-)casein gene should be used as basic data for preservation and improvement of genetic resources in Korean Native goat breeding.

        In<sub>2</sub>O<sub>3</sub>:Sn/TiO<sub>2</sub>/CdS heterojunction nanowire array photoanode in photoelectrochemical cells

        The design of photoanode with highly efficient light harvesting and charge collection properties is important in photoelectrochemical (PEC) cell performance for hydrogen production. Here, we report the hierarchical In<SUB>2</SUB>O<SUB>3</SUB>:Sn/TiO<SUB>2</SUB>/CdS heterojunction nanowire array photoanode (ITO/TiO<SUB>2</SUB>/CdS-nanowire array photoanode) as it provides a short travel distance for charge carrier and long light absorption pathway by scattering effect. In addition, optical properties and device performance of the ITO/TiO<SUB>2</SUB>/CdS-nanowire array photoanode were compared with the TiO<SUB>2</SUB> nanoparticle/CdS photoanode. The photocatalytic properties for water splitting were measured in the presence of sacrificial agent such as SO<SUB>3</SUB><SUP>2-</SUP> and S<SUP>2-</SUP> ions. Under illumination (AM 1.5G, 100 mW/cm<SUP>2</SUP>), ITO/TiO<SUB>2</SUB>/CdS-nanowire array photoanode exhibits a photocurrent density of 8.36 mA/cm<SUP>2</SUP> at 0 V versus Ag/AgCl, which is four times higher than the TiO<SUB>2</SUB> nanoparticle/CdS photoanode. The maximum applied bias photon-to-current efficiency for the ITO/TiO<SUB>2</SUB>/CdS-nanowire array and the TiO<SUB>2</SUB> nanoparticle/CdS photoanode were 3.33% and 2.09%, respectively. The improved light harvesting and the charge collection properties due to the increased light absorption pathway and reduced electron travel distance by ITO nanowire lead to enhancement of PEC performance.

        Power supply system for KSTAR neutral beam injector

        <P><B>Abstract</B></P> <P>The power supply system in KSTAR neutral beam injector consists of low voltage and high current DC power supplies for plasma generator of ion source and high voltage and high current DC power supply for accelerator grid system. The arc discharge is initiated by an arc power supply supplying the arc voltage between the chamber wall and 12 filaments which are heated by individual filament power supply. The negative output of arc power supply is common to each positive output of 12 filament power supplies. To interrupt the arc discharging for the fault condition of the arc current unbalance, DCCT current monitor is placed at the positive output cable of the filament power supply. The plasma grid (G1) power supply has the maximum capability of 120kV/70A which consists of low voltage regulator with IGBT-switched chopper array system for the voltage control in unit of 600V and the high voltage rectified transformers to supply DC voltage of 20kV, 30kV, and 50kV. The output voltage of the G1 power supply is also connected to the input of the voltage divider system which supplies the gradient voltage to the gradient grid (G2) in the range of 80–90% of G1 voltage by changing tap of winding resistors in unit of 1%. The charged G1 voltage is turned on and off by the high voltage switch (HVS) system consisting of MOSFET fast semiconductor switches which can immediately be opened less than 1μs when the ion source grid breakdown occurs. The decelerating grid (G3) power supply is inverter system using capacitor-charge power supply to supply maximum −5kV/5A. The important component in power supply system is the surge absorber near the ion source to limit the arc energy deposited to accelerator grid. This paper presents configuration and features of power supply system, main controller, and interlock system of KSTAR NBI.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The power supply system in KSTAR NBI consists of DC power supplies for ion source. </LI> <LI> For operation NBI, DC High Voltage based on the low voltage transformer with chopper. </LI> <LI> The surge absorber near the ion source limit the energy deposited to accelerator grid. </LI> </UL> </P>

        Purpose: The purpose of this study was to investigate correlation among sexual knowledge, attitude and Barriers to patient’s sexual health of nursing undergraduate students and to provide objective data to promote sexual health-care. Method: Using a descriptive correlation, 304 nursing undergraduate students were recruited through convenience random sampling from August 1st to December 20th, 2016. Data were analyzed using average, standard deviation, percentage, t-test, ANOVA, Scheffe test, Pearson’s correlation and Multiple linear regression analysis using the SPSS 21.0 program. Results: The correlation between Sexual knowledge and Sexual attitude is statistically significant(r=.289, p<.01). As the level of sexual knowledge is higher, the sexual attitude is more open. And, Sexual knowledge(r=-.307,p<.01) and Sexual attitude(r=.180,p=.002) had a negative relationship with Barriers to patient’s sexual health. Also, Sexual knowledge was the greatest influencing factor on Barriers to patient’s sexual health. Conclusion: This study showed as the level of Sexual knowledge is higher, the Sexual attitude is more open. As the degree of Sexual knowledge is lower, the Sexual attitude is more conservative, and Barriers to patient’s sexual health is more higher. Also, Sexual knowledge is the most influential factor on Barriers to patient’s sexual health.
