Enhanced Viral Replication by Cellular Replicative Senescence

Cellular replicative senescence is a major contributing factor to aging and to the development and progression of aging-associated diseases. In this study, we sought to determine viral replication efficiency of influenza virus (IFV) and Varicella Zoster Virus (VZV) infection in senescent cells. Primary human bronchial epithelial cells (HBE) or human dermal fibroblasts (HDF) were allowed to undergo numbers of passages to induce replicative senescence. Induction of replicative senescence in cells was validated by positive senescence-associated b-galactosidase staining. Increased susceptibility to both IFV and VZV infection was observed in senescent HBE and HDF cells, respectively, resulting in higher numbers of plaque formation, along with the upregulation of major viral antigen expression than that in the non-senescent cells. Interestingly, mRNA fold induction level of virus-induced type I interferon (IFN) was attenuated by senescence, whereas IFN-mediated antiviral effect remained robust and potent in virus-infected senescent cells. Additionally, we show that a longevity-promoting gene, sirtuin 1 (SIRT1), has antiviral role against influenza virus infection. In conclusion, our data indicate that enhanced viral replication by cellular senescence could be due to senescence-mediated reduction of virus-induced type I IFN expression.
HSV-1 ICP27 represses NF-κB activity by regulating Daxx sumoylation

Herpes simplex virus type 1 ICP27 is a multifunctional protein responsible for viral replication, late gene expression, and reactivation from latency. ICP27 interacts with various cellular proteins, including Daxx. However, the role of interaction between ICP27 and Daxx is largely unknown. Since Daxx is known to repress NF-κB activity, there is a possibility that ICP27 may influence the inhibitory effect of Daxx on NF-κB activity. In this study, we tested whether ICP27 affects the NF-κB activity through its interaction with Daxx. Interestingly, ICP27 enhanced the Daxx-mediated repression of NF-κB activity. In addition, we found that sumoylation of Daxx regulates its interaction with p65. ICP27 binds to Daxx, inhibits Daxx sumoylation, and enhances p65 deacetylation induced by Daxx. Consequently, ICP27 represses the NF-κB activity, by elevating the inhibitory effect of Daxx on NF-κB activity through desumoylation of Daxx.

This prospective randomized study was conducted to evaluate the efficacy of two common analgesic techniques, thoracic epidural patient-controlled analgesia (Epidural PCA), and intravenous patient-controlled analgesia (IV PCA), in patients undergoing lobectomy by the video-assisted thoracic surgical (VATS) approach. Fifty-two patients scheduled for VATS lobectomy were randomly allocated into two groups: an Epidural PCA group receiving an epidural infusion of ropivacaine 0.2%+fentanyl 5 ㎍/mL combination at a rate of 4 mL/hr, and an IV PCA group receiving an intravenous infusion of ketorolac 0.2 ㎎/㎏+fentanyl 15 ㎍/mL combination at a rate of 1 mL/hr. Pain scores were then recorded using the visual analogue scale at rest and during motion (VAS-R and VAS-M, 0-10) for five days following surgery. In addition, we measured the daily morphine consumption, forced vital capacity (FVC), forced expiratory volume in 1 second (FEV1), satisfaction score, and the incidence of side effects. Thirty-seven patients out of 52 completed the study (18 in the Epidural PCA group, 19 in the IV PCA group). There were no differences in the pain scores, analgesic requirements, pulmonary function, satisfaction score, and the incidence of side effects between groups. This indicates that IV PCA and Epidural PCA are equally effective to control the postoperative pain after VATS lobectomy, which suggests that IV PCA may be used instead of Epidural PCA.
A Patient with Coexisting Myasthenia Gravis and Lambert-Eaton Myasthenic Syndrome

Background The coexistence of myasthenia gravis (MG) and Lambert-Eaton myasthenic syndrome (LEMS) is very rare and remains controversial. Case Report A 48-year-old woman initially presented with noticeable right ptosis and intermittent diplopia. She then developed fluctuating proximal limb weakness and difficulty in swallowing. The serum titer of anti-acetylcholine-receptor antibody was elevated and the edrophonium (Tensilon) test was positive. However, repetitive nerve stimulation revealed abnormalities typical of LEMS. The patient exhibited a good response to treatment with anticholinesterase inhibitors and steroids, and long-term evaluation disclosed that she presented with the clinical,electrophysiological, and immunological characteristics of both diseases. Conclusions The reported clinical and electrophysiological features suggest that this patient was a very rare case of combined MG and LEMS.
이산화탄소 재호흡 방식에 의한 비침습적 심박출량 측정 방법의 신뢰성:열회석 방법과의 비교

Background: A noninvasive method for estimating cardiac output was tested in dogs. The technique is based on a differential CO_2 Fick equation applied during normal ventilation and 50 seconds of partial rebreathing using additional dead space. We compared the cardiac output measured by the CO_2 rebreathing method vs. the thermodilution technique. Methods: Seven mongrel-dogs (24.6 ±0.4 kg) were studied, anesthesia was induced and maintained with a pentobarbital 25 mg/kg IV bolus injection followed by an infusion of 5 mg/kg/h. Mechanical ventilation was accomplished with a Servo 900C ventilator with FiO_2 0.6 to maintain normocarbia. A fiberoptic pulmonary artery catheter was introduced via an external jugular vein for continuous monitoring of the cardiac output by the thermodilution method. Also cardiac output was measured by using partial CO_2 rebreathing method. A continuous infusion of 0.5% bupivacaine was started at 0.5 mg/kg/min via the venous infusion port of the pulmonary catheter. Bupivacaine was infcontinuously until mean arterial pressure decreased to 60 mmHg or less for at least 5 seconds. Results: The total measured cardiac outputs (n = 72) were distributed over the range of 1.03 7.72 L/min by thermodilution method and 1.6 7.3 L/min by CO_2 rebreathing. The correlation coefficients between both cardiac outputs was 0.6, the mean difference was 0.27 ±0.81 L/min. Conclusions: The cardiac output measured by CO_2 rebreathing method was well correlated with cardiac output by thermodilution method.

Rhois Vernicifluae Cortex (Rhus verniciflua Stokes Bark) as Anacardiaceae family plant doesn't have the standards on chemical characteristics in “Korean Herbal Pharmacopoeia” . Rhois Vernicifluae Cortex were not recorded on Chinese Pharmacopoeia and Japanese Pharmacopoeia as the medicinal herbs. To suggest the new standard on the quality control, we monitored domestic Rhois Vernicifluae Cortex according to the standard operating procedures for herbal drugs. Rhois Vernicifluae Cortex were studied on purity(%), heavy metals, loss on drying(%), total ash, acid-insoluble ash (%) and extract contents(%) and compared to each other. Also, Rhois Vernicifluae Cortex were profiled and compared on chemical patterns using HPLC. These results would be references of the suggestion for the quality control of Rhois Vernicifluae Cortex in Korea.
