        선모(仙茅) 열수(熱水) 추출물의 Collagen 유발 관절염에 대한 약리 효능 연구

        Objectives:In Korean medicine, Curculiginis Rhizoma was treated for arthritis in remedy. But efficacy of Curculiginis Rhizoma on collagen induced arthritis was not revealed. Methods:Anti inflammatory effect of Curculiginis Rhizoma was researched in vitro with RAW264.7 cell and cell toxicity, levels of proinflammatory cytokines (TNF-α, IL-1β, IL-6 and IL-12) and PGE2 were analyzed by ELISA assay. Inflammatory protein were analyzed by western blotting assay (JNK, ERK, COX-2, TNF-α and IL-1β). In vivo, collagen induced arthritis mice model was used to evaluate anti-inflammation effect through arthritis index, immune cell number and cytokine levels (TNF-α, IL-6 and IL-1β) in serum. Results:ECR(Extract of Curculiginis Rhizoma) has not shown cell toxicity in 200 ㎍/㎖ on RAW264.7 cell. ECR suppressed releases of NO, TNF-α, IL-1β, IL-6, IL-12 and PGE2 on RAW264.7 cell treated with lipopolysacharide (1 ㎍/㎖). And ECR inhibited regulation of TNF-α, IL-1β and IL-6 mRNA, reduced protein release of JNK, ERK, iNOS, COX-2, IL-1β and TNF-α. AI of group treated with ECR 200 ㎎/㎏ and 100 ㎎/㎏ were significantly decreased compared to vihicle arthritis mice, the number of immune cell in foot joint was increased on control mice but those of group treated with ECR 200 ㎎/㎏ and 100 ㎎/㎏ were significantly reduced. This results correspond with contens of cytokines (TNF-α, IL-1β and IL-6) in serum. Conclusions:Curculiginis Rhizoma has anti-inflammation effect on RAW264.7 cell in vitro and collagen induced arthritis in vivo . So it is necessary to research more mechanism for cascade imfact.

        Early Phase of UVB - induced GM - CSF Upregulation in Epithelial Cell Line is not Totally Dependent on IL - 1α

        Backgrounds : It was demonstrated that ultraviolet(UV) B light induces the release of IL-la in cultured human epithelial cell line and augmentation of GM-CSF production by UVB is reported to be mediated by IL-1α in the murine keratinocyte cell line Pam 212. Objective : The purpose of this study was to evaluate the effects of UVB on kinetic profile of IL-1 and GM-CSF mRNA expression and to see whether synthesis of GM-CSF by UVB can be completely inhibited by blocking IL-1α mediated pathway. Method : We used a competitive RT-PCR for measuring cytokine gene expression in epithelial cell line after UV radiation. Results : The IL-1α mRNA increased as early as 1h after UV irradiation, and then decreased at 3h after the irradiation. Thereafter, the response of IL-1α mRNA was upregulated with a second peak at 6h after the UV irradiation. However, mRNA for GM-CSF increased at 1h after UV light exposure and anti-IL-1α antibodies could only partially inhibit UV-augmented GMCSF production. Conclusion : UVB induced GM-CSF production seemed to be mainly mediated by UVB induced IL-1α but these results suggest that UVB may also induce GM-CSF production through an IL-1α independent pathway.

        Widespread use of BCG has not controlled tuberculosis, and more effective vaccines are clearly needed. Although chemotherapy will remain the mainstay of antituberculosis treatment, the use of adjunctive immunotherapeutic modalitites is attractive, particularly in persons with drug-resistant tuberculosis. Administration of IL-2 or IFN-γto tuberculosis patients enhance bacillary elimination. Cell-mediated immunity is the critical protective immune response in tuberculosis. Mycobacterial antigens are recognized by T cells and that elicit production of protective cytokines are potentially important vaccine antigens. The 30 kDa antigen is secreted in large quantities by growing mycobacteria. That antigen elicits greater proliferation in lymphocytes from healthy tuberculin reactors than healthy tuberculin nonreactors. In this study, the T lymphocyte proliferative responses to 30 kDa antigen from Mycobactrium tuberculosis H37Rv were examined by using tonsilar and peripheral blood lymphocytes from PPD(+) and PPD(-) tonsilectomized persons. When cultured with 30 kD antigen, tonsilar mononuclear leukocytes and T cells of PPD(+) demonstrated more ^3H-thymidine incorporation than PPD(-) persons (stimulation index was 2.5 and 1.9, 0.8 and 1.0, repectively). Peripheral blood mononuclear cells (PBMC) and peripheral blood T lymphocytes were shown the similar responses to this antigen. The combination of IL-2 and 30 kDa antigen elicited a significant proliferative responsiveness in tonsilar mononuclear leukocytes and T cells of PPD(+) persons (SI was 20 and 14.1). PBMC and peripheral blood T cells of PPD(+) persons were also shown a significant responsiveness, but PPD(-) persons did not show. These results demonstrate that the 30 kDa antigen and IL-2 have a synergistic stimulatory property in mycobacteria sensitizing lymphocytes.

      • Amelioration of inflammatory responses by <i>Socheongryong-Tang</i> , a traditional herbal medicine, in RAW 264.7 cells and rats

        <P>Socheongryong-Tang (SCRT) is a natural medicine prescription that has been mainly used in East Asia for the treatment of inflammatory disorders, including asthma and allergic rhinitis. The present study evaluated the anti-inflammatory effects of SCRT on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and in a rat model of carrageenan (CA)-induced paw edema. Levels of tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and prostaglandin E<SUB>2</SUB> (PGE<SUB>2</SUB>) in the culture supernatant were quantified and nitric oxide (NO) production was monitored. In addition, the effect of SCRT on the protein expression of nuclear factor-κB (NF-κB), mitogen-activated protein kinases (MAPKs), inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) was assessed by western blot analysis. Furthermore, the effects of SCRT on acute inflammation <I>in vivo</I> and changes in the histomorphometry and histopathology of paw skin were observed using CA-treated rats. SCRT (1 mg/ml) inhibited the LPS-induced changes in the protein expression of NF-κB, JNK, ERK1/2, iNOS and COX-2, as well as the production of NO, PGE<SUB>2</SUB> and cytokines. In the rat paw edema assay, administration of 1 g/kg of lyophilized powder obtained from the aqueous extracts of SCRT for 3 consecutive days inhibited the CA-induced increases in skin thickness, mast cell degranulation, and infiltration of inflammatory cells in the ventral and dorsal pedis skin within 4 h. These results demonstrated that SCRT exerts its anti-inflammatory activities in LPS-stimulated RAW 264.7 cells through decreasing the production of inflammatory mediators, including PGE<SUB>2</SUB>, NO and cytokines, via suppression of the NF-κB and JNK and ERK1/2 signaling pathways. In addition, the data of the CA-induced paw edema indicated an anti-edema effect of SCRT. SCRT (1 g/kg) reduced acute edematous inflammation through inhibition of mast cell degranulation and infiltration of inflammatory cells. Therefore, the present study provided scientific evidence for the anti-inflammatory activities of SCRT as well as the underlying mechanisms.</P>

        Hearing impairement in varying degrees constitutes one of the most common disabilites affecting school aged children. Eight out of every 1,000 students under the age of twenty have significant hearing loss. Hearing disabilities range from slight to total loss of sound perception. Statistics indicate that only one out of every ten hearing impaired children will become deaf. Most motor activities can be included students may possess low levels of static and/ or dynamic balance, but the greatest majority possess normal motor and physical capabilities. All the hearing impaired youngsters should be properly assessed and given the identical testing batteries as normal children. Hearing impaired students have, essentialy, a communication handicap and suffer from inability to understand verbal instructions. All physical educators should be able to use minimal finger spelling and signing for hearing impaired students.

      • Analysis of monitoring Road-kill in Odaesan National Park

        We surveyed mammalian roadkill at the National Route 6 (Jin-gogae to Songcheon, 18㎞) and a local road 446 (Odaesan National Park office to Sangwon-temple, 12㎞) in Odaesan National Park. Data of the surveys were analyzed by year, month, and species. Roadkills of 226 individuals were found at the survey areas. Of them, the road-killed 131 mammal individuals, 44 bird individuals, 40 reptile individual and 11 amphibian individuals were found during the survey period. High ratio of the road-kills was observed in mammals than the other animals. Among mammalian species, high ratio of the road-kills was observed in squirrels. There were road-kills of three endangered species, containing otters, yellow-throated marten and leopard cats. According to the analyses of annual road-kills, roadkills of 104 cases were occurred and highly frequent in 2014, following by roadkills of 81 cases in 2015, and then those of 41 cases in 2016. Most ases of the roadkills occurred in summer (June to September). The results will contribute to establishing management for reduction of roadkill in Odaesan National Park.

      • Reversal of serologic, immunologic, and histologic dysfunction in mice with systemic lupus erythematosus by long‐term serial adipose tissue–derived mesenchymal stem cell transplantation

        <P><B>Abstract</B></P><P><B>Objective</B></P><P>To investigate the efficacy of human adipose tissue–derived mesenchymal stem cell (AD‐MSC) transplantation in systemic lupus erythematosus (SLE) and to determine the optimal transplantation window for stem cells either before or after disease onset.</P><P><B>Methods</B></P><P>(NZB × NZW)F<SUB>1</SUB> mice with SLE were administered human AD‐MSCs (5 × 10<SUP>5</SUP>) intravenously every 2 weeks from age 6 weeks until age 60 weeks, while the control group received saline vehicle on the same schedule. Another experiment was carried out with a different initiation time point for serial transplantation (age 6 weeks or age 32 weeks).</P><P><B>Results</B></P><P>Long‐term serial administration (total of 28 times) of human AD‐MSCs ameliorated SLE without any adverse effects. Compared with the control group, the human AD‐MSC–treated group had a significantly higher survival rate with improvement of histologic and serologic abnormalities and immunologic function, and also had a decreased incidence of proteinuria. Anti–double‐stranded DNA antibodies and blood urea nitrogen levels decreased significantly with transplantation of human AD‐MSCs, and serum levels of granulocyte–macrophage colony‐stimulating factor, interleukin‐4 (IL‐4), and IL‐10 increased significantly. A significant increase in the proportion of CD4+FoxP3+ cells and a marked restoration of capacity for cytokine production were observed in spleens from the human AD‐MSC–treated group. In the second experiment, an early stage treatment group showed better results (higher survival rates and lower incidence of proteinuria) than an advanced stage treatment group.</P><P><B>Conclusion</B></P><P>Serial human AD‐MSC transplantation had beneficial effects in the treatment of SLE, without adverse effects. Transplantation of human AD‐MSCs before disease onset was preferable for amelioration of SLE and restoration of immune homeostasis.</P>

        상엽(桑葉)추출물의 LPS로 유도된 RAW 264.7 세포에서의 항염증 효과

        Objectives: Mori Folium is one of the traditional medicinal herb. It was commonly used for sericulture in the world and has been traditionally administered as natural therapeutic agent for the treatment of filariasis, diabetes and dropsy in East Asia. This study investigated an anti-inflammatory potential of Mori Folium ethanol extract (MFE). Methods: We examined the effects of MFE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) and prostaglandin E2 (PGE2) in a murine macrophage cell line, RAW 264.7. Results: MFE inhibited production of NO and PGE2 in a dose dependent manner and also decreased the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2, interleukin (IL)-1, IL-6 and tumor necrosis factor-a. As a plausible molecular mechanism, increased degradation of I-kBa and phosphorylation of I-kBa, NF-kB and MAP kinases by LPS were partly blocked by MFE treatment. Conclusions: These results suggest that MFE has an anti-inflammatory therapeutic potential, which may result from inhibition of NF-kB activation and MAPK phosphorylation, thereby decreasing the expression of pro-inflammatory genes.

        NF-κB 억제를 통한 대회향(大茴香)의 항염증효능

        Objectives : Illicium verum Hook. f. has been known to possess antimicrobial, antioxidant, antifungal, anti-inflammatory, insecticidal, analgesic, sedative, convulsive activities, it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Illicium verum Hook. f. water extracts (IVE) in vivo and in vitro. Methods : Cell viability was measured by MTT assay. The relative levels of NO were measured with Griess reagent. iNOS, COX-2, $NF-{\kappa}B$ and target proteins were detected by immunoblot analysis, and levels of cytokines were analyzed by ELISA kit. Anti-edema effect was determined in the carrageenan (CA)-induced paw edema model in rats. Results : All dosages of IVE used in MTT assay had no significant cytotoxicity. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control. However, these increases were attenuated by treatment with IVE. Also, IVE reduced the elevated production of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 by LPS. IVE inhibited the $p-I{\kappa}B$ and translocation of $NF-{\kappa}B$ to nuclear. Furthermore, IVE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations induced by CA injection. Therefore, IVE will be favorably inhibited the acute edematous inflammations. Conclusion : These results provide evidences that anti-inflammatory effect of IVE is partly due to the reduction of some inflammatory mediators by suppression of $NF-{\kappa}B$ pathway.

        히스티딘의 IgE 매개 알레르기 반응 억제 효과

        비만세포는 알레르기를 유발하는 중요한 세포로서 항원의 자극을 받으면 알레르기 유발 물질을 분비한다. 히스티딘은 비만세포가 관여하는 만성 장염 증세를 완화하는 기능이 있다. 그러나 히스티딘이 비만세포의 활성화에 미치는 영향에 대한 연구는 없다. 우리는 알레르기 반응 연구의 동물모델인 IgE 매개 수동 피부 아나필락시스 방법 그리고 비만세포가 알레르기 유발 물질을 분비하는 반응(탈과립 반응, 염증성 지질 및 사이토카인 분비)을 통해 히스티딘이 알레르기에 미치는 영향을 연구하였다. 생쥐의 복강으로 히스티딘을 100 mg/kg으로 투여하면 수동 피부 아나필락시스를 통계적으로 의미 있는 수준으로 억제하였고, 비만세포에서도 탈과립 반응과 알레르기 유발 불질의 분비를 억제하였다. 이러한 결과는 히스티딘 섭취는 IgE 매개 알레르기를 억제하는 데 유익할 것으로 생각할 수 있다. Mast cells are major immune cells in allergies that secrete allergic mediators in response to antigen stimulation. The amino acid, histidine, ameliorates the inflammation that mast cells are involved. On the other hand, there are no reports of the effects of histidine on the activation of mast cells. This study examined the anti-allergic effects of histidine with well-established experimental methods in allergy studies, including IgE-mediated passive cutaneous anaphylaxis (PCA) in mice and mast cell activation responses, such as degranulation, secretion of inflammatory lipid LTB4 and cytokines (TNF-α and IL-4). The intraperitoneal administration of 100 mg/kg histidine suppressed PCA significantly (P<0.05) in mice. In addition, histidine inhibited IgE-mediated mast cell activation, including degranulation and the production of LTB4 and the inflammatory cytokines, TNF-α and IL-4. Overall, histidine suppressed the IgE-mediated allergic responses in vivo and in vitro. Histidine supplementation is expected to beneficial for IgE-mediated allergies.
