      • 의약품중 잔류유기용매 시험법 확립에 관한 연구

        Headspace-GC(딘5-GC)를 이용하여 ICH 잔류웅매 가이드라인 class 1, 2에 속하는 28종의 잔류유기웅매 분석방법을 연군한 결과 SPB-5, BB-WAX column을 동시 이용하여 각각을 확인, 정량할 수 있었다.또한 염의 참가효과, 평형시간,평형온도가 headspace분석에 미치는 영향을 검토한 결과 평형 시간 30분,평형온도 85℃,Na₂SO₄Ig을 첨가하여 시험할 때 최적 부넉조건을 나타내었다. 회수율은 1,1-achloroethene(68.3%)를 제외하고 90.9 ~ 114.5%로 패체적으로 양호하였다. 따로 HS-GCJECD, DB-524 column을 이용하여 9종의 잔류유기용매에 대하여 각각의 검랸선을 작성하고 검출한계를 구하였다. 실제 의약품 원료 10종및 제제 5종에 적용시킨 결과 모두 불검출되었으며 본 시험법은 의약품중의 잔류유기용매 분석에 적용할 수 있는 방법이라고 판단된다. The effects of the addition of salts,equilibration time, and equilibration temperature on headspace analt·sis were invesugated. Theoptimum conditions weiFe obtained with addtion of Ifa2s04 19 as a salt, simultaneously. thetime and temperature of equitibration were 30min and 8i'f, respectivety. The recovery havefound between 90.9 and 114.5% except 1,1-dichloroethene(fi8.3%). Using DB-624 column &HS-GC/:ECD method, 9 kinds of residual solvents could be individually identified andfuantitated. This HS-GC method can be applied to test the residua3 organic solvent in thepharmaceuticals.

        Bacillus lentimorbus WJ5의 감마선유도 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자의 microarray 분석

        본 연구에서는 감마선으로 유도된 돌연변이체들에서 공통으로 발현되는 방사선 관련 유전자들의 발현을 연구하기 위하여, B. lentimorbus WJ5의 방사선 유도 돌연변이체에서 발현되는 유전자를 DNA microarray로 동시에 탐색하였다. DNA microarray는 B. lentimorbus WJ5 genome을 무작위로 절단하여 2,000단편으로 구성하였으며, 감마선 (^(60)Co)으로 유도된 7돌연변이체의 발현을 정량적으로 관찰하였다. 클러스터 분석결과 발현된 408 유전자 중 27개가 감마선 유도 돌연변이체 모두에서 유의하게 발현이 증가되었다. 특히, 복구(mutL, mutM)에너지 대사 (acsA, sdhB, p하, yhjB, citB), protease(npr), 산화자극에 대한 환원 (HMM)관련 유전자들이 동시에 증가되었다. 이는 감마선 유도 돌연변이체들에서 자발적인 직/간접 복구 관련 유전자의 발현 증가는 방사선 노출 직후 보이는 stress response와는 다른 현상임을 나타내는 것으로 생각된다. To study the radiation related gene expression in mutants of Bacillus lentimorbus WJ5 induced by gamm radiation, the simultaneous gene expression was analyzed by DNA microarray. We constructed DNA chips including two thousand randomly digested genome spots of B. lentimorbus WJ5 and compared its quantitative aspect with seven mutants induced by gamma radiation (^(60)Co). From the cluster analysis of gene expression pattern, totally 408 genes were expressed and 27 genes were significantly upregulated by the gamma radiation in all mutants. Especially, genes involved in repair (mutL, mutM), energy metabolism (acsA, sdhB, pgk, yhjB, citB), protease (npr), and reduction response to oxidative stress (HMM) were simultaneously upregulated. It seems that the induction of the direct and/or indirect repair related genes in mutants induced by gamma radiation could be remarkably different from the adaptive responses against acute exposure to radiation.

        The diagnostic requirements were suggested and explained regarding the systems of differentiation of symptoms and signs in the second year study of standardization and unification of the terms and conditions used for diagnosis in oriental medicine. The systems were as follows; - differential diagnosis according to condition of body fluid, differentiation of syndromes according to the state of qi and blood, differential diagnosis according to relative excessiveness or deficiency of yin and yang(氣血陰陽津液辨證) - differentiation of diseases according to pathological changes of the viscera and their interrelation - analyzing and differentiating of febrile diseases in accordance with the theory of the six channels(傷寒辨證) The individual diagnosis pattern was arranged by the diagnostic requirements in the following order : another name(異名), notion of diagnosis pattern, index of differentiation of symptoms and signs(辨證指標), the main point of diagnosis, analysis of diagnosis pattern(證候分析), discrimination of diagnosis pattern(證候鑑別), a way of curing a diseases(治法), prescription(處方), herb in common use(常用藥物), diseases appearing the diagnosis pattern(常見疾病), documents(文獻調査). This study was carried out on the basis of the Chinese documents and references.

        Background/Aims: In several previous study, insulin resistance has a harmful effect on treatment of chronic viral hepatitis C. We aimed to elucidate the impact of insulin resistance in treatment outcome of Korean Chronic hepatitis C patients. Methods: 152 Korean CVH-C patients were recruited and they were all treated with peginterferon plus ribavirin. All the patient`s ALT, triglyceride, cholesterol, fasting plasma glucose, body mass index were reviewed. Results: In 152 patients, 127 patients attained SVR and 25 did not. The mean FPG of SVR group was 118.3 ± 5.0 , whereas 142.0 ± 15.5 in non- SVR group and it has no significance (p=0.48). The mean TG level of SVR group and non-SVR group were 108.1 ± 4.8 and 116.6 ± 11.6, each. (p=0.08). In 25 DM patients, only 5 (3.9%) patients were succeed in attaining SVR (p=0.00). TG and fasting plasma glucose level were associated with attainment of SVR in CVH-C patients by multivariate logistic regression analysis ( TG ,Odds ratio = 2.65, p=0.049 ; FPG, Odds ratio=3.16, p=0.042). Conclusion: In Peginterferon treated Korean patients, who have high fasting plasma glucose and triglyceride level prone to fail in attainment of SVR.

        Process specific differential metabolomes for industrial <i>gochujang</i> types (pepper paste) manufactured using white rice, brown rice, and wheat

        <P><B>Abstract</B></P> <P>The metabolic perplexes for <I>gochujang</I> (GCJ) fermentative bioprocess, a traditional Korean pepper paste, has largely remain equivocal for preparative conditions and raw material (RM) additives exacerbating its commercial standardization. Herein, we outlined a differential non-targeted metabolite profiling for three GCJ (white rice-WR; brown rice-BR; wheat-WT) under varying processing steps (P1 – fermentation; P2 – <I>meju</I> addition; P3 – ripening; and P4 – red pepper addition). We correlated the process specific metabolomes with corresponding physicochemical factors, enzymatic phenotypes, and bioactivities for GCJ-types. The P1 was characterized by a uniform increase in the levels of RM-derived lysoPCs. In contrast, P2 was observed with proportionally higher levels of <I>meju</I>-released isoflavones and soyasaponins in WR-GCJ, followed by BR and WT-GCJ. The P3 involved a cumulative increase in primary metabolites in all GCJ samples except lower organic acid contents in WT-GCJ. The pepper derived flavonoids and alkaloids were selectively increased while P4 in all GCJ-types.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Metabolic alterations govern quality parameters for industrial <I>gochujang</I> (GCJ). </LI> <LI> Metabolic dynamics for GCJ-types were altered with production processes. </LI> <LI> P3 (ripening) crucially effects nutritional and functional aspects for GCJ. </LI> <LI> Process specific metabolic profiling for GCJ types revealed putative biomarkers. </LI> </UL> </P>

        Adenocarcinoma Occurring in a Gastric Hyperplastic Polyp Treated with Endoscopic Submucosal Dissection

        Gastric hyperplastic polyps are generally considered benign lesions, although rare cases of adenocarcinoma have been reported. Although, the underlying mechanism of carcinogenesis in gastric hyperplastic polyps is still uncertain, most malignant polyps are seen to originate from dysplastic epithelium rather than from hyperplastic epithelium. Herein, we report the case of a woman diagnosed with adenocarcinoma that originated from a hyperplastic gastric polyp that was successfully removed by endoscopic submucosal dissection. In this case, we observed adenomatous changes around the cancerous component.

        Renal Infarction Concurrent with Multiple Venous Thrombosis in Association with Inherited Antithrombin Deficiency

        Antithrombin deficiency is a rare condition among the numerous conditions that can lead to a hypercoagulable state, and can manifest as deep vein thrombosis, portal or mesenteric venous thrombosis, pulmonary thromboembolism and cerebrovascular accidents. In this report, we present a case of acute renal infarction and multiple venous thrombosis in a 36-year-old man with a family history of thromboembolism. He presented with a sudden onset of pain in the right flank and was admitted to the emergency room for evaluation. On computed tomography and renal angiography, the diagnosis of acute renal infarction concurrent with portal, splenic and superior mesenteric venous thrombosis was made. Laboratory data revealed parallel decreases in activity and antigen concentration of antithrombin despite normal liver and renal functions. He was treated with intravenous heparin and fresh frozen plasma followed by concomitant warfarin therapy. Taken together, the etiology of acute renal infarction and multiple venous thrombosis was considered to be associated with type I inherited antithrombin deficiency.

        Anthocyanin의 Delphinidin이 MDA-MB-231 유방암세포에 미치는 영향

        본 연구에서는 블루베리에 포함된 anthocyanin 중 delphinidin이 인간 유래의 MDA-MB-231 유방암세포의 성장을 억제시키고 apoptosis를 유발하는지 살펴보고 in vivo 실험에서도 항암효과가 나타나는지 확인하였다. 그 결과 cell viability를 보기 위한 MTT assay에서는 농도 의존적으로암세포의 성장을 억제시켰으며, apoptosis의 확인을 위한 DAPI stain에서 농도 의존적으로 chromatin condensation이 유의적으로 증가하는 것을 확인하였다. 또한 western blot에서 암 억제 유전자인 p53 단백질이 증가하였고, anti-apoptotic 분자인 Bcl-2 단백질과 p-GSK3β 단백질은 감소하였다. In vivo 실험에서는 대조군과 비교해 10 mg/kg delphinidin을 투여한 군에서 종양의 크기가 감소하였으며, TUNEL assay를 통해 apoptosis 세포 또한 통계학적으로 유의적인 증가가 관찰되어 종양 억제 효과를 확인하였다. 이상의 결과들로 볼 때, MDA-MB-231 유방암세포에서 delphinidin은 암세포의 증식을 억제시키고, apoptosis를 유발시키는 효과를 보이므로 암 예방제나 치료제로 개발될 수 있을 것으로 사료된다. This research showed that delphinidin, an anthocyanin present in blueberry, induced apoptosis in MDA-MB-231 breast cancer cells as well as mediated anti-cancer effects in vivo. As a result, growth of cancer cells as assessed by MTT assay decreased in a concentration-dependent manner. Chromatin condensation by DAPI staining significantly increased in a concentration-dependent manner, indicating apoptosis. The level of p53-protein increased while those of anti-apoptotic molecules (Bcl-2, p-GSK3β) decreased in the western blot. Tumor size decreased in cells treated with 10 mg/kg of delphinidin compared with the control group in vivo. Cell apoptosis assessed by TUNEL assay significantly increased, and tumor inhibitory effect was confirmed. Therefore, delphinidin can be developed for cancer preventive medicine due to its growth inhibitory effects and induction of apoptosis in human breast cancer cells.
