Thioacetamide에 의한 BALB/c 마우스 간의 시간별 약물대사효소 억제 양상 : A Time-Course Study

Thioacetamide is a potent hepatotoxicant which requires metabolic activation by cytochrome P450s (P450s) for toxicity. In the present study, the elevation kinetic of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities by thioacetamide treatment was investigated in male BALB/c mice. Inaddition, the inhibitory effects of thioacetamide on liver microsomal P450 enzymes were further investigated. Thioacetamide at 100 mg/kg/ was treated intraperitoneally for 6, 12, 24, 36, 48 and 72 hr. The blood was collected at the designated time for assaying the serum enzyme activities. To determine the P450 isozyme-specific activities. ethoxyresorufin O-deethylase (EROD), methoxyresorufin O-demethylase (MROD), and benzyloxyresorufin O-debenzylase (BROD) activities were determined for P450 1A1, 1A2 and 2B1, respectively, in liver microsomal fractions. The activities of ALT and AST were started to be elevated 6 hr after thioacetamide treatment andreached the maximun at 36 hr after the treatment. The elevated activities were dramatically recovered at 72 hr. The microscopic exmination of the liver specimen also showed a similar profile of hepatotoxicity. All P450-associated enzyme activities were time-dependently inhibited by the treatiment with thioacetamide. The maximum inhibition of P450 enzymes was observed 36 hr after the treatment. Because the inhibition of P450 enzymes by thioacetamide was time-dependent, our present results suggest that thioacetamide might inhibit P450 enzymes in mechanism-based inactivation.
口眼渦斜 患者의 Gadolinium-DPTA enhanced MRI 所見에 대한 臨床的 考察

This study is designed to evaluate the clinical implications of Gd-DPTA (Gadolinium-diethyl - enetriamine pentacetic acid) enhanced MRI(Magnetic resonance imaging) in Bell's palsy and find it's usefulness in Oriental Medicine In this study, 25 outpatients with Bell's palsy were studied that MRI was performed. To evaluate degree of facial palsy, H-B(House-Brackmann) Grade was used. In Oriental Medical therapy, Acupuncture and Herbal medicine were treated. Subjective causes was divided into exposure to chill, fatigue, stress, mixed cases. Enhanced site was compared with symptoms which were disorder of eye, hearing, taste, and facial muscle palsy. Also, Relation between time which was performed MRI and enhancement was analyzed. The enhanced lesion in MRI was divided into five segments; Internal auditory canal, Labyrinthine segment, Geniculate ganglion, Tympanic segment, Mastoid segment. In Bell's palsy, 20 of 25 patients(80%) had abnormal contrast enhancement of the facial nerve. The H-B grade and interval performed MRI from onset were directly proportionate to enhancement. That is to say, Severe facial palsy short interval show high possibility of enhancement. There was no relation between subjective causes and enhanced site of facial nerve in MRI. Also Clinical symptoms didn't coincide with MRI findings.

N/A HM10411 is a recombinant granulocyte-colony stimulating factor (rG-CSF) that has been developing as a drug for neutropenia. In this study, antigenic potential of HM 10411 was examined by active systemic anaphylaxis (ASA) in guinea pigs and passive cutaneous anaphylaxis (PCA) in guinea pig-guinea pig system. HM10411 was subcutaneously administered at 0, 5, and 50 ㎎/㎏ and also as a suspension with adjuvant (50 ㎎/㎏+FCA). Ovalbumin (OVA) as a suspension with adjuvant was used to induce positive control responses. In the ASA test, no symptoms except urination and evacuation that were considered as physiological phenomena were observed at 0 and 5 ㎎/㎏. Two of 5 animals at 50 ㎎/㎏ showed sneezing, dyspnea, or cyanosis. All animals in the adjuvant mixture group showed severe symptoms of anaphylatic shock and 3 of them died. In the PCA test, no antibody against HM10411 was detected in the sera from the animals sensitized with 0 or 5 ㎎/㎏. Only 1 serum sample from the animals immunized with 50 ㎎/㎏ showed positive reaction against HM10411, while all 5 sera collected from the HM10411 and FCA mixture group contained the HM10411 -specific antibodies. These results suggest HM10411 is considered to have antigenicity in guinea pig.

Futhermore, the hardware has been developed by completely domestic technology with knowledge from various actual experiences. In the long run, for uninterruptible power supply to train, building, and factory, real time tele-supervisory control to power facilities.

Background: Onychomycosis, especially toenail onychomycosis has become one of the common fungal infection and has historically been regarded as a cosmetic rather than medical problem by many patients, even by physicians. Recently, however, there are several reports that is a refractory disease which may cause a deleterious effect on patiets quality of life (QOL). Objective: The purpose of this study was to investigate the impact of toenail onychomycosis on QOL in Korea and to assess the chandes of QOL after treatment. Methods: Total 1004 patients with toenail onychomycosis which was confirmed by clinical findings and KOH preparation were were enrolled at 47 dermatologic centers in Korea, and interviewed with standardized QOL questionnaire before and after sytemic antifungal treatment. Responses to the questionnaire were scored by 5-point scale (1∼4) and averaged, and were analyzed for 5 dimensions of emotional impact, social impact, symptorn and functional impact, patients views concerning treatment, and relationship with doctor. Results: 1. Before and after treatment, the most serious impact was emotional dimension showing 1.90 and 1.30 in average score (AS), and social (AS: 1.14 and 0.83) and symptom and functional impact (AS: 1.05 and 0.92) was also affected. 2. In female rather than male, statistically more significant impact on patients QOL was observed in all dimensions. 3. After treatment, 3 of 5 dimensions were improved significantly - emotional dimension (AS : from 1.90 to 1.30), social dimension (AS: from 1.14 to 0.83), patients view concerning treatment (AS: from 1.34 to 1.02). 4. The drgree of patients satisfaction at the therapeutic effect was very high- 62.4% (immediately after treatment) and 65.8% (9 months affer initiation of treatment) of patients answered excellent or good. Conclusion: This study confirms that toenail onychomycosis has significant impact on the overall QOL of patients. Also the effect of antifungal therapy on patients QOL were watisfactory. Therefore, both doctor and patient should pay more attention to the treatment of onychomycosis. [Kor J Med Mycol 3(1): 115∼124]
Elevated cAMP level attenuates 2-deoxy-D-ribose-induced oxidative damage in pancreatic β-cells

Glucose toxicity to pancreatic β-cells is defined as irreversible β-cell damage, including apoptosis, caused by chronic exposure to high glucose levels in type 2 diabetes. Oxidative stress is an important mechanism for glucose toxicity to pancreatic β-cells. Reducing sugars produce reactive oxygen species through autoxidation and protein glycosylation. 2-Deoxy-D-ribose (dRib) is a reducing sugar with high reactivity. We investigated whether cAMP-stimulating agents could protect β-cells from dRib-induced oxidative damage. HIT-T15 cells were cultured with various concentrations of dRib for 24h. We measured cell survival, intracellular cAMP and H_(2)O_(2) levels, and apoptosis. dRib decreased cell survival in a dose-and time-dependent manner and markedly increased intracellular H_(2)O_(2) levels and apoptosis. N-Acetyl-L-cysteine decreased dRib-induced rises in intracellular H_(2)O_(2) and apoptosis to control levels. Forskolin, IBMX, and dbcAMP markedly elevated intracellular cAMP levels and significantly attenuated dRib-induced cytotoxicity and apoptosis, but had no influence on the dRib-induced rise in intracellular H_(2)O_(2) levels, These results demonstrate that dRib produced oxidative stress and apoptosis in pancreatic β-cells and that elevated intracellular cAMP levels reduced dRib-induced damage, independent of reactive oxygen species metabolism.
Antigenicity of HM10760 in Guinea Pigs

HM10760 is a recombinant human erythropoietin that has been developed as a drug for anemia. In this study, antigenic potential of HM10760 was examined by active systemic anaphylaxis in guinea pigs and passive cutaneous anaphylaxis in a guinea pig-guinea pig system. HM10760 was subcutaneously administered at 0, 2, and 20 ㎍/㎏ and also as a suspension with adjuvant (20 ㎍/㎏ + FCA). Ovalbumin as a suspension with adjuvant was administered to induce positive control responses. In the active systemic anaphylaxis test, no symptoms except rubbing or licking nose and urination that were considered as physiological phenomena were observed at 0 ㎍/㎏. Four of 5 animals at 2 ㎍/㎏ and all the 5 animals at 20 ㎍/㎏ showed cyanosis and lying on side. All animals in the adjuvant mixture group showed relatively mild symptoms such as rubbing or licking nose, urination, and evacuation. In the passive cutaneous anaphylaxis test, 0/5, 3/5, and 5/5 serum samples from the animals immunized with 0, 2, and 20 ㎍/㎏, respectively, showed positive reactions against HM10760. All 5 sera collected from the animals immunized with an adjuvant mixture contained HM10760-specific antibodies. These results suggest HM10760 have antigenicity in guinea pigs.
Rapidly growing giant pilomatricoma in the right parotid region of a pregnant woman

Pilomatricoma is a benign tumor arising from the primitive basal cells of the epidermis that differentiate into hair matrix cells. Mutations in the CTNNB1 gene, which encodes β-catenin (a protein involved in hair growth), play an etiological role in the development of pilomatricoma. A 34-year-old woman presenting with a mass in the right parotid region underwent an excisional biopsy. The mass was conclusively diagnosed as pilomatricoma. During pregnancy, the mass grew from 1 cm to 5 cm in diameter and was accompanied by pain and tenderness. The growth may have been facilitated by the increased production of estrogen and progesterone, which bind to receptors located in the outer root sheath cells of the hair follicles. No recurrence was observed during 6 months of follow-up.
