        Purification and Characterization of Thermostable β-Glucosidase from the Brown-Rot Basidiomycete Fomitopsis palustris Grown on Microcrystalline Cellulose

        An extracellular β-glucosidase was purified 154-fold to electrophoretic homogeneity from the brown-rot basidiomycete Fomitopsis palustris grown on 2.0% microcrystalline cellulose. SDS-polyacrylamide gel electrophoresis gel gave a single protein band and the molecular mass of purified enzyme was estimated to be approximately 138 kDa. The amino acid sequences of the proteolytic fragments determined by nano-LC- MS/MS suggested that the protein has high homology with fungal β-glucosidases that belong to glycosyl hydrolase family 3. The Kms for p-nitorophenyl-β-D-glucoside (p-NPG) and cellobiose hydrolyses were 0.117 and 4.81 mM, and the Kcat values were 721 and 101.8 per sec, respectively. The enzyme was competitively inhibited by both glucose (Ki= 0.35 mM) and gluconolactone (Ki= 0.008 mM), when p-NPG was used as substrate. The optimal activity of the purified β-glucosidase was observed at pH 4.5 and 70°C. The F. palustris protein exhibited half-lives of 97 h at 55°C and 15 h at 65°C, indicating some degree of thermostability. The enzyme has high activity against p-NPG and cellobiose but has very little or no activity against p-nitrophenyl-β-lactoside, p-nitrophenyl-β-xyloside, p-nitrophenyl-α-arabinofuranoside, xylan, and carboxymethyl cellulose. Thus, our results revealed that the β-glucosidase from F. palustris can be classified as an aryl- β-glucosidase with cellobiase activity.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        The objective of this study was to investigate the effect of AE activities during fatigue test. Laboratory experiment was carried out to identify AE characteristics of fatigue cracks for compact tension specimen. The relationship between a stress intensity factor and AE signals activity was discussed. From the experimental results, the features of specific parameters such as the length of crack growth, the AE energy and the AE peak amplitude, showed similar trend in their increase as the stress intensity factor increased. The fracture surface of test specimens revealed both intergranular fracture and transgranular fracture. Size and density of slip band were varied as the stress intensity factor increased and AE activities were different according to the slip band shape.

To serve this purpose, the meaning, characters and ideal teacher's role are observed. 1.The characters of human relations between kindergarten teachers and young children are ① Teaching and learning relations, ② Hardly have choice relations, ③ Giving and Taking influences relations, ④ Sharing love relations, ⑤ Complicated relations. 2.The idel teacher's role and attitudes are ① qualified teachers, ② Authoritative teachers, ③ Sharing love effectively, ④ Trying to develop themselves, ⑤ Growing with young children. 3.As conclusion, It is necessary to establish more effective ability of human relations for Kindergarten teachers. It is recommended to have lecture of human relations in teacher's college.

        The purpose of this study was to develop a in-service teacher education program for student teaching in early childhood education center in order to reinforce student teacher's educational role for improving their student teaching. In this paper, the sample consisted of 20 early childhood professionals who have experienced at least 3 years in students teaching program. The survey was administered using the Delphi method. The study explored the objectives, contents, administration and method, and evaluation of the developed program. We also attempt to outline the present conditions and problems on student teaching in early childhood education center. As a conclusion, the program have to assist the able kindergarten teacher for understanding student teacher correctly, to consist and administrate a practicum program with systematic support.

        악관절내장증을 동반한 Eagle's Syndrome

        A thirty two year-old male was referred with a chief complaint of mouth opening limitation, and maxillofacial pain including left TMJ area. The patient had been treated in a private clinic with medications and conservative treatments, without any improvements in symptoms. MRI findings showed a limitations in condylar head movements, and signs of disc adhesion without the Positional change. Panoramic views showed elongation of stylohyoid process. Brain C-T was taken due to the patient's complaint of headache, facial pain, and paresthesia of tongue, and the result showed no abnormalities. The first surgery included meniscoplasty and the removal of disc adhesion of left TMJ under the preliminary diagnosis of internal derangement with adhesion, and the patient displayed marked improvements in opening movements despite of persistent hemiparesthesia of left facial areas and tongue, and mild dysphasia. Therefore, the second surgery was carried out with the preliminary diagnosis of Eagle's Syndrome, and a resolution of neurologic symptoms was obtained. The final diagnosis of this case was Eagle's Syndrome with internal derangement, and the difficulties in diagnosis and treatment might be due to the existence of two separate diseases with similar symptoms.

        The purpose of this study is to get the foundational data for developing more effective Early Childhood Education program of Seamaul Nursery School(S.N.S) To serve this purpose, analysis was conducted about the aspects of group games and the intellectual values of it. Observation and interview were used at the 3 S.N.S selected as sample and located in Cheong-Ju. The major findings are follows: 1. Modern group games were played mostly more than traditional group games. 2. Children have learned traditional group games by their parent, brothers and peer. 3. According to their sex, group games differed remarkably in membership of it. 4. Tools used for the group games were mostly natural things or things easy to get. 5. Group games at S.N.S have important intellectual values.

The results of this study are as follows 1. Aspect of quantity, the education of Chung-buk was developed quickly through past 40years. But they have quality problems for developing more effective education. 2. Other characters of education in Chung-buk are complexity structure and many difference between each city or country in quality. So planning for city and country type educational model is needed. 3. The most problem is poor budget for education. change the tax system, contribution plan, issuance of public deventure are effective strategy to improve the educational structure in Chung-buk.

        A 7-year-old male castrated Shih-tzu dog was referred to the Veterinary Medical Teaching Hospital of Chungnam National University for severe pruritus. On physical examination were confirmed systemic erythema and alopecia, crust on the neck and flank region. Tape strip test identified numerous degenerative neutrophils and severe Malassezia infection. Treatment for severe pruritus was applied with systemic antihistamine therapy and topical therapy with antimicrobial shampoos. For the finding of appropriate treatment of atopic dermatitis, it was chosen six immune-related treatments. And clinical sign scores were evaluated by investigator with CADESI-03 after each application of six treatments. Application of human immunoglobulin G and leflunomide showed the remarkable regression of lesions.

      • 계열별 고등학생집단의 뇌기능특성화 경향에 대한 연구

        본 연구에서는 계열에 따른 고등학생들의 대뇌반구기능의 특성화 또는 편재화의 경향을 알아보고, 전공분야에 대한 적응력과의 상관관계를 검토하고자 했다. 일반고등학교 인문계열 학생 359명, 자연계열 학생 371명, 음악과 미술전공 학생을 포함한 예능계열 196명, 체육특기자를 포함한 체육계열 학생 151명 등 1,077명을 대상으로 각 계열별, 남녀별, 성적별 고등학생 집단들의 대뇌반구의 특성화, 편재화 경향을 시나가와(품川)질문지법을 이용하여 분석하였으며, 다음과 같은 결과를 얻었다. 1. 인문계열 고등학생의 뇌기능의 편재화 경향은 우뇌형 54.3%, 양쪽뇌형 32.6% 및 좌뇌형 13.1%였다. 남녀별로는 남학생은 우뇌형 62.9% 양쪽뇌형 27.9% 및 좌뇌형 9.5%로 나타났으며, 여학생은 각각 43.7%, 38.6% 및 17.7%로서 여학생이 남학생에 비하여 죄뇌화 경향을 보였다. 2. 자연계열 고등학생의 경우는 우뇌형 56.9%, 양쪽뇌형 29.9% 및 좌뇌형 13.2%로서 인문계열 고등학생과 비슷한 분포를 보였다 이를 남녀별로 구별하여 보면 남학생은 62.2%, 27.1% 및 10.2%, 여학생은 51.5%, 32.5%및 16.0%의 분포를 보여, 역시 여학생의 좌뇌화 경향이 뚜렷했다. 3. 예능계열 고등학생의 경우는 우뇌형 56.1%, 양쪽뇌형 35.2% 및 좌뇌형 8.7%의 분포를 보였으며, 이를 전공별로 세분해서 살펴보면 음악전공학생의 경우 53.7%, 35.5% 및 10.7%로서 전체 평균과 거의 비슷한 분포를 보였으나, 미술전공학생의 경우 60.0%, 34.7% 및 5.3%로서 , 특히 좌뇌형의 경우 음악전공에 비해 절반수준에도 못 미치는 경향을 보였다. 4. 체능계열 고등학생 집단의 경우 우뇌형 50.3%, 양쪽뇌형 37.7% 및 좌뇌형 11.9% 분포를 보였다. 5. 성적별로 보면 일반고등학교 학생 가운데 성적 우수 집단은 뚜렷하게 좌뇌형 경향(25.6% 및 25.0%)을 보였다. 6. 예능계열의 우수집단은 양쪽뇌형의 분포비율(50.0% 및 44.4%)이 특히 높았다. 7. 체육계열의 우수집단은 좌뇌형 분포(21.6%)가 높았다. According to the advanced study on differential theory of brain function, the left hemispheric functions are concerned more on the verbal, logical, sequential and arithmetic abilities, whereas the right hemisphere has characteristics of non-lingual, general, creative and aesthetic abilities. The authors compared the tendency of hemispheric specialization in high school students with different educational condition and facilities. Sinakawa's questionnaire method was the instrument for this study. The results obtained were as follows: Among 1,077 of whole subjects, right hemispheric dominant type was 55.0% (59.1% in male, 51.0% in female), bilateral type was 32.9% (10.6% in male, 35.1% in female) and left hemispheric dominant type was 12.2% (10.4% in male, 13.9% in female). 1. Of the 359 subjects in the field of humanity science, right hemispheric dominant type was 54.3% (62.9% in male, 43.7% in female), bilateral type was 32.6% (27.9% in male, 38.6% in female) and left hemispheric dominant type was 13.1% (9.5% in male, 17.7% in female). 2. Of the 371 subjects in the field of natural science, right hemispheric dominant type was 56.9% (62.2% in male, 51.5% in female), bilateral type was 29.9% (27.1% in male, 32.5% in female) and left hemispheric dominant type was 13.2% (10.2% in male, 16.0% in female). 3. Of the 196 art school students, 56.1% was regraded as right hemispheric dominant type, students majoring music was 53.7%, 35.5% and 10.7%, respectively, whereas the frequency for students majoring find art was 60.0%, 34.7% and 5.3%. 4. In students majoring physical education, 50.3% were shown in right hemispheric dominant type, 37.7% were bilateral type and 11.9% were left hemispheric dominant type. 5. Laterality patterns of students who adapt more successfully to their specific studying fields show following characteristics; Successful students of general high school show pronounced left shift in their laterality pattern, whereas successful students of art school exhibit more bilateral type. And successful students of physical education school show left shift in their laterality patterns.
