        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

      • 길항세균 Pseudomonas cepacia(P713 & P41)의 근권정착력 및 수박덩굴쪼김병 (Fusarium oxysporum f.sp.niveum)에 대한 생물적방제효과

        In order to find out effect of the antagonistic bacterium for Fusarium wilt of watermelon, the two strains of Pseudomonas cepacia were tested. Inhibition zone formed by the antagonist was not only confirmed with a dual culture but also examined conidial germination and mycelial growth of the fungus by using culture filterate. Root colonization of the two antagonistic Pseudomonas cepacia (P7l3 & P4l) were successfully confirmed on the cucurbit rhizospheres through DLF(double layered filterpaper method) and Ahmad and Baker methods, and there was no difference of root colonization between the two isolates of the antagonist. With regard to competition between the antagonists and pathogen, the two bacteria were not challanged by the casual fungus. Furthermore, numerous antagonistic bactera were observed at root tip, terminal root and leteral root through scanning electron microscope. Biocontrol effect of 40% to 100% forFusarium wilt of watermelon were confirmed by percent of diseased plants under the pots and polyethylene film house experiments. : Pseudomonas cepacia, root colonization, Fusarium oxysporum f. sp.         『東國金石評』의 자료적 가치

        이 논문은 아직까지 학계에 소개되지 않은 『東國金石評』의 자료적 가치를 밝히는 것을 목적으로 한다. 필사본 1책의 『東國金石評』은 현재 미국 버클리대학 아사미 문고에 소장되어 있다. 이 책에는 <東國金石評>, <輪回梅十箋>, <발합경> 등의 네 종류 저술이 함께 묶여져 있다. 이 가운데 <輪回梅十箋>은 李德懋의 저술로 『靑莊館全書』에 실려 있으며, <東國金石評>은 저자가 밝혀지지 않은 채 서유구의 『林園經濟志』에 그 일부만이 인용되어 있다. 그리고 <南方異木簿>와 <발합경>은 아직까지 알려지지 않은 신자료이다. <東國金石評>은 우리나라 역대 金石文을 대상으로 하여, 書筆者, 篇名, 書體, 品評 및 所在地 등을 기록해 놓은 것으로, 金石學 및 書藝史 연구의 기초 텍스트로서 널리 활용되어 왔다. 이 자료를 통해 그동안 일부분만이 알려져 왔던 <東國金石評>의 전모를 확인할 수 있게 되었다. <輪回梅十箋>은 밀랍으로 만든 梅花의 제작과 완상 방법 등을 10항목으로 유형화하여 서술해 놓은 것으로, 李德懋를 비롯하여 18세기 후반기 문인지식인들의 예술적 취향의 일단을 엿볼 수 있는 저술로 주목된다. 그리고 <南方異木簿>는 沈念祖의 부탁을 받은 저자가 15종의 기이한 나무의 명칭과 특징을 간략하게 적어놓은 저술이며, <발합경>은 집비둘기의 명칭, 종류, 외형, 습성 및 관련 사실 등을 매우 소상하게 설명해 놓은 흥미로운 저술이다. 특히 <발합경>은 심미적 감식과 세밀한 관찰에 근거하여 집비둘기의 품종, 생태, 특성을 상호 품평 및 유형화 방식을 통해 서술해 놓고 있다는 점에서, 18세기의 학술 및 예술 방면의 새로운 동향과 관련하여 주목되는 저술이다. 『東國金石評』의 編著者는 여러 관련 증거를 고려해 볼때 李德懋일 가능성이 현재로서는 가장 크다고 할 수 있는데, 좀더 면밀한 고증이 필요하다. Donggukgeumseokpyeong is new text which has not been introduced yet. This manuscript book is owned by Asami collection at the East Asian Library in UC Berkeley and supposed to be written by Yi Deokmu(1741-1793), namey one member of the positive school in the later Chosun dynasty. This manuscript book has four kinds of texts, namely Donggukgeumseokpyeong, Yunhoemaesipjeon, Nambangimokbu, Balhapgyeong. Donggukgeumseokpyeong estimates the handwriting and its style of the inscription on stone monuments. This text is important book on epigraphy in eighteen century. Yunhoemaesipjeon explains the method of production and appreciation on Japanese apricot tree which is made by beeswax. This text reflects artistic taste of literary men in eighteen century. Nambangimokbu explains species and its charateristic of tree. This text deals with fifteen kinds of peculiar trees. Finally Balhapgyeong is intersting text. Balhap means house pigeon. This text deals with the mode of life and habits of house pigeon. Donggukgeumseokpyeong reflects the new aspect of arts and science in eighteen century. Asami collection at the East Asian Library in UC Berkeley is evaluated as the invaluable materials in quality and quantity. Although there are some books known to Korean scholars, the collection has many important materials that one need to study closely and deeply.

      • Ortho-Chlorobenzylidene Malononitrile(CS) 이 흰쥐 피부조직에 미치는 조직학적 영향에 관한 연구

        This study was undertaken to describe the fine structural changes of cytoplasmic organelles in epidermal cells, and the changes of volume and localization of collagen type Ⅰ in the dermis of rat skin by CS(Ortho-chlorobenzylidene malononitrile) application. The experimental animals, 100∼150gm of body weight rats(Sprague-Dawley strain), were sacrificed at 12 hours, 24 hours, 3 day and 5 day after CS application on the hair-shaved back skin. The specimens were prepared for ultrastructural findings, ultrathin sections were stained with uranyl acetate and lead citrate double stain and observed with Hitachi-600 electron microscrope. The others were incubated with rabbit anti rat type Ⅰ collagen antibody as primary antibody, and Goat anti rabbit IgG antibodies as secondary antibody And advidin-biotin-conjugated(ABC) peroxidase procedure was used. All of specimens for immune reactions were observed with light microscope. The results obtained were as follows. 1 Keratinocytes appeared fully necrotized after 3 days of CS application. The epidermis showed degeneration of basal lamina, enlargement of intercellular spaces and necrotic keratinocytes with vacuole and vesicles. And large keratohyalin granules were seen in the shrunken epidermal cells. 2 At 3 or 5 day after CS treatment, the normal cytoplasmic organelles of keratinocytes began to appear on the basal and prickel cells. 3 At 12 hour an 24 hour after CS treatment. The immune reaction of type Ⅰ collagen showed weak reaction in reticular layer of dermis. But at 3 day and 5 day after CS treatment, moderate immune reaction were seen in throughout the dermis. These results suggest that CS application to rak skin may induce changes of keratinocytes and reductive changes of type Ⅰ collagen in the dermis.

      • CMC 溶液 氣泡塔에서의 Gas Hold-up 및 氣泡크기 分布

        Gas hold-ups, bubble frequency and bubble size distribution in a bubble column reactor were evaluated experimentally in CMC solutions (<1.0%) with the two-phase flow signal processor. Gas hold-ups were able to be described by the function of the superficial gas velocity, which was represented with Φ=aU_G^b where a=0.018 and b=0.764. Bubble frequency was increased with superficial gas velocity, but this phenomenon was reduced drastically in the higher concentrations of CMC by the results of bubble coalescence (slug flow). Bubble size distribution was corresponded quite well with the logarithmic normal distribution model.

      • 氣泡塔을 利用한 二酸化窒素의 吸收

        The absorption of nitrogen oxides into liquids is on industrial problem of major importance, not only for the manufacture of nitric acid but also for control of atmospheric pollution. The absorption process of nitrogen dioxide into water has been studied using a level controlled bubble column. An empirical correlation has been obtained for the gas hold-up of the form ∈_G=0.445(N_Bo)^1/8·(N_Go)^1/15·(N'_Fr)^1.0 where ∈_G is the gas hold-up, N_Bo is the Bond number, N_Ga is the Galileo number and N'_Fr is the modified Froude number. The absorption yield of nitrogen dioxide increased as its initial concentration increased up to 3%. In the range of 0-5% of nitrogen dioxide concentration, the rate equation of absorption of absorption in terms of partial pressure could be expressed as N_NO2=H√DK·P_i,_NO2 where H√DK was found to be (4.25±0.25)×10^-7gmol/(㎠)(atm)(s).

      • 미생물을 이용한 Gellan Gum 생성의 Kinetic에 관한 연구

        Polysaccharides produced microbially, have shown themselves to be commercially significant. The reasons for the commercial exploitation of these microbial polysaccharides is because of their unique physical and consistant chemical properties and regularity of supply. Gellan gum is a recently discovered extracellular polysaccaride. This is an anionic heteropolysaccharide that forms viscous solutions and can under certain conditions, form thermoreversible gels with characteristics similar to those formed by both carrigeenan and agar. The aspect of Gellan gum produced by Pseudomonas elodea were investigated under various concentration of carbon sources and nitrogen sources. Sucrose and glucose are used as carbon source, and Bacto-peptone, Promosoy and (NH_4)_2SO_4 are used as nitrogen source. The maximum Gellan gum production was obtained in the culture medium 30.0g/L glucose, 0.6g/L Promosoy at pH 6.6, 37℃. The molecular weight of Gellan gum was 5 × 10 exp (5) which was determined by gel permeation chromatography.
