      • 품종보호용 분자마커의 고추 내 도입

        본 연구는 쥐의 adenosine deaminase(ADA) 유전자를 이용하여 개발된 고추 품종보호용 분자마커로서의 실용 가능성을 검토하기 위하여 실시되었다. 513bp 크기의 개발된 품종보호용 DNA 마커를 운반하는 A. tumefaciens LBA4404를 이용한 형질전환 실험에서 공동배양한 후 선발배지로 옮겨 배양한 '135Q' 와 '276F' 고추자엽 절편체는 두 계통 모두에서 84%와 80%의 높은 재분화율을 보였다. '135Q'와 '276F' 고추 자엽 절편체로부터 재분화된 신초들에 대한 분자마커 특히 primer를 이용한 PCR검정과 PCR로 확인된 잠정적인 형질전환 신초에 대한 Southern hybridization 분석 결과 품종보호용으로 개발된 513bp DNA 마커의 삽입을 확인하였다. PCR 검정 결과 '135Q'와 '276F' 고추 자엽 절편체로부터 재분화된 신초의 62.6%, 62.9%가 각각 형질전환체로 확인되었다. 이러한 안정적인 DNA 마커의 전이는 고추에서의 품종보호를 위한 활용 가능성을 제시한다. This study was carried out to investigate the possibility for the practical application of a molecular marker which was developed from mouse adenosine deaminase (ADA) gene for cultivar protection in hot pepper (Capsicum annuum L.). In the transformation of hot peppers using Agrobacterium tumefaciens LBA4404 carrying a 513 bp DNA marker for cultivar protection, the regeneration percentage from cotyledon explants of '135Q' and '276F' hot pepper lines after co-cultivation were 84 and 80%, respectively. Results of PCR analysis on regenerated shoots using marker-specific primers and Southern hybridization analysis on putative transgenic shoots confirmed by PCR indicated that the 513 bp DNA marker for cultivar protection was successfully transferred into the regenerated shoots. Twenty three and twenty four out of 33 and 37 regenerated shoots were transgenic in '135Q' and '276F' hot peppers, respectively. This stable transfer of the DNA marker suggested the possibility of practical application for cultivar protection in hot peppers.

        The purpose of this retrospective study was to define platelet indices in severe preeclampsia(N=98) and to compare them to normal nonpregnant values(N=93) during the years 1990-1993 in the Department of Obstetrics and Gynecology, College of medicine, University of Ulsan, Asan Medical Center. Indices evaluated included platelet count, mean platelet volume, and platelet distribution width. Means platelet volume was statistically different between two groups, however mean platelet count was significantly decreased(p=0.029) and platelet distribution width was significantly increased(p=0.0001) and platelet distribution width was significantly increased(p=0.0001) in severe preeclampria. Mean platelet volume versus platelet count in normal pregnancy showed a significant inverse nonlinear relationship(r=-0.39, 9<0.001) and that in preeclampsia was congruent with notmal nonpregnant valus. With same mean platelet volume, platelet distribution width showed tendency to shift to right in severe preeclampsia. These findings support the concept of severe preeclampsia as a compensated state of increased platelet comsumption comparing with normal pregnancy. We concluded from above results that quantification of platelet indices may have some value in screening for and following severe preeclampsia.

        Objective: Occupational medicine specialists in the Busan, Ulsan and Kyung-Nam areas established an area-based occupational disease surveillance system and used this system to collected case information for the purpose of preventing occupational diseases Methods: l l hospital participated in this system. The authors selected five main diseases, which were hand-arm vibration syndrome (HAVS), work-related musculoskeletal disorder (WRMSD), occupational asthma, occupational skin disease and occupational and occupational lung cancer and established their case definitions. All cases were reported on the web, and real time analysis was conducted. Results: Between April 2001 and April 2003, 192 cases of HAVS, 118 cases of WRMSD, 33 cases of occupational asthma, 17 cases of occupational skin disease and 17 cases of occupational lung cancer (for a total of 377 cases of these five main diseases) were reported. most of the HAVS cases came from shipbuilding, and 172 of them (92.7%) were associated with grinding. Of the four main types of WRMSD, carpal tunnel syndrome (CTS) was the most prevalent with 46 cases and shipbuilding was also the main industry involved (83.9%). The ergonomic risk factors involved mainly associated with the hand. In 19 (57.6%) and 4 (12.1%) cases of occupational asthma, the agents involved. The causative agents of occupational lung cancer included Cr, welding fume, PAH, Ni, etc. 10(58.8%) of the cases were diagnosed as squamous cell carcinoma and 5(29.4%) as adenoma. Conclusion: This result showed that an area-based occupational disease surveillance system might provide an effective method of evaluating the prevalence of such diseases, however the Busan, Ulsan and Kyung-Nam provinces are too large to be treated as individual areas. Therefore, the authors suggest that each province should establish its own surveillance system.

        In this paper, we test the electrical resistance of flat wire in the PV module. normally solar cell has two kind of flat wire(inter connection ribbon and bus bar ribbon). we found the phenomenon that has a unbalance with resistance when we make a wiling between of string. So, we measurement the resistance of flat wire each other. and analysis of missmatch with resistance between flat wires on PV module. next to survey of IR picture on missmatch flat wire samples for analyze of missmatch with current in the wire. and we perform IR test with solar cell that has a connection with flat wire for test the effect of missmatch resistance on solar cell. Finally we perform the Dark I-V test for survey of effect by the unbalance of resistance. By the result of Dark I-V test, the series resistance of existing connection sample is large more then innovated connection sample.

        음성 기반 심리상담 에이전트의 활용 가능성 탐색 연구

        본 연구에서는 상담 및 심리치료 분야에서 대화형 에이전트의 활용이 증가하는 가운데, 음성 기반 심리상담 에이전트 설계 시 중요하게 고려해야 하는 요인을 탐색하기 위해 실험 연구를 진행하였다. 이를 위해 실제 심리상담 맥락에서 음성 기반 대화형 에이전트를 구현하여, 20~30대 성인 남녀 48명을 대상으로 상담 대화를 진행하였다. 연구 참여자는 4가지 유형의 에이전트 목소리(청년여성, 청년남성, 중년여성, 중년남성) 중 선호하는 목소리를 선택한 후 대화하였으며, 목소리 선택에 대한 이유, 기분 변화, 에이전트의 특성에 대한 지각, 상담 성과 등을 평가하였다. 연구 결과, 사용자 성별에 따른 에이전트의 목소리 유형 선택은 통계적으로 유의하지 않았다. 그러나 목소리 선택 이유로 ‘편안함’이 가장 많이 언급됨으로써, 심리상담 에이전트에게 요구되는 목소리 특성이 탐색되었다. 다음으로, 사용자는 에이전트와의 대화 후, 기분이 유의미하게 개선되는 것으로 나타남으로써 심리상담 에이전트의 개입 효과를 확인하였다. 마지막으로, 사용자가 심리상담 에이전트에게 지각하는 특성 중, 전문성과 호감이 상담 성과를 긍정적으로 평가하는데 중요한 요소임이 확인되었다. 본 연구결과를 바탕으로 심리상담 에이전트의 활용 가능성과 추후 연구에 대한 제언을 제시하였다. This study was conducted to explore important factors to consider when designing voice-based psychological counseling agents amid the increasing use of conversational agents in counseling and psychotherapy. 48 participants selected their preferred agent’s voice among four types (young women and men, middle-aged women and men) and had a conversation with a psychological counseling agent. They also evaluated the reasons for voice selection, mood changes, perception of the agents characteristics, and counseling outcomes. As a results, the agents voice type selected according to the users gender was not statistically significant. However, the qualitative analysis showed ‘comfort of the voice was an important factor. Next, the users mood improved significantly after the conversation with the agent, which confirmed the intervention effect. Finally, it was found that expertness and attractiveness perceptions toward the agent contributed to the counseling outcomes. The implications of the study and suggestions for future research were discussed.

        서로 다른 인공광원 하에서 기내 발아 및 배양된 새싹 황기의 생육 특성 및 생리활성 검정

        Background: Astragalus membranaceus Bunge is a plant belonging to the Leguminosae family that is used in oriental medicine as a diuretic or a tonic and has blood pressure-lowering properties. Several other propertieds, including reduction of blood sugar, antitumor, and antiviral properties, have also been reported. Methods and Results: A. membranaceus treated exposed to blue light showed the maximum value for plant height, root length, and leaf number. Antioxidant activity was also observed under white light. The maximum values of total phenol and total flavonoid content were observed under white light and blue light, respectively. To assess the whitening and anti-wrinkle activities, the tyrosinase inhibiting activity was determined, which showed the highest value at the maximum concentration of 5,000 ㎍/㎖ in blue light-treated A. membranaceus. Conclusions: The use of LED light will benefit the commercial production of A. membranaceus for cosmetic and food purposes.

        F-18 FDG 자동분주기의 분주 최적화에 관한 연구

        본 연구는 PET/CT 종사자의 피폭선량 감소 및 방사성의약품의 정확한 방사능량 투여를 목적으로 사용 중인 자동분주기의 분주 최적화에 관한 연구이다. 연구방법은 주사기 종류 및 분주 속도, Vial 압력에 따른 평가 결과를 통해 분주 횟수에 따른 오차 값을 알아보고 보정 값을 적용하여 최적화된 분주방법을 찾고자 하였다. 연구결과 5 ml 주사기에서는 평균 9.38 mCi가 분주되었고, 3 ml에서는 9.55 mCi가 분주되어 3 ml 주사기에서 10 mCi에 근접한 재현성을 보였다. 분주 속도에 따른 평가에서는 속도를 5, 10, 15, 20 mm/min 으로 증가시켜 10회씩 측정한 결과, 5 mm/min의 속도에서 10 mCi 정량에 가까운 방사성의약품이 분주되었다. Needle필터 사용 전/후 Vial 압력에 따른 평가 결과에서는 3 ml 주사기의 경우 사용 전 9.53 mCi, 사용 후 9.84 mCi로 측정되어 Needle필터를 사용한 후 분주하는 것이 최적화된 값으로 확인되었다. 또한 분주 횟수 증가에 따른 보정 값 적용 전/후 방사능 평가에서는 보정 전 9.53 mCi, 보정 후 10.07 mCi로 측정되어 보정 값을 적용한 실험에서 정량 값에 가까운 것으로 확인되었다. 따라서, 방사성의약품 분주 시 최적화된 분주방법은 주사기는 3 ml를 사용하고, 분주 속도는 5 mm/min로 설정하며 분주 시 Needle필터를 사용하고, 장비의 분주 횟수(χ)에 따른 보정 값은 [y = 0.097 × χ]로 설정하는 것이 좋은 것으로 확인되었다. This is a study on the optimized dispensing of the auto dispenser used for the purpose of reducing the exposure dose and accurate radiation dose of radioisotope with regard to the PET/CT practitioners. The research method was to find the optimized dispensing method through evaluating the results according to the syringe type, dispensing rate, and vial pressure and through the application of corrected values. As a result of this study, 9.38 mCi has been dispensed on average in the case of 5 ml syringe, and the reproducibility close to 10 mCi was shown at the dispense of 9.55 mCi in the case of 3 ml syringe. In the evaluation according to the dispensing rate, the quantity of radioisotope close to 10 mCi was dispensed at the rate of 5 mm/min when the measurement was carried out by increasing the rate by 5 mm/min units in the order of 5, 10, 15 and 20 mm/min . In the evaluation result according to the vial pressure before/after the use of Needle filter, it was measured to be 9.53 mCi before use and 9.84 mCi after use confirming that the dispensing after using Needle filter showed the optimal value. In addition, in the evaluation of radioactivity before/after the application of corrected values according to the increase in dispense frequency, it was measured 9.53 mCi before correction and 10.07 mCi after correction confirming that the value with correction applied was closer to the quantitative value. Thus, a good optimized method was confirmed to use a 3 ml syringe with dispensing rate of 5 mm/min, to use a Needle filter at dispensing, and to set the corrected value of [y = 0.097 × χ] according to the dispensing frequency of equipment.
